ABSTRACT

Reddit is a forum-based social media and message board platform that has been used in the social sciences as a recruitment source of human subject data. In addiction science, Reddit remains a viable but underutilized tool, compared to other websites (e.g., Amazon's Mechanical Turk, Prolific). The purpose of this commentary is to provide a rationale and recommendations for the successful use of Reddit for addiction science researchers interested in adding it as a recruitment tool. We provide an example of how Reddit can be used to target specific populations of interest, such as individuals struggling with depression or alcohol use disorder. Last, we discuss the limitations of Reddit as a research tool and some considerations for future research to help promote effective use of the platform.

ABSTRACT

The lateral ordering of arrays of self-assembled InAs-GaAs quantum dots (QDs) has been quantified as a function of growth rate, using the Hopkins-Skellam index (HSI). Coherent QD arrays have a spatial distribution which is neither random nor ordered, but intermediate. The lateral ordering improves as the growth rate is increased and can be explained by more spatially regular nucleation as the QD density increases. By contrast, large and irregular 3D islands are distributed randomly on the surface. This is consistent with a random selection of the mature QDs relaxing by dislocation nucleation at a later stage in the growth, independently of each QD's surroundings. In addition we explore the statistical variability of the HSI as a function of the number N of spatial points analysed, and we recommend N > 103 to reliably distinguish random from ordered arrays.

ABSTRACT

Various concepts of steerable needles have been developed in order to reduce placement errors during insertions and to enable complex procedures through curved trajectories in minimally invasive surgery. When inserted into soft tissue, motion of the targeted location ahead of the needle tip has to be taken into account for controlling such tools. This paper investigates the motion caused by flexible bending needles in comparison to rigid straight ones when inserted into a homogeneous tissue phantom. A laser based experimental setup is used to measure displacements of the substrate around the needles. Displacements are transformed into the local frame in order to quantify the relative substrate motion. It is shown that the radial contribution of the displacements is higher for bending needles and that this effect increases with higher path curvatures. This motion must be taken into account for controlling steerable needles along curved trajectories to reduce placement errors in applications such as multi-targeting or reinsertions in soft tissue.

ABSTRACT

The deposition of Mn on to reconstructed InSb and GaAs surfaces, without coincident As or Sb flux, has been studied by reflection high energy electron diffraction, atomic force microscopy and scanning tunnelling microscopy. On both Ga- and As-terminated GaAs(0 0 1), (2 × n) Mn-induced reconstruction domains arise with n = 2 for the most well ordered reconstructions. On the Ga-terminated (4 × 6), the Mn-induced (2 × 2) persists up to around 0.5 ML Mn followed by Mn nano-cluster formation. For deposition on initially ß2(2 × 4)-reconstructed GaAs(0 0 1), the characteristic trench structure of the reconstruction is partially preserved even beyond 1 monolayer Mn coverage. On both the ß2(2 × 4) and c(4 × 4) surfaces, MnAs-like nano-clusters form alongside the reconstruction changes. In contrast, there are no new Mn-induced surface reconstructions on InSb. Instead, the Sb-terminated surfaces of InSb (0 0 1), (1 1 1)A and (1 1 1)B revert to reconstructions characteristic of clean In-rich surfaces after well defined coverages of Mn proportional to the Sb content of the starting reconstruction. These surfaces are decorated with self-assembled MnSb nanoclusters. These results are discussed in terms of basic thermodynamic quantities and the generalized electron counting rule.

