ABSTRACT

Neosporosis, an infectious disease caused by the protozoan Neospora caninum, has been associated with economic losses in cattle rearing worldwide. However, previous studies have not presented any evidence regarding the association between serological status of neosporosis and alteration of the reproductive parameters. Thus, this study aimed to determine whether N. caninum is associated with reproductive disorders and to evaluate the possible risk factors of the infection. Blood samples from 202 dairy cows, 51 with a history of reproductive disorders (case group) and 151 without (control group), were collected from different farms in Brazil. Epidemiological questionnaires were conducted with all the farmers. Serum samples were subjected to an indirect fluorescent antibody test to detect antibodies against the parasite. In total, 28.22% (57/202) of the cows were seropositive: 47.06% (24/51) from the case group and 21.85% (33/151) from the control group. By logistic regression, cows aged ≥48 months and cows with history of abortion were 4.8 (95% confidence interval [CI] = 1.91-12.05; p = 0.001) and 2.3 (95% CI = 1.06-5.1; p = 0.034) times more likely to be seropositive, respectively. Furthermore, our results show an association between N. caninum seropositivity and abortion in dairy cows from Brazil with poor management conditions and N. caninum seropositivity risk factors for reproductive disorders.

ABSTRACT

Dairy production and cattle reproductive parameters exhibit a strong relationship, which can be disrupted by infectious agents, such as Neospora caninum, that affect the reproductive tract of these animals. Neosporosis is a major cause of abortion in cattle worldwide and is responsible for substantial economic losses. We evaluated the impact of N. caninum infection on the reproductive parameters of 434 Holstein dairy cows from a herd in Brazil that had been closed for 4 years through the assessment of serological testing and reproductive parameters. Dairy cows older than 24 months and that required at least 2.2 inseminations to become pregnant had a higher frequency of seropositive results. Analysis of the medians of positive and negative cows revealed that seropositive cows tended to require more insemination efforts to achieve pregnancy and showed a 2-month increase in age at first calving. According to the reproductive parameters, the correlation coefficient was higher in seronegative animals, while no such correlation was detected in seropositive cows. Thus, we concluded that N. caninum negatively affects the reproductive parameters of dairy cows kept in a closed herd.

ABSTRACT

ABSTRACT: The presence of anti-Neospora caninum antibodies in beef cattle slaughtered in the northern region of the state of Paraná, Brazil, was evaluated. A total of 401 blood samples were collected; 281 samples from the municipality of Rolândia and 120 from the municipality of Borrazópolis, between April 2015 and November 2016. Of the total samples, 289 were from females and 112 from males, aged one and a half to eight years. Indirect fluorescent antibody test (IFAT) was performed, using a cut-off of 1:100. Variables were tabulated for statistical analyses (Fisher's exact test and chi-square tests, p≤0.05). The analysis showed that of the 401 samples, 37 were positive for N. caninum, indicating a prevalence of 9.2 %, and observed titers were 1:100 (16), 1:200 (14), and 1:400 (7). The variables sex, age, and location did not differ statistically (p>0.05). Our results showed a sero-occurrence of N. caninum in cattle slaughtered in the northern region of the state of Paraná.


RESUMO: A presença de anticorpos anti-Neospora caninum em bovinos de corte, abatidos na região norte do estado do Paraná, Brasil, foi avaliada. Foram coletadas 401 amostras de sangue, sendo 281 amostras no município de Rolândia e 120 no municipio de Borrazopolis, entre os meses de abril de 2015 e novembro de 2016. Do total de amostras, 289 eram de fêmeas e 112 amostras de machos, na faixa etária de um ano e meio até oito anos de idade. Foi realizada a reação da imunofluorescência indireta (RIFI) utilizando ponte de corte de 1:100. Em seguida, foram tabulados as variáveis para análise estatística (testes exato de Fisher e do qui-quadrado, p≤0,05). A análise mostrou que das 401 amostras, 37 foram positivas para N. caninum, indicando uma prevalência de 9,2 % e os títulos observados foram 1:100 (16), 1:200 (14) e 1:400 (7). As variáveis sexo, idade e local não diferiram estatisticamente (p>0,05). Nossos resultados demonstram uma soro-ocorrência de N. caninum em bovinos abatidos na região norte do Paraná.

