ABSTRACT

This study evaluated the genotoxicity of lyophilized glycolic extract of Theobroma cacao Linné seeds (TCL), using the micronucleus assay in bone marrow of mice. The interaction between TCL and doxorubicin (DXR) was also analyzed. Experimental groups were evaluated 24-48 h after treatment with N-Nitroso-N-ethylurea (NEU: 50 mg/kg), DXR (5 mg/kg), NaCl (145 mM), TCL (0.5-2 g/kg), and TCL (2 g/kg) in combination with DXR (antigenotoxic assays). Analysis of micronucleated polychromatic erythrocytes (MNPCEs) showed no significant differences between all the treatment doses of TCL and NaCl control. Mice experimentally treated with DXR and NEU significantly induced MNPCEs. However, a significant reduction of MNPCEs was also observed when TCL was administered in combination with the chemotherapeutic agent DXR. The analysis of the PCE/NCE ratio revealed no significant differences between the NaCl control, all doses of TCL, and DXR. However, there were significant differences in the PCE/NCE ratio between positive NEU control and all other treatments. The PCE/NCE ratio observed after treatment with TCL and DXR showed significant differences and intermediate values to controls (NaCl and NEU). This study suggests absence of genotoxicity and cytotoxicity of TCL, regardless of dose, sex, and time. TCL reduced genotoxic effects induced by DXR, suggesting potential antigenotoxic effects.(AU)

ABSTRACT

Choline is an essential nutrient in poultry diets because it performs various important metabolic functions. The objective of this study was to re-evaluate the choline requirements of male broiler chickens from 1 to 21 days of age at two levels of methionine. Two assays using 2,160 Cobb® chickens (1,080 in each assay) were conducted. The study design was completely randomized and consisted of six treatments and six replicates, with 30 animals per experimental unit. The semipurified basal diet was formulated with corn, soya bean meal, soya protein concentrate, starch and sugar, providing 390 mg/kg choline and 0.593% digestible methionine (requirement level) in Assay 1 and a reduction of about one-quarter in the requirement level of digestible methionine (0.440%) in Assay 2. Choline chloride (62.5%) was added by a supplementation technique to both basal diets to compose crescent levels of choline supplementation (715, 1,040, 1,365, 1,690 and 2,015 mg/kg). The weight gain responses were fitted using quadratic polynomial (QP) and broken-line (BL) models. The ideal intake of choline (mg/bird.day) was estimated from the first intercept of the QP with the BL plateau (BL + QP). The results showed that the diet with the 25% reduction in digestible methionine limited the maximum weight gain by approximately 10%. The choline requirements of broilers from 1 to 7, 1 to 14 and 1 to 21 days of age were 27,013, 44,458 and 62,535 mg/bird.day, respectively, for the requirement level of digestible methionine and 26,796, 41,820 and 56,578 mg/bird.day for the broilers receiving the diet with the 25% reduction in digestible methionine.

Handroanthus impetiginosus has long been used in traditional medicine and various studies have determined the presence of bioactive chemical compounds and potential phytotherapeutics. In this study, the genotoxicity of the lyophilized tincture of H. impetiginosus bark (THI) was evaluated in mouse bone marrow using micronucleus assays. The interaction between THI and genotoxic effects induced by the chemotherapeutic agent, doxorubicin (DXR), was also analyzed. Experimental groups were evaluated 24 to 48 h after treatment with N-nitroso-N-ethylurea (NEU; 50 mg/kg), DXR (5 mg/kg), sodium chloride (NaCl; 150 mM), and THI (0.5-2 g/kg). Antigenotoxic assays were carried out using THI (0.5 g/kg) in combination with NEU or DXR. Analysis of the micronucleated polychromatic erythrocytes (MNPCEs) indicated no significant differences between treatment doses of THI (0.5-2 g/kg) and NaCl. Polychromatic erythrocyte (PCE) to normochromatic erythrocyte (NCE) ratios did not indicate any statistical differences between DXR and THI or NaCl, but there were differences between THI and NaCl. ABSTRACT