ABSTRACT

Percutaneous needle insertions are a common part of minimally invasive surgery. However, the insertion process is necessarily disruptive to the substrate. Negative side effects are migration of deep-seated targets and trauma to the surrounding material. Mitigation of these effects is highly desirable, but relies on a detailed understanding of the needle-tissue interactions, which are difficult to capture at a sufficiently high resolution. Here, an adapted Digital Image Correlation (DIC) technique is used to quantify mechanical behaviour at the sliding interface, with resolution of measurement points which is better than 0.5mm, representing a marked improvement over the state of the art. A method for converting the Eulerian description of DIC output to Lagrangian displacements and strains is presented and the method is validated during the simple insertion of a symmetrical needle into a gelatine tissue phantom. The needle is comprised of four axially interlocked quadrants, each with a bevel tip. Tests are performed where the segments are inserted into the phantom simultaneously, or in a cyclic sequence taking inspiration from the unique insertion strategy associated to the ovipositor of certain wasps. Data from around the needle-tissue interface includes local strain variations, material dragged along the needle surface and relaxation of the phantom, which show that the cyclic actuation of individual needle segments is potentially able to mitigate tissue strain and could be used to reduce target migration.

ABSTRACT

The effects of bacitracin methylene disalicylate (BMD) and scours on the fecal microbiome, animal performance, and health were studied in Holstein bull calves. Holstein bull calves (n = 150) were obtained from a single source at 12 to 24 h of age. Bull calves were randomly assigned to 1 of 2 treatments including CON (no BMD; n = 75 calves) and BMD (n = 75 calves). Starting 3 d after arrival, BMD was added into milk replacer (0.5 g/feeding; twice daily) and fed to the calves for 10 consecutive d. No differences (P > 0.10) were observed in ADG for d 0 to 28 and d 0 to 56, DMI for d 0 to 28, d 29 to 56, and d 0 to 56, or G:F for d 0 to 28, d 29 to 56, and d 0 to 56; ADG for d 29 to 56 tended to increase (P < 0.10) for BMD-treated calves compared with controls. Fecal samples were collected from 15 scouring calves and 10 cohorts (nonscouring calves received on the same day and administered the same treatment as the scouring calves). Animal morbidity and fecal score did not vary between the 2 treatments. Mortality was not influenced by the treatments in the BMD administration period or throughout the experiment. Fecal samples were subjected to pyrotagged 454 FLX pyrosequencing of 16S rRNA gene amplicon to examine compositional dynamics of fecal microbes. Escherichia, Enterococcus, and Shigella had greater (P < 0.05) populations in the BMD group whereas Dorea, Roseburia, Fecalibacterium, Papillibacter, Collinsella, Eubacterium, Peptostreptococcus, and Prevotella were decreased (P < 0.05) by BMD treatment. Genus populations were also compared between scouring and nonscouring calves. Streptococcus was the only genus that had notable increase (P < 0.05) in fecal samples from scouring calves whereas populations of Bacteroides, Roseburia, and Eubacterium were markedly (P < 0.05) greater in nonscouring calves. These results show that BMD has the ability to alter the composition of the fecal microbiome but failed to improve performance in Holstein bull calves. Discrepancy of microorganism profiles between scouring and nonscouring calves might be associated with the occurrence of scours and bacterial genera identified might be potential target of treating diarrhea.

ABSTRACT

Depth-dependent magnetism in MnSb(0001) epitaxial films has been studied by combining experimental methods with different surface specificities: polarized neutron reflectivity, x-ray magnetic circular dichroism (XMCD), x-ray resonant magnetic scattering and spin-polarized low energy electron microscopy (SPLEEM). A native oxide ∼4.5 nm thick covers air-exposed samples which increases the film's coercivity. HCl etching efficiently removes this oxide and in situ surface treatment of etched samples enables surface magnetic contrast to be observed in SPLEEM. A thin Sb capping layer prevents oxidation and preserves ferromagnetism throughout the MnSb film. The interpretation of Mn L(3,2) edge XMCD data is discussed.

