ABSTRACT

UNLABELLED: Several authors have shown the effect of LH or hCG gonadotrophins on steroid secretion in the chick embryo ovary. However, the effects of luteinising hormone or human chorionic gonadotropin on chick embryo ovary innervation have not been studied. The purpose of the present work was to study the subcellular aspects of interstitial cells innervation in chick embryo ovaries cultured with LH or hCG. Coob's White Rock female chick embryos at 7, 11, 15 and 19 days of incubation (in ovo) were employed. Explants of the left and right ovaries were cultured separately for 4 days in: 1--Basic medium (control): minimum essential medium (MEM-GIBCO) with 10% bovine fetal serum, 2--Basic medium with added LH or hCG (problems). The cultures were processed to be studied ultrastructurally (TEM). Left and right ovary explants were fixed two hours at room temperature in Karnovsky's fixative. The tissues were then post-fixed in osmium tetroxide in 0.1 M cacodylate buffer for one hour and after a rinse in buffer were dehydrated in acetone: afterwards they were embedded in Araldite resin. Thick sections stained with 1% Toluidine blue were examined and photographed using a Zeiss Photo II microscope. Thin sections were stained with uranyl acetate and lead citrate and examined in a Siemens ElmisKop 101 electron microscope. CONTROLS: In both ovaries the isolated interstitial cells and the appearance of nerve fibers and nerve endings were observed in the explants of 11 day cultured during four days and in a similar way at the age of 15 days of in ovo development. PROBLEMS: With LH or hCG nerve fibers and nerve endings were observed both in the right gonad and in the medulla of the left ovary in close contact with the steroid-producing interstitial cells. Ultrastructurally, the interstitial cells are recognized by their typical organoids: SER, Golgi apparatus, mitochondria with tubular crests and abundant lipidic droplets. The innervation of these cells were seen in ovaries at 7 days development, cultured during 4 days. The present state corresponds to that of 11 days development in ovo. The nerve endings had mitochondria, microtubules and small and medium-sized vesicles, some with an electrodense content. These results in vitro suggest that innervation of the ovaries is controlled by an indirect mechanism via the hypothalamic-pituitary system and another local with production neurotrophic factors modulated by LH.

Diversos autores han demostrado el efecto de LH y hCG sobre la secreción esteroidea del ovario del embrión de pollo. Sin embargo, no se ha investigado el efecto de las hormonas luteinizante y gonadotrofina coriónica humana sobre su inervación. El propósito del presente trabajo fue estudiar los aspectos subcelulares de la inervación de las células intersticiales de ovarios de embrión de pollo cultivados con LH o hCG. Explantos de ovarios izquierdo (funcionante) y derecho (regresivo) de 14 embriones de pollo de 7, 11, 15 u 19 días de desarrollo in ovo de raza Cobb's White Rock fueron cultivados por separado durante 4 días en : 1 - Medio básico (contro); minimun essential medium (MEM-GIBCO)CON 10 de suero fetal bovino, 2-Medio básico con el agregado de LH o hCG (problemas). Los ovarios fueron seccionados en trozos de 2 a 3 mm de diámetro, lo que permitió realizar aproximadamente 30 cultivos de cada ovario. Controles: las características de las células intersticiales y la aparición de las fibras y terminaciones nerviosas observadas en los explantos de ambos ovarios 11 días cultivados durante cuartro días eran similares a los 15 días "in ovo". Problemas: con LH o hCG las fibras y terminaciones nerviosas fueron observadas en el ovario derecho y la médula del ovario izquierdo de 7 días cultivados durante cuatro días, en íntimo contacto con las células intersticiales productoras de esteroides. El presente estadio se correspondía con los 11 días de desarrollo "in ovo"Estos hallazagos sugieren que la inervación de los ovarios estaría controlada por un mecanismo indirecto vía hipotálamo-hipoficiario y por otro local con la producción de factores neurotróficos modulada por LH

ABSTRACT

The morpho-histochemical alterations that occur in the chicken ovary at 7 days of incubation after irradiation with He-Ne laser of a potency of 5 mw and at a wavelength of 632.8 nm were studied. The embryos were irradiated for 5 minutes through a window opened in the eggshell and aseptically maintained in incubator for 24 hours. The gonads were dissected out and processed for the following techniques: H/E, PAS, alcian blue, and toluidine blue. Controls: The ovaries were formed by a germinative or superficial epithelium, with germ and epithelial cells, and by primary sex cords compressed between them, although separated by a reduced stroma. The cords contained germ cells The surface coat of the germinative epithelium presented a thin layer of PAS positive, alcianophilic at pH 25 and orthochromatic material. Basement membranes and intercord extracellular substance were also PAS positive. Problems: Disorganization of the tissue structure was well manifest in irradiated gonads, accompanied by negativization of the histochemical reactions. A lymphocytic infiltration was also found. No structural alterations were observed in germ or epithelial cells. It is concluded that laser radiations would act producing decrease of the mucosubstances associated to the plasma membrane and basement membrane. They would also provoke the appearance of an inflammatory mononuclear infiltration.
