ABSTRACT

Diastase is used internationally as a quality monitor for excessive heat treatment and prolonged storage of honey; honey must contain an activity of at least 8 diastase numbers (DN) for it to be considered export quality. Freshly harvested manuka honey can have diastase activity close to the export threshold of 8 DN without excess heating, increasing susceptibility for export failure. This research investigated the effect of compounds unique to or high in concentration in manuka honey on diastase activity. Investigation of the effect of methylglyoxal, dihydroxyacetone, 2-methoxybenzoic acid, 3-phenyllatic acid, 4-hydroxyphenyllactic acid and 2'-methoxyacetophenone on diastase activity was carried out. Manuka honey was stored at 20 and 27 °C and clover honey spiked with compounds of interest were stored at 20, 27 and 34 °C and monitored overtime. Methylglyoxal and 3-phenyllactic acid were found to accelerate the loss of diastase above the loss normally observed with time and elevated temperature.

ABSTRACT

The diagnostic process for fetal alcohol spectrum disorder (FASD) involves a multi-disciplinary team and includes neurodevelopmental, physical, and facial assessments and evidence of prenatal alcohol exposure during the index pregnancy. With the increased use of virtual care in health care due to the pandemic, and desire of clinics to be more efficient when providing timely services, there was a need to develop a virtual diagnostic model for FASD. This study develops a virtual model for the entire FASD assessment and diagnostic process, including individual neurodevelopmental assessments. It proposes a virtual model for assessment and diagnosis of FASD in children and evaluates the functionality of this model with other national and international FASD diagnostic teams and caregivers of children being assessed for FASD.

ABSTRACT

The M2-1 protein of human respiratory syncytial virus (HRSV) is a transcription anti-terminator that regulates the processivity of the HRSV RNA-dependent RNA polymerase (RdRP). Here, we report a crystal structure of HRSV M2-1 bound to a short positive-sense gene-end RNA (SH7) at 2.7 Å resolution. We identified multiple critical residues of M2-1 involved in RNA interaction and examined their roles using mutagenesis and MicroScale Thermophoresis (MST) assay. We found that hydrophobic residue Phe23 is indispensable for M2-1 to recognize the base of RNA. We also captured spontaneous binding of RNA (SH7) to M2-1 in all-atom simulations using a robust Gaussian accelerated molecular dynamics (GaMD) method. Both experiments and simulations revealed that the interactions of RNA with two separate domains of M2-1, the zinc-binding domain (ZBD) and the core domain (CD), are independent of each other. Collectively, our results provided a structural basis for RNA recognition by HRSV M2-1.