ABSTRACT

'Candidatus Mycoplasma haemobos' is a haemotropic mycoplasma that can produce various clinical signs in cattle, but abortive potential of the parasite is unknown, as well as the frequency of transplacental transmission in cattle. Thus, the objective of this work was to evaluate the frequency of detection of 'C. M. haemobos' in aborted fetuses and the blood of dairy cows. Blood samples of 22 dairy cows that aborted and pool tissues (brain, lung, heart and liver) of their respective aborted fetuses were tested by conventional PCR. The occurrence of 'C. M. haemobos' DNA in adult animals was 40.9% (9/22) and in the fetuses was 18.2% (4/22). Two fetuses that contained 'C. M. haemobos' DNA were derived from cows which were PCR negative. When stratifying by breed, it was observed that Jersey cows had a higher proportion of positive animals (8/11; 72.7%) as compared to Holstein (1/9; 11.1% P<0.01). The results of this study suggest that this parasite can be transferred via the placenta, but it is not certain if the abortions were due to 'C. M. haemobos'.

ABSTRACT

The objective of this study was to determine the gamma-glutamyl transferase (GGT) levels and its relationship with diarrhea and passive transfer of immunity in Holstein calves within 24 hours and 30 days of life from Leopolis municipality, the north Parana region. Colostrum is rich in immunoglobulins and vital for immunity to newborn calves, since bovine placenta does not allow the passage of immunoglobulin to the fetus. Calves undergo various challenges that can lead to disease and death in the first month of life, including diarrhea. Diarrhea has a multifactorial etiology, and the passive immunity transferred through ingestion of colostrum is able to protect the calf against many of these etiologic agents. GGT measurements indirectly infer the amount of immunoglobulin ingested by the calf. Higher serum GGT levels (381.72 IU / L) were found at 24 hours, and a significant reduction was observed at 30 days (66.22 IU / L). When the presence or absence of diarrhea was associated with GGT levels above and below 200 IU / L, no statistical significance (P> 0.05) was observed, since 80% of animals with diarrhea had serum GGT levels higher than 200 IU / L. Under the conditions of this study, there was no relationship between the GGT concentration and the occurrence of diarrhea, and no mortality was observed despite some animals presented diarrhea.


O colostro é rico em imunoglobulinas e vital para a imunidade de bezerros recém-nascidos, uma vez que placenta bovina não permite a passagem de imunoglobulinas para o feto. Os bezerros são submetidos a diversos desafios que podem levar à doença e morte no primeiro mês de vida, dentre as quais a diarreia. A etiologia da diarreia é multifatorial e a imunidade passiva transferida através da ingestão de colostro é capaz de proteger o bezerro contra muitos desses agentes etiológicos. O objetivo deste estudo foi determinar a dosagem de gama-glutamil transferase (GGT) e sua relação com diarreia e a transferência de imunidade passiva em novilhas da raça Holandesa nos tempos de 24 horas e 30 dias de vida, no município de Leópolis, na região norte do Paraná. As mensurações de GGT infere indiretamente a quantidade de imunoglobulinas ingeridas pelo bezerro. Os níveis séricos de GGT demonstraram valores médios maiores 24 horas após o nascimento (381,72 UI / L) e uma redução significativa foi observada aos 30 dias de vida (66,22 UI/L). Quando a presença ou ausência de diarreia foi associada com níveis de GGT acima e abaixo de 200UI/L, não foi observado significância estatística (p>0,05), uma vez que 80% dos animais com diarreia tinham níveis séricos de GGT superior a 200 UI/L. Sob as condições que foi realizado o presente estudo, não houve relação entre a concentração de GGT e a ocorrência de diarréia, e nenhuma mortalidade foi observada apesar de alguns animais apresentarem diarreia.

The aim of the present study was to evaluate anti-Neospora caninum antibodies and the vertical transmission rate in naturally infected pregnant zebu beef cows (Bos indicus) reared on pasture. The present study began with 200 cows from four farms (50 cows from each farm), and these animals were submitted to timed artificial insemination (TAI). After ultrasonography, 76 pregnant cows were selected, 22, 15, 22, and 17, respectively, from farms 1, 2, 3, and 4. Blood samples were taken from cows thrice during the first, second, and third trimester of gestation, and a blood sample was collected from 31 calves before colostrum milking. From 76 cows 23 (30.3%) had anti-N. caninum antibodies detected by indirect ELISA (Idexx), and 53 (69.7%) did not. Sixty-four cows that initiated the experiment were negative to N. caninum and 11 became positive either during the second or third trimester of gestation, this mean an infection incidence of 17.2% (11/64). OD for ELISA was higher (OD=2.08) during the second and third (OD=2.10) trimesters of pregnancy when compared with the first (OD=1.81), however, there were no statistical differences (P=0.45). The vertical transmission was calculated to be 29.0% (9/31), and the risk of vertical transmission of N. caninum in seropositive dams was 26.25 times higher than seronegative animals (OR=26.25, 2.38

