ABSTRACT

Chronic mitral regurgitation (MR) in dogs results in pulmonary congestion and increased cardiac angiotensin-converting enzyme (ACE) activity and angiotensin (ANG) II levels. ACE could contribute to altered pulmonary vasomotion in heart failure, and ACE inhibitor (ACEI) therapy may normalize pulmonary vasomotion. We evaluated pulmonary artery (PA) responses to ANG II and bradykinin (BK) in control dogs, in dogs with 4 mo of MR, in MR dogs treated with the ACEI ramipril (MR + R), and in control dogs treated with ramipril (C + R). Mean PA systolic pressure increased in MR dogs (21 +/- 4 mmHg) but was normal in MR + R dogs (13 +/- 1 mmHg). Constriction of PA rings to ANG II was depressed in MR dogs. ACEI treatment (MR + R) restored ANG II responsiveness, but peak ANG II response (3.6 +/- 0.2 g) in MR + R dogs remained lower than in C + R dogs (4.7 +/- 0.2 g). Endothelium-dependent relaxation to BK was decreased (-87 +/- 4% C, -65 +/- 4% MR; P < 0.05). Ramipril (MR + R) restored relaxation to BK. This demonstrates that pulmonary congestion results in impaired pulmonary vasomotion to ANG II and BK, which ACEIs could normalize, supporting the use of ACEIs in clinical management of chronic congestive heart failure.

ABSTRACT

We tested the hypothesis that angiotensin-converting enzyme (ACE) inhibitor therapy prevents volume-overload hypertrophy in dogs with chronic mitral regurgitation (MR). Seven adult mongrel dogs receiving ramipril (R; 10 mg orally, twice/day) for 4 mo were compared with 11 dogs receiving no R (N) for 4 mo after induction of MR. Cine-magnetic resonance imaging demonstrated that left ventricular (LV) mass increased in the R-MR dogs [80 +/- 4 (SE) to 108 +/- 7 g, P < 0.01] and in the N-MR dogs (92 +/- 7 to 112 +/- 8 g, P < 0.001). LV myocyte cell length was greater in the R-MR and N-MR dogs (203 +/- 6 and 177 +/- 10 microns, respectively) than in normal (144 +/- 4 microns, P < 0.05) dogs. There was significant loss of the collagen weave pattern by scanning electron microscopy in both R-MR and N-MR dogs. LV ACE and chymase activities were significantly elevated in R-MR and N-MR compared with normal dogs. LV angiotensin II (ANG II) levels in the R-MR dogs (28 +/- 12 pg/g) were reduced to levels seen in normal dogs (28 +/- 4 pg/g) compared with N-MR dogs (72 +/- 11 pg/g, P < 0.05). Steady-state AT1-receptor mRNA levels decreased 66% in N-MR compared with normal dogs (P < 0.001) and increased 1.5-fold in R-MR compared with normal dogs (P < 0.01). Thus upregulation of the AT1 receptor in the R-MR hearts may provide a mechanism by which normal intracardiac ANG II levels could continue to mediate LV hypertrophy. However, the mechanism of dissolution collagen weave in both N-MR and R-MR hearts may be related to the stretch of volume overload.

ABSTRACT

The current study was designed to test the hypothesis that intracardiac angiotensin-converting enzyme (ACE) activity, chymase-like activity, and angiotensin (ANG) peptide levels are increased and are positively related to wall stress estimates in response to the chronic low pressure volume overload of mitral regurgitation produced by percutaneous chordal rupture in the dog. Chronic mitral regurgitation (MR) resulted in an increase in left ventricular (LV) end-diastolic volume [59 +/- 11 (SD) to 103 +/- 32 ml, P < 0.001], LV mass (96 +/- 17 to 114 +/- 23 g, P < 0.001), and a decrease in the LV mass-to-end-diastolic volume ratio (1.64 +/- 0.22 to 1.16 +/- 0.23 g/ml, P < 0.001) measured by magnetic resonance imaging. In vitro studies of heart tissue extracts demonstrated that the majority of ANG II-forming activity was from chymase-like activity rather than from ACE activity in five normal (83.5 +/- 7.5 vs. 6.04 +/- 5.2%) and seven MR hearts (86 +/- 3.9 vs. 2.6 +/- 1.7%). ACE activity (1.22 +/- 0.22 vs. 3.55 +/- 0.62 mU/g, P < 0.05) and chymase-like activity (9.42 +/- 4.64 vs. 20.60 +/- 8.41 nmol.g-1.min-1, P < 0.05) were increased in MR compared with normal hearts. ACE activity correlated with the LV mass-to-volume ratio (r = -0.93, P < 0.001) and LV diastolic wall stress ( r = 0.71, P < 0.05); however, chymase-like activity did not correlate with any hemodynamic parameter. ANG II levels were significantly higher in the midwall of the left ventricle in MR hearts than in normal controls (85 +/- 39 vs. 27 +/- 16 pg/g, P < 0.01). Our results demonstrate a positive correlation between LV diastolic wall stress and increased ACE activity with increased ANG II stores, suggesting that mechanical wall stress activated intracardiac ACE. Although chymase accounted for most ANG II formation in vitro in extracts of both normal and MR dog hearts, the significance of this enzyme in vivo remains unclear.