ABSTRACT

As the dairy industry continues to grow, more dairy calves are available for dairy, beef, and veal purposes. Rearing systems must be highly efficient to make this industry cost efficient, making the evaluation of rearing methods important to establish the most practical method. A study was designed and conducted to evaluate effects of housing and feeding systems on performance of neonatal Holstein bull calves. Treatments (2 × 2 factorial arrangement) consisted of: 1) individually housed, bottle-fed (n = 5 bull calves); 2) individually housed, bucket-fed (n = 5 bull calves); 3) group-housed, bottle-fed (n = 5 pens; 4 bull calves/pen); and 4) group-housed, bucket- (trough) fed (n = 5 pens; 3 or 4 bull calves/pen). Feeding treatments began on d 7 when calves had been acclimated to their new environment. Body weight measurements were collected every 7 d and blood samples were collected on d 0, 28, 55, and 66 for ß-hydroxybutyrate (BHBA) concentration as a gross indicator of ruminal development. No housing × feeding interactions or feeding treatment effects were observed (P > 0.10). Average DMI (dry feed plus milk replacer) was increased (P < 0.05) for group-housed vs. individual animals after d 41, and final BW was greater (P < 0.05) for group-housed calves compared with individually housed calves. Feed efficiency and ADG, however, remained similar (P > 0.10) for all treatments. Fecal scores (P > 0.26), CV for BW (P > 0.26), and BHBA concentrations (P > 0.14) showed no differences among treatments. Housing system had greater effect on calf performance compared with milk feeding regimen.

ABSTRACT

We present results from composite plasmonic nanostructures designed to achieve the cascaded enhancement of electromagnetic fields at optical frequencies. Our structures comprise a small metallic nanodisc suspended above a larger disk. We probe the optical properties of these structures by coating them with a layer of a visible-light fluorophore and observing fluorescence signals with the help of scanning confocal microscopy. A 43 +/- 5-fold increase in the far-field fluorescence signal has been observed for two-tier composite nanostructures, when compared to the signal obtained from individual nanodiscs. Our results offer the prospect of using such nanostructures for field concentration, optical manipulation of nanoobjects, chemical and biological sensing.

ABSTRACT

We present composite plasmonic nanostructures designed to achieve cascaded enhancement of electromagnetic fields at optical frequencies. Our structures were made with the help of electron-beam lithography and comprise a set of metallic nanodisks placed one above another. The optical properties of reproducible arrays of these structures were studied by using scanning confocal Raman spectroscopy. We show that our composite nanostructures robustly demonstrate dramatic enhancement of the Raman signals when compared to those measured from constituent elements.

ABSTRACT

The migration of monovacancies (V0) and self-interstitials (I) has been observed in ion-implanted low-doped float-zone silicon by variable-energy positron annihilation spectroscopy. V0 and I were created by the in situ implantation of approximately 20 keV helium ions below 50 K. Monitoring the time evolution of the vacancy response during isothermal heating enabled the measurement of activation energies for I and V(0) [corrected] migration of 0.078(7) and 0.46(28) eV, respectively. In highly As-doped Si, partial V annihilation occurs via free I migration, with a second stage of annealing, probably associated with V-As complexes, above room temperature.

Estimates of terrestrial Broad Habitat cover for Wales from the Countryside Survey 2000 stratified sample field mapping programme in Britain are compared with the findings of a full census field mapping project, the Habitat Survey of Wales. The Countryside Survey sampling regime comprised a stratified random sample of 1 km squares [corrected] covering <0.5% of the land surface. Comparative assessment indicates that although few of the sample-derived estimates for individual Broad Habitats are within 30% of the full census survey results, relative extents accord with data from the complete census survey for all Broad Habitats apart from Arable & Horticultural. The accuracy of this estimate is improved when the national boundary of Wales is taken into account in the sample stratification scheme. It is suggested that cultural land-use differences between countries render cropland habitat extent less predictable from physical environmental parameters than semi-natural habitat extent. It is also shown that the precision of sample-derived cover estimates is influenced by habitat pattern: the error term associated with habitats of broadly equal extent is greater for those with the most clumped distributions.

