ABSTRACT

Since the 1970s, maternal corticosteroid therapy has been used successfully to induce labor. This allows for better monitoring of parturients and provision of first aid to neonates, improving neonatal viability, as this treatment induces maturation in a variety of fetal tissues, thereby reducing morbidity and mortality. Although the effects of corticosteroids are well known, few studies have investigated the influence of this therapy in Santa Inês sheep. This study aimed to evaluate the efficacy of dexamethasone at two doses (8 and 16 mg) to induce lambing in Santa Inês ewes at 145 days of gestation and assess its effects on neonatal vitality. For this study, 58 ewes raised in an extensive system were investigated. Pregnancy was confirmed after artificial insemination at a set time or after controlled mounting. Ewes were separated into three groups: an untreated control group (G1: 0 mg) and groups treated with two doses of dexamethasone (G2: 8 mg and G3: 16 mg). In total, 79 lambs were born. Their vitality was assessed based on their Apgar score, weight, temperature, and postnatal behavior. SAS v9.1.3 (SAS Institute, Cary, NC) was used to analyze data, considering a 5% significance level for all analyses. The births in the induced groups occurred 48.4 ± 22.1 h after induction, while the ewes that underwent non-induced labor gave birth 131.96 ± 41.9 h after placebo application (p < 0.05), confirming the efficacy of dexamethasone to induce and synchronize labor. The induced and non-induced neonates had similar Apgar scores, temperatures, weights, and postnatal behavioral parameters (p > 0.05). This study showed that inducing labor in Santa Inês ewes at 145 days of gestation with a full (16 mg) or half dose (8 mg) of dexamethasone is an effective technique and does not compromise neonate vitality.

ABSTRACT

This study was conducted to compare the effectiveness of two methodologies in evaluating B- and Doppler-mode ultrasonic images: analysis using ultrasonic software and utilizing a computer with ImageJ software. To determine if ImageJ software utilization is an efficacious alternative to ultrasonic software device- analysis, there were comparisons of values when using the two methods for morphological and vascular characteristics of follicular dynamics and luteal function in 18 crossbred cattle. From day 8 of an ovarian dynamics synchronization treatment regimen period until the time of ovulation (Day 10), B-mode and power-flow ultrasonic cineloops were obtained every 12 h to assess follicular diameter, wall area, and wall blood perfusion area. On Day 14 after ovulation, US cineloops of ovaries were obtained in B mode and power flow to evaluate various morphological and vascular characteristics of the corpus luteum (CL), including luteal diameter, luteal area, and CL blood perfusion area. Cineloops were evaluated and analyzed using ultrasonic software, and in a computer with ImageJ software. To evaluate consistency in results between the two methods, there was evaluation utilizing paired t-test, Pearson correlation coefficient, Bland-Altman plot, and Linear Regression Test procedures to calculate proportion of bias between values for measurements of variables evaluated. Results indicated none of the values for variables before and after ovulation differed (P > 0.05). It, therefore, was concluded that utilization of ImageJ software is an efficacious biomedical technique to analyze ultrasonic images of morphological and vascular characteristics before and after ovulation in cattle.

ABSTRACT

Hematological and serum biochemical characteristics and prevalence of microorganisms in the uterine content of bitches with pyometra were described, according to type and age. This study was carried out at the Veterinary Medicine Hospital Renato Rodenburg de Medeiros Netto of the UFBA with 132 patients diagnosed with pyometra through imaging and laboratory tests, divided into two groups: Group 1 - less than eight years (n=72) and Group 2 - more than eight years (n=60). Both groups were submitted to Ovariohysterectomy (OH), in which blood and uterine lumen secretion samples were collected. The highest incidence of pyometra was found in bitches under 8 years (54.5%), Poodle breed (31.8%) and mixed-breed (27.3%). Open pyometra represented 80.3% of all cases. The death rate (6.0%) did not show significant difference between the types. Anemia (73.0%), thrombocytopenia (41.2%) and leukocytosis (77.0%) were observed. Urea and ALP were above the reference values in 41.8% and 52.2% of each group, respectively, while ALT (94.0%) and creatine (82%) remained normal in most animals. Serum biochemistry did not show significant differences (P<0.05). Escherichia coli and Klebsiella sp. grew in 33.3% and 16.7% of the samples, respectively. Therefore, no significant hematological and serum biochemical differences were observed in bitches with pyometra in both age categories.