ABSTRACT

The present study assessed the canine prostate gland using B-mode real-time ultrasonography and correlated these findings with corresponding histologic sections. The prostate glands of 10 dogs were suspended in a saline bath and were scanned with an ultrasound probe. The glands were fixed and histologic sections were made. The sonograms were visually compared with the histologic sections. Areas having high collagenous tissue content appeared hyperechoic on the ultrasound scans. This hyperechogenicity was seen in the diffusely hyperechoic immature gland, the hyperechoic periurethral stroma, and the hyperechoic butterfly-shaped area seen on transverse scans. The butterfly-shaped area was isoechoic, with an elongated oval area seen on transverse scans. Areas of glandular epithelial content appeared hypoechoic on ultrasound scans. No zones comparable to the human prostate were observed.

ABSTRACT

After a preliminary finding of ultrasonographically detectable ring structure within the yolk of avian eggs, a study was initiated to investigate these properties. Ultrasonographically detectable alternating hyperechoic and hypoechoic rings within the egg yolk were found to be consistent but variability occurred in ring numbers, thickness, and clarity. Rings were best seen at two days post oviposition and were not affected by refrigeration or heating of the yolks. The usefulness of rings in experimental studies of yolk content of lipoprotein is postulated.

ABSTRACT

Thirty superovulatory treatments were administered to 19 mixed-breed, nonlactating cows. In 10 superovulatory treatments, the cows were primed with follicle stimulating hormone (FSH) on the second and third day of the estrous cycle, and in another 10 superovulatory treatments, the cows received no priming dosage of FSH. Initiation of the superovulatory treatments in both groups was determined by ultrasonically monitoring for regression of the dominant anovulatory follicle. Still another 10 superovulatory treatments were begun on Day 10 without regard for regression of the dominant anovulatory follicle and without a priming dosage of FSH. The mean days for starting the superovulatory treatment in the FSH-primed cows, in the nonprimed cows and in the controls were 10.5, 11.9 and 10 days, respectively. All cows were treated with eight injections of FSH at 12-hour intervals in a declining dosage (36 mg total). Cows were bred naturally and embryos collected nonsurgically seven days later. There was no significant difference (P>0.05) between the total number of embryos or transferable embryos in the three treatment groups. In this study neither priming on Days 2 or 3 nor initiating the superovulatory treatment, based on the morphologic regression of the dominant anovulatory follicle, was an effective means for improving the superovulatory response in cattle.

ABSTRACT

The relative antiandrogen-induced prostate involution activity of the newly synthesized hydroxyflutamide pro-drug was compared with that of flutamide in 25 Beagles. Secondary antiandrogen activity of both drugs on the testes and mammary tissue was investigated. Daily oral administration of both compounds at 2 dosages (ie, 2.5 and 5.0 mg/kg of body weight) during a 7-week period was monitored by transabdominal ultrasonography of the prostate twice a week. Cross-sectional area estimates of the prostate gland calculated from oblique dorsoventral, and transverse sonographic measurements were diminished significantly in some of the treated dogs as early as day 14 of drug administration. All treated dogs had significant differences in reduction by day 47. Involution was related directly to dose (P less than 0.05), but no difference was observed between test compounds. Differences in secondary antiandrogen activity were not remarkable. Flutamide was not found to have any activity advantage in vivo over hydroxyflutamide. It was concluded that ultrasonography can be a highly effective means of monitoring prostate size, and of monitoring drug-induced involution over time.

ABSTRACT

Ten adult rams were used in a study to compare ultrasonographic and physical measurements of testicles. Scrotal circumference measurements were correlated with both physical and ultrasonographic measurements and with semen analysis results. Scrotal circumference correlated well with most of the physical and ultrasonographic measurements and with semen motility and percent abnormalities. Ultrasonographic measurements correlated with scrotal circumferences but not with the semen analysis. There were significant differences in physical and ultrasonographic measurements of testicle circumference and diameter, especially when the ultrasonographic measurements were made through the intact scrotal wall. There appear to be problems in the accuracy of ultrasonographic measurements of testicle size in rams.

ABSTRACT

The carotid artery of clinically normal goats was examined, using duplex ultrasonography and arteriography. The diameter of the carotid artery was measured by use of two-dimensional ultrasonography and Doppler ultrasonography, respectively, before and after xylazine administration. The diameter of the artery was also measured by use of an arteriography technique in halothane-anesthetized goats. There was no significant difference between the mean diameter of the carotid artery measured by ultrasonography in conscious nonsedated goats and that determined by arteriography in goats under halothane anesthesia. On the other hand, ultrasonography of xylazine-sedated goats revealed an increase of carotid artery diameter of 20 to 30%. There was no change in the velocity of blood flow after xylazine administration.

