ABSTRACT

PURPOSE: Current evidence suggests a high co-prevalence of hypertension and cervical cancer. Accordingly, blood pressure control is indicated during anti-tumor drug therapy in this patient population. Over the past few years, immunotherapy has made great strides in treating different cancers. However, the role and clinical significance of verapamil as a first-line anti-hypertensive drug during immunotherapy remain poorly understood, emphasizing the need for further studies. METHODS: Murine cervical cancer models were employed to assess the effect of verapamil monotherapy and combination with PD1ab. Immunohistochemistry was conducted to quantify the abundance of CD8+ T cell and Ki67+ cells. Several in-vitro and in-vivo assays were used to study the effects of verapamil and explore the preliminary mechanism. RESULTS: Monotherapy with verapamil or PD1ab immune checkpoint inhibitor significantly suppressed the growth of subcutaneously grafted U14 cells in WT BABL/c mice, respectively, with increased survival time of mice. Consistent results were observed in the melanoma model. Furthermore, we substantiated that verapamil significantly impaired tumor proliferation and migration of SiHa human cervical cancer cells in vitro and in vivo. In silico analysis using TCGA data revealed that NFAT2 expression negatively correlated with patient survival. The CCK8 assay revealed that verapamil abrogated the stimulatory effect of NFAT2 after knockdown of NFAT2. CONCLUSIONS: Our results suggest that verapamil inhibits tumor growth by modulating NFAT2 expression and enhancing tumor immune responses to PD1ab, which can be harnessed for cervical cancer therapy, especially for patients with comorbid hypertension. Indeed, further clinical trials are warranted to increase the robustness of our findings.

ABSTRACT

Objective To investigate the laboratory examination,diagnosis and treatment of 57 cases of Acute Promyelocytic Leukemia.Methods The clinical manifestation,laboratory morphology-immunology-cytogenetics-molecular (MICM) classification,treatment and follow-up were analyzed retrospectively among 57 newly diagnosed cases of acute pmmyelocytic leukemia (APL) patients from January 2011 to December 2013 in Xiangya Hospital,Central South University.Results In 57 patients,the first symptom was given priority to with bleeding (73.7%),fever (26.3%),and the organ infiltration accounted for 29.8%.The phenotype of CD13 + CD33 + CD34-HLD-RA-was 59.6% in 57 patients,CD13 + CD33 +CD34 + HLD-RA + was 8.8%,and the remaining 18 cases were not classic phenotypes.The positive percentage of PML/RARa was 93.0% by fluorescence in situ hybridization (FISH),and the remaining 4 cases were other rare abnormal.Except one case gave up the treatment,the other 56 cases of APL patients reached 100% complete remission rate under early screening and diagnosis,relatively standardized and effective treatment.Conclusions Hemorrhage is the most common clinical symptoms of M3.MICM classification,especially the FISH technology,is the main laboratory basis for diagnosis of M3.After the treatment of early specifications currently,M3 can almost be cured.

ABSTRACT

Objective:To review the treatment of life threatening head and neck space infection.Methods:Retrospective was con-ducted to analyse the patient characterization,signs and symptoms at the time of presentation,presence of concomitant systemic co-morbidities,treatment,prognosis and bacteriology.Results:32 patients from 2007.01 to 2013.01(mean age was 57.71 years)were included.The cases were increased year by year.1 1 (34.37%)patients had breathing difficulty requiring tracheostomy and 19 (59.3%)had concomitant systemic comorbidities.23 cases recovered completely,8 with severe mediastinal infection were trans-ferred to the department of thoracic surgery or the department of respiratory medicine.1 patient died.Pus submission rate was 96.87%,the bacteria detection rate was 37.50%.Pus culture showed mixed infection dominated by streptococci.Staphylococcus au-reus,pseudomonas aeruginosa,enterococcus faecalis,prevotella and neisseria were detected.Conclusion:Annually incidence of life threatening head and neck space infection is increasing.Treatment of the infection should include:①Ensuring the airway patency;②Surgical incision and drainage as early as possible;③ Hypoglycemic control,maintenance of hemodynamic,nutritional support,and multi-disciplinary approach are the key points in the management of such infections;④ Systemic antibiotics.

