ABSTRACT

Transporting livestock at high stocking density by ship presents significant risks to their welfare, especially if it is over long distances. Previous research has investigated small variations in density for long periods or a moderate variation for short periods. The objective of this study was to assess the effects of a doubling of space allowance during two types of simulated ship movement, regular and irregular floor motion, on the welfare of sheep for a short one-hour period. Six 25 kg sheep were restrained in pairs in a crate on a programmable platform that generated roll and pitch motion typical of that experienced on board ship. Sheep were subjected to regular or irregular movement or a control treatment at high and low stocking densities (0.26 and 0.52 m2/sheep) in a multilevel changeover design. Irregular movement was programmed as a sequence of 30 different amplitude and duration values for pitch and roll movements, which were randomly selected by computer software controlling the movement. Regular movement was the mean of these values, which represented approximately 33% of the recommended maximum tolerance for livestock carriers. Behaviour was recorded by six cameras positioned around the crate. The low space allowance increased sheep pushing each other (Low: 4.51 events/h, High: 1.37 events/h, p < 0.001), affiliative behaviour, with their heads one on top of the other (Low 8.64, High 3.75 s/h, p = 0.02) and standing supported by the crate (Low 96, High 3.2 s/h, p < 0.001). Sheep stepped more frequently when more space was provided, particularly in the forward (Low 6.4, High 8.4 steps/h, p = 0.02) and left (Low 4.0, High 4.7 steps/h, p = 0.03) directions. The low space allowance group also had i heart rates, providing evidence of physiological stress. Irregular movement reduced rumination (Irregular 288, Control 592, Regular 403 s/h, p = 0.02), which was evidence of reduced welfare, but balance corrections by stepping were more common if the motion was regular. Thus, there was evidence that the low space allowance increased interactions between sheep and was stressful, and that irregular floor motion in simulated ship transport limited balance control and reduced welfare.

Veterinarians regularly face animal ethics conflicts, and research has identified the moral reasoning methods that they utilize to solve these. It is unclear whether students' sensitivity to animal ethics conflicts influences their reasoning methods, and the recent development of appropriate tests allows this to be assessed. We compared the moral reasoning methods, intended action and sensitivity of 112 first-year veterinary students in two contrasting veterinary schools, in Australia and Turkey. Students were presented with two animal ethics issues: breeding blind hens to address welfare concerns in intensive housing, for moral reasoning evaluation; and a video of a lame dairy cow walking, for sensitivity assessment. The sensitivity score was not related to the principal moral reasoning methods, which are Personal Interest (PI), Maintaining Norms (MN), and Universal Principles (UP). However, less sensitive students were more concerned about professional criticism of emotional reactions when addressing the hen scenario. Turkish students, mostly males, used more MN reasoning when deciding the hen dilemma. Australian, mostly female, students did not. Overall, female students were more likely to consider the universal moral principles in moral reasoning than male students and were more likely to recommend against breeding blind hens. This suggests that females are more likely to consider the ethical implications of their actions than males. This study demonstrates relationships between ethical sensitivity (ES) and moral reasoning, and cultural and gender effects on moral action choices. Students placing greater importance on professional criticism about having an emotional reaction are more likely to be those who have less ES.

ABSTRACT

The aim of this study was to investigate the presence of Staphylococcus aureus and staphylococcal enterotoxin (SE) genes in Urfa cheese samples and to characterize the enterotoxigenic potential of these isolates. From a total of 127 Urfa cheese samples, 53 isolates (from 41.7% of the samples) were identified by a species-specific PCR assay as S. aureus. Of these isolates, 40 (75.5%) gave positive PCR results for the 3' end of the coa gene. The coa-positive S. aureus strains were characterized for their population levels and enterotoxigenic properties, including slime factor, ß-lactamase, antibiotic susceptibilities, production of the classical SEs (SEA through SEE), in both cheese and liquid cultures by enzyme-linked immunosorbent assay (ELISA) and for the presence of specific genes, including classical SE genes (sea through see), mecA, femA, and spa, by PCR. The genetic relatedness among the coa-positive S. aureus isolates was investigated by PCR-based restriction fragment length polymorphism (RFLP) analysis and the 23S rRNA gene spacer. The 23S rRNA gene spacer and coa RFLP analysis using AluI and Hin6I revealed 14 different patterns. SEB, SEC, and SEA and SEE were detected by ELISA in three cheese samples. Fourteen S. aureus strains harbored enterotoxin genes sea through see, and three strains carried multiple toxin genes. The most commonly detected toxin gene was sec (25% of tested strains). Of the 40 analyzed S. aureus strains, 3 (7.5%) were mecA positive. Based on tandem repeats, four coa and spa types were identified. The results of this study indicate that S. aureus and SEs are present at significant levels in Urfa cheese. These toxins can cause staphylococcal food poisoning, creating a serious hazard for public health.

ABSTRACT

Recombinant interleukin-10 (rIL10) has been found to suppress the synthesis of tumour necrosis factor (TNF), interleukin-1 (IL-1), interleukin-6 (IL-6) and tissue factor and to improve survival from experimental sepsis. The aim of this study was to evaluate the protective effect of rIL-10 on lipopolysaccharide-(LPS-) induced haematological and biochemical disturbances in rats. In the present study, 40 rats were used and divided equally into four groups. Group 1 (control group, C) was treated with 0.9% saline. Group 2: LPS was injected intravenously (1.6 mg/100 g), Group 3 received rIL10 treatment (125 µg/kg) 2 min before 0.9% saline injection, Group 4 received rIL10 treatment 2 min before endotoxin treatment. When compared with the controls, platelet count, leukocyte count (with a marked neutrophilia and lymphopenia) and fibrinogen were decreased, while activated partial thromboplastin time (APTT) and prothrombin time (PT) were prolonged in the endotoxaemic rats. In addition, LPS caused statistically significant increases in plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities as well as creatinine, cholesterol and triglyceride concentrations, while it caused a statistically significant decrease in glucose, total protein and albumin levels as compared to the control group. On the other hand, rIL10 significantly suppressed disturbances in the haematological and biochemical parameters associated with endotoxaemia. As a result, rIL10 may be efficacious in preventing haematological disorders, tissue damage and changes in lipid, protein and carbohydrate metabolism in endotoxaemia.
