ABSTRACT

Sea water desalination is regarded as a major solution that could alleviate the water scarcity problem. Reverse osmosis (RO) is typically employed to recover fresh water from sea and brackish water via economical means. RO membrane fouling remains a critical issue restricting their widespread application. In this work, a tertiary thiophenal quaternary ammonium salt-based antibacterial agent was covalently reacted with cellulose acetate (CA) to obtain contact-active antibacterial quaternized CA-RO membrane (QCA-RO). The membrane was characterized by Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, scanning electron microscopy, water contact angle testing, and X-ray diffraction spectroscopy. The obtained QCA-RO membrane displayed good antibacterial activity against Pseudomonas aeruginosa and Staphylococcus aureus and had bactericidal rates of 99 % in the presence of visible light. Results showed that embedding the quaternary ammonium salt did not cause any significant changes to the morphology, mechanical performance, and thermal stability of the RO membrane. The method described in this work not only produces QCA-RO membranes with good anti-biofilm performance but also presents great potential in seawater desalination.

ABSTRACT

This Letter proposes a method that balances miniaturization and high performance of fiber optic speckle spectrometers. The periodically tapered coreless fiber is used as the scattering element to excite more higher-order modes in the coreless fiber. As a result, a remarkable spectral resolution of 0.03 nm in the near-infrared spectrum can be achieved with a 5-cm-long fiber. Narrow linewidth and broadband spectra in the wavelength of 1540-1560 nm are reconstructed separately, demonstrating the excellent performance of the designed all-fiber spectrometer. The spectral resolution of our proposed spectrometer is comparable to that of a 2-m multimode fiber spectrometer and has a significant improvement in miniaturization.

ABSTRACT

Curcumin is a polyphenolic compound that has been widely investigated for its health benefits. However, the clinical relevance of curcumin is limited due to its low water solubility and inefficient absorption. Therefore, curcumin is often encapsulated in nanocarriers to improve its delivery and function. In this study, composite nanoparticles composed of stearic acid-modified chitosan (SA-CS) and sodium caseinate (NaCas) were formed using sodium periodate-oxidized dextran with different molecular weights as a crosslinking agent. The effects of oxidized dextran (Odex) with different molecular weights on the composite nanoparticles were compared. The optimal SA-CS/NaCas/Odex composite nanoparticle (NPO) was obtained using an Odex (150 kDa)-to-SA-CS mass ratio of 2:1. Its size, polydispersity index (PDI), and zeta potential (ZP) were 130.2 nm, 0.149, and 25.4 mV, respectively. The particles were highly stable in simulated gastric fluid (SGF) in vitro, and their size and PDI were 172.3 nm and 0.263, respectively. The encapsulation rate of NPO loaded with curcumin (Cur-NPO) was 93% under optimal ultrasonic conditions. Compared with free curcumin, the sustained release of Cur-NPO significantly reduced to 17.9%, and free-radical-scavenging ability improved to 78.7%. In general, the optimal prepared NPO exhibited good GI stability and has potential applications in the formulation of orally bioactive hydrophobic drugs.

ABSTRACT

Respiratory syncytial virus (RSV) infection can cause life-threatening pneumonia and bronchiolitis, posing a significant threat to human health worldwide, especially to children and the elderly. Currently, there is no specific treatment for RSV infection. The most effective measures for preventing RSV infection are vaccines and prophylactic medications. However, not all population groups are eligible for the approved vaccines or antibody-based preventive medications. Therefore, there is an urgent need to develop novel vaccines and prophylactic drugs available for people of all ages. High-throughput assays that evaluate the efficacy of viral entry inhibitors or vaccine-induced neutralizing antibodies in blocking RSV entry are crucial for evaluating vaccine and prophylactic drug candidates. We developed an efficient entry assay using a lentiviral pseudovirus carrying the fusion (F) protein of type A or B RSV. In addition, the essential parameters were systematically optimized, including the number of transfected plasmids, storage conditions of the pseudovirus, cell types, cell numbers, virus inoculum, and time point of detection. Furthermore, the convalescent sera exhibited comparable inhibitory activity in this assay as in the authentic RSV virus neutralization assay. We established a robust pseudovirus-based entry assay for RSV, which holds excellent promise for studying entry mechanisms, evaluating viral entry inhibitors, and assessing vaccine-elicited neutralizing antibodies against RSV.

