ABSTRACT

Since citrus flavonoids have antioxidant and anti-inflammatory properties, it was hypothesized that these compounds would become a suitable alternative to the use of therapeutic doses of zinc oxide at weaning. A total of 252 weaned pigs ([LargeWhite × Landrace] × Pietrain) were distributed according to BW (5.7 kg ± 0.76) into 18 pens (6 pens per diet, 14 pigs/pen). Three experimental diets for the prestarter (0-14 d postweaning) and starter (15-35 d postweaning) period were prepared: (i) a nonmedicated (CON) diet, (ii) a CON diet supplemented with zinc oxide at 2500 mg/kg, amoxicillin at 0.3 mg/kg and apramycin at 0.1 mg/kg (ZnO), and (iii) CON diet with the addition of a commercial citrus flavonoid extract at 0.3 mg/kg and amoxicillin at 0.3 mg/kg (FLAV). Pig BW, ADG, ADFI, and FCR were assessed on d7, d14, and d35. Samples of intestinal tissue, cecal content, and serum were collected on day seven (18 piglets). FLAV treatment achieved greater BW and ADG during the starter and for the entire experimental period compared with the CON diet (p < 0.05), whereas ZnO pigs evidenced intermediate results. Jejunum tissue analysis showed that pigs fed the FLAV diet overexpressed genes related to barrier function, digestive enzymes, and nutrient transport compared to those pigs fed the CON diet (p < 0.05). An increase in the abundance of bacterial genera such as Succinivibrio, Turicibacter, and Mitsuokella (p < 0.05) was observed in the FLAV compared with the CON and ZnO piglets. ZnO and FLAV increased the expression of TAS2R39, while ZnO pigs also expressed greater TAS2R16 than CON (p < 0.05) in the intestine. FLAV treatment improved the gut function, possibly explaining a higher performance at the end of the nursery period. Consequently, citrus flavonoids supplementation, together with amoxicillin, is a promising alternative to the use of zinc oxide plus amoxicillin and apramycin in weanling pigs, minimizing the use of antibiotics.

ABSTRACT

We propose a novel biomarker that can identify patients at high risk of early progression after chimeric antigen receptor (CAR) T cell therapy. Calculation of cell-free DNA (cfDNA) with a pre-apheresis (PA) and pre-lymphodepletion (PL) sample allows monitoring of tumor dynamics (∆cfDNA). In the present study, ∆cfDNA and other biomarkers and clinical variables were evaluated in 58 patients with relapsed/refractory diffuse large B cell lymphoma (DLBCL). ∆cfDNA (>11 ng/mL plasma; P =.003), C-reactive protein (CRP) PL (>1.06 mg/dL; P = .004), lactate dehydrogenase (LDH) PL (>304; P = .006), disease status PL (progressive disease; P = .035) and sex (male; P = .016) were highly correlated with 1 month progression. After adjusting for ∆cfDNA, CRP PL, and LDH PL, disease status PL, and sex, ∆cfDNA remained associated with 1-month progression after CAR T cell infusion.

ABSTRACT

Enterolobium cyclocarpum and Piscidia piscipula are two important tree Fabaceae species distributed from the Yucatan Peninsula, Mexico. Our aims were focused on the E. cyclocarpum and P. piscipula seeds for: (1) to examine the seed permeability and imbibition rate, (2) to evaluate the effect of seed pre-germinative treatments, and (3) to characterize the structures involved on the presence of physical dormancy (PY). We used fresh seeds to determine seed permeability and imbibition rate, seed viability by means of tetrazolium test, furthermore, we applied mechanical scarification and boiler shocks for 5 s, 10 s and 15 s treatments. Morphological characterization of the seed coat was by Scanning Electron Microscope (SEM). Seed viability in E. cyclocarpum and P. piscipula were 100% and 96%, respectively. Seed permeability and imbibition rate in E. cyclocarpum were low. The highest germination in E. cyclocarpum was in the mechanical scarification (92%), while in P. piscipula, this parameter was in the 10 s boiling water treatment (76.0%). The presence of PY was confirmed in both species because they showed low seed permeability, and imbibition rate; furthermore, exhibited macrosclereids cells. The present research seeks to promote the sustainable use of E. cyclocarpum and P. piscipula.

ABSTRACT

Background: The role of homework in students’ academic performance is a widely debated topic about which there is no definitive answer. The objective of this study was to analyse the importance of parental help with homework in academic achievement, testing its cultural invariance (by country), academic invariance (by subject), and the stability of its effects over time. Method: A meta-analysis was performed using the results of PISA evaluations from 2009, 2012, 2015, and 2018 from countries which applied the family PISA questionnaire. We analysed 180 effects and confirmed the fit of the model and the heterogeneity of the effects, performing an analysis of moderators and multimodal inference. Results: Students who had more help with homework had lower academic achievement, with an overall effect (d) = 0.23, 95% CI [0.21, 0.25]. The effects were greater in Europe than in Asia. We did not find differences by subject type, and the results were stable over time. Conclusions: Family help with homework does not ensure students’ academic success, and it is more important how that help is given than how much. This conclusion is valid for different types of subjects and is stable over time, with some variation between cultures.