Abstract Handroanthus impetiginosus has long been used in traditional medicine and various studies have determined the presence of bioactive chemical compounds and potential phytotherapeutics. In this study, the genotoxicity of the lyophilized tincture of H. impetiginosus bark (THI) was evaluated in mouse bone marrow using micronucleus assays. The interaction between THI and genotoxic effects induced by the chemotherapeutic agent, doxorubicin (DXR), was also analyzed. Experimental groups were evaluated 24 to 48 h after treatment with N-nitroso-N-ethylurea (NEU; 50 mg/kg), DXR (5 mg/kg), sodium chloride (NaCl; 150 mM), and THI (0.5-2 g/kg). Antigenotoxic assays were carried out using THI (0.5 g/kg) in combination with NEU or DXR. Analysis of the micronucleated polychromatic erythrocytes (MNPCEs) indicated no significant differences between treatment doses of THI (0.5-2 g/kg) and NaCl. Polychromatic erythrocyte (PCE) to normochromatic erythrocyte (NCE) ratios did not indicate any statistical differences between DXR and THI or NaCl, but there were differences between THI and NaCl. A significant reduction in MNPCEs and PCE/NCE ratios was observed when THI was administered in combination with DXR. This study suggested the absence of THI genotoxicity that was dose-, time-, and gender-independent and the presence of moderate systemic toxicity that was dose-independent, but time- and gender-dependent. The combination of THI and DXR also suggested antigenotoxic effects, indicating that THI reduced genotoxic effects induced by chemotherapeutic agents.


Resumo Handroanthus impetiginosus tem sido usada durante um longo período pela medicina tradicional e vários estudos têm demonstrados a presença de compostos químicos e potencial fitoterapêutico. Esta pesquisa avaliou a genotoxicidade da tintura da casca liofilizada de H. impetiginosus (THI) usando o ensaio do micronúcleo em medula óssea de camundongos. A interação entre THI e os efeitos genotóxicos induzidos pelo quimioterápico doxorrubicina (DXR) também foram analisados. Grupos experimentais foram analisados a 24-48 h após o tratamento com N-Nitroso-N-etiluréia (NEU; 50 mg/kg), DXR (5 mg/kg), NaCl (150 mM) e THI (0,5-2 g/kg). O ensaio antigenotóxico foi conduzido utilizando THI (0,5 g/kg) em combinação com NEU ou DXR. Os seguintes sites sob Cerrado sensu stricto (Css) foram avaliados: preservado (UND) e não preservado (DIS). O C orgânico total e da biomassa microbiana foram maiores na área preservada do que na não preservada em ambas estações seca e chuvosa. Entretanto, o C da biomassa microbiana foi maior na estação chuvosa do que na seca. A respiração do solo não variou entre as áreas, mas foi maior na estação chuvosa. ABSTRACT

The objective of this study was to evaluate the effect of the dietary addition of different levels of glycerin on the performance, litter moisture, pododermatitis incidence, and carcass and parts yield of broilers. In total, 1,610 broilers were reared in 35 pens with 46 birds each. A completely randomized experimental design, with five treatments with seven replicates was applied. The experimental treatments were: T1: control diet; T2: dietary inclusion of 5% glycerin from 1-42 days of age; T3: dietary inclusion of 10% glycerin from 1-42 days of age; T4: dietary inclusion of 5% glycerin from 7-42 days of age; T5: dietary inclusion of 10% glycerin from 7-42 days of age. The diets containing glycerin fed since the pre-starter period improved broiler weight gain and feed conversion ratio, but did not influence feed intake or livability. At the end of the experiment, the production efficiency index of the broilers fed 10% glycerin during the entire rearing period was significantly reduced compared with the other treatments. Litter moisture in the pens of broilers fed 10% glycerin during the entire rearing period was higher compared to the other treatments since day 21.Diets containing 10% glycerin, both for the entire rearing period (1-42 days) or only after the pre-starter phase (7-42 days), influenced broiler performance and incidence of severe pododermatitis, reducing the production efficiency indexes at 42 days. Glycerin may be added up to 5% in broiler´s diets with no effect on performance, litter moisture and carcass yield, indicating that this co-product of the biodiesel industry can be used as an alternative feedstuff for broilers.(AU)