ABSTRACT

Fourier-transform (FT) Raman spectra were measured for 7 kinds of different polypropylene copolymers and the assignments of all Raman bands were made through a detailed analysis. Ultimately, by choosing 2 700-3 100 cm(-1) as the characteristic spectral, partial least squares (PLS) regression models were set up for xylene solubles content, ethylene content and ethylene content of xylene solubles in polypropylene copolymers. The values of correlation coefficient (r) between predicted results and actual values were all greater than 0.94, and the values of average relative error (ARE) were all less than 5%. The second PLS factor was also analyzed in this paper, and it was concluded that the original information of the sample was accurately extracted by using PLS regression. This paper offered a kind of possibility for on-line measurement of xylene solubles content, ethylene content and ethylene content of xylene solubles in polypropylene copolymers.

ABSTRACT

Quantitative determination of polyvinyl chloride (PVC) concentration by Raman spectrum was studied in the present work. According to partial least squares (PLS) analysis, it was found that scores of PLS factor 1 were proportional to the concentrations of the sample solutions. Meanwhile, the loadings of factor 1 could reflect the contents of PVC and cyclohexanone simultaneously. The PLS regression model for PVC concentration prediction was built. The values of r and root mean square error (RMSE) between predictive results and actual values were 0.9963 and 2.775, respectively. The Raman characteristic peaks of PVC and cyclohexanone were found, including the C-Cl bond for PVC (620 and 695 cm(-1)) and the alicyclic ketone for cyclohexanone (1709 cm(-1)). By using internal standard method, another model for PVC concentration prediction was established, and the values of r and RMSE were 0.9941 and 3.151, respectively. The results indicated that it is feasible to use Raman spectrum to detect the PVC concentration, which is of significant importance to PVC recycling.

ABSTRACT

Ethylene content in polypropylene was studied by Raman spectrum, combined with partial least squares (PLS) method. The comparison between Raman spectra for polyethylene and polypropylene was carried out, and the spectra between 50 and 600, 600 and 1600, and 2700 and 3100 cm(-1) were analyzed respectively. The models for ethylene content prediction were built, while the model based on 50-3600 cm(-1) spectra gave the best performance. The experiment indicated that Raman spectrum gave the similar predictive results as the near infrared (NIR) spectrum; the values of correlation coefficient (r), relative average deviation (RAD) and root mean square error (RMSE) between predictive results and actual values were 0.995, 2.65% and 0.319, respectively. According to PLS analysis, the loadings of factor 1 could reflect the relationship between the composition of polypropylene molecular chain and ethylene content, and ethylene content had a positive correlation with CH2 content, but a negative correlation with content of CH3, C-H, and C-C. The results indicated that it was feasible to detect the ethylene content in polypropylene by Raman spectrum.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the application of digital surgical technology in reconstruction of orbital frame and assess the treatment outcomes.</p><p><b>METHODS</b>Seven patients with post-traumatic orbital defect were included in this study. Images of the orbit were obtained for each individual through computed tomography (CT). Preoperative design was finished according to rapid prototyping, computer-aided design and computer-aided manufacturing (CAD-CAM) and other digital surgical techniques. Surgical fracture reductions with internal fixation and implant of Medpor were used in operation to reconstruct orbit as well as correct enophthalmos and diplopia.</p><p><b>RESULTS</b>Accurate realignment of the displaced orbital rim was obtained in all the 7 patients, and enophthalmos and diplopia were corrected in 4 and 2 patients, respectively.</p><p><b>CONCLUSIONS</b>Digital techniques provide a precise means for preoperative design and operation implementation during orbital reconstruction. As a result, complications can be reduced, and the patient's facial appearance can be maximally improved.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To discuss the early surgical treatment of fronto-orbital retrusion in craniosynostosis.</p><p><b>METHODS</b>Operative techniques including "C" fronto-orbital osteotomies and advancement, frontal radially oriented osteotomies and remoulding were performed to correct the fronto-orbital retrusion. Ten patients aged from 6-9 months were included. There were two metopic synostosis, three unilateral coronal synostosis, three bilateral coronal synostosis, one Apert syndromic synostosis and one Crouzon syndromic synostosis.</p><p><b>RESULTS</b>Fronto-orbital deformities of all patients were corrected. There were no complications happened in these patients. The post-operation results were kept well for 3-11 months follow-up in 8 cases.</p><p><b>CONCLUSIONS</b>Fronto-orbital osteotomies and advancement and frontal reshaping were effective to correct fronto-orbital retrusion in craniosynostosis.</p>

