ABSTRACT

Low protein (LP) diets may increase the occurrence of damaging behaviours, like tail biting, in pigs. We investigated the effect of supplementing a LP diet with indispensable amino acids (IAA) or environmental enrichment on tail biting. Undocked pigs (n = 48 groups of 12) received either a normal protein diet (NP), a LP, LP with supplemented IAA (LP+), or LP diet with extra environmental enrichment (LP-E+) during the starter, grower, and finisher phase. Performance, activity, behaviour, and body damage were recorded. LP and LP-E+ had a lower feed intake, growth, and gain-to-feed ratio, and were more active than NP and LP+ pigs. LP-E+ pigs interacted most often with enrichment materials, followed by LP, LP+, and NP pigs. LP pigs showed more tail biting than all other groups during the starter phase and the finisher phase (tendency) compared to NP and LP+ pigs. Thus, LP-E+ only reduced tail biting in the starter phase, whereas LP+ tended to do so throughout. Tail damage was more severe in LP pigs than in NP and LP+, with LP-E+ in between. In conclusion, IAA supplementation was more effective than extra environmental enrichment in countering the negative effects of a low protein diet on tail biting in pigs.

ABSTRACT

BACKGROUND: Third-party interference in agonistic contests entails a deliberate intervention in an ongoing fight by a bystanding individual (third party) and may be followed by post-conflict social behaviour to provide support to a specific individual. The mechanisms behind third-party intervention are, however, still largely understudied. The aim of this study was to investigate third-party interference, with the predictions that (1) the interferer derives benefits from its action by winning a fight, (2) that patterns of intervention depend on familiarity, (3) that dyadic fights last longer than triadic fights, and (4) that interferers engage in non-agonistic social behaviours afterwards. Pre-pubertal pigs (Sus scrofa) (n = 384) were grouped with one familiar and four unfamiliar conspecifics (all non-kin) to elicit contests for dominance rank. Third-party interference was analysed for the first 30 min after grouping, along with the behaviour (nosing or aggression), contest duration, contest outcome, and interferer behaviour after the fight (post-conflict social behaviour). RESULTS: Three types of interference were observed: non-agonistic involvement (nose contact) by the interferer in a dyadic fight; a triadic fight with each of three contestants fighting one opponent at a time; and triadic fights with two opponents jointly attacking the third one (two-against-one fights). The likelihood of a third-party intervention to occur did not depend on the presence of a familiar animal in the fight. However, once intervention was triggered, interferers attacked unfamiliar fight initiators more than familiar ones. Two-against-one fights lasted longer than other triadic fights and occurred more often when both initial contestants were females. Results of 110 triadic fights (out of 585 fights in total) revealed that interferers were more likely to win compared to the initial opponents at equal body weight. The most common post-conflict behaviour displayed by the interferer was agonistic behaviour towards another group member, independently of familiarity. CONCLUSIONS: The general lack of discrimination for familiarity suggests interference is not driven by support to familiar individuals in pigs. The results show that intervening in an ongoing fight gives the interferer a high chance of contest success and may be a strategy that is beneficial to the interferer to increase its dominance status.

ABSTRACT

Welfare issues in broilers include inactivity and poor leg health. Activity can be stimulated by providing whole dried Black Soldier Fly Larvae (BFSL), but it is unknown whether this effect changes over time. Therefore, effects of BSFL provisioning on broiler activity per minute were assessed in detail. Additionally, the use of pixel changes as a proxy for broiler activity was explored. Broilers were housed in groups of 28 (n = 4 pens, <33 kg/m2). Dried BSFL were scattered through the pen of treatment groups at 08:00, 11:00, 14:00, and 17:00. Activity was assessed per minute both manually using scan sampling, and automatically as percent pixel changes for 30 min around BSFL provisioning, and at 14, 21 and 28 days of age. Both methods were moderately positively correlated and showed that BSFL provisioning increased activity. Activity as assessed by manual observations decreased at 17:00, at 21, and 28 days in both groups. The pixel changes indicated that this could be due to low activity levels in the control group. Using pixel changes seems to be a promising, timesaving tool to score broiler activity, but future research is necessary to validate this method and determine how high activity can be maintained over time.

