ABSTRACT

Electron cryomicroscopy of large macromolecular complexes is becoming an increasingly powerful tool for revealing three-dimensional structures without the need for crystallization. The execution of image processing, however, requires experience and is error-prone due to the need for a human operator to carry out interactive and repetitive processes. We have designed an approach which is intended to make image processing simple and rapid, both for experts and for novice users. We demonstrate this approach using the well-established reconstruction scheme for icosahedral particles. Finally, we implement semi-automated virus reconstruction (SAVR), an expert system that integrates the most CPU intensive and iterative steps using the scripting language Python. SAVR is portable across platforms and has been parallelized to run on both shared and distributed memory platforms. SAVR also allows the incorporation of new algorithms and facilitates the management of the increasingly large data sets needed to achieve higher resolution reconstructions. The package has been successfully applied to several data sets and shown capable of generating icosahedral reconstructions to sub-nanometer resolutions (7-10 A ).

ABSTRACT

Ground-based measurements of solar UV spectral irradiance made from Ushuaia, Argentina at latitude 55 degrees S reveal a large degree of variability among corresponding months of different years over the period from September 1990 through April 1998. The magnitude and wavelength dependence of year-to-year changes in monthly spectral UV-B irradiation are consistent with expectations based on the behavior of column ozone and cloudiness. When combined with satellite measurements of column ozone, a regression model fit to the ground-based data set allows estimates of monthly UV-B irradiation over a time frame of two decades, 1978-1998, during several months of the year. Results show a general increase in ground-level irradiation at 305.0 nm from the end of the 1970s to the early 1990s during calendar months from September through December. This is followed by generally smaller irradiances through the middle to late 1990s for all months except November, where the increase continues through the end of the data record. The long-term variability in monthly irradiation over the time period studied is more complicated than can be described by a simple linear trend.

ABSTRACT

A high-speed cine-camera system utilizing holographic principles to circumvent the restrictive depth of field limitations of photomicrography is being used for behavioral studies of rapidly moving marine zooplankton. High resolution is achieved from small format, flexible 35-mm movie film by the development of a lens-assisted holocamera. The lens images subjects close to the film in the recording process in order to reduce flatness requirements of the film. Aberrations introduced by the lens are cancelled by positioning it in precisely the same relationship to the film during reconstruction as it occupied in recording. New alignment and pathlength matching techniques for complex holocameras were developed.