ABSTRACT

BACKGROUND: Shenmai injection is frequently utilized in China to clinically treat granulocytopenia in oncology patients following chemotherapy. Despite this, the drug's therapeutic benefits remain a topic of contention, and its active components and potential treatment targets have yet to be established. The present study utilizes a network pharmacology approach to investigate the drug's active ingredients and possible therapeutic targets, and to evaluate the effectiveness of Shenmai injection in treating granulocytopenia through meta-analysis. METHODS: In our subject paper, we utilized the TCMID database to investigate the active ingredients present in red ginseng and ophiopogon japonicus. To further identify molecular targets, we employed SuperPred, as well as OMIM, Genecards, and DisGeNET databases. Our focus was on targets associated with granulocytopenia. The DAVID 6.8 database was utilized to perform gene ontology functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis. Additionally, a protein-protein interaction network was established. The resulting "drug-key component-potential target-core pathway" network was used to predict the mechanism of action of Shenmai injection in the treatment of granulocytopenia. In order to evaluate the quality of the studies included in our analysis, we utilized the Cochrane Reviewers' Handbook. We then conducted a meta-analysis of the clinical curative effect of Shenmai injection for granulocytopenia, utilizing the Cochrane Collaboration's RevMan 5.3 software. RESULTS: After conducting a thorough screening, the study identified 5 primary ingredients of Shenmai injection - ophiopogonoside a, ß-patchoulene, ginsenoside rf, ginsenoside re, and ginsenoside rg1-that can potentially target 5 essential proteins: STAT3, TLR4, PIK3CA, PIK3R1, and GRB2. Additionally, Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed that Shenmai injection can be beneficial in treating granulocytopenia by interacting with pathways such as HIF-1 signaling, T-cell receptor signaling, PI3K-Akt signaling, chemokine signaling, and FoxO signaling. The results of meta-analysis indicate that the treatment group exhibited superior performance in terms of both efficiency and post-treatment leukocyte count when compared to the control group. CONCLUSION: In summary, studies in network pharmacology demonstrate that Shenmai injection exerts an impact on granulocytopenia via various components, targets, and mechanisms. Additionally, evidence-based studies provide strong support for the effectiveness of Shenmai injection in preventing and treating granulocytopenia.

ABSTRACT

Diabetes mellitus (DM) is a chronic metabolic disorder characterized by hyperglycemia resulting from insulin secretion defects or insulin resistance. The global incidence of DM has been gradually increasing due to improvements in living standards and changes in dietary habits, making it a major non-communicable disease that poses a significant threat to human health and life. The pathogenesis of DM remains incompletely understood till now, and current pharmacotherapeutic interventions are largely inadequate, resulting in relapses and severe adverse reactions. Although DM is not explicitly mentioned in traditional Chinese medicine (TCM) theory and clinical practice, it is often classified as "Xiaoke" due to similarities in etiology, pathogenesis, and symptoms. With its overall regulation, multiple targets, and personalized medication approach, TCM treatment can effectively alleviate the clinical manifestations of DM and prevent or treat its complications. Furthermore, TCM exhibits desirable therapeutic effects with minimal side effects and a favorable safety profile. This paper provides a comprehensive comparison and contrast of Xiaoke and DM by examining the involvement of TCM in their etiology, pathogenesis, treatment guidelines, and other relevant aspects based on classical literature and research reports. The current TCM experimental research on the treatment of DM by lowering blood glucose levels also be generalized. This innovative focus not only illuminates the role of TCM in DM treatment, but also underscores the potential of TCM in DM management.