ABSTRACT

BACKGROUND: : The aim of this study was to develop a tool which can be used to do a collaborative and prospective survey on the epidemiologic characteristics, treatment and outcome of maxillofacial trauma in China and to ameliorate the existing score and treatment methods of maxillofacial trauma. METHODS: : Chinese Maxillofacial Trauma Registry, Analysis and Injury Severity Score System were developed. Functional modules of this system include clinical data, user management, data inspection, data retrieval, statistic analysis, follow-up survey, injury severity score, system maintenance. The Client/Server and Browser/Server database structure were applied to achieve telematic. The Abbreviated Injury Scale code rules of facial region were revised in detail to embody the characteristics of maxillofacial trauma. Maxillofacial Injury Severity Score (MISS) was designed to evaluate the severity of function derangement in maxillofacial trauma. Thousand nine hundred fifty-nine patients with facial injuries were classified into different groups by the number of fractures and site of injuries. The severity of different groups were evaluated with Injury Severity Score (ISS), Revised Facial Injury Severity Score (RFISS) and MISS, respectively. The results of ISS, RFISS, and MISS were statistically analyzed. RESULTS: : By distributed applying of the system, the survey networks of Chinese maxillofacial trauma had been founded. The classification of trauma etiology, injury type, and severity were meliorated. With the help of the system, the input, management, retrieval and statistically analysis of maxillofacial trauma data can be conveniently and accurately accomplished. Furthermore, the ISS, RFISS, and MISS can be automatically calculated and analyzed according to the code rules of Abbreviated Injury Scale and Revised Abbreviated Maxillofacial Injury Scale. CONCLUSION: : The system has satisfactory stability, security, compatibility, and specialty. It can be used to collect standard cases of maxillofacial trauma and to do a collaborative research on the epidemiologic characteristics, treatment, and outcome of facial injuries. MISS is more accurate, objective, and credible than ISS and RFISS in reflecting the severity of maxillofacial trauma and evaluating the curative effect and prognosis.

ABSTRACT

<p><b>OBJECTIVE</b>To treat complicated midface deficiencies with external distraction device.</p><p><b>METHODS</b>Four patients (one with acquired midface hypoplasia, two with Crouzon syndrome and one with Parry Romberg) with severe midface deficiencies underwent external distraction osteogenesis. After Le Fort III or Le Fort III plus I osteotomies were performed, the external distraction devices were activated on the 7th postoperative day with a rate of 1 mm per day. The distraction devices were removed after consolidation for 3 months.</p><p><b>RESULTS</b>All patients completed the distraction treatment. The distraction distance ranged from 10 to 14 mm. The appearance of all patients was significantly improved.</p><p><b>CONCLUSIONS</b>External distraction osteogenesis provides an effectively treatment for patients with severe and complicated midface deficiencies.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the feasibility of growing tissue-engineered cartilage using chondrocytes seeded onto a biodegradable porous bioceramic, the beta-tricalcium phosphate (beta-TCP).</p><p><b>METHODS</b>A porous bioceramic template of beta-TCP was created in the shape of a disc. Chondrocytes isolated from rabbit articular cartilage were seeded on the beta-TCP template and then kept in rotatory cell culture system (RCCS) for 1 week prior to subcutaneous transplantation into athymic mice. The three-dimensional structure was well-maintained 16 weeks after implantation. After 4, 8, 16 weeks, the specimens were harvested and examined macroscopically, histologically and immunohistochemically.</p><p><b>RESULTS</b>Gross morphological and histological analysis of the specimens from the chondrocyte-beta-TCP complex demonstrated new cartilage construction. The overall configuration of the experimental specimens closely resembled the structure of beta-TCP template.</p><p><b>CONCLUSION</b>These findings suggest that porous bioceramic (beta-TCP) is a good "matrix" for chondrocyte, and can be used for cartilage engineering.</p>

ABSTRACT

Objective:To study the biomechanical characters of human mandible under compression load and to deduce the constitutive equation.Methods: Specimens taken from five mandible of fresh human cadavers were examined on INSTRON by a group of tensile tests at different strain rates (0.000 5,0.005,0.05 and 0.5 s-1).Bone mineral density of every individual mandible was measured.The effects of strain rate(**),bone mineral density (ρ) on Young'Modulus (E),strength(σu)and ultimate strain(εu) were studied and the constitutive equations were deduced using the following model:Y=aρb**c.Results:(1)σu=216.27ρ1.23**0.055(P<0.01),(2)E(Pa)=5 604.024**0.083(P<0.01),(3)εu =0.045**-0.045(P<0.01),(4)σu=1 798.87ρ-1.17**0.083ε0.82(P<0.01).Conclusions:Strain rate and bone mineral density are important factors for the biomechanical characters of human mandible under compression load.Based on the constitutive equations,the stress level,the possibility and degree of mandible injury may be estimated.
