ABSTRACT

The budding yeast Saccharomyces cerevisiae is a proven model organism for elucidating conserved eukaryotic biology, but to date its extracellular vesicle (EV) biology is understudied. Here, we show yeast transmit information through the extracellular medium that increases survival when confronted with heat stress and demonstrate the EV-enriched samples mediate this thermotolerance transfer. These samples contain vesicle-like particles that are exosome-sized and disrupting exosome biogenesis by targeting endosomal sorting complexes required for transport (ESCRT) machinery inhibits thermotolerance transfer. We find that Bro1, the yeast ortholog of the human exosome biomarker ALIX, is present in EV samples, and use Bro1 tagged with green fluorescent protein (GFP) to track EV release and uptake by endocytosis. Proteomics analysis reveals that heat shock protein 70 (HSP70) family proteins are enriched in EV samples that provide thermotolerance. We confirm the presence of the HSP70 ortholog stress-seventy subunit A2 (Ssa2) in EV samples and find that mutant yeast cells lacking SSA2 produce EVs but they fail to transfer thermotolerance. We conclude that Ssa2 within exosomes shared between yeast cells contributes to thermotolerance. Through this work, we advance Saccharomyces cerevisiae as an emerging model organism for elucidating molecular details of eukaryotic EV biology and establish a role for exosomes in heat stress and proteostasis that seems to be evolutionarily conserved.

ABSTRACT

Despite research showing that many college men and women have experienced misperception of their friendliness as sexual intent, such research has focused on this type of misperception only as a correlate of men's sexual aggression. In fact, regardless of methodology used many researchers seem to suggest women do not misperceive men's sexual intent, and in some instances may actually under-perceive it. We used a hypothetical scenario to determine whether men (n = 324) and women (n = 689) college students perceived similar sexual intent from a character who is not the same gender as they are, as depicted in a story about a man and woman on a "date." Our results revealed that men and women in our sample reported similar levels of perceived sexual intent on the part of the character with a different gender as described in the scenario, even after that character clearly indicated to the partner that they "think they do not want to have sex." In addition, the perceived level of the character's sexual intent as solicited in response to this scenario design was related to sexual coercion intentions among both men and women (though it appears more strongly related among men), and these relationships remained even after controlling for other known correlates of sexual coercion (e.g., rape myth acceptance, level of sexual arousal). Implications for the study of misperception and its origins are discussed.

ABSTRACT

The current study explores the relationship between psychopathy, cognitive distortions that could promote sexual coercion, i.e., rape myth acceptance (RMA) and misperception of sexual intent (MPSI), and one's level of sexual arousal on the intentions to use sexual coercion. Using a sample of 218 college men (aged 18-69) and a hypothetical vignette design, we examine potential direct and indirect effects of psychopathy on coercion intentions. Psychopathy directly related to coercion intentions and also operated indirectly through RMA. Furthermore, RMA was related to coercion intentions through MPSI. Implications for understanding sexual coercion pathways and prevention efforts are discussed.

ABSTRACT

Synthetic biologists have made great progress over the past decade in developing methods for modular assembly of genetic sequences and in engineering biological systems with a wide variety of functions in various contexts and organisms. However, current paradigms in the field entangle sequence and functionality in a manner that makes abstraction difficult, reduces engineering flexibility and impairs predictability and design reuse. Functional Synthetic Biology aims to overcome these impediments by focusing the design of biological systems on function, rather than on sequence. This reorientation will decouple the engineering of biological devices from the specifics of how those devices are put to use, requiring both conceptual and organizational change, as well as supporting software tooling. Realizing this vision of Functional Synthetic Biology will allow more flexibility in how devices are used, more opportunity for reuse of devices and data, improvements in predictability and reductions in technical risk and cost.

ABSTRACT

The increased accessibility and use of pornography in Western society highlights the emergent need to understand the relationship between its use and sexual coercion. Decades of research have demonstrated a consistent relationship between pornography use and engaging in sexually aggressive behavior, although what drives this relationship remains largely unexplained. Researchers have recently presented potential explanations for these relationships, such as the use of violent pornography types, the development of aberrant sexual scripts, and the frequency of pornography use. This study seeks to contribute to the explanation by examining the potential mediating effects of sexual arousal on the relationship between pornography use frequency and willingness to engage in verbal and illegal sexual coercive behaviors by examining a sample of male and female college adults. This population reports some of the highest rates of pornography use. The sample of 745 college students were exposed to either an exotic video presentation or a criminal justice lecture, and provided a dating scenario and sexual arousal assessments. Results indicated those young adults that consume pornography more frequently were more likely to experience higher levels of sexual arousal to the erotic video than those who reported little or no use. However, after controlling for several variables significantly related to sexual coercion, arousal did not mediate willingness to engage in verbal or illegal sexual coercive behaviors. Other significant results and implications are discussed.