ABSTRACT

To compare the pancreatic proteomics and autophagy between Rehmanniae Radix-and Rehmanniae Radix Praeparata-treated mice with type 2 diabetes mellitus(T2DM). The T2DM mouse model was established by high-fat diet coupled with streptozotocin(STZ, intraperitoneal injection, 100 mg·kg~(-1), once a day for three consecutive days). The mice were then randomly assigned into a control group, low-(5 g·kg~(-1)) and high-dose(15 g·kg~(-1)) Rehmanniae Radix groups, low-(150 mg·kg~(-1)) and high-dose(300 mg·kg~(-1)) catalpol groups, low-(5 g·kg~(-1)) and high-dose(15 g·kg~(-1)) Rehmanniae Radix Praeparata groups, low-(150 mg·kg~(-1)) and high-dose(300 mg·kg~(-1)) 5-hydroxymethyl furfuraldehyde(5-HMF) groups, and a metformin(250 mg·kg~(-1)) group. In addition, a normal group was also set and each group included 8 mice. The pancreas was collected after four weeks of administration and proteomics tools were employed to study the effects of Rehmanniae Radix and Rehmanniae Radix Praeparata on protein expression in the pancreas of T2DM mice. The expression levels of proteins involved in autophagy, inflammation, and oxidative stress response in the pancreatic tissues of T2DM mice were determined by western blotting, immunohistochemical assay, and transmission electron microscopy. The results showed that the differential proteins between the model group and Rehmanniae Radix/Rehmanniae Radix Prae-parata group were enriched in 7 KEGG pathways, such as autophagy-animal, which indicated that the 7 pathways may be associated with T2DM. Compared with the control group, drug administration significantly up-regulated the expression levels of beclin1 and phosphorylated mammalian target of rapamycin(p-mTOR)/mTOR and down-regulated those of the inflammation indicators, Toll-like receptor-4(TLR4) and Nod-like receptor protein 3(NLRP3), in the pancreas of T2DM mice, and Rehmanniae Radix showed better performance. In addition, the expression levels of inducible nitric oxide synthase(iNOS), nuclear factor erythroid 2-related factor 2(Nrf2), and heine oxygenase-1(HO-1) in the pancreas of T2DM mice were down-regulated after drug administration, and Rehmanniae Radix Praeparata demonstrated better performance. The results indicate that both Rehmanniae Radix and Rehmanniae Radix Praeparata can alleviate the inflammatory symptoms, reduce oxidative stress response, and increase the autophagy level in the pancreas of T2DM mice, while they exert the effect on different autophagy pathways.

ABSTRACT

BACKGROUND: The high-altitude-adapted frog Rana kukunoris, occurring on the Tibetan plateau, is an excellent model to study life history evolution and adaptation to harsh high-altitude environments. However, genomic resources for this species are still underdeveloped constraining attempts to investigate the underpinnings of adaptation. RESULTS: The R. kukunoris genome was assembled to a size of 4.83 Gb and the contig N50 was 1.80 Mb. The 6555 contigs were clustered and ordered into 12 pseudo-chromosomes covering ~ 93.07% of the assembled genome. In total, 32,304 genes were functionally annotated. Synteny analysis between the genomes of R. kukunoris and a low latitude species Rana temporaria showed a high degree of chromosome level synteny with one fusion event between chr11 and chr13 forming pseudo-chromosome 11 in R. kukunoris. Characterization of features of the R. kukunoris genome identified that 61.5% consisted of transposable elements and expansions of gene families related to cell nucleus structure and taste sense were identified. Ninety-five single-copy orthologous genes were identified as being under positive selection and had functions associated with the positive regulation of proteins in the catabolic process and negative regulation of developmental growth. These gene family expansions and positively selected genes indicate regions for further interrogation to understand adaptation to high altitude. CONCLUSIONS: Here, we reported a high-quality chromosome-level genome assembly of a high-altitude amphibian species using a combination of Illumina, PacBio and Hi-C sequencing technologies. This genome assembly provides a valuable resource for subsequent research on R. kukunoris genomics and amphibian genome evolution in general.

