ABSTRACT

OBJECTIVE: To establish fluorescence microscopy combined with HPLC method for rapid detection the distribution and content of resveratrol tissues in different growth stages of Polygonum cuspidatum. METHODS: Used sequential experiment to design conditions of frozen and observe of the section by fluorescence microscopy; Resveratrol was extracted by ultrasonic-assisted extraction and its content was detected by HPLC. RESULTS: The results showed that frozen condition for concentration of gum Arabic was from 20% (dipping time was 5 - 6 h) to 40% (2 - 5 min), the freezer temperature was -5 degrees C, and the thickness was 15 microm. Resveratrol in polygonum cuspidatum was mainly accumulated in the organs, tissues and cells of fiber and cellulose, its content in rhizomes declined as the following sequence: spinal cord > xylem > phloem > periderm; Its content declined in organ as the following sequence: buds > rhizomes > ground stem > leaves; The content of resveratrol in root increased with age. CONCLUSION: The results of fluorescence microscopic observation is in accordance with the HPLC results, indicating that the method is simple, fast and reliable, and provides a fast and reliable detection method for the determination of optimum harvesting period of Polygonum cuspidatum and acquisition of quality.

ABSTRACT

OBJECTIVE: To optimize the aqueous two-phase extraction and separation technology of flavonoids from the leaves of Eucommia ulmoides. METHODS: The system was prepared by dissolving suitable amounis of polyethylene glycol 4000 (PEG4000) and dextran 40000 ( D40) in aqueous solution. Phase map of the system, the mass fraction of PEG4000 and D40, the effects of amount of added sample, pH value and temperature on target compounds were investigated systematically. RESULTS: The optimum extraction conditions were PEG4000 11%, N40 8%, 8 g sample which contains 6.85% of flavonoids, temperature 60 t and pH value 8. The highest extraction rate could reach to 75.82%. CONCLUSION: This method has the advantages of mild conditions, solvent-free residues and less time-consumed, and the obvious efficiency in single-stage separation and purification of flavonoids.

ABSTRACT

The present paper reports the determination of mineral elements by FAAS, including K, Ca, Na, Mg, Fe, Zn, Mn and Cu. The best condition for the determination was investigated. The best ionization inhibitor and acidity media were confirmed. The experiment used CsCl to prevent ionization interference, and the concentration of HCl was controlled below 2%. At the same time SrCl prevented interference of P to Ca. So the value of absorbance was stable. The results were obtained by calibration curve method. The analysis method is simple, rapid, accurate and fitting to the determination of mineral elements in plants. The recoveries of the method are 90.5%-108.2%. The relative standard deviations are 0.3%-0.7%. The result show that large amounts of mineral elements are embodied in polygonum cuspidatum. The contents of K and Ca are in conformation with the medical effects of polygonum cuspidatum, and each element is related closely to its growing.