Descreveu-se características hematológicas, bioquímicas séricas e prevalência de microrganismos no conteúdo uterino de cadelas com piometra, de acordo com o tipo e idade. Realizou-se este estudo no Hospital de Medicina Veterinária Renato Rodenburg de Medeiros Netto da UFBA com 132 pacientes diagnosticadas com piometra, por exames de imagem e laboratoriais, divididas em dois grupos: Grupo 1 ­ menos de oito anos (n=72) e Grupo 2 ­ mais de oito anos (n=60), submetidas a Ovariectomia (OH), quando se coletou amostras de sangue e secreção do lúmen uterino. A incidência maior deu-se em cadelas abaixo de 8 anos (54,5%), da raça Poodle (31,8%) e sem raça definida (SRD) (27,3%), sendo 80,3% de piometra aberta. A bioquímica sérica não apresentou diferenças significativas (P<0,05). Observou-se crescimento de Escherichia coli e Klebsiella sp. em 33,3% e 16,7% das amostras, respectivamente. Sendo assim, não se observou diferenças significativas hematológicas e na bioquímica sérica de cadelas com piometra nas categorias de idade.

ABSTRACT

Dimethylacetamide has been included in different extenders for the cryopreservation of semen from species with promising results. The objective of this study was to evaluate the use of dimethylacetamide (DMA) in different concentrations, associated or not with glycerol (GLY), for the cryopreservation of ovine semen, and its effects on in vitro sperm parameters and post-thaw in vivo fertility. Five semen samples of five adult Santa Ines sheep (n=25) were used. The collected ejaculates were divided among the seven treatments for subsequent cryopreservation. The treatments presented different concentrations of DMA and GLY, being divided as G1: GLY 6%; G2: DMA 3%; G3: GLY 5% + DMA 1%; G4: GLY 4% + DMA 2%; G5: GLY 3% + DMA 3%; G6: GLY 2% + DMA 4%; G7: GLY 1% + DMA 5%. %. Post-thawing of the straws, aliquots were evaluated for computerized sperm kinetics (CASA) and plasma membrane integrity, using fluorescent probes and flow cytometry. After the in vitro evaluation of the sperm parameters, in vivo testing was performed by laparoscopic artificial insemination of 72 females. The post-thaw total motility (%) evaluated by CASA were 51.4, 51.4, 50.1, 53.6, 52.3, 52.8 and 46.9, respectively, for the seven groups. And the plasma membrane integrity (%) were 19.7, 28.4, 22.3, 29.4, 24.3, 17.9 and 16.9, respectively. There were no differences (P> 0.05) between the treatments for the parameters of spermatic kinetics and membrane integrity. For females inseminated with semen from the control group (G1, GLY6%), the percentage of pregnant females was 36.1%, a result similar to that obtained with G3 treatment (GLY5% + DMA1%). In conclusion, dimethylacetamide, either alone or in combination with glycerol, can be used for cryopreservation of ovine semen.

ABSTRACT

This Research aimed to evaluate the metabolic status and behavior changes during the transition period in dairy goats from three breeds, under tropical conditions. Thirty multiparous female goats were kept in pens, distributed randomly by breeds. Infrared cameras were fitted in the pens to monitor the animals, and its activities were recorded. Goats displayed varied idle, standing, walking, and feeding behaviors at kidding day (P < 0.10) when compared with the days after and before. Agnostic interactions prevailed between 3.33 and 7.98% of the time on the day of kidding. There was a day effect for the exploratory activities (P < 0.10), where the most exploratory activities were observed on the day of kidding. The milk production and fat content differed according to breed and lactation week. Collective pens for lactating goats, kept in a tropical environment, do not compromise the social behavior and welfare.

