ABSTRACT

Objective:To observe the effects of sparrow-pecking moxibustion at Shenque(CV8)and Guanyuan(CV4)on the writhing reaction and score,the temperature and blood flow perfusion of moxibustion point area and uterus,the serum levels of arginine vasopressin(AVP),prostaglandin(PG)F2α,and thromboxane(TX)B2 in rats with primary dysmenorrhea(PD)due to cold-dampness stagnation,and to explore the possible mechanism of sparrow-pecking moxibustion in treating PD.Methods:Thirty-two healthy non-pregnant female Wistar rats were randomly divided into a normal group,a model group,an ibuprofen group,and a sparrow-pecking moxibustion group,with 8 rats in each group.Except for the normal group,the other three groups were subjected to modeling with cold water bath combined with estradiol benzoate and oxytocin injection.Rats in the sparrow-pecking moxibustion group were treated with sparrow-pecking moxibustion at Shenque(CV8)and Guanyuan(CV4)on the 8th day of modeling,30 min/time,once a day for 3 d;those in the ibuprofen group were treated with 0.8 mL ibuprofen solution(a specification of 125 mg in 10 mL)on the 8th day of modeling,once a day for 3 d;those in the normal group and the model group were given 0.8 mL normal saline,once a day for 3 d.On the 11th day,rats in each group were intraperitoneally injected with oxytocin(2 U/rat),and the writhing incubation period and writhing score in 20 min were observed;the temperature and the blood perfusion of Shenque(CV8),Guanyuan(CV4),and uterus in vivo were detected;the serum levels of AVP,PGF2α,and TXB2 were determined.Results:The writhing incubation period was significantly longer(P<0.01)and the writhing score was significantly lower(P<0.01)in the sparrow-pecking moxibustion group and the ibuprofen group than in the model group;compared with the ibuprofen group,the writhing incubation period was prolonged(P<0.01)and the writhing score was decreased(P<0.01)in the sparrow-pecking moxibustion group;compared with the normal group,the temperature and the blood perfusion of Shenque(CV8),Guanyuan(CV4),and uterus were significantly decreased,while the serum PGF2α,AVP,and TXB2 levels were significantly increased(P<0.01)in the model group;compared with the model group,the temperature and the blood perfusion of Shenque(CV8),Guanyuan(CV4),and uterus were significantly increased,and the serum levels of PGF2α,AVP,and TXB2 were significantly decreased in the ibuprofen group and the sparrow-pecking moxibustion group(P<0.05 or P<0.01);compared with the ibuprofen group,the temperature and the blood perfusion of Shenque(CV8),Guanyuan(CV4),and uterus were significantly increased(P<0.05),the serum AVP and TXB2 levels were significantly decreased(P<0.05),while the serum PGF2α level had no statistical difference in the sparrow-pecking moxibustion group(P>0.05).Conclusion:Sparrow-pecking moxibustion had a remarkable analgesic effect on the rats with PD due to cold-dampness stagnation,and the mechanism may be related to the increased temperature and blood perfusion of the moxibustion point area and uterus,as well as the decreased serum PGF2α,AVP,and TXB2 levels.