ABSTRACT

Four polled Hereford bulls were found to have satisfactory breeding soundness examinations. They were examined by B-mode ultrasonography, at which time the testicular diameter was measured by ultrasound. The testicles were removed, measured physically and this data was compared with the ultrasound measurements and correlated with other parameters of the breeding soundness examination. The testicles from bulls with normal breeding soundness examinations appeared ultrasonographically identical with the normal testicles from other species such as the pig and dog. Results indicated that the bull testicle diameter could be accurately measured by ultrasonography. Neither ultrasonographic nor physical testicular diameter measurements correlated statistically with scrotal circumference, but they did correlated well with testicular circumference, weight and volume.

ABSTRACT

To provide a noninvasive means for studying individual macroscopic blood vessels, an ultrasound scanner was modified to provide a recording of blood vessel diameter. The instrument has A- and B-modes of signal display. The B-mode is used to position the probe and the A-mode to measure vessel diameter. The A-mode has two electronic gates for tracking the echo from each of two structures, i.e., near and far wall of vessel. The front gate was modified to pick up the falling rather than rising edge of a peak generated by the vessel wall. An analog signal proportional to the distance between gates was derived for recording with a strip-chart recorder. Probe holders were constructed to optimize positioning and holding of probe. Stability was excellent (reading varied 0.05 mm over 1 h). Axial resolution was between 0.3 and 0.73 mm. Discrepancy of measurements of plastic tubing made by ocular and ultrasound varied from 1.1 to 4.6%. Discrepancy with lightly fixed vessels was 2.7-8.2%. Ex vivo measurements on vessels with viable smooth muscle were more variable, perhaps because of actual change during measurements. Changes in vessel diameter induced by change in hydrostatic pressure and exposure to histamine were recorded.

ABSTRACT

The testicles of 14 Landrace boars (7 at 9 months of age and 7 at 15 months of age) were measured and examined by routine physical examination. Semen analysis was done. The testicles were then examined and measured by B-mode ultrasonography. The ultrasonographic appearance and measurements were compared with the physical measurements and the semen analysis. Seminiferous epithelial area (SEA) data were obtained after the testicles were surgically removed. These data were compared with the ultrasonographic data and the other physical data. A correlation could not be found between semen analysis and either physical or ultrasonographic data or between SEA data and semen analysis. Significant differences were found between the 9-month-old and the 15-month-old boars relative to 5 measurements: SEA, tunica albuginea thickness, testicular diameter, testicular length, and epididymal diameter. A ratio of 4:1 was found between each of the average physical measurements (scrotal diameter, testicular-epididymal circumference, testicular-epididymal length) and ultrasonographic measurements of testicular diameter. A ratio of 2.6:1 was found between testicular diameter and epididymal tail diameter (cranial to caudal).

ABSTRACT

The udders and teats of cows were examined by brightness mode (B-mode) ultrasonography. A 5-MHz linear array transducer and a 5-MHz or 10-MHz mechanical sector transducer were used in the examinations. In lactating animals, the teat sinus, gland sinus, and lactiferous ducts were imaged easily. The scans also allowed visualization of the layers of the teat wall. The annular fold and the folding of the mucosa at the junction of the teat sinus and the papillary duct were seen best with the 5-MHz and 10-MHz mechanical sector transducers. In one cow lacking milk flow from one quarter, a mass at the junction of the papillary duct and the teat sinus was observed ultrasonographically. Surgical removal of the mass resulted in a return of milk flow from that quarter.

ABSTRACT

A noninvasive technique, B-scan ultrasonography, was used to obtain detailed cross-sectional images of ocular and orbital structure and form. When a low-frequency probe (5 MHz) was used, tissue penetration was excellent; however, axial resolution was limited. The technique was used in a study of 2 cats and 1 dog. The use of low-frequency B-scan ultrasound should aid in the diagnostic examinations of space-occupying masses in the posterior portion of the ocular bulb and the orbit.

ABSTRACT

The results from water-bath studies on equine muscle in which foreign material had been inserted and then scanned ultrasonographically were compared with the results from scanning of the muscles of horses hospitalized for diagnosis of suspected foreign objects embedded in muscle. The water-bath studies showed that bone, wood, and large tendons all will demonstrate acoustic shadowing, but that subtle differences in the characteristics of the relative hyperechogenicity and acoustic shadows facilitate elimination of certain considerations in the differential diagnosis. Ultrasonography was effective in locating fistulous tracts and bone sequestra. A protocol for diagnosis of suspected foreign objects imbedded in muscle was proposed.

ABSTRACT

A total of 18 dogs were examined by ultrasound during their gestation period. At gestational day 7, a uterus indicative of possible pregnancy was observed. At gestational day 10, the embryo was observed. Fetal and cardiac activities appeared at 28 days. Rapid increases of the crown-rump length and biparietal diameter occurred between the 5th and the 6th weeks of gestation and of the body diameter between the 4th and 5th weeks and again between the 6th and the 7th weeks of gestation.