ABSTRACT

Bovine hemoplasmas are bacteria found on the erythrocyte surface or free in the plasma of cattle. The aim of the present study was to evaluate the occurrence of 'Candidatus Mycoplasma haemobos' ('C. M. haemobos') in Holstein and Jersey cattle raised in Londrina and surroundings, northern region of the State of Parana, Southern Brazil. PCR testing directed to 16S rRNA gene fragment was performed to investigate the occurrence and characterize the molecular identity of 'C. M. haemobos'. A total of 264/433 (60.97%) blood samples were positive by PCR. Further alignment of 500-bp amplicons to available sequences at the GenBank database showed high identity (100%) to 'C. M. haemobos'. To the author's knowledge, this is the first molecular confirmation of the hemoplasma 'C. M. haemobos' in cattle from Brazil. Moreover, 'C. M. haemobos' was observed in high occurrence in dairy cattle, and may have significant impact in livestock production.

During this study, cats were immunized by the intranasal and rectal routes with crude rhoptry proteins of Toxoplasma gondii admixed with Quil-A. Twenty-five domestic short hair cats divided into five groups (n=5) were used during this evaluation: G1 and G3 cats received 200 µg of the rhoptry proteins with Quil-A (20 µg) by the intranasal and rectal routes, respectively; G2 and G4 cats received bovine serum albumin (BSA, 200 µg/dose) with Quil-A (20 µg); and G5 animals served as unvaccinated controls. All treatments were performed at days 0, 21, 42, and 63. The challenge was done with 800 cysts of the ME49 of T. gondii strain at day 70 (challenge day). The serum IgG, IgM, IgA, and fecal IgA antibody levels were evaluated by using the indirect enzyme-linked immunosorbent assay (ELISA). Some animals produced antibody levels beyond cut-off; however, two animals from G1 (OD(mean)=0.308, OD(cut-off)=0.200) and three from G3 (OD(mean)=0.254) demonstrated IgG levels on being challenged, with similar results occurring in two cats from G1 to IgM (OD(mean)=0.279, OD(cut-off)=0.200). Fecal IgA levels were detected in all G1 cats (OD(mean)=0.330, OD(cut-off)=0.065), and in one cat from G3 (OD(mean)=0.167). The serum and fecal humoral immune responses did not correlate with oocyst shedding. Oocyst shedding varied from 98.4% (G1), 87.5% (G2), 53.0% (G3), to 58% (G4), and was lower than that of G5 cats. The prepatent period of cats vaccinated intranasally (G1) was reduced from 6-9.6 to 2.8 days, suggesting protection of environmental contamination, considering cats as the primary source of contamination. The intranasally and rectally administered rhoptry vaccines were able to partially protect cats against T. gondii cysts on being challenged; however, the intranasal method of vaccination yielded better results relative to the rectal route.

ABSTRACT

We evaluated the humoral and cellular immune responses in pigs immunized intranasally with crude rhoptry proteins of Toxoplasma gondii plus Quil-A. The experiment used 13 mixed-breed pigs divided into the following three groups: G1 (vaccinated-challenged, n=6), which received the rhoptry vaccine (200(g/dose); G2 (adjuvant-challenged, n=4), which received PBS plus Quil-A; and G3 (unvaccinated-challenged, n=3), which was the control group. The treatments were performed intranasally at days 0, 21, and 42. Three pigs from G1 produced IgG and IgM antibody levels above the cut-off in the ELISA on the challenge day. Partial protection was observed in G1 at the chronic phase of infection when compared with G3. The preventable fractions were 41.6% and 6.5%, in G1 and G2, respectively. The results of this study suggest that rhoptry proteins plus Quil-A stimulated humoral, local, and systemic immune responses, which were able to partially protect the brain from cyst formation.

Hemoplasmas de bovinos são bactérias encontradas na superfície de hemácias, ou livre no plasma de bovinos. O objetivo do presente estudo foi avaliar a ocorrência de Candidatus Mycoplasma haemobos' ('C. M. haemobos') em bovinos das raças Holandesa e Jersey da região de Londrina, norte do Paraná, sul do Brasil. Para investigar a ocorrência e caracterizar a identidade molecular do 'C. M haemobos' uma PCR baseada no fragmento do gene 16S rRNA foi realizada. A PCR identificou como positivas 264/433 (61%) amostras de sangue testadas. O alinhamento deste fragmento de 500 pb com seqüências disponíveis no GenBank mostrou 100% de identidade 'C. M. haemobos'. Pela bibliografia consultada, esta é a primeira confirmação molecular do hemoplasma 'C. M. haemobos' em bovinos no Brasil. Além disso, foi observada uma alta prevalência deste hemoplasma em bovinos de leite, que pode ter um impacto importante na pecuária bovina.