Introduction: Diet-microbiota-host interactions are increasingly studied to comprehend their implications in host metabolism and overall health. Keeping in mind the importance of early life programming in shaping intestinal mucosal development, the pre-weaning period can be utilised to understand these interactions in suckling piglets. The objective of this study was to investigate the consequences of early life feeding on the time-resolved mucosal transcriptional program as well as mucosal morphology. Methods: A customised fibrous feed was provided to piglets (early-fed or EF group; 7 litters) from five days of age until weaning (29 days of age) in addition to sow's milk, whereas control piglets (CON; 6 litters) suckled mother's milk only. Rectal swabs, intestinal content, and mucosal tissues (jejunum, colon) were obtained pre- and post-weaning for microbiota analysis (16S amplicon sequencing) and host transcriptome analysis (RNA sequencing). Results: Early feeding accelerated both microbiota colonisation as well as host transcriptome, towards a more "mature state", with a more pronounced response in colon compared to jejunum. Early feeding elicited the largest impact on the colon transcriptome just before weaning (compared to post-weaning time-points), exemplified by the modulation of genes involved in cholesterol and energy metabolism and immune response. The transcriptional impact of early feeding persisted during the first days post-weaning and was highlighted by a stronger mucosal response to the weaning stress, via pronounced activation of barrier repair reactions, which is a combination of immune activation, epithelial migration and "wound-repair" like processes, compared to the CON piglets. Discussion: Our study demonstrates the potential of early life nutrition in neonatal piglets as a means to support their intestinal development during the suckling period, and to improve adaptation during the weaning transition.

ABSTRACT

The aim of this study was to evaluate the effect of 16L:8D photoperiod with green (GREEN) or white (WHITE) lights during incubation on hatching performance, blood melatonin, corticosterone, and serotonin levels, hypothalamic expressions of genes related to photoreception, serotonin, and stress systems in layers in relation with feather pecking behavior. Dark incubation (DARK) was the control. Eggs (n = 1,176) from Brown Nick breeders in 2 batches (n = 588/batch) were incubated in the experiment. A total of 396 female chicks and 261 hens were used at rearing and laying periods until 40 wk. Incubation lighting did not affect hatchability, day-old chick weight, and length, but resulted in a more synchronized hatch as compared with the DARK. The effect of incubation lighting on blood hormones was not significant except for reduced serotonin in the GREEN group at the end of the experiment. There was no effect of incubation lighting on gentle, severe, and aggressive pecking of birds during the early rearing period. From 16 wk, GREEN hens showed increased gentle pecking with increasing age. WHITE hens had the highest gentle pecking frequency at 16 wk while they performed less gentle but higher severe and aggressive pecks at 24 and 32 wk. At hatching, the hypothalamic expression of CRH, 5-HTR1A, and 5-HTR1B was higher for the WHITE group compared with both GREEN and DARK, however, 5-HTT expression was higher in GREEN than WHITE which was similar to DARK. Except for the highest VA opsin expression obtained for WHITE hens at 40 wk of age, there was no change in hypothalamic expression levels of rhodopsin, VA opsin, red, and green opsins at any age. Although blood hormone levels were not consistent, results provide preliminary evidence that incubation lighting modulates the pecking tendencies of laying hens, probably through the observed changes in hypothalamic expression of genes related to the serotonin system and stress. Significant correlations among the hypothalamic gene expression levels supplied further evidence for the associations among photoreception, serotonin, and stress systems.