Antecedentes: el papel de los deberes escolares en el rendimiento académico es un tema ampliamente debatido, no existiendo una respuesta definitiva. El presente trabajo analiza el efecto de la ayuda parental con los deberes sobre los resultados educativos, comprobando la invarianza cultural (por países) y académica (por asignaturas), así como la estabilidad temporal de los efectos. Método: se realizó un meta-análisis con los resultados PISA de los años 2009, 2012, 2015 y 2018, incluyendo los países que aplicaron el cuestionario PISA para las familias. Se analizaron 180 efectos, comprobando el ajuste del modelo y la heterogeneidad de los efectos, y realizando análisis de moderadores y análisis de inferencia con modelos múltiples. Resultados: los estudiantes que reciben más ayuda con los deberes obtienen resultados más bajos: d = 0.23, IC 95% [0.21, 0.25]. Los efectos son mayores en Europa que en Asia. No se encontraron diferencias en función de la asignatura, y los resultados son estables a lo largo del tiempo. Conclusiones: la ayuda familiar con los deberes no garantiza el éxito académico, siendo más importante cómo se hace que el cuánto. Esta conclusión es válida en todas las asignaturas y se muestra estable en el tiempo, observándose variaciones interculturales.

ABSTRACT

Background: Academically resilient students are those who exhibit highperformance starting from a disadvantaged socioeconomic situation. Thisstudy aims to identify the personal, school, and national factors that areassociated with that resilience in the European Union (EU). Method:The sample comprised 96556 fourth grade students from 21 EU countriesparticipating in TIMSS-2019. Two three-level logistic regression modelswere specified for the overall sample. Results: The EU has an average of25.67% resilient students in mathematics and 24.16% in science. Studentconfidence and having done prior linguistic tasks at school were thevariables with the most predictive power after accounting for gender andstudents’ immigrant background. The European countries analyzed largelycompensated for the doubly-disadvantaged situation of immigrant students.Those countries with higher proportions of low-performing students hadfewer resilient students. Conclusions: The educational policies in the EUmember states are able to largely compensate for unfavorable startingpositions; fundamentally, policies of a social nature such as support forimmigrant students, families, or schools.


Antecedentes: el alumnado académicamente resilientees aquel que obtiene un alto rendimiento partiendo de una situaciónsocioeconómica desaventajada. Esta investigación pretende identificarlos factores personales, escolares y nacionales que están asociados a laresiliencia académica en la Unión Europea (UE). Método: la muestra fuede 96.556 estudiantes de 4º grado de 21 países de la UE participantes enTIMSS-2019. Para el conjunto de la muestra se ajustaron dos modelos deregresión logística multinivel de tres niveles. Resultados:la UE tiene unpromedio de 25,67% de alumnado resiliente en matemáticas y 24,16% enciencias. La confianza de los estudiantes y haber realizado tareas lingüísticasprevias a la escuela son las variables con mayor poder predictivo después detener en cuenta el género y los antecedentes inmigrantes de los estudiantes.Los países europeos analizados compensan en buena medida la situacióndoblemente desaventajada del alumnado inmigrante. Aquellos países queposeen un mayor porcentaje de alumnado con bajo rendimiento tienenmenos estudiantes resilientes. Conclusiones: las políticas educativas delos estados miembros de la UE son capaces de compensar en gran medidalas situaciones desfavorecidas de partida. Fundamentalmente aquellas decarácter social como el apoyo al alumnado inmigrante, a la familia o lasinstituciones educativas.

ABSTRACT

Although next-generation sequencing (NGS) data on lymphomas require further validation before being implemented in daily practice, the clinical application of NGS can be considered right around the corner. The aim of our study was to validate an NGS lymphoid panel for tissue and liquid biopsy with the most common types of non-Hodgkin's lymphoma [follicular lymphoma (FL) and diffuse large B-cell lymphoma (DLBCL)]. In this series, 372 somatic alterations were detected in 93.6% (44/47) of the patients through tissue biopsy. In FL, we identified 93 somatic alterations, with a median of 7.4 mutations per sample. In DLBCL, we detected 279 somatic variants with a median of 8.6 mutations (range 0-35). In 92% (24/26) of the cases, we were able to detect some variant in the circulating tumor DNA. We detected a total of 386 variants; 63.7% were detected in both types of samples, 13.2% were detected only in the circulating tumor DNA, and 23% were detected only in the tissue biopsy. We found a correlation between the number of circulating tumor DNA mutations, advanced stage, and bulky disease. The genetic alterations detected in this panel were consistent with those previously described at diagnosis. The liquid biopsy sample is therefore a complementary tool that can provide new genetic information, even in cases where a solid biopsy cannot be performed or an insufficient sample was obtained. In summary, we describe and analyze in this study the findings and difficulties encountered when incorporating liquid biopsy into clinical practice in non-Hodgkin's lymphoma at diagnosis.