ABSTRACT

According to recent statistics, as many as one in five female college students are victims of sexual assault during their college career. To combat what has been called the "Campus Rape Crisis," researchers have attempted to understand what variables are associated with sexually coercive behaviors in college males. Although investigators have found support for the relationship between pornography consumption and sexually coercive behavior, researchers typically operationalize pornography use in terms of frequency of use. Furthermore, frequency of use has been assessed vaguely and inconsistently. The current study offered a more concrete assessment of frequency of use and an additional variable not yet included for pornography use: number of modalities. Beyond examining the relationship between pornography use and sexual coercion likelihood, the current study was the first to use pornography variables in a threshold analysis to test whether there is a cut point that is predictive of sexual coercion likelihood. Analyses were conducted with a sample of 463 college males. Results indicated that both pornography use variables were significantly related to a higher likelihood of sexually coercive behaviors. When both frequency of use and number of modalities were included in the model, modalities were significant and frequency was not. In addition, significant thresholds for both pornography variables that predicted sexual coercion likelihood were identified. These results imply that factors other than frequency of use, such as number of modalities, may be more important for the prediction of sexual coercive behaviors. Furthermore, threshold analyses revealed the most significant increase in risk occurred between one modality and two, indicating that it is not pornography use in general that is related to sexual coercion likelihood, but rather, specific aspects of pornography use.

ABSTRACT

PIEZO1 and PIEZO2 are newly identified mechanosensitive ion channels that exhibit a preference for calcium in response to mechanical stimuli. In this study, we discovered the vital roles of pezo-1, the sole PIEZO ortholog in Caenorhabditiselegans, in regulating reproduction. A number of deletion alleles, as well as a putative gain-of-function mutant, of PEZO-1 caused a severe reduction in brood size. In vivo observations showed that oocytes undergo a variety of transit defects as they enter and exit the spermatheca during ovulation. Post-ovulation oocytes were frequently damaged during spermathecal contraction. However, the calcium signaling was not dramatically changed in the pezo-1 mutants during ovulation. Loss of PEZO-1 also led to an inability of self-sperm to navigate back to the spermatheca properly after being pushed out of the spermatheca during ovulation. These findings suggest that PEZO-1 acts in different reproductive tissues to promote proper ovulation and fertilization in C. elegans.

ABSTRACT

Fam20C is a secreted protein kinase mutated in Raine syndrome, a human skeletal disorder. In vertebrates, bone and enamel proteins are major Fam20C substrates. However, Fam20 kinases are conserved in invertebrates lacking bone and enamel, suggesting other ancestral functions. We show that FAMK-1, the Caenorhabditis elegans Fam20C orthologue, contributes to fertility, embryogenesis, and development. These functions are not fulfilled when FAMK-1 is retained in the early secretory pathway. During embryogenesis, FAMK-1 maintains intercellular partitions and prevents multinucleation; notably, temperature elevation or lowering cortical stiffness reduces requirement for FAMK-1 in these contexts. FAMK-1 is expressed in multiple adult tissues that undergo repeated mechanical strain, and selective expression in the spermatheca restores fertility. Informatic, biochemical, and functional analysis implicate lectins as FAMK-1 substrates. These findings suggest that FAMK-1 phosphorylation of substrates, including lectins, in the late secretory pathway is important in embryonic and tissue contexts where cells are subjected to mechanical strain.

ABSTRACT

Contractility of the nonmuscle and smooth muscle cells that comprise biological tubing is regulated by the Rho-ROCK (Rho-associated protein kinase) and calcium signaling pathways. Although many molecular details about these signaling pathways are known, less is known about how they are coordinated spatiotemporally in biological tubes. The spermatheca of the Caenorhabditis elegans reproductive system enables study of the signaling pathways regulating actomyosin contractility in live adult animals. The RhoGAP (GTPase--activating protein toward Rho family small GTPases) SPV-1 was previously identified as a negative regulator of RHO-1/Rho and spermathecal contractility. Here, we uncover a role for SPV-1 as a key regulator of calcium signaling. spv-1 mutants expressing the calcium indicator GCaMP in the spermatheca exhibit premature calcium release, elevated calcium levels, and disrupted spatial regulation of calcium signaling during spermathecal contraction. Although RHO-1 is required for spermathecal contractility, RHO-1 does not play a significant role in regulating calcium. In contrast, activation of CDC-42 recapitulates many aspects of spv-1 mutant calcium signaling. Depletion of cdc-42 by RNA interference does not suppress the premature or elevated calcium signal seen in spv-1 mutants, suggesting other targets remain to be identified. Our results suggest that SPV-1 works through both the Rho-ROCK and calcium signaling pathways to coordinate cellular contractility.

