ABSTRACT

Avaliou-se a influência do manejo de fornecimento de colostro nas concentrações de imunoglobulina G, glicose, proteína total, triglicerídeos, lactato e cortisol de 18 bezerros da raça Holandesa. Os animais foram distribuídos em três tratamentos: T1 - os bezerros foram separados das vacas 6h após o nascimento, mamando em suas mães, por 30 minutos, duas vezes ao dia, nos três primeiros dias; T2 - os bezerros foram separados 6h após o nascimento e nos três primeiros dias receberam o colostro em mamadeiras, duas vezes ao dia, quando receberam 2l/refeição; T3 - permaneceram o tempo todo com as vacas durante os três primeiros dias de vida. Foram colhidas amostras de sangue nos tempos de 0; 12; 24; 48; 72 e 96h após o nascimento. Os valores de IgG do T3 foram mais altos que os encontrados no T1, às 24h (P<0,10). No T3, os níveis de glicose foram mais elevados que os verificados nos demais tratamentos (P<0,10). Os níveis de proteína total no T2 foram maiores que no T1 (P<0,10). As concentrações de cortisol e lactato foram maiores ao nascimento nos três tratamentos (P<0,10). Conclui-se que o T2 seria o manejo mais indicado por sua praticidade.


The influence of colostrum supply on immunoglobulin G, glucose, total protein, triglycerides, lactate and cortisol concentrations of 18 Holstein calves, was evaluated. Calves were allotted to three treatments, T1 - calves were separated from cows 6h after birth, nursing 30 minutes a day in their mothers, twice a day in the first three days; T2 - calves were separated 6h after birth, and in the first three days they received colostrum through nipple bottles twice a day, 2l/meal; and T3 - calves remained all the time with cows during the first three days. Blood samples were taken at times 0; 12; 24; 48; 72 and 96h after birth. IgG levels were higher in T3 than in T1 at 24h (P<0.10). Glucose levels were higher in T3 than in the other treatments (P<0.10). Total protein levels in T2 were higher than in T1 (P<0.10). Cortisol and lactate concentrations at birth were higher in all treatments (P<0.10). Howewer, T2 would be more indicated for its feasibility.

ABSTRACT

BACKGROUND: Tissue accumulation of high amounts of D-2-hydroxyglutaric acid (DGA) is the biochemical hallmark of the inherited neurometabolic disorder D-2-hydroxyglutaric aciduria (DHGA). Patients affected by this disease usually present hypotonia, muscular weakness, hypertrophy and cardiomyopathy, besides severe neurological findings. However, the underlying mechanisms of muscle injury in this disorder are virtually unknown. MATERIALS AND METHODS: In the present study we have evaluated the in vitro role of DGA, at concentrations ranging from 0.25 to 5.0 mM, on total, cytosolic and mitochondrial creatine kinase activities from skeletal and cardiac muscle of 30-day-old Wistar rats. We also tested the effects of various antioxidants on the effects elicited by DGA. RESULTS: We first verified that total creatine kinase (CK) activity from homogenates was significantly inhibited by DGA (22-24% inhibition) in skeletal and cardiac muscle, and that this activity was approximately threefold higher in skeletal muscle than in cardiac muscle. We also observed that CK activities from mitochondrial (Mi-CK) and cytosolic (Cy-CK) preparations from skeletal muscle and cardiac muscle were also inhibited (12-35% inhibition) by DGA at concentrations as low as 0.25 mm, with the effect being more pronounced in cardiac muscle preparations. Finally, we verified that the DGA-inhibitory effect was fully prevented by preincubation of the homogenates with reduced glutathione and cysteine, suggesting that this effect is possibly mediated by modification of essential thiol groups of the enzyme. Furthermore, alpha-tocopherol, melatonin and the inhibitor of nitric oxide synthase L-NAME were unable to prevent this effect, indicating that the most common reactive oxygen and nitrogen species were not involved in the inhibition of CK provoked by DGA. CONCLUSION: Considering the importance of creatine kinase activity for cellular energy homeostasis, our results suggest that inhibition of this enzyme by increased levels of DGA might be an important mechanism involved in the myopathy and cardiomyopathy of patients affected by DHGA.

ABSTRACT

Setenta e cinco bezerras cruzadas Angus x MARC II foram usadas para determinar os efeitos da idade à desmama sobre a performance e sobre os constituintes do sangue. As bezerras foram desmamadas com 150, 210 ou 270 dias de idade e foram sempre comparadas com um grupo de não desmamadas. Alterações plasmáticas nas características hematológicas e químicas (cortisol, glicose, T3, proteínas totais, contagem diferencial de células brancas) foram determinadas no dia da desmama (dia-0) e nos dias 2, 7, 14 e 28 após a desmama. As bezerras desmamadas aos 210 dias de idade apresentaram as maiores concentrações de cortisol, glicose e proteínas totais no início do período pós-desmama. Bezerras desmamadas aos 150 dias de idade apresentaram valores de T3 significativamente menores. As características hematológicas (PCV e contagem diferencial de células brancas) não apresentaram relação direta com a desmama. As bezerras desmamadas aos 210 dias de idade apresentaram resposta mais rápida de crescimento no período pós-desmama, comparadas com as desmamadas aos 150 e 270 dias de idade

Os autores relatam um caso de glioblastoma multiforme em uma crianca com 12 anos de idade e com sintomatologia ha um mes. Os autores tentaram correlacionar s sintomatologia e o quadro clinico com os trabalhos anteriormente publicados. O diagnostico foi constatado post-mortem e parece ser o primeiro caso descrito na literatura brasileira nesta localizacao e faixa etaria
