ABSTRACT

BACKGROUND/AIM: Significant biological differences in platelet-rich plasma (PRP) preparations have been highlighted and could explain the large variability in the clinical benefit of PRP reported in the literature. The scientific community now recommends the use of classification for PRP injection; however, these classifications are focused on platelet and leucocyte concentrations. This presents the disadvantages of (1) not taking into account the final volume of the preparation; (2) omitting the presence of red blood cells in PRP and (3) not assessing the efficiency of production. METHODS: On the basis of standards classically used in the Cell Therapy field, we propose the DEPA (Dose of injected platelets, Efficiency of production, Purity of the PRP, Activation of the PRP) classification to extend the characterisation of the injected PRP preparation. We retrospectively applied this classification on 20 PRP preparations for which biological characteristics were available in the literature. RESULTS: Dose of injected platelets varies from 0.21 to 5.43 billion, corresponding to a 25-fold increase. Only a Magellan device was able to obtain an A score for this parameter. Assessments of the efficiency of production reveal that no device is able to recover more than 90% of platelets from the blood. Purity of the preparation reveals that a majority of the preparations are contaminated by red blood cells as only three devices reach an A score for this parameter, corresponding to a percentage of platelets compared with red blood cells and leucocytes over 90%. CONCLUSIONS: These findings should provide significant help to clinicians in selecting a system that meets their specific needs for a given indication.

ABSTRACT

In Gram-positive bacteria, CodY is an important regulator of genes whose expression changes under conditions of nutrient limitation. Bacillus anthracis CodY represses or activates directly or indirectly approximately 500 genes. Affinity purification of CodY-DNA complexes was used to identify the direct targets of CodY. Of the 389 DNA binding sites that were copurified with CodY, 132 sites were in or near the regulatory regions governing the expression of 197 CodY-controlled genes, indicating that CodY controls many other genes indirectly. CodY-binding specificity was verified using electrophoretic mobility shift and DNase I footprinting assays for three CodY targets. Analysis of the bound sequences led to the identification of a B. anthracis CodY-binding consensus motif that was found in 366 of the 389 affinity-purified DNA regions. Regulation of the expression of the two genes directly controlled by CodY, sap and eag, encoding the two surface layer (S-layer) proteins, was analyzed further by monitoring the expression of transcriptional lacZ reporter fusions in parental and codY mutant strains. CodY proved to be a direct repressor of both sap and eag expression. Since the expression of the S-layer genes is under the control of both CodY and PagR (a regulator that responds to bicarbonate), their expression levels respond to both metabolic and environmental cues.

ABSTRACT

Due to the poor results obtained in the treatment of Peyronie's disease by ionophoresis and ultrasound, using the classical buffer solution of the cortisone thiomucase type, the authors have experimented ionophoresis with a new product: orgoteine a dismutase superoxide. Austrian authors have already demonstrated the value of this product. Its mechanism of action is still uncertain but it has an experimentally well demonstrated antiinflammatory activity. The good results are obvious not only on the pain, but also on the induration and incurvation, permitting the resumption of sexual intercourse in more than 75% of cases.

ABSTRACT

In face of the double reflex of fascination and defiance which is right away created by statistical methods by computers, the authors have attempted by means of precise examples to demystify these methods by recalling: 1) That computer statistics can be defined as the association of a service and a machine, the combination should be considered as an instrument and only an instrument. 2) That the application of computer statistics to odonto-stomatological research of course evidently enables the resolution of problems which would be otherwise insoluble, but above all considerably enlarges the field of research by rendering visible phenomena which would otherwise remain hidden. In this way, statistical methods by computer can be compared with the microscope; a low magnification with the optical microscope justifies only an overall view of the section while changing to higher magnifications permits the demonstration of details which would remain unsuspected. Use of the electromicroscope opens up another world for research. 3) In the same way as the biologist who uses the microscope, if he knows the principle of it is not in so far a specialist in optics, neither does the research woker who uses statistics necessarily have to be a trained statistician, on the one condition that evidently he disposes of professional computer logistics. This is the case of the ACB (Nantes) rush technical units, who cosigned this work, where the engineers who pose the problems to be resolved and emit the working hypotheses from the results supplied by the statistical studies are not the same people who carry out these very studies. 4) An essential fact, the research worker's independance is complete, only he can pose the problems and emit the hypothesis from the correlation, i. e. point of fact concomitance between the various isolated parameters. It therefore does not seem that weighty statistical techniques presently occupy the place of choice which should be theirs in stomatological research and more particularly in research concerning growth and development of the face where their use seems to be electively implied in face of the number and complexity of the parameters looked at.