ABSTRACT

IGF-binding protein (IGFBP)-3 is generally considered to have actions that counterbalance those of IGFs and is therefore being developed as a cancer treatment. In breast tumors, however, high levels are associated with aggressive tumors and poor prognosis. Consistent with this we have demonstrated that although IGFBP-3 and a non-IGF-binding fragment (serine phosphorylation domain peptide) reduced attachment and enhanced apoptosis of Hs578T breast cancer cells cultured on collagen or laminin, it promoted their attachment and survival on fibronectin, which is abundant in the matrix of aggressive tumors. We have now examined the factors that determine whether IGFBP-3 has positive or negative actions on breast epithelial cells. IGFBP-3 also promoted survival of Hs578T cells in the presence of an antibody to the beta1-integrin subunit or when cholesterol-stabilized complexes were disrupted. These actions were blocked by IGF-I or a MAPK inhibitor. Serine phosphorylation domain peptide had similar actions on MCF-7 cells that were again reversed on fibronectin or with disruption of cholesterol-stabilized complexes and blocked by the beta1-integrin antibody. In contrast, IGFBP-3 promoted growth and survival for nonmalignant MCF-10A cells, but these effects were again reversed on fibronectin and blocked by the beta1 antibody or a MAPK inhibitor or by disruption of cholesterol-stabilized complexes. On Hs578T cells, IGFBP-3 bound to caveolin-1 and beta1-integrins, enhancing their aggregation, the recruitment of focal adhesion kinase, and the activation of MAPK. In summary, with three breast epithelial cell lines, IGFBP-3 had positive or negative effects on growth and survival dependent upon the status of cholesterol-stabilized integrin receptor complexes.

ABSTRACT

The effect of vacancies on the behavior of F in crystalline Si has been elucidated experimentally for the first time. With positron annihilation spectroscopy and secondary ion mass spectroscopy, we find that F retards recombination between vacancies (V) and interstitials (I) because V and I trap F to form complexes. F diffuses in the V-rich region via a vacancy mechanism with an activation energy of 2.12+/-0.08 eV. After a long annealing time at 700 degrees C, F precipitates have been observed by cross-section transmission electron microscopy which are developed from the V-type defects around the implantation range and the I-type defects at the end of range.

ABSTRACT

Since the sequencing of the first two chromosomes of the malaria parasite, Plasmodium falciparum, there has been a concerted effort to sequence and assemble the entire genome of this organism. Here we report the sequence of chromosomes 1, 3-9 and 13 of P. falciparum clone 3D7--these chromosomes account for approximately 55% of the total genome. We describe the methods used to map, sequence and annotate these chromosomes. By comparing our assemblies with the optical map, we indicate the completeness of the resulting sequence. During annotation, we assign Gene Ontology terms to the predicted gene products, and observe clustering of some malaria-specific terms to specific chromosomes. We identify a highly conserved sequence element found in the intergenic region of internal var genes that is not associated with their telomeric counterparts.

ABSTRACT

'The troubles' over the last 33 years in Northern Ireland have claimed the lives of 3,636 people, including 302 police-officers and 644 soldiers. Of these deaths, 315 were attributed to the military and 52 to the police.(1) Formative experience of public disorder within the early parts of the conflict is reviewed. This article places the evolution, development and use of baton rounds (rubber and plastic bullets) in context and describes the training of police in their use. The development of a conflict management strategy which provides synergy with community based policing is discussed and the importance of understanding the underlying causes of a conflict for law enforcement personnel is emphasized.

ABSTRACT

In the experiment reported here, the lactulose/rhamnose urinary excretion test was used to compare intestinal permeability between four breeds of healthy adult dogs and a group of healthy adult cats. A significant difference in permeability was found between dogs and cats (P <0.001) and between different breeds of dogs (P <0.005). The range of urinary lactulose/rhamnose ratios in the dogs in this study (0.07-0.61) was wider than previously reported (0.03-0.12). The mean value for dogs was 0.19. The range in cats was 0.41-1.25 and the mean 0.52. The results of this study suggest that breed or some other factor such as environment, diet or sexual status as well as species should be taken into account when assessing intestinal permeability using the lactulose/rhamnose urinary excretion test.