ABSTRACT

Applying the concept of residential co-reasoning, this research explored the role of the next of kin in the residential decision-making of childless older adults in rural China. We examined research questions regarding who, if anyone, had been the significant other(s) during the residential decision-making, and how they conducted the co-reasoning process. A constructivist grounded theory approach was applied, with 27 childless older adults being interviewed. We found that participants tended to choose to age in place if they had a continuous, reliable, and trustworthy relationship with certain members of next of kin, which they referred to as "I have someone at home." Acquiring care from kin was an ongoing negotiation process. Participants applied three types of negotiations: exchanging properties for care from kin; maintaining relational intimacy with kin; and counting on the filial obligation of kin. Otherwise, if care from kin was unavailable, participants tended to relocate to rural institutions.

ABSTRACT

We propose a refractive index (RI) sensor based on a tapered hole-assisted dual-core fiber (HADCF). The sensor is fabricated by splicing a tapered HADCF between two single-mode fibers and operates on the coupling between the fundamental mode and the low-order mode in two cores. The HADCF is tapered to meet the phase matching condition between the fundamental mode (LP01) in the central core and the low-order mode (LP11) in the eccentric core. The tapered waist of the fiber becomes thinner; the coupling wavelength has a blue shift. Glycerin solutions of different RIs were injected into the air hole. The RI sensitivity of 936.69 nm/RIU is obtained in the RI range of 1.335-1.360. The multi-channel RI sensor cascaded by HADCFs with different taper lengths is obtained and can simultaneously measure the RI of different solutions. The proposed device has the advantages of high sensitivity, simple structure, and stable performance. The special microfluidic channel in the HADCF can protect the tested solution from external environmental pollution.

ABSTRACT

Classification of the genus Rhacophorus has been problematic. In particular there has been considerable controversy surrounding the phylogenetic relationships among Rhacophorus rhodopus, R. bipunctatus, and R. reinwardtii. To examine the relationship among these Rhacophorus species, we assembled the complete mitochondrial genome sequence of R. rhodopus. The R. rhodopus genome is 15,789 bp in length with 12 protein-coding genes (PCGs) (losing ND5), two ribosomal genes, 22 transfer RNA genes, and a control region (D-loop). Base composition of the overall sequence was 60.86% for A + T content and 39.14% for C + G content. Most of the PCGs used ATG as a start codon, except for the COX I gene, which used the ATA start codon. COX I and ND6 used AGG and ATP8 stop codons respectively, while ND3 and ND4L used the TAA stop codon. For the remaining seven genes, the stop codons was incomplete. In addition, both 5' and 3' of the control areas had distinct repeating regions. Based on three datasets and two methods (Bayesian inference (BI) and maximum likelihood (ML)), we reconstructed three phylogenetic trees to explore the taxonomic status of the species and the phylogenetic relationship among R. rhodopus, R. bipunctatus and R. reinwardtii. Our results indicated that these three species are non-monophyletic; thus, the phylogenetic relationship among them is complex and difficult to determine. Further, R. rhodopus is divided into three lineages from different parts of China. The two Rhacophorus samples showed very close phylogenetic relationship with R. rhodopus. Our results add to the mitochondrial genome database of amphibians and will help to disentangle the phylogenetic relationships within the Rhacophoridae.

Fusarium head blight(FHB)caused by Fusarium graminearum species complex (FGSC) is one of the most important diseases around the world. Deoxynivalenol (DON) is a type of mycotoxin produced by FGSC when infecting cereal crops. It is a serious threat to the health of both humans and livestock. Trehalose-6-phosphate phosphatase (TPP), a conserved metabolic enzyme found in many plants and pathogens, catalyzes the formation of trehalose. N-(phenylthio) phthalimide (NPP) has been reported to inhibit the normal growth of nematodes by inhibiting the activity of TPP, but this inhibitor of nematodes has not previously been tested against F. graminearum. In this study, we found that TPP in F. graminearum (FgTPP) had similar secondary structures and conserved cysteine (Cys356) to nematodes by means of bioinformatics. At the same time, the sensitivity of F. graminearum strains to NPP was determined. NPP exhibited a better inhibitory effect on conidia germination than mycelial growth. In addition, the effects of NPP on DON biosynthesis and trehalose biosynthesis pathway in PH-1 were also determined. We found that NPP decreased DON production, trehalose content, glucose content and TPP enzyme activity but increased trehalose-6-phosphate content and trehalose-6-phosphate synthase (TPS) enzyme activity. Moreover, the expression of TRI1, TRI4, TRI5, TRI6, and TPP genes were downregulated, on the contrary, the TPS gene was upregulated. Finally, in order to further determine the control ability of NPP on DON production in the field, we conducted a series of field experiments, and found that NPP could effectively reduce the DON content in wheat grain and had a general control effect on FHB. In conclusion, the research in this study will provide important theoretical basis for controlling FHB caused by F. graminearum and reducing DON production in the field.

