ABSTRACT

The objective of this paper was to report the isolation of two fluoroacetate degrading bacteria from the rumen of goats. The animals were adult goats, males, crossbred, with rumen fistula, fed with hay, and native pasture. The rumen fluid was obtained through the rumen fistula and immediately was inoculated 100 µL in mineral medium added with 20 mmol L(-1) sodium fluoroacetate (SF), incubated at 39°C in an orbital shaker. Pseudomonas fluorescens (strain DSM 8341) was used as positive control for fluoroacetate dehalogenase activity. Two isolates were identified by 16S rRNA gene sequencing as Pigmentiphaga kullae (ECPB08) and Ancylobacter dichloromethanicus (ECPB09). These bacteria degraded sodium fluoroacetate, releasing 20 mmol L(-1) of fluoride ion after 32 hours of incubation in Brunner medium containing 20 mmol L(-1) of SF. There are no previous reports of fluoroacetate dehalogenase activity for P. kullae and A. dichloromethanicus. Control measures to prevent plant intoxication, including use of fences, herbicides, or other methods of eliminating poisonous plants, have been unsuccessful to avoid poisoning by fluoroacetate containing plants in Brazil. In this way, P. kullae and A. dichloromethanicus may be used to colonize the rumen of susceptible animals to avoid intoxication by fluoroacetate containing plants.

ABSTRACT

The aim of this work was to isolate and identify bacteria able to degrade sodium fluoroacetate from soil and plant samples collected in areas where the fluoroacetate-containing plants Mascagnia rigida and Palicourea aenofusca are found. The samples were cultivated in mineral medium added with 20 mmol L(-1) sodium fluoroacetate. Seven isolates were identified by 16S rRNA gene sequencing as Paenibacillus sp. (ECPB01), Burkholderia sp. (ECPB02), Cupriavidus sp. (ECPB03), Staphylococcus sp. (ECPB04), Ancylobacter sp. (ECPB05), Ralstonia sp. (ECPB06), and Stenotrophomonas sp. (ECPB07). All seven isolates degraded sodium-fluoroacetate-containing in the medium, reaching defluorination rate of fluoride ion of 20 mmol L(-1). Six of them are reported for the first time as able to degrade sodium fluoroacetate (SF). In the future, some of these microorganisms can be used to establish in the rumen an engineered bacterial population able to degrade sodium fluoroacetate and protect ruminants from the poisoning by this compound.

ABSTRACT

This article reports a new case of protothecosis by Prototheca wickerhamii in goats. The animal presented severe respiratory difficulty and nodules, sometimes ulcerated, in the nasal vestibule, mucocutaneous junction of the nostrils and skin of the face. Prototheca wickerhamii was isolated from the lesions. The animal had no clinical or haematologiccl evidence of immunodepression. The agent was highly resistant to antimicrobial drugs. The goat was treated unsuccessfully with fluconazole and euthanised 10 months after the diagnosis of the disease. Histological lesions were necrotising pyogranulomatous dermatitis, rhinitis and osteomyelitis with myriads of walled sporangia characteristic of P. wickerhamii. It is suggested that in goats, protothecosis is characterised by a chronic, slowly progressive infection, which affects immunologically competent goats, causing multifocal, ulcerative, pyogranulomatous and necrotising lesions of the mucosa of the nasal vestibule, mucocutaneous junctions of the nostrils and skin of the face.

ABSTRACT

Foi realizado um estudo da mastite subclínica em nove rebanhos de cabras leiteiras no semiárido paraibano com o objetivo de determinar a ocorrência da infecção, avaliar o perfil microbiológico e celular do leite, testar a sensibilidade dos microorganismos isolados frente a antimicrobianos além de identificar os fatores de risco. Foram utilizadas 131 cabras leiteiras das quais foram colhidas 261 amostras de leite para exame microbiológico e 131 para contagem de células somáticas (CCS). Na ocasião das colheitas foi realizado o California Mastitis Test (CMT) e aplicado um questionário epidemiológico por propriedade. Houve crescimento bacteriano em 30 amostras (11,49 por cento) com 25 (83,33 por cento) dos isolados identificados como Staphylococcus coagulase negativa e cinco (16,66 por cento) Staphylococcus aureus. A média de CCS foi de 1,39x10(6) células/ml. O CMT apresentou baixa sensibilidade (46,7 por cento) e baixa especificidade (60,6 por cento) quando comparado ao exame microbiológico. A gentamicina e a associação da neomicina, bacitracina e tetraciclina foram os antimicrobianos contra os quais os microrganismos isolados apresentaram 100 por cento de sensibilidade. Penicilina e ampicilina foram os de maiores índices de resistência (66,67 por cento e 63,89 por cento, respectivamente). A caprinocultura não ser a atividade principal da propriedade e o não isolamento de animais doentes, foram identificados como fatores de risco para a mastite subclínica caprina nas propriedades estudadas. Programas de controle e profilaxia da mastite devem ser implementados enfocando as medidas de higiene na ordenha e correção dos fatores de risco identificados nesse estudo.


