ABSTRACT

A saccharolytic action of insulin given orally with various synthetic polymers to rabbits was studied. Blood sugar level gave the greatest fall when insulin mixtures with strong electrolytes were used. In this case when the weight proportion of insulin-polymers was 1 : 5 insulin action was prolonged to 5 hours, addition of surfactants permitted to get an analogous saccharolytic effect with reduction of the insulin dose by half (50 AU per animal).

ABSTRACT

Prolonged overfeeding of rats with a highly caloric mixed food led in the majority of the animals to a more intensive increase in body weight and of fat depôt than in control animals given conventional laboratory diet. Rats with adiposity caused by prolonged overfeeding displayed disturbances of carbohydrate and lipid metabolism expressed in elevation of blood sugar level on fasting stomach, a fall of glucose tolerance and of insulin sensitivity, and also in increase of blood insulin activity and of free fatty acids and blood triglycerides on fasting stomach. The number of fat cells in the adipose tissue remained unchanged, but their volume increased. In the animals with adiposity the relative area of the islar tissue of the pancreas and the mass of beta-cells increased significantly; the count of the islar cells in the glandular parenchyma increased as well.

ABSTRACT

An experimental study of the hypoglycemic action of polymeric derivatives of insulin was carried out by its enteral administration through a gastric tube into the stomach and into the oral cavity on 105 intract rabbits. A marked hypoglycemic effect significantly differing from the action of crystalline insulin was produced by enteral administration of insulin modified with polymeres. Changes in the character of binding of the protein with the polymere, of the molecular weight of the polymere and of its chemical nature proved to influence the value and the duration of action of insulin modified by polymeres.