ABSTRACT

The global and local climate changes determine the producing of highly-adaptive common (bread) wheat commercial cultivars of a new generation whose optimal earliness matches the climatic features of the territory where the cultivars are farmed. Principal component analysis involving our own and published data has been applied to investigate 98 commercial common wheat cultivars from Western and Eastern Siberia comparing their morphotypes; cultivar zoning time; length of the vegetation period; 1000-grain weight, and inheritance of spring growth habit. It demonstrated that the dominant Vrn gene polymorphism determining the spring growth habit of the Siberian cultivars was minimally polymorphic. In 75 % of the tested cultivars, the spring growth habit was controlled by digenic, namely dominant Vrn-A1 and Vrn-B1 genes. In 25 % of them (24 cultivars), spring growth habit is controlled by a single gene. In 19 and 5 of these cultivars spring growth habit is controlled by only one dominant gene, Vrn-B1 or Vrn-A1, respectively. In cv. Tulun 15, a trigenic control was identified. A conclusion about the optimality of the digenic control for the climatic conditions of both Western and Eastern Siberia has been confirmed. However, since none of the tested cultivars had the dominant Vrn-D1 gene typical of the regions of China and Central Asia bordering Siberia, it can be considered as an additional argument in favor of the European origin of Siberian common wheat cultivars. The revealed high frequency of the Vrn-B1c allele in the Western Siberian cultivars and the Vrn-B1a allele in the Eastern Siberian cultivars suggests their selectivity. The analysis also confirmed the dominance of red glume (ferrugineum, milturum) and awned spike (ferrugineum, erythrospermum) varieties in the Eastern Siberian cultivars, and white glume and awnedless spike (lutescens and albidum) ones in the Western Siberian cultivars. Small grain size cultivars are more typical of Eastern than Western Siberia. The retrospective analysis based on the cultivars' zoning time included in the "State Register for Selection Achievements Admitted for Usage" brought us to the conclusion that the earliness/lateness of modern Siberian commercial cultivars was not regionally but rather zonally-associated (taiga, subtaiga, forest-steppe and steppe zones).

ABSTRACT

Objective: To observe the effects of ginger-partitioned moxibustion at Shenque (CV8) and Guanyuan (CV4) on the expression levels of endocrine-related molecules and their receptors in rats with primary dysmenorrhea (PD) due to cold-dampness stagnation, thus to explore their analgesic mechanisms. Methods: Thirty-two female Wistar rats were divided into a normal group, a model group, a ginger-partitioned moxibustion group, and a Western medicine group according to the random number table method, with 8 rats in each group. Except for rats in the normal group, all other rats were treated with oxytocin combined with ice-water bath to establish the rat models of PD due to cold-dampness stagnation. After successful modeling, rats in the normal group and the model group did not receive treatment; rats in the ginger-partitioned moxibustion group received treatments with ginger-partitioned moxibustion at Shenque (CV8) and Guanyuan (CV4); rats in the Western medicine group received ibuprofen by intragastric administration. The writhing response of rats was compared among groups, and the serum levels of prostaglandin F2α (PGF2α), estrogen (estradiol, E2), progesterone (P), and the mRNA expression of PGF2α and E2 receptors in the uterine tissues were detected. Results: No writhing behavior was observed in the normal group; compared with the normal group, the serum PGF2α and E2 levels in the model group were increased (P<0.01), while the P level was decreased (P<0.01), and the mRNA expression levels of the uterine PGF2α and E2 receptors were increased (P<0.01, P<0.05). Compared with the model group, the writhing behavior latency was prolonged, and the writhing response score was decreased in the ginger-partitioned moxibustion group and the Western medicine group (P<0.01); the serum PGF2α and E2 levels in the ginger-partitioned moxibustion group and the Western medicine group were decreased, while the P level was increased (P<0.05 or P<0.01); the mRNA expression levels of the uterine PGF2α and E2 receptors in the ginger-partitioned moxibustion group and the Western medicine group were decreased (P<0.05). Compared with the Western medicine group, the ginger-partitioned moxibustion group showed a prolonged writhing behavior latency, reduced writhing response score (P<0.05), and decreased serum E2 level (P<0.05), while no statistical differences in the serum PGF2α and P levels, or the mRNA expression levels of uterine PGF2α and E2 receptors (P>0.05).Conclusion: The analgesic effect of ginger-partitioned moxibustion on PD due to cold-dampness stagnation may be related to regulating the mRNA expression levels of PGF2α and E2 receptors in the uterine tissues.