ABSTRACT

Improving welfare is still a critical issue in pig husbandry. Upgrades of the housing environment seem to be a promising solution to optimise resilience as a whole, and therefore improve animal welfare. The objective of this study was to evaluate the effect of an alternative housing system to enhance cognitive resilience and also to promote the pigs' welfare. A total of 96 piglets from two contrasted housing systems [alternative housing system (AHS) vs. conventional system (CONV)] was used. The major upgrades of the alternative system were multi-litter housing during lactation, delayed weaning, extra space allowance, and environmental enrichment from birth onwards. To estimate welfare, weight, and feed intake (as a general indicator of performances), the tear staining area (as a chronic stress indicator), behavioural postures, heart rate traits, and saliva cortisol concentration were measured over a 21 h-isolation. To assess cognitive resilience, the pigs were subjected to a maze with a social reward both before and after the isolation challenge and indicators of cognitive abilities were followed. The AHS pigs showed lower cortisol levels and tear staining area before the challenge, demonstrating overall better welfare due to the alternative housing conditions. During the challenge, AHS pigs had a lower heart rate, higher heart rate variability, and higher vagal activity than the CONV pigs, which might indicate a reduced sensitivity to the stressor. AHS pigs appeared to have a better long-term memory tested in a maze. Providing social and environmental enrichments, that fit the satisfaction of the essential needs of the pigs better, appears to be beneficial for pig welfare as a whole. Its effects on cognitive resilience still need to be proven.

ABSTRACT

Resilience, the capacity of animals to be minimally affected by a disturbance or to rapidly bounce back to the state before the challenge, may be improved by enrichment, but negatively impacted by a high allostatic load from stressful management procedures in pigs. We investigated the combined effects of diverging environmental conditions from weaning and repeated mixing to create high allostatic load on resilience of pigs. Pigs were either exposed to barren housing conditions (B) from weaning onwards or provided with sawdust, extra toys, regular access to a "play arena" and daily positive human contact (E). Half of the pigs were exposed to repeated mixing (RM) and the other half to one mixing only at weaning (minimal mixing, MM). To assess their resilience, the response to and recovery from a lipopolysaccharide (LPS) sickness challenge and a Frustration challenge were studied. In addition, potential long-term resilience indicators, i.e. natural antibodies, hair cortisol and growth were measured. Some indications of more favorable responses to the challenges in E pigs were found, such as lower serum reactive oxygen metabolite (dROM) concentrations and a smaller area under the curve of dROM after LPS injection. In the Frustration challenge, E pigs showed less standing alert, escape behaviors and other negative behaviors, a tendency for a smaller area under the curve of salivary cortisol and a lower plasma cortisol level at 1 h after the challenge. Aggression did not decrease over mixings in RM pigs and was higher in B pigs than in E pigs. Repeated mixing did not seem to reduce resilience. Contrary to expectations, RM pigs showed a higher relative growth than MM pigs during the experiment, especially in the week of the challenges. Barren RM pigs showed a lower plasma cortisol concentration than barren MM pigs after the LPS challenge, which may suggest that those RM pigs responded less detrimentally than MM pigs. Enriched RM pigs showed a higher level of IgM antibodies binding keyhole limpet hemocyanin (KLH) than enriched MM and barren RM pigs, and RM pigs showed a sharper decline in IgG antibodies binding Bovine Serum Albumin (PC-BSA) over time than MM pigs. Hair cortisol concentrations were not affected by enrichment or mixing. To conclude, enrichment did not enhance the speed of recovery from challenges in pigs, although there were indications of reduced stress. Repeated as opposed to single mixing did not seem to aggravate the negative effects of barren housing on resilience and for some parameters even seemed to reduce the negative effects of barren housing.

Weaning is a stressful event for piglets, involving substantial changes to their nutritional and social environment. Providing edible enrichment around weaning may ease the weaning transition by increasing pre-weaning feed intake and improving post-weaning performance, health, behavior, and affective state. In this study, we investigated the effects of providing live black soldier fly larvae (BSFL) as edible enrichment pre- and/or post-weaning. Pre-weaning, piglets received either only creep feed (Pre-C, n = 14 litters) or creep feed and live BSFL (Pre-L, n = 15 litters) ad libitum, and post-weaning piglets either had no access to live BSFL (Post-C, n = 24 pens) or they could rotate tubes that released BSFL (Post-L, n = 24 pens) at levels up to 20% of their expected daily dry matter intake, resulting in treatments CC, CL, LC, and LL. No interaction between pre- and post-weaning treatment was found for any of the measured parameters. Before weaning, Pre-L piglets preferred to interact with larvae over creep feed, and Pre-C piglets interacted more with creep feed than Pre-L piglets. Total time spent on feed-directed behaviors did not differ. Continuous larvae provisioning increased caecum length and proximal stomach digesta pH, while it decreased the passage of glucose and fluorescein isothiocyanate through the colon wall on d3 post-weaning (CC vs. LL, n = 12 piglets/treatment). Post-weaning diarrhea and final body weight were not affected by treatment. After weaning, Pre-C piglets tended to eat more and grew marginally faster than Pre-L piglets. Post-C piglets spent more time eating and had a higher feed intake post-weaning than Post-L piglets. Based on home-pen behavioral observations, Post-L piglets actively explored and ate the larvae. Post-C piglets spent more time on exploring the environment and nosing pen mates, and they spent more time on manipulating pen mates on d8 and played more on d8 & 15 compared to Post-L piglets. Piglet responses to a novel environment and an attention bias test on d4 & 5 post-weaning were not influenced by larvae provisioning. In conclusion, pre-weaning larvae provisioning did not improve pre-weaning feed intake and post-weaning performance, however post-weaning larvae provisioning did benefit piglet behavior as less manipulation of pen mates was observed.

