ABSTRACT

A highly efficient and operationally simple method for the synthesis of ß-sulfinyl alkenylsulfones through a BF3·OEt2-promoted reaction of alkynes and sodium sulfinates is developed, successfully avoiding the complicated anhydrous treatment before the reaction and greatly simplifying the reaction conditions. As a facile and selective route to the targets, it features good functional group compatibility, mild conditions, easily available starting materials, and excellent yields. Notably, the trace water in solvent plays a key role in promoting the reaction, which provides a more practical pathway for the utilization of the BF3·OEt2 catalytic system.

ABSTRACT

Developing potentially toxic metal ion probes is significant for environment and food safety. Although Hg2+ probes have been extensively studied, small molecule fluorophores that can integrate two applications of visual detection and separation into one unit remain challenging to access. Herein, by incorporating triphenylamine (TPA) into tridentate skeleton with an acetylene bridge, 2,6-bisbenzimidazolpyridine-TPA (4a), 2,6-bisbenzothiazolylpyridine-TPA (4b) and 2,6-bisbenzothiazolylpyridine-TPA (4c) were first constructed, expectably showing distinct solvatochromism and dual-state emission properties. Since the diverse emission properties, the fluorescence detection of 4a-4b can be achieved with an ultrasensitive response (LOD = 10-11 M) and efficient removal of Hg2+. More interestingly, 4a-4b can not only be developed into paper/film sensing platform, but also reliably detect Hg2+ in real water and seaweed samples, with recoveries ranging from 97.3% to 107.8% and a relative standard deviation of less than 5%, indicating that they have excellent application potential in the field of environmental and food chemistry.

ABSTRACT

Dual-state emission luminogens (DSEgens) as fluorophores emit efficiently in solution and solid forms have gained increasing concern in the field of chemical sensing. Recent efforts by our group led to the identification of DSEgens as an easy-to-visualize nitroaromatic explosives (NAEs) detection platform. However, none of the previously studied NAEs probes show effective improvement in sensitivity. Here, we designed a series of benzoxazole-based DSEgens through multiple strategies driven by theoretical calculations, revealing their improved detecting performance on NAEs. Compounds 4a-4e exhibit thermal- and photo-stability, large Stokes shift as well as sensitivity solvatochromism (except for 4a and 4b). A subtle balance between rigid conjugation and distorted conformation endows these D-A type fluorophores 4a-4e with DSE properties. Furthermore, 4d and 4e show aggregation-induced emission phenomenon caused by distorted molecular conformation and restricted intramolecular rotation. Interestingly, DSEgen 4e displays anti-interference and sensitivity towards NAEs with a detection limit of 10-8 M. It can be applied for expedient and distinct visual identification of NAEs not only in solution but also on filter paper and film, supporting this new DSEgen as reliable NAEs chemoprobe.

ABSTRACT

PURPOSE: To identify the expression profile of novel microRNAs (miRNAs) in colon cancer and evaluate their clinical applicability. METHODS: Differences in the expression of serum miRNAs in patients with colon cancer and healthy controls were identified using miRNA microarrays. Differentially expressed miRNAs were verified via real-time polymerase chain reaction (PCR) using sera from 50 patients with colon cancer and 44 healthy controls. These miRNAs were also verified in a double-blind validation experiment using sera from 30 patients with colon cancer, 30 patients with colonic polyps, and 30 healthy controls. RESULTS: Microarray hybridization revealed that 87 miRNAs were differentially expressed between the sera of patients with colon cancer and healthy controls. Among these miRNAs, 39 miRNAs were up-regulated, whereas 48 miRNAs were down-regulated. Verification of the expression of these miRNAs using real-time PCR revealed that the expression levels of miR-31, miR-141, miR-224-3p, miR-576-5p, and miR-4669 were significantly different between patients with colon cancer and healthy controls. Using these five miRNAs to construct a miRNA expression profile (or miRNA panel) will facilitate more effective diagnosis of colon cancer. CONCLUSION: Clinical analysis of miR-31, miR-141, miR-224-3p, miR-576-5p, and miR-4669 expression in patients with colon cancer may facilitate the diagnosis of colon cancer.