ABSTRACT

Flavonoid supplementation may modify the behavior and rumen inflammatory response of fattening bulls, and this could be related to the concentrate presentation (mash or pellet) form. In the present study, 150 Holstein bulls (183.0 ± 7.53 kg BW and 137 ± 1.8 d of age) were randomly allocated to one of eight pens and assigned to control (C) or (BF) (Citrus aurantium, Bioflavex CA, HealthTech Bio Actives, Spain, 0.4 kg per ton of concentrate of Bioflavex CA, 20% naringin). Concentrate (pellet) intake was recorded daily, and BW and animal behavior fortnightly. Animals were slaughtered after 168 d of study, and ruminal epithelium samples were collected for gene expression analyses. Treatment did not affect animal performance; however, BF supplementation reduced agonistic interactions and oral non-nutritive behaviors and increased the time devoted to eating concentrate and ruminating activity (p < 0.05). The gene expression of some genes in the rumen epithelium was greater or tended to be greater in BF than C bulls (bitter taste receptor 16, cytokine IL-25, ß-defensin; p < 0.10; pancreatic polypeptide receptor 1 and tumor necrosis factor alpha; p < 0.05). In conclusion, flavonoid supplementation modifies the expression of genes in the rumen epithelium that could be related to inflammation and animal behavior modulation.

BACKGROUND: Students' academic achievement depends on their personal, family and sociocultural characteristics. This study aims to identify the personal and family factors of European students who do not perform as expected, whether by surpassing expectations (resilient) or by failing to meet them (low performers). METHOD: The sample was composed of 117,539 fourth grade students who completed the Progress in International Reading Literacy Study (PIRLS) test, from 23 European Union countries. Academic performance was evaluated via a reading comprehension test. For each country, two binary logistic regression models were used, one for resilient students and the other for low performers. Variables related to the students and their families were used as predictor variables. RESULTS: Significant differences were found between European countries in terms of the proportion of resilient and low performing students. The two variables with the most predictive power were student confidence with reading, and having done early literacy activities. CONCLUSIONS: Students' personal characteristics and family conditions are instrumental in students being considered resilient or low performers


ANTECEDENTES: el rendimiento académico de los estudiantes depende de sus características personales, familiares y socioculturales. El presente trabajo trata de identificar los factores personales y familiares de los estudiantes europeos que no obtienen el rendimiento esperado, bien sea porque superan las expectativas previas (resilientes), o porque rinden por debajo de lo esperado (low performers). MÉTODO: la muestra está formada por 117.539 estudiantes de cuarto grado que realizaron la prueba de Progress in International Reading Literacy Study (PIRLS), pertenecientes a 23 países de la Unión Europea. El rendimiento académico se evaluó mediante una prueba de compresión lectora. Para cada país se utilizaron dos modelos de regresion logística binaria, uno para los alumnos resilientes y otro para los low performers. Como variables predictoras se utilizaron variables asociadas al alumno y a su familia. RESULTADOS: se han obtenido diferencias importantes entre los países europeos en relación a la proporción de alumnos resilientes y low performers. La confianza de los estudiantes en la lectura y el haber realizado actividades de alfabetización temprana son las dos variables con mayor poder predictivo. CONCLUSIONES: las características personales del alumnado y sus condiciones familiares son determinantes para que los estudiantes sean considerados resilientes y low performers

ABSTRACT

Clinical and molecular prognostic factors in gliomas include age, IDH mutation, the glioma CpG island methylator phenotype (G-CIMP+) and promoter methylation of the O(6)-methylguanine DNA-methyltransferase (MGMT) gene. Among these markers, a predictive value was reported in glioblastomas (GBM) for MGMT promoter methylation, in particular in elderly GBM patients. In this study, methylation data from 46 glioma samples with the Illumina 450K platform were obtained and extended using external data to include a total of 247 glioma samples. Methylation analysis of the whole MGMT gene with this platform revealed two strongly survival-associated CpG regions within the promoter and the gene body, which were confirmed in a reported dataset of high grade-gliomas. Methylation at the promoter (CpG 25, cg12981137 and the prognostic model MGMT-STP27) and at the gene body CpG 165 (cg07933035), were significantly associated with better overall survival, and strongly correlated with G-CIMP+ status. In this series, the prognostic value of MGMT methylation at the promoter was not observed in G-CIMP- cases, although around 50 % of them were MGMT-methylated. These results were also obtained in an homogeneously-treated series of chemoradiated G-CIMP- GBMs analyzed by MSP and qMSP, and confirmed in a reported pyrosequencing-analyzed series of gliomas. Interestingly, in contrast to the MGMT promoter, gene body methylation was of prognostic value in G-CIMP-patients older than 65 years. Our study highlights the relevance of the prognostic value of the different regions of methylation throughout the MGMT gene that could be affected by specific G-CIMP profiles and age groups.
