ABSTRACT

Migration of metastatic tumor cells is similar to the traffic of leukocytes and has been reported that can be guided by chemokines and their receptors, through the circulation to distant organs. The chemokine CXCL12 and its receptor CXCR4 play an essential role in hematopoietic stem cell homing and the activation of this axis supports malignant events. Binding of CXCL12 to CXCR4 activates signal transduction pathways, with broad effects on chemotaxis, cell proliferation, migration and gene expression. Thus, this axis serves as a bridge for tumor-stromal cell communication, creating a permissive microenvironment for tumor development, survival, angiogenesis and metastasis. Evidence suggests that this axis may be involved in the colorectal cancer (CRC) carcinogenesis. Therefore, we review emerging data and correlations between CXCL12/CXCR4 axis in CRC, the implications for cancer progression and possible therapeutic strategies that exploit this system.

ABSTRACT

O papel dos produtos naturais, em especial os nutracêuticos, vem ganhando destaque nos últimos anos devido aos efeitos positivos em parâmetros relacionados à saúde. O açafrão é uma espécie originária do sudeste da Ásia e considerado uma preciosa especiaria. Além da principal utilização como condimento, possui substâncias com atividades antioxidante, antimicrobiana e corante, que lhe conferem possibilidade de emprego nas áreas de cosméticos, têxtil e alimentícia, sendo também muito utilizado nas medicinas tradicionais chinesa e indiana. Seu principal composto fenólico, a curcumina, tem sido amplamente estudado, por apresentar uma variedade de propriedades para a saúde, como efeitos antioxidantes, anti-inflamatórios, antivirais, antibacterianos, antidepressivos e anticancerígenos e, assim, potencial de ação contra várias doenças crônicas. Portanto, o objetivo do presente estudo foi revisar os efeitos do açafrão nos parâmetros biológicos e comportamentais, bem como sua utilização na manutenção da saúde e qualidade de vida dos indivíduos.


The role of natural products, especially nutraceuticals, has been gaining prominence in recent years due to their positive effects on health-related parameters. Turmeric is a species native to Southeast Asia and considered a precious spice. In addition to its main use as a condiment, it has substances with antioxidant, antimicrobial, and coloring activity, which give it the possibility of being used in the areas of cosmetics, textiles, and food; It is also widely used in traditional Chinese and Indian medicine. Its main phenolic compound, curcumin, has been widely studied, as it has a variety of health properties, such as antioxidant, anti-inflammatory, antiviral, antibacterial, antidepressant and anticancer effects and, thus, potential for action against various chronic diseases. Therefore, the objective of the present study was to review the effects of saffron on biological and behavioral parameters, as well as its use in maintaining the health and quality of life of individuals.

ABSTRACT

Introduction: some plants such as turmeric, cinnamon, and okra are known to have therapeutic functions such as antioxidant and anti-inflammatory activity. Furthermore, an immunomodulatory role has been observed in the production of antibodies, in particular immunoglobulin A (IgA), which mediates a variety of protective functions for the organism. Objective: the aim of the present study was to investigate the effect of dietary plants on the production of IgA in healthy Wistar rats. Methods: thus, 48 male Wistar rats of 90 days of age were allocated to four groups. The animals were treated for 14 days with dried turmeric, cinnamon, or okra (50, 50, 12.5 mg/day, respectively) in phosphate buffered saline, or with only phosphate buffered saline by gavage. The animals received water and feed ad libitum. Body mass and relative weight ofperitoneal fat, adrenal gland, kidney, spleen, liver and thymus, biochemical parameters, and IgA levels were analyzed. Results: no significant changes were observed in the body mass, relative weight of organs and tissues, and biochemical parameters. An increase in serum IgA levels was observed in animals treated with turmeric or cinnamon. Conclusion: we conclude that the treatment with turmeric and cinnamon increased IgA production. Therefore, our study supports the idea that dietary supplementation with these plants may improve humoral immunity.