ABSTRACT

Objectives were to evaluate effects of a smaller than typically used dose of equine chorionic gonadotropin (eCG) during a fixed-time artificial insemination (FTAI) treatment regimen. Transrectal ultrasonic (US) examinations were conducted on dairy cows on Day 0 (D0) and the treatment regimen was initiated with administrations of an intravaginal progesterone (P4) implant, estradiol benzoate (im), and prostaglandin F2α (PGF2α; im). On D8, the P4 implant was removed and PGF2α and estradiol cypionate were administered to all animals. Subsequently, cows were randomly assigned to three groups and eCG was administered to Groups 1, 2, and 3 in doses of 300 (im); 100 (im); and 100 (Baihui acupoint) IUs, respectively. The B-mode and power-flow US cineloops were performed to assess follicular dynamics and evaluate various morphological and vascular characteristics of the corpus luteum. Blood samples were collected to quantify serum P4 concentrations. There were no differences between the ovulation synchronization treatment regimens for all follicular dynamic variables tested; however, cows in Group 3 differed from Group 2 having a larger follicle diameter (FD) on D10 (P = 0.06) and larger preovulatory FD (P = 0.09), as well as a blood perfusion area of the dominant follicle wall on D8 (P = 0.07). There were no differences in responses to the ovulation synchronization treatment regimens for the luteal variables evaluated subsequent to ovulation. In conclusion, the Baihui acupoint was effective as an alternative route for eCG dose reduction when FTAI treatment regimens were imposed without detrimentally affecting values for reproductive variables.

ABSTRACT

Due to the peculiarities of the reproductive cycle of the female dog, which makes it difficult to accurately ascertain the date of conception, it may be challenging to precisely estimate the gestational age in bitches. Using fetal measurements obtained by ultrasound, it is possible to estimate the gestational age in dogs; however, due to the differences in size of the various breeds, such estimates may have a significant standard deviation, which represents less accuracy when specifying the date of birth. The purpose of this study was to evaluate pregnant female Chihuahuas, establishing relations between the fetal dimensions measured by ultrasound and the remaining time until delivery. Using 13 pregnant female Chihuahuas, weekly ultrasound assessments and measurements were performed, of the inner chorionic cavity, cranial-caudal length, biparietal diameter, abdominal diameter and thoracic diameter. Such parameters were retroactively correlated to the date of delivery, and linear regressions were established between the gestational measurements and remaining days until delivery. The fetal measurement presenting the highest correlation (r = 0.99; P<0.0001) and reliability (R2 = 0.98, P<0.0001) with the probable date of delivery was the biparietal diameter, a measurement that can be easily and safely obtained and, when applying a specific formula (Y = -15.46X1 + 38.72), can accurately predict the date of delivery in Chihuahua female dogs.

ABSTRACT

The objective of this study was to evaluate the effects of an hCG subdose applied at the Hou Hai acupoint as an ovulation inducer in donkeys. Eleven donkeys were distributed in randomized blocks in T1 = application of 1,500 IU of hCG intravenous (IV); T2 = 450 IU of hCG applied at the false acupoint (IV), and T3 = 450 IU of hCG applied at the Hou Hai acupoint. There was no difference (P > 0.05) between the treatments regarding the mean diameter of the pre-ovulatory follicle (34.5 ± 1.3 mm), the ovulation rate (96.97%), the interval between induction and ovulation (58.07 ± 16.82 h), the mean diameter of the CL (D0 = 23.0 ± 0.6; D2 = 27.7 ± 1.9 and D8 = 28.2 ± 0.8mm), and serum P4 concentrations (10.50 ± 2.99 ng.mL-1). The application of 450 IU of hCG at the Hou Hai acupoint increased ovulation rate (72.73%) more than 48 h after induction (P = 0.03) and a larger diameter of the CL on D4 (30.7 ± 5.1 mm) (P = 0.04). The vascularization area of the CL on D8, obtained by minimum number of colored pixel (NCP), was greater (P < 0.05) in the donkeys that received 1,500 IU of IV hCG (T1, 41.91 ± 1.17), and we found a positive correlation (P < 0.05) between mean NCP and P4 concentration in the donkeys that received 450 IU of hCG IV at the false acupoint (T2) or at the Hou Hai acupoint (T3). The application of 450 IU of hCG by IV route at the false acupoint or the Hou Hai acupoint was sufficient to induce ovulation in donkeys, demonstrating that the average dosage commonly used for this species is too high.