Abstract This study evaluated the genotoxicity of lyophilized glycolic extract of Theobroma cacao Linné seeds (TCL), using the micronucleus assay in bone marrow of mice. The interaction between TCL and doxorubicin (DXR) was also analyzed. Experimental groups were evaluated 24-48 h after treatment with N-Nitroso-N-ethylurea (NEU: 50 mg/kg), DXR (5 mg/kg), NaCl (145 mM), TCL (0.5-2 g/kg), and TCL (2 g/kg) in combination with DXR (antigenotoxic assays). Analysis of micronucleated polychromatic erythrocytes (MNPCEs) showed no significant differences between all the treatment doses of TCL and NaCl control. Mice experimentally treated with DXR and NEU significantly induced MNPCEs. However, a significant reduction of MNPCEs was also observed when TCL was administered in combination with the chemotherapeutic agent DXR. The analysis of the PCE/NCE ratio revealed no significant differences between the NaCl control, all doses of TCL, and DXR. However, there were significant differences in the PCE/NCE ratio between positive NEU control and all other treatments. The PCE/NCE ratio observed after treatment with TCL and DXR showed significant differences and intermediate values to controls (NaCl and NEU). This study suggests absence of genotoxicity and cytotoxicity of TCL, regardless of dose, sex, and time. TCL reduced genotoxic effects induced by DXR, suggesting potential antigenotoxic effects.


Resumo Este estudo avaliou a genotoxicidade do extrato glicólico liofilizado de sementes de Theobroma cacao Linné (TCL), usando o ensaio do micronúcleo em medula óssea de camundongos. A interação entre TCL e doxorrubicina (DXR) foi também analisada. Grupos experimentais foram avaliados 24-48 h após tratamento com N-Nitroso-N-etilureia (NEU: 50 mg/kg), DXR (5 mg/kg), NaCl (145 mM), TCL (0,5-2 g/kg), e TCL (2 g/kg) em combinação com DXR (ensaio antigenotóxico). As análises de eritrócitos policromáticos micronucleados (EPCMNs) não mostraram diferenças significantes entre todas as doses de tratamento do TCL e o controle NaCl. Camundongos experimentalmente tratados com DXR e NEU induziram significativamente EPCMNs. Contudo, uma redução significante de EPCMNs foi também observada quando TCL foi administrada em combinação com o agente quimioterapêutico DXR. As análises da relação EPC/ENC (eritrócito policromático/eritrócito normocromático) revelaram ausência de diferenças significantes entre o controle NaCl, todas as doses de TCL e DXR. Contudo, houve diferenças significantes na relação EPC/ENC entre o controle positivo NEU e todos os outros tratamento. A relação ECP/ENC observada após o tratamento com TCL e DXR mostrou diferenças significantes e valores intermediários aos controles (NaCl e NEU). Este estudo sugere ausência de genotoxicidade e citotoxicidade de TCL, independentemente da dose, sexo e tempo. TCL reduziu os efeitos genotóxicos induzidos por DXR, sugerindo potencial efeitos antigenotóxicos.