Introdução: algumas plantas como a cúrcuma, a canela e o quiabo são conhecidas por apresentar funções terapêuticas, como atividade antioxidante e anti-inflamatória. Além disso, tem sido observado um papel imunomodulador sobre a produção de anticorpos, em especial a imunoglobulina A (IgA), a qual medeia uma variedade de funções protetoras para o organismo. Objetivo: o objetivo do presente estudo foi investigar o efeito de plantas dietéticas na produção de IgA em ratos Wistar saudáveis. Métodos: destarte, 48 ratos machos Wistar com 90 dias de idade foram alocados em quatro grupos. Os animais foram tratados por 14 dias com cúrcuma seca, canela ou quiabo (50, 50, 12,5 mg/dia, respectivamente) em solução salina tamponada com fosfato ou apenas solução salina tamponada com fosfato, por gavagem. Os animais receberam água e ração ad libitum. Foram analisados a massa corporal e o peso relativo da gordura peritoneal, glândula adrenal, rim, baço, fígado e timo, parâmetros bioquímicos e níveis de IgA. Resultados: não foram observadas alterações significativas na massa corporal, no peso relativo dos órgãos e tecidos e nos parâmetros bioquímicos. Foi observado aumento dos níveis séricos de IgA nos animais tratados com cúrcuma ou canela. Conclusão: podemos concluir que o tratamento com cúrcuma e canela aumentou a produção de IgA. Portanto, nosso estudo suporta a ideia de que a suplementação alimentar com essas plantas pode melhorar a imunidade humoral.

ABSTRACT

The clinical course of breast cancer (BC) and survival depend on a wide range of risk factors. From the psychosomatic point of view, BC is one of the most studied type of cancer but there is no evidence available for this relation. Therefore, in the present study we evaluate the impact of chronic life stressors in BC patients. A total of 100 BC patients were invited to participate in an interview, when information about social parameters and emotional changes in the period prior to diagnosis were collected. The emotional changes were evaluated by the Holmes and Rahe's Stress Scale, which analyzes the difficulty required for a person to readjust to society after significant changes in their life. Clinicopathological parameters were obtained from the medical records. For all data, the level of significance adopted was p <0.05. It was observed that 55.2 % of the patients have a medium and 13.8 % were at high risk for disease development related to stressful events in the period prior to the BC diagnosis. The highest stress levels were presented by separated, divorced, or widowed patients compared to married (p <0.01) and single (p = 0.037) patients. The high-risk (HR) group had a lower proportion of positivity for estrogen receptor when compared to the low (LR) and moderate risk (MR) groups (p= 0.001). In addition, a binary logistic regression analysis was performed, and we found that the relationship between the estrogen receptor and the HR of chronic stress was independently associated with the histological type of BC and lymph nodes involvement. The relationship of stressful life experiences and BC is not well established, so our study collaborates with the literature to demonstrate the importance of stress as a factor associated with the development of BC.

BACKGROUND: Obesity is considered a chronic inflammatory disease and transforming growth factor beta 1 (TGFß1) might exert important roles in disease pathogenesis regulating adipocyte differentiation and immune-inflammatory environment. However, the role of this cytokine as a biomarker in obesity is poorly addressed. Therefore, the present study aimed to evaluate the impact of TGFB1 polymorphisms and TGFß1 plasmatic levels in obesity METHODS AND RESULTS: TGFB1 promoter region polymorphisms (rs1800468, G-800A and rs1800469, C-509 T) were evaluated in 75 obese patients and 45 eutrophic patients through PCR-RFLP and plasmatic TGFß1 was quantified through ELISA from 37 of the obese patients, and correlations with clinical and biochemical parameters were tested. Despite no association was found between TGFB1 polymorphisms and obesity susceptibility, several correlations with clinical data were noted. Among others, AC haplotype negatively correlated with plasmatic TGFß1, while plasmatic TGFß1 negatively correlated with C-reactive protein and positively correlated with liver abnormalities on ultrasound and, specifically, with steatosis presence and degree. Conversely, GT haplotype, which associates with higher TGFß1 production, was also positively correlated with the same parameters of liver abnormalities. Further, plasmatic vitamin D negatively correlated with TGFß1, while positively correlated with AC haplotype. CONCLUSION: Overall, the results indicate that TGFß1 might exert important roles in obesity pathophysiology and correlate with biochemical and clinical parameters both at systemic protein as well as at genetic level. Importantly, the consistent positive correlation at both levels with steatosis might suggest this cytokine as a biomarker for this hepatic abnormality in obese patients.

