ABSTRACT

Bisphenols and phthalates affect androgen receptor-mediated signaling that directly regulates Kallikrein-Related serine Peptidase 3 (KLK3) secretion, indicating that environmental factors may play a role in KLK3 secretion. With the aim of obtaining preliminary data on whether KLK3 could serve as an early marker of environmental pollution effects, in 61 and 58 healthy women living in a high environmental impact (HEI) and low environmental impact (LEI) area, respectively, serum KLK3 levels at different phases of menstrual cycle were measured. KLK3 values resulted in always being higher in the HEI group with respect to the LEI group. These differences were particularly relevant in the ovulatory phase (cycle day 12°-13°) of the menstrual cycle. The differences in KLK3 values during the three phases of the menstrual cycle were significant in the LEI group differently from the HEI group. In addition, higher progesterone levels were observed in the LEI group with respect to the HEI group in the luteal phase, indicating an opposite trend of KLK3 and progesterone in this phase of the menstrual cycle. Although changes in KLK3 could also depend on other factors, these preliminary data could be an early indication of an expanding study of the role of biomarkers in assessing early environmental effects for female reproductive health.

ABSTRACT

Os eventos que compõem a carreira do servidor foram redimensionados, após a implantação dos planos de cargos, carreiras e salários da Prefeitura Municipal. Frente a isto foi necessária uma análise de situação e da legislação, um processo de informação e um planejamento de ações para uma efetiva gestão de carreiras. O objetivo foi instrumentalizar o trabalhador frente aos eventos de carreira, antecipar a realização de capacitações para a melhoria dos processos de trabalho, atingir metas e articular possibilidades de evolução na carreira. Através de oficinas temáticas desenvolvemos os temas pertinentes à proposta e o resultado final foi a transformação de uma proposta em um processo contínuo de análise de contexto e capacitações.

ABSTRACT

Um processo de avaliação de desempenho representa uma ferramenta importante para o desenvolvimento de uma cultura voltada para o alinhamento dos objetivos individuais e das equipes às metas da organização implicando assim no maior envolvimento de todos os níveis, os quais passam a se sentir pessoalmente responsáveis pelo processo. Outra possibilidade é a integração com a política de capacitação, oferecendo oportunidades de desenvolvimento aos profissionais. Justificativa A diretriz é a integração das ações na gestão da saúde, incluindo a gestão de pessoas e focando a avaliação de desempenho;foi proposto um processo de avaliação de desempenho único, pactuado entre a administração direta e uma das organizações sociais de saúde da região. Foi possível inserir outras estratégias de avaliação, tal como avaliação dialogada acerca de potencialidades, habilidades e competências. Projeto Ermelino Matarazzo Território:Supervisão Técnica de Saúde Ermelino Matarazzo Trabalhadores avaliados: 834trabalhadores do SUS Gestores de unidades: 14 Participantes: Supervisão Técnica de Saúde Ermelino Matarazzo, SAS – Superintendência de Atenção à Saúde/ OSS -SECONCI e Gerência de Aperfeiçoamento de Carreiras Período: 2013 Objetivo Contribuir para o desenvolvimento das competências dos trabalhadores do SUS,necessárias à excelência da gestão da SMS; estimular a criação de uma cultura quevalorize o feedback; Reforçar o comprometimento do gestor com o desenvolvimento de sua equipe utilizando a ferramenta como instrumental de análise dos processos de trabalho, possibilitar o levantamento das necessidades de PDI* e PDC**. Metodologia utilizar um único formulário para todos os trabalhadores do SUS, propusemos reuniões periódicas e oficinas locais, quando necessário o envolvimentos de outras áreas técnicas para discussões ampliadas das necessidades encontradas.

ABSTRACT

PURPOSE: Embryo kinetics analysis is an emerging tool for selecting embryo(s) for transfer. The aim of the present study was to determine morphokinetic parameters easily usable in the laboratory and predictive of embryo development and, most importantly, of embryo competence in producing a clinical pregnancy after day 5 transfer. METHODS: A retrospective time-lapse monitoring analysis of morphokinetic parameters for 72 fully implanted embryos (group A) were compared to 106 non-implanted embryos (group B), and to 66 embryos with arrested development from the same pool of group A. All the embryos were from 78 patients undergoing ICSI treatment and day 5 embryo transfers. RESULTS: A day 3 embryo will develop into a viable blastocyst if the following ranges of morphokinetic parameters are met: t1 (between 18.4 h and 30.9 h post-ICSI), t2 (21.4-34.8 h), t4 (33.1-57.2 h), t7 (46.1-82.5 h), t8 (46.4-97.8 h), tC-tF (7.7-22.9 h) and s3 (0.7-30.8 h). On day 5 embryos with the highest probability to implant are those with a cc3 between 9.7 h and 21 h. CONCLUSIONS: Morphokinetic parameters are helpful to make appropriate decisions for the disposition of each embryo. It is recommended that each laboratory should determine its own ranges of in vitro development (IVD-MKP) and implantation-associated (IMP-MKP) morphokinetic parameters.

ABSTRACT

A novel cationic peptide based on L-lysine and L-diaminobutyric acid was prepared for the first time by solid phase synthesis. After HPLC purification and ESI MS characterization, we studied by CD and IR spectroscopy the structural features of the novel basic peptide, which is able to form a ß-turn-like structure. Furthermore, its interaction with DNA and RNA was investigated by CD and UV spectroscopy, which revealed a preferential RNA-binding ability of the sequential peptide, whereas its inhibitory activity toward HIV and Moloney murine leukemia virus (MMLV) reverse transcriptase action was evaluated by semiquantitative PCR. The cationic sequential peptide was able to inhibit the reverse transcriptase activity in both cases, even if our PCR data suggested a major activity in the case of HIV-RT, probably due to the stronger cationic peptide-protein interaction evidenced by UV spectroscopy.

ABSTRACT

In this work, we report the synthesis of a novel Fmoc-protected nucleoaminoacid, based on 4-piperidinyl glycine, carrying the DNA nucleobase on the secondary amino group, suitable for the solid-phase synthesis of nucleopeptides. After ESI-MS and NMR characterization this building block was used for the assembly of a thymine-functionalized tetrapeptide, composed of 4-piperidinyl glycine and L-arginine moieties alternated in the backbone. The ability to interact with RNA and the efficiency in interfering with the reverse transcription of eukaryotic mRNA of the novel nucleo-tetrapeptide found in this study are in favour of the employment of chiral nucleopeptides based on alternate 4-piperidinyl glycine/L-arginine backbone in biomedicine.