Negative ions powders (NIP) have been widely applied in many fields because of their natural electric field and far infrared radiation, especially in wastewater treatment. In this study, the NIP was first introduced into Fe3O4/H2O2 system to degrade methylene blue (MB). The MB removal was completely achieved at 5 h via a non-photochemical pathway and the degradation rate constant of this system is about 0.565 h-1, which is about 16 times higher than in Fe3O4/H2O2 Fenton-like system (0.035 h-1). In addition, the results of quenching experiments indicate that the electron (e-) and negative oxygen ion (•O2-) are the main reactive species. It was determined that Fe3O4@NIP is the effective component that leads to the activation of H2O2 to produce •OH, which derive from the pathway: NIP acts as an electron donor to reduce Fe(III) into Fe(II). Moreover, NIP can produce negative ions, which is also conductive to degradation. This study suggests a promising direction for the practical application of NIP based catalysis by integrating it with the Fe(III)/Fe(II) transformation process.

ABSTRACT

Particle size significantly affects the brightness of luminescent nanocrystals. Herein we firstly adopt a 1530 nm CW laser as the optical heating source to increase the particle size of Er3+ heavily doped nanocrystals, leading to giant enhancement of the luminescent intensity. The advantages of this method are mainly feature along the facile route, with an ultrafast process, and low threshold of the laser power density. The detailed mechanisms of the laser annealing are carefully investigated. In addition, fluorescence intensity ratio behaviours using different emission bands are comparatively investigated.

Subjects may voluntarily implement an internal or external strategy during mental rotation (MR) task. However, few studies have reported the spatiotemporal differences of brain activation between the two MR strategies. This study aims to compare the two strategies from the perspective of behavioral performance and spatiotemporal brain activations in each cognitive sub-stage using EEG measurements. Both the internal (IN) and external (EX) groups showed a significant 'angle effect' on reaction time (RT) and accuracy (ACC). However, a smaller increase of RT with rotation angle was observed in the EX group. Event-related (de)synchronization in the beta band revealed similar temporal patterns of brain activation in the two groups, but with a stronger activation in the MR sub-stage in the EX group. We speculate that MR of 3D abstract objects is easier when an external strategy is used, and would be promoted by an additional visual-spatial process involving the parietal-occipital areas. Our results suggested that the differences between the two strategies were mainly induced by main MR rather than other cognitive processes.

ABSTRACT

The influence of micronization on improving the phytoestrogenic effects of wheat bran was studied. Wheat bran samples were prepared by ball milling, and an animal experiment was carried out by feeding 8-month-old female rats wheat bran. The effect of wheat bran samples on serum estradiol (E2) and progesterone (P) levels of female 8-month-old rats was investigated. The wheat bran with a median diameter of 392.1 µm was micronized to 91.1 and 9.7 µm in median diameter by dry milling and wet milling for 5 hours, respectively. Microscopic observation and X-ray diffraction revealed more potential damage from wet milling than dry milling on the crystal structure of wheat bran granules. Almost all particles were dissolved and there was no obvious crystal peak in the 5-hour wet-milled wheat bran. The serum E2 and P levels of the 8-month-old rats fed wet-milled bran were increased significantly, 2.2 times higher than that of the same aged control group. The experimental results indicated that wet milling could destroy the crystal structure of wheat bran granules and improve the phytoestrogenic effects of wheat bran.