ABSTRACT

Better methods to interrogate host-pathogen interactions during Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) infections are imperative to help understand and prevent this disease. Here we implemented RNA-sequencing (RNA-seq) combined with the Oxford Nanopore Technologies (ONT) long-reads to measure differential host gene expression, transcript polyadenylation and isoform usage within various epithelial cell lines permissive and non-permissive for SARS-CoV-2 infection. SARS-CoV-2-infected and mock-infected Vero (African green monkey kidney epithelial cells), Calu-3 (human lung adenocarcinoma epithelial cells), Caco-2 (human colorectal adenocarcinoma epithelial cells) and A549 (human lung carcinoma epithelial cells) were analysed over time (0, 2, 24, 48 hours). Differential polyadenylation was found to occur in both infected Calu-3 and Vero cells during a late time point (48 hpi), with Gene Ontology (GO) terms such as viral transcription and translation shown to be significantly enriched in Calu-3 data. Poly(A) tails showed increased lengths in the majority of the differentially polyadenylated transcripts in Calu-3 and Vero cell lines (up to ~136 nt in mean poly(A) length, padj = 0.029). Of these genes, ribosomal protein genes such as RPS4X and RPS6 also showed downregulation in expression levels, suggesting the importance of ribosomal protein genes during infection. Furthermore, differential transcript usage was identified in Caco-2, Calu-3 and Vero cells, including transcripts of genes such as GSDMB and KPNA2, which have previously been implicated in SARS-CoV-2 infections. Overall, these results highlight the potential role of differential polyadenylation and transcript usage in host immune response or viral manipulation of host mechanisms during infection, and therefore, showcase the value of long-read sequencing in identifying less-explored host responses to disease.

ABSTRACT

SARS-CoV-2 uses subgenomic (sg)RNA to produce viral proteins for replication and immune evasion. We applied long-read RNA and cDNA sequencing to in vitro human and primate infection models to study transcriptional dynamics. Transcription-regulating sequence (TRS)-dependent sgRNA was upregulated earlier in infection than TRS-independent sgRNA. An abundant class of TRS-independent sgRNA consisting of a portion of ORF1ab containing nsp1 joined to ORF10 and 3UTR was upregulated at 48 hours post infection in human cell lines. We identified double-junction sgRNA containing both TRS-dependent and independent junctions. We found multiple sites at which the SARS-CoV-2 genome is consistently more modified than sgRNA, and that sgRNA modifications are stable across transcript clusters, host cells and time since infection. Our work highlights the dynamic nature of the SARS-CoV-2 transcriptome during its replication cycle. Our results are available via an interactive web-app at http://coinlab.mdhs.unimelb.edu.au/.

ABSTRACT

Objective To investigate the nursing experts' perspectives on evaluation index and evaluation mode of core competence of postgraduate in Master of Nursing Specialist (MNS).Methods The evaluation indexes as well as the evaluation modes of core competences of postgraduates in MNS were summed up on the basis of the national relevant policies,site investigation and literature analysis.From February 2017 to April 2017,online survey was carried out in experts with experience of cultivating MNS using convenience sampling.Results A total of 85 nursing experts completed the questionnaires.The results showed that the importance scores of 5 first-class,and 17 second-class indexes of the core competences were higher than 4.5.The top three evaluation modes were:Mini Clinical Evaluation Exercise,Objective Structure Clinical Examination and Nursing Ward Round.It was suggested that the clinical nursing competence and the ethical decision-making were mainly evaluated at the examination of clinical ability,the clinical research and education ability were mainly evaluated at the end of the course,and the nursing management ability was mainly assessed during the clinical study.Besides,the evaluations of the clinical nursing competence,the ethical decision-making competence and the nursing management ability were largely relying on clinical teachers,the clinical research and the education ability were mainly relying on supervisor group.Conclusion This research reaches consensus on the evaluation indlicators and the relative evaluation modes for the core competences of postgraduates in MNS,and can provide references for evaluation of MNS in China.

ABSTRACT

Large number of cystic fibrosis transmembrane conductance regulator (CFTR) gene mutations are responsible for a wide spectrum of disease expressions in CF (cystic fibrosis). They includs not only the malfunction of the lung and pancreas, intestinal obstruction, and incapable of salt reabsorption, but also some kinds of male infertility, such as congenital bilateral absence of the vas deferens (CBAVD),poor sperm quality, the obstruction and agenesis of the epididymis. More recent studies have indicated that CFTR gene mutations have a far-reaching effect on human reproduction.