Eighteen semen samples from nine Santa Ines rams were cryopreserved in order to study the cryoprotective capacity of dimethylacetamide (DMA) at two concentrations (3% and 6%), and its interaction with trehalose (TRE, 100 mOsmol). A further objective of this study was to evaluate in vivo fertility of ram semen cryopreserved with dimethylacetamide. The control treatment was an extender medium with 6% glycerol (GLY). Five treatments were examined: 6% GLY, 3% DMA, 6% DMA, 3% DMA + TRE, and 6% DMA + TRE. After thawing, the kinetic sperm parameters were analyzed using a computerized system. Sperm viability was observed using a multiple parameter sperm staining with propidium iodide (for plasmatic membrane integrity), JC-1 (for mitochondrial membrane potential), and FITC-PSA (for acrosomal integrity). The isosmotic extender with GLY was the most effective medium for the maintenance of sperm characteristics, compared to extenders containing DMA as cryoprotectant. Regarding fertility, the extender medium with 3% DMA may be a safe alternative for sperm cryopreservation, and does not compromise the fertility rates. The addition of TRE decreased the kinetic sperm parameters; however, a positive effect on the integrity of the plasmatic and acrosomal membranes was observed.

Objetivou-se categorizar a contagem de folículos antrais (CFA) em fêmeas Nelore e posteriormente determinar o efeito de alguns parâmetros reprodutivos e do escore de condição corporal (ECC) sobre a CFA, assim como, comparar a taxa de concepção entre fêmeas com distintas categorias de CFA submetidas a um protocolo de inseminação artificial em tempo fixo (IATF). Para tanto, 595 fêmeas Nelore foram submetidas a um protocolo para IATF; no D4 do protocolo, foi determinado o diâmetro ovariano (DOV), a presença de corpo lúteo (CL) e a CFA. As inseminações foram executadas utilizando-se sêmen criopreservado. O diagnóstico de gestação foi realizado por ultrassonografia. As fêmeas foram dividas conforme a categoria animal, dias pós-parto (DPP), DOV, presença de CL e ECC. De acordo com a média da CFA, sugeriu-se uma categorização da CFA, a qual considerou baixa CFA ≤32, intermediária CFA entre 32 e 48 e alta CFA ≥48 folículos/animal. A categoria animal, os DPP e o ECC não afetaram a CFA dos animais. Entretanto, houve diferença entre a média de CFA na classe com maior DOV em comparação com as demais. A presença de CL influenciou na CFA dos animais. Em relação à fertilidade, não foi demonstrado efeito da CFA sobre a taxa de concepção das fêmeas submetidas à IATF. Os resultados encontrados sugerem que a CFA parece ser uma característica intrínseca, mantendo-se constante independente do status fisiológico do animal. Houve uma inter-relação positiva entre o DOV e CFA. A presença de CL influenciou a CFA. No entanto, a CFA não afeta a performance reprodutiva em programas de IATF.