BACKGROUND: In recent years, jurisdictions have recognized the strain placed on limited existing resources by criminal offenders with mental illness who frequently cycle through local jail facilities. In response, many locales have developed and implemented specialized programs to more effectively and efficiently manage these offenders, particularly the process of assigning defense attorneys to these often indigent defendants. METHODS: The current study examined the impact of an Indigent Defense Counsel (IDC) program designed to provide specially trained defense attorneys, and enhanced case management services to 257 indigent jail inmates with a qualifying, major mental health diagnosis (e.g., major depression). These offenders were compared to 117 similar offenders who did not receive these services, on both their length of stay in the jail, and their likelihood of recidivism after release to the community. RESULTS: Survival analyses revealed that program participants spent about 17 fewer days in jail; however, recidivism rates between groups, measured as return to the same county jail or as statewide re-arrest, did not differ. CONCLUSIONS: These results suggest that defendants with mental illness can potentially be managed effectively in the community, with little added risk to public safety and at potential savings in jail bed days/costs. Implications for the processing of indigent criminal defendants with mental illness are presented.

BACKGROUND: Hirschi recently revised the measurement of self-control to include the number and salience of costs an individual considers in an offending situation. Evidence is mixed on its predictive utility relative to other self-control measures but suffers from different studies operationalising the measure in different ways and mostly examining it with non-offenders. AIMS: This study aimed to investigate Hirschi's reconceptualisation of self-control. Our research question was whether the number and salience of Hirschi's 'costs' are independently related to offending. METHODS: Data on self-perceptions of likelihood of driving while drunk and various self-control and social control measures were collected by researchers during an orientation class for convicted offenders newly received into correctional facilities--one for men and one for women--during January to May 2011. RESULTS: Eight hundred and nineteen men and 194 women completed the ratings. Both attitudinal and situational self-control measures were independently associated with self-rated likelihood of driving while drunk. These findings were confined to the male offenders. CONCLUSIONS: Findings revealed mixed support for Hirschi's conceptualisation of self-control and its relevance. Situational and attitudinal measures of self-control share some common ground but relate differently to offending according to gender. Further research is needed to find out if these results are generalisable to other crime types and whether race/ethnicity could modify the findings. Even among convicted offenders whose crimes are serious and/or frequent enough to result in imprisonment, high self-control may inhibit offending. This has implications for intervention programmes. This is the first study to compare attitudinal and self-control measures in relation to offending among a sample of incarcerated offenders and across gender.

Deterrence represents the central theoretical core of the American criminal justice system, yet relatively little attention has been paid to how emotions like fear and anger may relate to deterrence. Psychological research has debated whether negative emotions each have similar impacts on decision making (valence approaches) or if distinct emotions have unique impacts (appraisal tendency approaches). This study explores the direct and indirect influences of fear and anger on hypothetical drunk driving likelihood, including their impact on cost perceptions. Surveys were administered to 1,013 male and female incarcerated felony offenders in the Southwestern United States. Using a multivariate path model and controlling for a number of other individual factors, current fear related to increased cost perceptions and anger to decreased costs. Anger also maintained a direct influence on drunk driving, whereas fear did not. Despite their shared negative valence, fear and anger appear to have dissimilar influences on cost perceptions and criminal decision making. A better understanding of these processes may lead to improved crime prevention approaches.

ABSTRACT

Much of the criminological literature testing rational choice theory has utilized hypothetical scenarios presented to university students. Although this research generally supports rational choice theory, a common criticism is that conclusions from these studies may not generalize to samples of actual offenders. This study proceeds to examine this issue in two steps. First, a traditional sample of university students is examined to determine how various costs and benefits relate to their hypothetical likelihood of offending. Then the same data collection procedures are employed with a somewhat different sample of younger, adjudicated, and institutionalized offenders to determine whether the conclusions drawn from the student sample generalize to this offender sample. Results generally suggest that the content and process of hypothetical criminal decision making differ in the sample of known offenders relative to the university students. Limitations of the current study, as well as suggestions for future research, are discussed.