ABSTRACT

In livestock breeding, continuous and objective monitoring of animals is manually unfeasible due to the large scale of breeding and expensive labour. Computer vision technology can generate accurate and real-time individual animal or animal group information from video surveillance. However, the frequent occlusion between animals and changes in appearance features caused by varying lighting conditions makes single-camera systems less attractive. We propose a double-camera system and image registration algorithms to spatially fuse the information from different viewpoints to solve these issues. This paper presents a deformable learning-based registration framework, where the input image pairs are initially linearly pre-registered. Then, an unsupervised convolutional neural network is employed to fit the mapping from one view to another, using a large number of unlabelled samples for training. The learned parameters are then used in a semi-supervised network and fine-tuned with a small number of manually annotated landmarks. The actual pixel displacement error is introduced as a complement to an image similarity measure. The performance of the proposed fine-tuned method is evaluated on real farming datasets and demonstrates significant improvement in lowering the registration errors than commonly used feature-based and intensity-based methods. This approach also reduces the registration time of an unseen image pair to less than 0.5 s. The proposed method provides a high-quality reference processing step for improving subsequent tasks such as multi-object tracking and behaviour recognition of animals for further analysis.

ABSTRACT

During weaning, piglets experience concurrent social, physical, and nutritional stressors. Consequently, piglets often have poor feed intake and display increased oral manipulative behaviours post-weaning, indicative of compromised welfare. Black soldier fly larvae (BSFL) possess many attractive properties for pigs and could therefore function as effective edible enrichment, potentially alleviating weaning stress by facilitating exploration and promoting feed intake. In this study, pairs of piglets received a small amount of either live BSFL or wood shavings (8 pens/treatment) scattered throughout the pen twice a day for 11 days after weaning. Home-pen behaviour was scored by instantaneous scan sampling on day 2, 5 and 8, and behavioural responses to a novel environment and novel object were scored on day 10/11. Performance-related parameters were observed regularly. Larvae provisioning increased floor-directed exploration and decreased object-directed exploration, pig-directed oral manipulation, fighting and eating of pellets, and reduced neophobia towards a novel object. Pellet intake was significantly decreased by BSFL provisioning during day 4-11 post-weaning, although feed and net energy intake including BSFL never differed between treatments. BSFL provisioning did not influence piglet growth, feed efficiency, energy efficiency, and faecal consistency. To conclude, live BSFL provisioning positively affected post-weaning piglet behaviour while maintaining performance.