Hematological parameters were evaluated in broilers immunized and challenged with Eimeria tenella. Broiler chickens of Hubbard strain, females, coccidian-free, were kept in wire cages and inoculated on the third day. The experiment was designed to include five sorts of treatment with three replicates each. T1 was the negative control group, T2 received 500 attenuated sporulated oocysts by gavage, T3 was the positive control, T4 received 50 µg of sporozoite protein + Quil A vaccine, and T5 received Quil A without sporozoite protein + PBS, the last two through nasal route on days 0, 7, and 21. On the 31st day, all treatments were challenged with homologous virulent strain of E. tenella in the dose of 8.0 x 104 oocysts, with the exception of T1. One week later, blood sampling, lesion scores, and cecal oocyst count were carried out. The parasitological parameters showed statistical significance (p < 0.05) and there was no damage to the hematological parameters of birds (p > 0.05) by ANOVA test. The correlations suggest that the blood parameters were impaired by effects of the parasite on tissue, showing levels of hemorrhage and/or hydration.

Foram avaliados os parâmetros hematológicos em frangos de corte imunizados e desafiados com Eimeria tenella. Pintos de corte fêmeas da linhagem Hubbard, livres de coccídios, foram mantidos em baterias metálicas e inoculados no terceiro dia. O experimento foi delineado por cinco tratamentos com três repetições cada, sendo: T1 controle negativo, T2 recebeu 500 oocistos esporulados atenuados via oral, T3 controle positivo, T4 recebeu vacina contendo 50 µg de proteínas de esporozoítos + Quil A e T5 recebeu Quil A + PBS, sendo os dois últimos por via nasal nos dias 0, 7 e 21. No dia 31, todos os tratamentos foram desafiados com cepa virulenta homóloga de E. tenella na dose de 8,0 × 10(4) oocistos, exceto T1. Uma semana depois, foi realizada amostragem de sangue, escore de lesão e contagem de oocistos cecais. Os parâmetros parasitológicos apresentaram significância estatística (p < 0,05), sem que causassem prejuízos aos parâmetros hematológicos das aves (p > 0,05), pelo teste ANOVA. As correlações sugerem que os parâmetros sanguíneos foram afetados pelos efeitos do parasita no tecido, apresentando níveis de hemorragia e/ou hidratação.

ABSTRACT

The aim of this study was to evaluate a nasal vaccine using antigens derived from sporozoites of Eimeria tenella incorporated into Iscom to protect broiler chicks. Forty-five one-day-old chickens (Cobb), unvaccinated against coccidiosis, were used in this experiment. The birds were maintained in separated battery cages and divided into three groups: G1 (n=15), G2 (n=15), and G3 (n=15). G1 received 50 microg of sporozoites+Iscom vaccine, G2 received Iscom without antigens, and G3 received only PBS. The treatments were administered by nasal route on days 0, 7, and 21 of the experiment. On the 28th day, all birds were challenged with 105 sporulated oocysts of E. tenella. On the challenge day, three birds from each group were euthanized to evaluate lymphocyte proliferation. Lesion scores were obtained from five birds from each group, 7 days after challenge. The remaining animals were euthanized on the 50th day. The mean lymphocyte proliferation responses were significantly different (P=0.03); G1 was 2.3-2.6 times more elevated than G2, and G3 (P<0.001). 83% of the birds from G1 showed an IgY antibody reaction by ELISA at challenge. The means for oocysts shedding were 16,890+/-20,511, 48,080+/-50,047, and 65,020+/-74,461, for G1, G2 and G3 birds, respectively. There was no significant difference (P>0.17) in oocysts shedding between groups. However, the G1 and G2 chicks demonstrated reduction in percentage of oocyst shedding when compared to control birds (G3) by 74.02% and 26.05%, respectively. The average lesion scores were G1=0.4, G2=1, and G3=2. This study demonstrated that the lowest lesion score and oocyst shedding were observed in the birds from the group that received antigens derived from sporozoite with an Iscom adjuvant (G1). These results suggest that this vaccine can induce protection against avian coccidiosis.