ABSTRACT

A Anatomia da região cervical é de particular importância para os clínicos e cirurgiões de diversas especialidades médicas. Em se tratando da organização nervosa presente na região anterolateral do pescoço, encontra-se o plexo cervical, formado pelos ramos anteriores dos nervos cervicais C1 a C4, cuja função é promover a sensibilidade da pele anterolateral cervical, torácica anterossuperior e do couro cabeludo posterior, na cabeça, além de controlar a musculatura infra-hióidea e diafragmática. Logo, lesões a essa estrutura nervosa e aos seus ramos podem causar graves complicações ao corpo humano. Esse trabalho objetivou utilizar a dissecção da região anterolateral do pescoço como uma ferramenta pedagógica para o estudo das relações anatômicas dos nervos do plexo cervical observados durante essa prática, assim como relatar algumas de suas importâncias clínicas e cirúrgicas. O presente estudo é caracterizado como qualitativo/descritivo. A dissecção foi realizada semanalmente, durante o segundo semestre de 2018 e o primeiro semestre de 2019, com a supervisão do professor responsável e auxílio do técnico de laboratório, no Laboratório de aulas práticas da Universidade Estadual de Londrina (UEL). Considera-se que a dissecção da região anterolateral do pescoço permitiu a visualização de diversos nervos cutâneos e musculares do plexo cervical, assim como de alguns de seus ramos e suas relações anatômicas. Também contribuiu para o conhecimento da topografia em que se encontravam tais estruturas e sua organização em camadas. Esse conhecimento anatômico é essencial para a prática médica, tanto clínica quanto cirúrgica.(AU)


The anatomy of the cervical region is of particular importance for surgeons and physicians of different medical specialties. The cervical plexus can be found in the anterolateral region of the neck, formed by the anterior branches of the cervical nerves C1 to C4, whose function is to promote the sensitivity of the cervical anterolateral skin, anterosuperior thoracic skin, and posterior scalp, on the head, in addition to controlling the infrahyoid and diaphragmatic muscles. Therefore, injuries to this nervous structure and its branches may cause serious complications to the human body. This work aimed at using the dissection of the anterolateral neck region as a pedagogical tool for the study of the anatomical relationships of the cervical plexus nerves observed during this practice, as well as to highlight some of its clinical and surgical importance. This is a qualitative/descriptive study. The dissection was performed weekly, during the second semester of 2018 and the first semester of 2019, with the supervision of the professor in charge and the assistance of the laboratory technician at the Laboratory of Practical Classes at the State University of Londrina (UEL). It is considered that the dissection of the anterolateral neck region allowed the visualization of several cutaneous and muscular nerves present in the cervical plexus. It also contributed to the knowledge of the topography in which these structures were found and their organization in layers. This anatomical knowledge is essential for both clinical and surgical medical practice.(AU)

Background: The increase in breast cancer (BC) cases in young women is of great importance since the tumor behavior in this group is generally more aggressive than in their older counterparts, and strategies for early diagnosis and prognostication are needed. Therefore, this work sought to investigate prognostic markers associated with young (<44 years old) BC patients. Methods: Two hundred thirty-six primary tumor tissues from 232 BC patients, of which 44 had less than 44 years at diagnosis were evaluated regarding the expression of estrogen and progesterone receptors (ER and PR), human epidermal growth factor receptor 2 (HER2), Ki67, and p53 (used as an indicator of p53 mutations) through immunohistochemistry. Also, data regarding tumor size, histopathological grade (HG), lymph node metastasis disease stage, and patients' survival status were collected. Results: Early age tumors had higher Ki67 expression and p53 mutations, and these markers were positively correlated with each other and associated worse prognosis parameters, such as negativity for ER and PR and positivity for HER2, and with higher HG, tumor size, and disease stage. In young patients, Ki67 correlated with ER, PR, and HG, whereas p53 correlated with HER2 and disease stage. Also, Ki67 associated with BC death independently of time from diagnosis, patients age, tumor size, and disease stage, and showed a trend toward a positive correlation with death in young patients, but not in the older group. Conclusion: Young BC patients were more likely to have intensely proliferative tumors with p53 mutations and these markers may hold prognostic relevance in BC, especially in this subgroup of patients.