This study aimed to categorize the antral follicle count(AFC) in Nelore females and subsequently determine the effect of some reproductive parameters and body condition score (BCS) on the AFC, as well as compare conception rates among Nelore females with different categories of AFC subjected to a fixed-time artificial insemination (FTAI) protocol. For that, 596 Nelore females were submitted a FTAI protocol, D4 protocol females were determined the ovarian diameter (OVD), the presence of corpus luteum (CL) and the AFC. The inseminations were performed using cryopreserved semen. The pregnancy diagnosis was performed by ultrasonography. The females were divided according to animal category (ACT), the postpartum days(PPD), OVD, presence of CL and BCS. According to this the average of the AFC and the largest population follicular intrinsically presented by the Nelore females, it was suggested that a categorization, which was considered low AFC ≤32, intermediate AFC between 32 and 48and high AFC≥48folliclesanimal-1. The ACT, the PPD and the BCS did not affect AFC of the animals, however, there were differences between the mean AFC in the class with the largest OVD compared the others.The presence of CL affected the AFC animals. Regarding fertility has not been demonstrated effect of AFC on conception rates of females inseminated. The results suggest that the AFC seems to be an intrinsic characteristic remained constant regard less of the physiological status of the animal. There was a positive interrelationship between DOV and the AFC. The presence of CL affected the AFC animals. However the AFC doesn't affect reproductive performance in FTAI programs.

This study evaluated the use of hipoosmotic swelling test (HOST) with deionized water (0 mOsmol), as a method of post thaw ram semen evaluation and correlate their findings with different techniques of semen evaluation. Therefore, twenty semen samples of 20 different adult rams were assessed as for kinetic sperm parameters through computerized system (IVOS 12, Hamiton Thorn Biosciences, Beverly, MA, EUA) and subjective analysis. The sperm membranes viability was carried out by the association of fluorescent probes (propidium iodide, JC-1 and FITC-PSA). The structural integrity of the plasma membrane was also studied through supravital test with eosin and the functional integrity of membrane evaluated by doing the hipoosmotic swelling test with deionized water (0 mOsmol), in the following proportions: One part of semen for 10 (HOST 10), 50 (HOST 50) and 100 (HOST 100) parts of water. After semen dilution in the different proportions it was fixed in formalin-buffered saline and analyzed with regard to percentage of HOST reactive sperm (bent/coiled). The percentage of reaction obtained for HOST 10 (33,1%); HOST 50 (32,8%) and HOST 100 (31,8%) did not differ significantly. HOST 10 presented positive correlation with the plasma membrane integrity by the EOS (r = 0,80; p 0,05). Positive correlations between HOST 50 and HOST 100 with sperm subpopulation with membrane integrity by fluorescence were observed (r = 0,83 and r = 0,85; p 0,01). The findings suggest that the HOST with deionized water can provide additional information for post thawing ram sperm viability evaluation.


O presente trabalho analisou o emprego do choque osmótico (HOST) com o uso da água deionizada (0 mOsmol), como método de avaliação do sêmen ovino descongelado e correlacionou os resultado obtidos com aqueles encontrados em diferentes técnicas de avaliação de sêmen. Para tanto, vinte amostras de sêmen criopreservado (20 reprodutores), foram avaliadas quanto aos parâmetros de cinética espermática pelo sistema computadorizado (IVOS 12, Hamilton Thorne Biosciences, Beverly, MA, EUA) e análise subjetiva. A análise da viabilidade das membranas espermáticas foi efetuada com a associação de sondas fluorescentes (PI, JC-1 e FITC-PSA). A integridade estrutural da membrana plasmática foi estudada com o teste supravital com eosina (EOS) e, por fim, a integridade funcional da membrana foi avaliada pelo choque osmótico, com a utilização da água deionizada nas seguintes proporções: uma parte de sêmen para 10 (HOST 10), 50 (HOST 50) e para 100 (HOST 100) partes de água. Após a diluição do sêmen, nas diferentes proporções, as amostras foram analisadas quanto ao percentual de espermatozoides reativos ao HOST. Os valores de positividade obtidos para o HOST 10 (33,1%), HOST 50 (32,8%) e HOST 100 (31,8%) não diferiram significativamente. O HOST 10 apresentou importante correlação positiva com a integridade da membrana plasmática pela EOS (r = 0,8; p 0,05). Os HOST 50 e 100 demonstraram correlações positivas com subpopulações espermáticas com membrana plasmática íntegra pela fluorescência (r = 0,83 e r = 0,85; p 0,01). Os resultados obtidos revelaram que o HOST com água deionizada pode fornecer informações complementares para a avaliação da viabilidade do sêmen ovino pós-descongelação.