A subclinical mastitis study was conducted in nine dairy goat herds in the semi-arid region of Paraíba state, Northeastern Brazil, to determine the occurrence of infection, to evaluate microbiological and cellular profiles of the milk, to test the sensitivity of isolated microorganisms to antimicrobials, and to identify risk factors. One hundred thirty-one dairy goats were used, 261 samples were collected for microbiological culture and 131 samples for somatic cells count (SCC). During collection, the California Mastitis Test (CMT) was conducted and an epidemiological questionnaire was applied for each herd. There was bacterial growth in 30 samples (11.49 percent), with 25 (83.33 percent) coagulase-negative Staphylococcus and five (16.66 percent) Staphylococcus aureus isolated. The SCC mean was 1.39x10(6) cells/ml. CMT presented low sensitivity (46.7 percent) and low specificity (60.6 percent) compared with microbiological culture. Gentamicin and the association of neomycin, bacitracin and tetracyclin were the antimicrobials against which the microorganisms isolated showed 100 percent sensitivity. Penicillin and ampicillin had the greatest resistance rates (66.67 percent and 63.89 percent, respectively). Goat breeding is not the main activity on the farms and do not isolate diseased animals were identified as risk factors for caprine subclinical mastitis. Program for the control and prophylaxis of mastitis must be implemented focusing primarily on hygiene cares at milking and correction of the risk factors identified in this study.

ABSTRACT

Prototecose, causada por Prototheca zopfii ou P. wickerhamii, é uma doença emergente em animais e humanos. Em bovinos, P. zopfii é uma importante causa de mastite ambiental. Em cães e gatos, a prototecose é causada principalmente por P. zopfii. Em cães, causa infecção cutânea ou uma forma sistêmica envolvendo diversos órgãos. Em gatos, predominam as lesões tegumentares na região da face e plano nasal. No homem, a prototecose, causada principalmente por P. wickerhamii, manifesta-se sob três formas: cutânea, articular com bursite do olécrano e sistêmica. Pode ocorrer em indivíduos imunocompetentes, os quais podem apresentar bursite e/ou infecções cutâneas localizadas, ou em indivíduos imunossuprimidos, nos quais a enfermidade pode ser disseminada e/ou com envolvimento visceral. A prototecose causada por P. wickerhamii foi descrita recentemente em caprinos como causa de rinite afetando o vestíbulo nasal, união mucocutânea, pele da face e orelha. Nesta revisão são abordadas as características microbiologias e susceptibilidade a antimicrobianos do agente, e a epidemiologia, sinais clínicos, patologia, diagnóstico, tratamento e controle da prototecose em animais domésticos e no homem.


Protothecosis, caused by Prototheca zopfii or P. wickerhamii, is an emergent disease of human and animals. In cattle, P. zopfii is an important cause of environmental mastitis. In dogs and cats protothecosis is caused mainly by P. zopfii, causing cutaneous infections or a systemic form affecting many organs in dogs, and cutaneous infection affecting mainly the skin of the face and nose in cats. In humans, protothecosis, caused mainly by P. wickerhamii, occurs in three forms: cutaneous; olecran bursitis; and disseminated. The lesion is usually localized in the site of inoculation in immunocompetent individuals; however, in immunocompromised patients, it can become widespread. Protothecosis caused by P. wickerhamii was recently reported in goats causing rhinitis and dermatitis of the face and pinna. This paper reviews microbiologic characteristics and susceptibility to antimicrobials of Prototheca spp., and the epidemiology, clinical signs, pathology, diagnosis, treatment and control of protothecosis.