ABSTRACT

The riboflavin-catalyzed photooxidation of 2',3',5'-tri-O-acetyl-8-oxo-7,8-dihydroguanosine generates a radical intermediate that is competitively trapped by H(2)O, O2(-)(*), or O(2). The products of H(2)O trapping have been previously described as the spiroiminodihydantoin (pH >or= 7) and iminoallantoin/guanidinohydantoin (pH < 7) nucleosides. Trapping by O2(-)(*) leads to the oxaluric acid (pH or= 8.6) pathways (R' ', R' ' = H or 2,3,5-tri-O-Ac-ribofuranosyl). The pH-dependent role of superoxide was probed using Mn-SOD and compared to guanosine and 8-methoxyguanosine photooxidation.

ABSTRACT

Texturized vegetable protein from soy (TVP) is widely used in canned dog foods, but its nutritional value remains in doubt. This study compared apparent prececal and total intestinal digestibility when four canned diets containing reciprocal proportions of protein from TVP (0 to 57%) and from beef (100 to 43%) were fed to eight cannulated dogs. As dietary TVP increased, the following linear changes were observed (P < 0.05): prececal and total intestinal protein digestibility decreased slightly from 77 to 71% and 86 to 80%, respectively; prececal amino acid digestibility decreased in parallel with protein; prececal carbohydrate digestibility decreased markedly from 80% to 62% and carbohydrate disappearance in the large intestine increased from 8 to 22%; prececal sodium digestibility decreased markedly (from 37 to 4%); and prececal potassium digestibility decreased (from 93 to 85%). Total intestinal digestibility of sodium and potassium decreased little (from 97 to 95% and from 98 to 97%, respectively); fecal mass and water content increased markedly (from 98 to 174 g/d and from 61 to 72%, respectively); and fat digestibility was unaffected. Prececal phosphorus digestibility from two diets containing TVP was lower than that from the all-beef diet (13 and 17%, vs 26%, P < 0.05). In conclusion, TVP is a useful source of protein in canine canned diets because amino acids from TVP are almost as digestible as those from beef in the canine intestine. Nevertheless, soy carbohydrate is poorly digested and large amounts of TVP inhibit small intestinal electrolyte digestibility and increase fecal water content.

ABSTRACT

Modified and chimeric cytokines have been developed to aid in the recovery of hematopoietic precursor cells after myeloablative chemotherapy. The interleukin-3 (IL-3) receptor agonist, daniplestim, binds to the IL-3 receptor-alpha subunit with 60-fold greater affinity and induces cell proliferation and colony-forming unit formation 10- to 22-fold better than native IL-3. A chimeric cytokine, myelopoietin-1, composed of daniplestim and a G-CSF receptor agonist binds both the IL-3 and G-CSF receptors. While the in vivo effects of daniplestim and myelopoietin-1 are well described, the mechanisms by which they stimulate growth are not well understood. We have investigated the effects of daniplestim and myelopoietin-1 on the prevention of apoptosis in two human hematopoietic cell lines, OCI-AML.5 and AML 193. Daniplestim and myelopoietin-1 prevented apoptosis to a greater degree than native recombinant IL-3 or G-CSF as determined by annexin V/propidium iodide binding and TUNEL assays. Daniplestim and myelopoietin-1 promoted the maintenance of the mitochondrial membrane potential better than native IL-3 or G-CSF. These cytokines promoted a lower redox potential as higher levels of free radicals were detected after cytokine treatment than in cytokine-deprived cells implying increased respiration. These results indicate that daniplestim and myelopoietin-1 are able to prevent apoptosis in hematopoietic cells more effectively than native IL-3 and G-CSF. These effects of daniplestim and myelopoietin-1 may contribute to their effective ability to repopulate hematopoietic precursor cells after chemotherapy.
