ABSTRACT

[Objective] To investigate the expression of microRNA-200c (miR-200c) in colorectal carcinomas (CRC),and analyze its role on tumor cell migration and invasion.[Methods] The expression levels of miR-200c in CRC tissues and adjacent normal mucosa were assessed by real-time quantitative RT-PCR (qRT-PCR).miR-200c mimics were transiently transfected into human colorectal cancer cells,and their roles on cell migration and invasion were analyzed by Transwell assay.Cell proliferation was measured using the Cell Counting kit-8.The expression levels of epithelial and mesenchymal markers as well as related transcription factor ZEB1 were detected by Western blotting.[Results] Lower miR-200c expression was found in primary CRC tissues with lymph node metastasis compared to those without lymph node metastasis and adjacent normal mucosa.Transfection of miR-200c mimics suppressed proliferation,and reduced invasion and migration in SW620 cells.Furthermore,up-regulation of miR-200c inhibited ZEB1,and resulted in increased E-cadherin and reduced Vimentin gene expression.[Conclusion] miR-200c was associated with invasive and metastatic behavior of CRC.These effects may be mediated through regulation of epithelial-mesenchymal transition.

ABSTRACT

[Objective]To analyze the feasibility and clinical efficacy of one-stage debridement,bone grafting and in-ternal fixation for the treatment of single-segmental lumbar spinal tuberculosis with extreme lateral approach.[Methods]Thirteen patients of single-segmental lumbar spinal tuberculosis that received the surgeries from April 2013 to August 2016 were included.The operation duration and the amount of intraoperative blood loss were recorded.The VAS and ODI of the back pain,lumbar kyphosis angle,segment height restoration,and vertebral fusion rate were used to analyze the clinical efficacy.[Results]Thirteen patients were successfully followed up for 13-32 months(mean,20.3 months);the operation duration was 160-280 min(average,214±96)min;the amount of intraoperative blood loss was 150-350 mL, average(average,263±63)mL. At the final follow-up,ESR and CRP were normal and lower back pain(VAS)and Oswestry disability index(ODI)were significantly reduced(7.2±1.6 vs 2.5±1.2 and 63.3±5.4 vs 31.9±3.7,respectively)compared to preoperative values;there were no significant difference in the lumbar kyphosis angle,segment height resto-ration between preoperation(segmental lordosis,7.1°±4.7°;segmental height,64.8 mm±9.3 mm)and the values at final follow-ups(segmental lordosis,5.2°±3.5°;segmental height,69.4 mm±10.5 mm;P>0.05). All cases acquired good lumbar interbody fusion with no internal fixation failure or recurrence of tuberculosis.[Conclusions]Under systemic and routine antituberculosis chemotherapy,one-stage extreme lateral approach debridement,bone graft and internal fixation is effective and feasible for single-segmental lumbar spinal tuberculosis.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the clinical phenotype and genotype in a Chinese family affected with early-onset familial Alzheimer's disease (EOFAD).</p><p><b>METHODS</b>Potential mutation of beta-amyloid precursor protein (APP) gene, presenilin 1 (PSEN1) gene and apolipoprotein E (APOE) gene was detected with polymerase chain reaction (PCR) and direct sequencing.</p><p><b>RESULTS</b>Homozygous APOE ε 2 allele and no gene mutation of APP gene were detected in the proband (III1). A 488A>G mutation (His163Arg) of the PSEN1 gene was found in the proband and other 4 family members (IV1, IV12, IV21, V2).</p><p><b>CONCLUSION</b>A mutation (c.488A>G, p.His163Arg) of PSEN1 gene was found in a Chinese family affected with EOFAD.</p>

ABSTRACT

The association of lead toxicity with delta -aminolevulinic acid dehydratase (ALAD) and vitamin D receptor gene polymorphisms was examined in 216 workers occupationally exposed to lead. Urinary N-acetyl- beta-glucosaminidase (UNAG) activity and albumin concentration, adjusted by creatinine, were measured as biomarkers of nephrotoxicity. Genotyping was performed using polymerase chain reaction and restricted fragment length polymorphisms. Blood pressure and blood and urinary lead concentrations were measured. No statistically significant difference in blood lead concentrations was observed with the ALAD genotype, although the geometric mean of blood lead concentrations was higher in battery workers with the ALAD-12 genotype (50.95 mug/dL) than in workers with the ALAD-11 genotype (42.95 mug/dL). Battery workers with the vitamin D receptor (VDR)-B allele had higher mean blood lead concentrations (60.54 mug/dL) than did the workers with the VDR-bb genotype (51.65 mug/dL) (p <. 05) . Based on multiple linear regression analysis, the VDR genotype was associated with a blood lead level (p <. 021). The ALAD genotype was associated with a urinary lead level (p <. 009). Workers with the ALAD-12 genotype who also had blood lead concentrations equal to or greater than 40 mug/dL had higher UNAG activity (4.17 U/mmol.Cr) than the workers with the ALAD-11 genotype (1.27 U/mmol.Cr) (p =. 06). Compared with workers with the VDR-bb genotype, workers with the VDR-B allele had higher systolic blood pressure when the blood lead level was equal to or greater than 40 mug/dL (133.06 +/- 19.19 mmHg versus 123.12 +/- 15.69 mmHg, p =. 01). Lead workers with ALAD-12 had larger elevations in UNAG activity and increasing blood lead concentrations compared to lead workers with the ALAD-11 genotype. Lead workers with the VDR-B allele had greater elevations in systolic blood pressure and higher blood lead levels than did workers with the VDR-bb genotype. In summary, the ALAD-2 and VDR-B alleles may modify lead absorption and toxicity.