PURPOSE: To compare complications of percutaneous vertebroplasty (PVP) and balloon kyphoplasty (BKP) for the treatment of osteoporotic vertebral compression fractures (OVCFs). BACKGROUND: PVP and BKP are two minimally invasive procedures for treating OVCFs, while few studies emphases attention to intra- and post-operative complications about the two procedures. METHODS: Online databases were searched for studies comparing complications of PVP and BKP for OVCFs, the randomized controlled trials, clinical controlled trials and cohort studies that provided related data were identified. Demographic characteristics and complications related to procedures were extracted and analysed from all of the included studies. RESULTS: Nineteen studies encompassing 1,787 patients in total, of whom 887 received PVP and 900 received BKP, met the inclusion criteria. For subsequent fractures, our meta-analysis detected no significant difference between the two procedures, both for adjacent fractures (p = 0.29) and non-adjacent fractures (p = 0.37). For cement extravasations, there was no significant difference between the two interventions if considering disc spaces extravasations only (p = 0.24), while considering total extravasations and paravertebral extravasations, the cement leakage rate in the PVP group was significantly higher than the BKP group (total: p < 0.01; paravertebral: p < 0.01). CONCLUSIONS: The two procedures suffer from equal risk of subsequent spinal fractures; PVP has a significant higher cement leakage rate compared to BKP, mainly caused by a higher paravertebral leakage, patients with extremely poor pulmonary function or unstable haemodynamic are better candidates for BKP.

ABSTRACT

This study investigated mental rotation for identical stimuli and mirrored stimuli by both behavior response and event-related desynchronization (ERD) of EEG signals. Results showed that subjects had longer response time for mirrored stimuli than identical stimuli. Beta-band desynchronization appeared in whole brain with the parietal-occipital dominance. The ERD in beta band recovered slowly in an angular order after 450-600 ms of stimulus onset except the sharp rebound in the case of identical stimuli with rotation at 0 °. This temporal difference of beta ERD between the identical and the mirrored stimuli at 0° rotation and the ERD topographic difference in left fronto-parietal regions, together with the behavior difference, may imply an extra flip process in mirrored condition.

ABSTRACT

Numerous studies have shown that conjugated linoleic acid (CLA) can inhibit cancer cells growth and induce apoptosis in vitro and in vivo. The aim of the present study was to investigate the effects of CLA, including cis9, trans11-conjugated linoleic acid (c9, t11-CLA) and trans10, cis12-conjugated linoleic acid (t10, c12-CLA), on apoptosis of human endometrial cancer RL 95-2 cells and its related mechanisms. The MTT analysis was used to evaluate the effect of CLA isomers on the viability of endometrial cancer RL 95-2 cells. We then estimated the apoptosis by Morphological observation and Annexin V-FITC/PI staining and flow cytometry. We also used Western blot analysis to assess the expression of caspase-3, Bax, Bcl-2 proteins and the activation of Akt/p-Akt and ERα/p-ERα. Propylpyrazole-triol (PPT), a selective ERα agonist was used to confirm the induction of apoptosis by c9, t11 CLA may relate to ERα-mediated pathway. In CLA-treated RL 95-2 cells, we found that c9, t11-CLA inhibited viability and trigged apoptosis, as judged from nuclear morphology and flow cytometric analysis. The expression of caspase-3 and the ratio of Bax/Bcl-2 were significant increased, but no obvious change was observed about Akt and p-Akt in c9, t11-CLA-treated cells. However, the expression of total ERα level in RL 95-2 cells-treated with c9, t11-CLA was unchanged, while in the concentration of 80 mM, c9, t11-CLA down-regulated the protein expression level of p-ERα. Then PPT has the antagonistic action on growth inhibitory effect in RL 95-2 cells incubated with c9, t11-CLA. This study demonstrated that c9, t11- CLA could induce apoptosis in RL 95-2 cells, and may involve in ERα-mediated pathway. These results indicated that c9, t11- CLA could induce apoptosis of endometrial cancer cells and may be potential agents for the treatment of endometrial cancer.