Concentrations of 75 µg.L(-1) and 100 µg.L(-1) of MC-LR equivalent induced a greater occurrence of abnormal seedlings in lettuce, due to necrosis of the radicle and shortening of this organ in normal seedlings, as well as the reduction in total chlorophyll content and increase in the activity of the antioxidant enzyme peroxidase (POD). The MC- extract caused no harmful effects to seed germination and initial development of seedlings of arugula. However, in lettuce, it caused elevation of POD enzyme activity, decrease in seed germination at concentrations of 75 µg.L(-1) (MC-75) and 100 µg.L(-1) (MC-100), and shortening of the radicle length, suggesting that other compounds present in the cyanobacteria extracts contributed to this result. Crude extracts of M. aeruginosa (MC-) may contain other compounds, besides the cyanotoxins, capable of causing inhibitory or stimulatory effects on seed germination and initial development of salad green seedlings. Analisamos os efeitos de extratos brutos da cianobactéria M. aeruginosa, produtora de microcistinas (MC+), na germinação de sementes e no desenvolvimento de plântulas de alface e rúcula, em concentrações de 0,5 a 100 μg.L–1de MC–LR equivalente e comparamos com extrato brutos da mesma espécie sem a toxina (MC–). Extratos brutos de cianobactérias com MC (+) e sem MC (–) causaram efeitos diferentes na germinação de sementes e desenvolvimento de plântulas de hortaliças, sendo que a alface apresentou maior sensibilidade a ambos os extratos comparando-se com a rúcula. Extratos brutos de M. aeruginosa (MC+) causaram efeitos mais evidentes sobre a germinação de sementes e desenvolvimento de plântulas de hortaliças do que os (MC–). Concentrações de 75 e 100 μg.L–1 de MC–LR equivalente induziram maior ocorrência de plântulas anormais na alface devido ao aparecimento de necrose na radícula e seu encurtamento nas plântulas normais, bem como a redução no teor de clorofila total e aumento na atividade da enzima antioxidante peroxidase (POD). O extrato (MC–) não provocou efeitos inibitórios na germinação de sementes e desenvolvimento de plântulas para a rúcula, no entanto, provocou elevação da atividade da enzima POD, redução na germinação de sementes nas concentrações de 75 e 100 μg.L–1, e no comprimento da radícula na alface, sugerindo a ação de outros compostos presentes nos extratos da cianobactéria. Extratos brutos de M. aeruginosa (MC–) podem conter outros compostos além de cianotoxinas capazes de provocar efeitos inibitórios ou estimulatórios na germinação de sementes e no desenvolvimento de plântulas de hortaliças. A rúcula apresentou menor sensibilidade aos extratos brutos de M. aeruginosa (MC+) e (MC–) e outros possíveis compostos produzidos por estas cianobactérias.(AU)

This study evaluated the metabolic response of broilers fed diets containing increasing crude glycerine levels in two bioassays. Birds were house in metabolic cages, and were distributed according to a completely randomized experimental design with five treatments of 4 replicates each (1st assay: 5 birds/ cage; 2nd assay: 1-20 days = 8 birds/ cage, and 21-42 days = 4 birds/cage). Treatments consisted of a control diet based on corn and soybean meal, and four other diets containing 2.5%, 5.0%, 7.5% and 10.0% glycerine derived from biodiesel. In experiment I, there was no effect (p>0.05) of glycerol level on liver weight or blood parameters. Serum blood glycerol levels of the birds fed 10% crude glycerine increased during the first nine days of diet intake (p 0.05). In experiment II, water intake increased (p 0.05) in the birds fed 7.5 and 10.0% crude glycerine at 4 and 8 days of age. Feed intake increased (p 0.05) on days 8 and 12 in birds fed 2.5 and 7.5% glycerine. Fecal moisture increased (p 0.05) in birds fed diets with 5.0, 7.5, and 10.0% crude glycerine on days 16 and 20. Ileal content moisture was not different (p>0.05) among treatments when birds were 42 days old. High dietary glycerine levels may induce metabolic change in broilers, such as increased blood glycerol level, water intake and fecal moisture.(AU)

ABSTRACT

UNLABELLED: This study correlated the composition of the spoilage bacterial flora with the main gaseous and volatile organic compounds (VOCs) found in the package headspace of spoiled, chilled, vacuum-packed meat. Fifteen chilled, vacuum-packed beef samples, suffering from blown pack spoilage, were studied using 16S rRNA clone sequencing. More than 50% of the bacteria were identified as lactic acid bacteria (LAB), followed by clostridia and enterobacteria. Fifty-one volatile compounds were detected in the spoiled samples. Although the major spoilage compounds were identified as alcohols and aldehydes, CO2 was identified as the major gas in the spoiled samples by headspace technique. Different species of bacteria contribute to different volatile compounds during meat spoilage. LAB played an important role in blown pack deterioration of the Brazilian beef studied. SIGNIFICANCE AND IMPACT OF THE STUDY: The data generated by this study provided useful information to correlate the microbial contamination of Enterobacteriaceae, lactic acid bacteria and Clostridium with the VOC and gaseous compound production to define, in a faster manner, not only the type of contamination, but also to prevent it.