Nove amostras de sêmen de dois caprinos adultos, colhidas com a utilização de vagina artificial, foram submetidas a criopreservação, com o objetivo de verificar o efeito do Equex STM Paste e do EDTA, adicionados a um diluidor a base de Tris-gema de ovo, sobre a viabilidade espermática pós-descongelação. Também foi objetivo desse trabalho, avaliar a utilização de um diluidor comercial, a base de lecitina de soja (Bioexcell® - IMV, L'Aigle, França), para a congelação do sêmen caprino. Assim, foram formados cinco grupos experimentais: TRIS; TRIS+EDTA; TRIS+EQUEX; TRIS+EQUEX+EDTA e Bioexcell. Logo depois da avaliação, o sêmen foi diluído nos diferentes meios e envasado em palhetas de 0,25 mL, com dose inseminante de 100 x 10 elevado a 6 espermatozóides. As amostras foram submetidas ao resfriamento, com uma curva média de 0,46°C/min, até atingirem a temperatura de 5°C, mantidas por mais 75min em tempo de equilíbrio e, posteriormente, congeladas em nitrogênio líquido. A descongelação foi realizada em banho-maria a 37°C por 50s. Não houve diferença (P>0,05) entre as médias de motilidade total e progressiva, em relação aos grupos TRIS, TRIS+EQUEX e TRIS+EQUEX+EDTA. O grupo Bioexcell obteve o menor (P<0,05) percentual de espermatozóides com motilidade total e progressiva após a descongelação. Após o teste de termoresistência, as melhores (P<0,05) taxas de motilidade total e progressiva foram observadas para os grupos com Equex STM (TRIS+EQUEX e TRIS+EQUEX+EDTA). Assim, pode-se afirmar que a adição do Equex promove uma maior manutenção das taxas de viabilidade espermática após a descongelação, quando comparado com os diluidores que não o continham.


The aim of this paper was verifying the effect of the Equex STM Paste and EDTA addition to a Tris-egg yolk extender, on the post-thaw goat sperm viability. Nine semen samples of two adult goats were collected by artificial vagina and cryopreserved. It was also objective of this study, to evaluate the utilization of a soybean lecithin based commercial extender (Bioexcell® - IMV, L'Aigle, French) for the goat semen freezing. They were formed five experimental groups: TRIS; TRIS+EDTA; TRIS+EQUEX; TRIS+EDTA+EQUEX e Bioexcell. After evaluation, the semen was diluted in the five extenders and packed in 0.25mL straws with 100 million of motile spermatozoa. The samples were cooled at 0,46°C/min to 5°C, submitted at 75min of equilibration time and frozen in liquid nitrogen vapour. The thawing was accomplished in 37°C water bath for 50s. There were no differences (P>0,05) on the means of post-thaw total and progressive sperm motility among the groups TRIS, TRIS+EQUEX and TRIS+EQUEX+EDTA. The Bioexcell group obtained the least (P<0,05) percentage of post-thawing total and progressive sperm motility. After the thermotolerance test, it was observed the greatest (P<0,05) rates of total and progressive sperm motility in the Equex STM groups (TRIS+EQUEX and TRIS+EQUEX+EDTA). Thus, it can be affirmed that the Equex addition promotes better maintenance rates in the pos-thaw sperm viability, when compared with the extenders that did not contain it.