ABSTRACT

Damaging behaviors (DB) such as tail and ear biting are prevalent in pig production and reduce welfare and performance. Anecdotal reports suggest that health challenges increase the risk of tail-biting. The prevalence of tail damage and health problems show high correlations across batches within and between farms. There are many common risk factors for tail-biting and health problems, notably respiratory, enteric and locomotory diseases. These include suboptimal thermal climate, hygiene, stocking density and feed quality. The prevalence of tail damage and health problems also show high correlations across batches within and between farms. However, limited evidence supports two likely causal mechanisms for a direct link between DB and health problems. The first is that generalized poor health (e.g., enzootic pneumonia) on farm poses an increased risk of pigs performing DB. Recent studies indicate a possible causal link between an experimental inflammation and an increase in DB, and suggest a link between cytokines and tail-biting. The negative effects of poor health on the ingestion and processing of nutrients means that immune-stimulated pigs may develop specific nutrient deficiencies, increasing DB. The second causal mechanism involves tail-biting causing poor health. Indirectly, pathogens enter the body via the tail lesion and once infected, systemic spread of infection may occur. This occurs mainly via the venous route targeting the lungs, and to a lesser extent via cerebrospinal fluid and the lymphatic system. In carcasses with tail lesions, there is an increase in lung lesions, abscessation, arthritis and osteomyelitis. There is also evidence for the direct spread of pathogens between biters and victims. In summary, the literature supports the association between poor health and DB, particularly tail-biting. However, there is insufficient evidence to confirm causality in either direction. Nevertheless, the limited evidence is compelling enough to suggest that improvements to management and housing to enhance pig health will reduce DB. In the same way, improvements to housing and management designed to address DB, are likely to result in benefits to pig health. While most of the available literature relates to tail-biting, we suggest that similar mechanisms are responsible for links between health and other DB.

Creep feed provision may ease weaning, hence we determined the impact of providing fibrous creep feed before weaning and adding this feed to the post-weaning diet on piglet behavior and performance. Pre-weaning, litters with on average 12 piglets were given creep feed (CF, n = 12 litters) or not (NF, n = 10 litters). Post-weaning, piglets (n = 8 pens with 4 piglets/treatment) were given a weaner diet (CON) or weaner diet supplemented with creep feed (CS). Behaviors were scored in the home pen at d11, 16, 22 and 27 after birth and at wk 1 and 2 post-weaning. Feed intake, growth and fecal consistency were measured up to d14 post-weaning. A blood sample was taken at d2, 15 and 29 after birth and d2, 5 and 14 post-weaning. CF-piglets consumed on average 397 ± 71 g creep feed before weaning. CF-piglets grew faster in the last week before weaning than NF-piglets (249 ± 7 vs. 236 ± 11 g/d, F (1, 18) = 5.81, P = 0.03). However, CF- and NF-piglets did not differ in weaning weight, within-litter coefficient of variation in weaning weight, behaviors in the farrowing and weaner pen, and haptoglobin concentrations. Creep feed supplementation enhanced feed exploration at wk 2 post-weaning (0.29 ± 0.08 vs. 0.11 ± 0.03%, F (1, 27) = 5.27, P = 0.03), but did not affect other post-weaning behaviors. Pre-weaning creep feed provision and post-weaning creep feed supplementation did not affect overall feed intake, growth, feed efficiency and fecal consistency for the first 14 days post-weaning, neither body weight at d14 post-weaning. Nevertheless, CF-piglets had a lower within-pen coefficient of variation in body weight at d14 post-weaning than NF-piglets (13.6 ± 1.9 vs. 15.1 ± 1.5%, F (1, 26) = 6.89, P = 0.01). In conclusion, pre-weaning creep feed provision and post-weaning creep feed supplementation had no clear effects on piglet behavior and performance.

ABSTRACT

Sickness can change our mood for the worse, leaving us sad, lethargic, grumpy and less socially inclined. This mood change is part of a set of behavioral symptoms called sickness behavior and has features in common with core symptoms of depression. Therefore, the physiological changes induced by immune activation, for example following infection, are in the spotlight for explaining mechanisms behind mental health challenges such as depression. While humans may take a day off and isolate themselves until they feel better, farm animals housed in groups have only limited possibilities for social withdrawal. We suggest that immune activation could be a major factor influencing social interactions in pigs, with outbreaks of damaging behavior such as tail biting as a possible result. The hypothesis presented here is that the effects of several known risk factors for tail biting are mediated by pro-inflammatory cytokines, proteins produced by the immune system, and their effect on neurotransmitter systems. We describe the background for and implications of this hypothesis.

