ABSTRACT

BACKGROUND: Glioma is a primary intracranial malignancy with poor prognosis, of which the pathogenesis remains to be elucidated. Therein, the aim of this study is to discuss the impacts of lncRNA plasmacytoma variant translocation 1 (PVT1)/microRNA-128-1-5p (miR-128-1-5p)/polypyrimidine tract-binding protein 1 (PTBP1) axis on the biological characteristics of glioma cells. METHODS: Glioma tissue samples (72 cases) and normal brain tissue samples (35 cases) were harvested. The expression of PVT1, miR-128-1-5p and PTBP1 in glioma tissues and cells was detected. Glioma cells were transfected with sh-PVT1, miR-128-1-5p mimics or miR-128-1-5p inhibitors to verify the impacts of PVT1 and miR-128-1-5p on DNA damage, cell colony formation, invasion, proliferation, migration and apoptosis of glioma U87 and U251 cells. The growth of transplanted tumor was tested by tumor xenograft in nude mice. The combination of PVT1 and miR-128-1-5p and the targeting relationship between miR-128-1-5p and PTBP1 were verified. RESULTS: PVT1 and PTBP1 expression was enhanced and miR-128-1-5p expression was degraded in glioma tissues and cells. Overexpressed miR-128-1-5p and lowly-expressed PVT1 promoted DNA damage, suppressed colony formation, invasion, proliferation and migration as well as boosted apoptosis of U251 and U87 cells. Up-regulating miR-128-1-5p and down-regulating PVT1 reduced transplanted tumor volume and weight of glioma in mice. Low expression miR-128-1-5p reversed the effect of low expression PVT1 on the biological characteristics of glioma cells. PVT1 specifically bound to miR-128-1-5p and PTBP1 was the target gene of miR-128-1-5p. CONCLUSION: This study suggests that down-regulated PVT1 or up-regulated miR-128-1-5p boosts apoptosis and attenuates proliferation of glioma cells by inhibiting PTBP1 expression. This study is essential for finding new therapeutic targets for glioma.

ABSTRACT

Objective To study the relationship between expression of Caspase 3 and regression of hemangioma. Methods Caspase 3 expression was detected in 79 hemangiomas and 5 normal skin tissues by immunohistochemistry (SP). Results In 79 hemangiomas, the positive rates of Caspase 3 expression in proliferating, involuting and involuted phases were 52.4%, 91.2% and 83.3%, respectively. The expression of Caspase 3 in involuted and involuting phases was higher than proliferating phase( P 0.05). Conclusion Caspase 3 is involved in the converting of hemangiomas from proliferating phase into involuting phase. The activation of Caspase 3 may induce apoptosis of endothelial cells, thus leading to the regression of hemagioma.

ABSTRACT

<p><b>OBJECTIVE</b>To compare the difference in local recurrence Dixon and Miles groups after total mesorectal excision.</p><p><b>METHODS</b>One hundred and seventy-three patients with rectal cancer were divided into two groups (Dixon group, 123 patients; Miles group 50 patients). Total mesorectal excision was made according to Heald's method.</p><p><b>RESULTS</b>The local recurrence in the Dixon and Miles groups was 4.8% (6/123) and 18.0% (9/50) respectively (P < 0.05), and in the Miles group before and after 1997 was 36.8% (7/19) and 6.5% (2/31) respectively (P < 0.05), 4.8% in LRR (P < 0.01). No significant difference was observed in the local recurrence between the two groups (P > 0.05).</p><p><b>CONCLUSION</b>Total mesorectal excision is an important factor reducing local recurrence after radical operation for rectal cancer.</p>

ABSTRACT

ObjectiveTo study the treament of arterial occlusion of lower extrimities. MethodsSeventy-three cases(78 limbs) of chronic ischemia o f lower extremities were diagnosed as artroial occlusion by color u ltrasounography, DSA or MRA. Arterial bypass using grafting,endarterectomy and percuteneous transluminal angioplasty (PTA) were used for the artery segmental occlusion in 44 limbs. Venous arterialization and reconstruc tion of profunda femoris were performed for extensive occlusion in 29 limbs. ResultsAll the cases showed satisfactory r esults 6 months to 8 years after operation except 8 limbs (segmenta l occlusion in 3 limbs and extensive occlusion in 5 limbs) were amputated. ConclusionsThe key points of improvement of the therapeu tic effects of the arteral occlusion of lowe extremity lie on the cho ice of the therapeutic methods accoding to the results of DSA or MRA.

ABSTRACT

Objectives To study the feasibility of total mesorectal excision (TME) by laparoscopy for rectal cancer.Methods The mesorectum in the resected samples and the number of lymph nodes resected were compared between laparoscopic (n=20) and open surgery group (n=20).Results ①The mesorecum of resected specimen was all complete in two groups. ②The lymph node yield (N 1) in laparoscopic and open group was 8.5?3.6 vs. 10.6?6.2(P=0.334), respectively. That in N 2 was 3.7?2.2 vs. 2.4?2.1(P=0.328). That in N 3 was 2.4?1.7 vs. 2.0?1.8(P=0.590).③The distal margin was all tumor-free in the two groups. ConclusionLaparoscopy is capable of performing TME in patients with rectal carcinoma undergoing radical resection.

ABSTRACT

Objective To investigate the biopathologic characteristics of transitional mucosa (TM) adjacent to rectal cancer and its relationship to the anastomotic recurrence after curative surgery of rectal cancer. Method The expression of p53,p21 ras and PCNA proteins was determined immunohistochemically in primary rectal adenocarcinomas (n=67), the proximal margins (n=67), the distal margins (n=67) and rectal mucosa 1~3?cm distant to the anastomosis in patients one year after radical resection (n=33), and two years after (n=31).Results The expression of TM, p21 ras and p53 proteins,and PCNA index in primary rectal adenocarcinomas was higher than that in two resection margins (P0.05). Positive TM was often found in the area of chronic inflammation. conclusion TM adjacent to the rectal cancer is a nonspecific, not cancer precursor event.