ABSTRACT

Human papillomavirus infection is a major health problem and caused substantial benign and malignancy diseases among female and male worldwide. We aim to investigate the epidemiology of high-risk human papillomavirus (HR-HPV) and related diseases in Suzhou population. As well as evaluating the potential benefit of a nine-valent HPV vaccine (regardless of HPV-6 and -11) in Suzhou. A total of 40,108 people aged 13-89 years were retrospectively examined by database retrieval from 2010 to 2015. Thirteen genotypes (16, 18, 31, 33, 35, 39, 45, 51, 52, 56, 58, 59, and 66) of HR-HPV were detected using Tellgenplex™ xMAP™ HPV DNA Test assay. The overall prevalence of HR-HPV was 21.1%, the female and male account for 96.4% and 3.6%, respectively. The infection rate among male (25.6%, 367/1,432) was significantly higher than that among female (20.9%, 8,100/38,676), X2 = 17.341 (P < 0.001), with OR = 1.293, 95% CI (1.146-1.460). The five most frequent HR-HPV genotypes were HPV-16 (5.12%), -52 (5.07%), -58 (3.02%), -39 (2.00%), and -18 (1.74%). HR-HPV infection rate was peak in person aged <20 years, and second higher in person aged 51-60 years. Infection modes as HPV-16, -18, -31, -33, -45, -52, -58 alone or mixed accounted for 63.2%. The top three prevalent diseases in HR-HPV infected women were cervicitis, vaginitis, and cervical lesions, and in men were verruca, urethritis, and balanitis, respectively. This is the first study to demonstrate HPV infection status in Suzhou population. Both women and men had a large burden of HPV infection. Objective@#To compare the clinical effects of transperitoneal (Tp) versus extraperitoneal (Ep) robot-assisted radical prostatectomy (RARP) in the treatment of localized prostate cancer.@*METHODS@#We searched PubMed, EMBASE, Web of Science, EBSCO, Cochrane Library, Wanfang, CNKI, and CBM for the articles comparing the clinical effect Tp-RARP with that of Ep-RARP in the treatment of localized prostate cancer published from January 2000 to November 2016. All the articles must meet the inclusion criteria, that is, dealing with at least one of the following aspects: operation time, intraoperative blood loss, postoperative catheterization time, length of bed confinement, perioperative complications, positive surgical margins, bowel-related complications, postoperative anastomotic leakage, and postoperative urinary continence. We subjected the data obtained to statistical analysis using the RevMan5.3 software.@*RESULTS@#Two randomized controlled trials and six case-control studies were included in this meta-analysis, involving 451 cases of Tp-RARP and 676 cases of Ep-RARP. Compared with Tp-RARP, Ep-RARP showed significantly shorter operation time (WMD = 21.39, 95% CI: 7.54-35.24, P = 0.002), shorter length of bed confinement (WMD = 0.85, 95% CI: 0.61-1.09, P <0.001), and lower rate of bowel-related complications (RR = 9.74, 95% CI: 3.26-29.07, P <0.001). However, no statistically significant differences were found between the two strategies in intraoperative blood loss (WMD = -8.12, 95% CI: -27.86-11.63, P = 0.42), postoperative catheterization time (WMD = 0.17, 95% CI: -0.55-0.21, P = 0.38), or the rates of perioperative complications (RR = 1.34, 95% CI: -0.97-1.87, P = 0.08), positive surgical margins (RR = 1.24, 95% CI: 0.95-1.61, P = 0.12), anastomotic leakage (RR = 0.98, 95% CI: 0.46-2.10, P = 0.95), urinary continence at 3 months (RR = 0.96, 95% CI: 0.91-1.00, P = 0.05) and urinary continence at 6 months (RR = 1.00, 95% CI: 0.97-1.02, P = 0.82).@*CONCLUSIONS@#Ep-RARP has the advantages of shorter operation time, shorter length of bed confinement and lower rate of bowel-related complications over Tp-RARP, and therefore may be a better option for the treatment of localized prostate cancer. However, more multi-centered randomized controlled clinical trials are needed for further evaluation of these two approaches.

ABSTRACT

Data quality management system is essential to ensure accurate, complete, consistent, and reliable data collection in clinical research. This paper is devoted to various choices of data quality metrics. They are categorized by study status, e.g. study start up, conduct, and close-out. In each category, metrics for different purposes are listed according to ALCOA+ principles such us completeness, accuracy, timeliness, traceability, etc. Some general quality metrics frequently used are also introduced. This paper contains detail information as much as possible to each metric by providing definition, purpose, evaluation, referenced benchmark, and recommended targets in favor of real practice. It is important that sponsors and data management service providers establish a robust integrated clinical trial data quality management system to ensure sustainable high quality of clinical trial deliverables. It will also support enterprise level of data evaluation and bench marking the quality of data across projects, sponsors, data management service providers by using objective metrics from the real clinical trials. We hope this will be a significant input to accelerate the improvement of clinical trial data quality in the industry.

ABSTRACT

Based on the integrated assessment methods of ecological quality status (EcoQS) in coastal waters abroad as well as the domestic related research status, 11 indices were selected from the biotic and physicochemical elements of water and sediment to built an integrated EcoQS assessment index system, and, with the comprehensive consideration of domestic and international evaluation standards, 5 levels of EcoQS classified as "high, good, moderate, poor, and bad" were determined. Then, an integrated assessment method of EcoQS in coastal waters was established by using analytic hierarchy process (AHP) and fuzzy mathematics method, and tested by the analysis of the correlations between the EcoQS grade and the major anthropogenic disturbances and pollutant concentrations of Tong' an Bay. The EcoQS of Tong' an Bay was assessed as moderate, i.e., the Bay was moderately disturbed and in transitional to poor status. The established integrated assessment method could not only reflect the major anthropogenic environmental pressure and risk factors, but also give an early warning of the pollutants satisfied by law in the study area.

ABSTRACT

The feasibility of separating intact bacterial cells by high-performance ion exchange chromatography typical for macromolecular study was investigated. A common HPLC system coupled with a light scattering detector was employed; TOYOPEARL SuperQ-650C, a beaded anion exchanger, was used as the column media; bacterial cell samples were prepared by suspending colonies in the equilibrium buffer. Piperazine-hydrochloric acid buffer (0.02 M, pH 7.0) was chosen to be the equilibrium buffer after screening different buffer systems. The absorbed cells were eluted by a linear gradient of NaCl from 0 to 1.00 M in the equilibrium buffer within 30 min, at a flow rate of 1.0 mL/min. A distinctive chromatographic profile with high reproducibility and accuracy was obtained with all of the six kinds of bacteria tested. The eluted fractions for each sample were confirmed by microscopic analysis and physiological or biochemical identification to the bacterial cells applied. More importantly, the eluted cells retained full viability. It is apparent that living bacteria cells exhibited similar behaviors and properties with molecules in anion exchange chromatography, implying that chromatographic methods can become an effective approach in the studies of intact bacterial cells.