Commercially housed broilers frequently experience limited environmental stimulation and various health issues, compromising their welfare. Providing environmental enrichment can alleviate these problems by facilitating natural behaviour and activity. We investigated the effect of providing live black soldier fly larvae (BSFL) to broilers housed at commercial densities (33 kg/m2) on behaviour, fearfulness, health and performance. One-day-old broilers were distributed over five treatments with eight pens/treatment: a control treatment without BSFL; two treatments where 5% of the daily nutrient intake was replaced with live BSFL, provided four or seven times a day; and two treatments where 10% of the daily dietary intake was replaced with live BSFL provided four times a day or in transparent, movable tubes with holes. In all BSFL treatments foraging behaviour, and thereby broiler activity, was increased. Prolonged access to live BSFL, either by providing larvae seven times a day or in tubes, caused the largest increase in activity while also decreasing the time spend in tonic immobility, indicating reduced fearfulness. Broiler final weight and health were not affected. Overall, long-term access to live BSFL seems most effective in improving broiler welfare by facilitating natural behaviour and reducing fearfulness, without hindering broiler performance and health.

ABSTRACT

Contact dermatitis, both on the foot pads and hocks, is a well-known health issue in broilers. Less is known about contact dermatitis in broiler breeders, however, although they have many risk factors for developing leg health problems in common with broilers. This study aimed to describe the prevalence and severity of contact dermatitis during the production cycle in 5 lines of broiler breeders, investigate possible causes of contact dermatitis, and study its relationship with gait, egg production, and floor egg percentage. Five commercially available genetic lines of broiler breeders were housed in 21 pens of 550 females and 50 males from 20 to 60 wk of age. Every 10 wk litter quality, leg health measurements (foot pad dermatitis, hock burn, and gait) and body weight were assessed of 50 random hens per pen. Total number of eggs, number of eggs laid outside the nest (floor eggs), and mortality were recorded daily per pen. Prevalence of foot pad dermatitis, hock burn, and gait problems increased with age. Litter quality started to decrease at 50 wk of age. Prevalence of foot pad dermatitis was affected by litter quality, whereas genetic line had little effect. One genetic line was more prone to developing hock burns, though generally the prevalence of hock burn (13%) was much lower than that of foot pad dermatitis (74%). The percentage of broiler breeders with gait problems increased up to 24% with age, but this was not related to the prevalence of contact dermatitis. The lines differed in body weight from 32 wk of age onwards, and a higher body weight was related to lower egg production and higher cumulative mortality. The percentage of floor eggs was not related to leg health parameters or genetic line. Broiler breeders thus have similar leg health problems as broilers, but these problems are not related to the percentage of floor eggs, suggesting that other factors are involved in the undesirable behavior of floor laying.

ABSTRACT

Pigs are faced with various perturbations throughout their lives, some of which are induced by management practices, others by natural causes. Resilience is described as the ability to recover from or cope with a perturbation. Using these data, activity patterns of an individual, as well as deviations from these patterns, can potentially be used to quantify resilience. Dynamic indicators of resilience (DIORs) may measure resilience on a different dimension by calculating variation, autocorrelation and skewness of activity from the absolute activity data. The aim of this study was to investigate the potential of using DIORs of activity, such as average, root mean square error (RMSE), autocorrelation or skewness as indicators of resilience to infection with the Porcine Reproductive and Respiratory Syndrome Virus (PRRSV). For this study, individual activity was obtained from 232 pigs equipped with ear tag accelerometers and inoculated with PRRSV between seven and 9 weeks of age. Clinical scores were assigned to each individual at 13 days post-challenge and used to distinguish between a resilient and non-resilient group. Mortality post-challenge was also recorded. Average, RMSE, autocorrelation and skewness of activity were calculated for the pre- and post-challenge phases, as well as the change in activity level pre- vs. post-challenge (i.e., delta). DIORs pre-challenge were expected to predict resilience to PRRSV in the absence of PRRSV infection, whereas DIORs post-challenge and delta were expected to reflect the effect of the PRRSV challenge. None of the pre-challenge DIORs predicted morbidity or mortality post-challenge. However, a higher RMSE in the 3 days post-challenge and larger change in level and RMSE of activity from pre- to post-challenge tended to increase the probability of clinical signs at day 13 post-infection (poor resilience). A higher skewness post-challenge (tendency) and a larger change in skewness from pre- to post-challenge increased the probability of mortality. A decrease in skewness post-challenge lowered the risk of mortality. The post-challenge DIOR autocorrelation was neither linked to morbidity nor to mortality. In conclusion, results from this study showed that post-challenge DIORs of activity can be used to quantify resilience to PRRSV challenge.

Enriched environments are known to beneficially affect the behavior of pigs, as compared with barren pens. The influence of enrichment may, however, depend on pigs' early life housing experiences. The aim of this study was to investigate the long-term effects of early and later life environmental enrichment on behavior and growth in pigs with different coping styles. Pigs were housed in either barren pens or in larger pens enriched with rooting substrates from birth, and half of them experienced a housing switch, i.e., a loss or gain of enrichment, at 7 weeks of age, creating four treatment groups. Home pen behavior and body weight were recorded until 19 weeks of age. Pigs were classified as reactive or proactive based on a backtest at 2 weeks of age. Enrichment increased time spent exploring, chewing, and play and decreased oral manipulation of penmates and pen-directed exploring and chewing. Behavior of pigs that switched from barren to enriched pens or vice versa reflected not only their actual environment, but also their early life housing. As early and later life enrichment affected most behaviors in opposite directions, effects of enrichment, or lack thereof, after the switch were more pronounced in pigs that had experienced a different early life condition. For instance, pigs experiencing an upgrade from barren to enriched pens seemed to "catch-up" by showing more exploration and play. Conversely, pigs exposed to a downgrade displayed more oral manipulation of penmates than ones kept barren throughout, which particularly held for pigs with a reactive coping style. Effects of early life and current housing on several other behaviors depended on coping style too. Pigs housed in enriched conditions appeared better able to cope with weaning than barren housed pigs, as they gained more weight and had higher feed intake post-weaning. Barren housed pigs had a lower body weight than enriched pigs just before the switch, after which growth was mainly determined by actual housing, with enriched kept pigs having a higher feed intake and body weight. Thus, not only current housing conditions, but also a (mis)match with the early life environment may affect behavior and growth of pigs.

ABSTRACT

This study addressed the impact of early and later life environmental enrichment, and their combination, on specific antibody responses and peripheral blood leukocyte subpopulations in pigs. Pigs were kept in either barren (B1) or enriched (E1) housing from birth, and half of the pigs switched to barren or enriched housing on day 47, resulting in four treatment combinations: B1B2, B1E2, E1B2, E1E2). Pigs were immunized with keyhole limpet hemocyanin-conjugated trinitrophenyl (KLH-TNP) on day 74 and 109 to induce primary and secondary antibody responses. Blood samples were taken weekly until day 130, and IgM and IgG antibody responses were measured. Leukocyte subpopulations were measured on day 74 and 130. Time course of the antibody responses was not affected by housing. Early life enrichment increased the IgG response to KLH, particularly the primary one. At day 74 the relative frequency of lymphocytes, DC and SLA-II expression on monocytes were higher in E1 pigs, whereas the percentage of granulocytes tended to be lower in E1 pigs at day 74. Early life enrichment increased the SLA-II expression on monocytes, the granulocyte to lymphocyte ratio, and tended to increase the percentage of granulocytes, but tended to decrease the percentage of monocytes at day 130. Later life enrichment reduced percentages of CD4+CD8α+ T cells before and after immunization and the SLA-II expression on monocytes at day 74, the percentage of granulocytes and the granulocyte to lymphocyte ratio at day 130. Notably, early and later life housing interacted in their effects on several immune parameters. KLH-IgM responses (both primary and secondary) were affected by the interaction between early and later life housing. IgM titers were higher for B1B2 than for E1E2, with the switched animals (B1E2 and E1B2) moving towards the titers of the animals kept in their later life environment from birth onwards. At day 130 the percentage of gamma delta T cells, CD8α+ cytotoxic T cells and DC were not different between pigs kept in B1B2 and E1E2, but there was a clear impact of the switch in housing conditions, particularly for the pigs that changed from barren to enriched housing. We also found effects of coping style (personality) and sex on some immune parameters. In conclusion, both early life and later life enrichment, and, notably a switch in housing conditions influenced specific antibodies and leukocyte subpopulations in pigs. The current study implies that the early life history of animals and the (mis)match with their current environment could thus be of major importance for their immune system. Further research is needed to investigate potential consequences for the pigs' health.
