ABSTRACT

INTRODUCTION: Epidermal growth factor (EGF) has various important physiological functions, which it exerts by binding to the epidermal growth factor receptor (EGFR). Reports show that EGF expression is strongly correlated with the occurrence and development of many types of tumour. To date, however, the relationship between EGF/EGFR and the occurrence and development of thyroid carcinoma remains unclear. MATERIAL AND METHODS: In the current study, we investigated this phenomenon using human anaplastic thyroid carcinoma cell lines (SUN-80). RESULTS: The results indicated that EGF triggered the EGFR-mediated intracellular signalling pathway, including signal transducers and activators of transcription 1/3/5 (STAT1/3/5) and protein kinase B (AKT) in a time- and dose-dependent manner. In addition, results from EGF-induced EGFR internalization and co-localization analyses showed that clathrin, Rab5/7, and EEA1 play critical roles in the intracellular trafficking of EGF/EGFR. Interestingly, EGF triggered EGFR translocation into the nucleus, while nuclear-localized EGFR affected cell cycle distribution, thereby significantly promoting the ration of S phase. Overall, these findings indicated that nuclear EGFR exerts biological activity and physiological functions, including changing cell cycle, which in turn promotes proliferation and migration of SUN-80 cells. CONCLUSION: These findings lay a foundation for further explorations seeking to understand the biological effects of the EGF/EGFR system on the occurrence and development of thyroid cancer.

ABSTRACT

Objective:In order to establish a painless delivery detection system in plateau area, promote the concept of painless delivery, reduce the cesarean section rate, the clinical study of painless childbirth in Tibet was carried out.Methods:150 primiparas in Lhasa People's Hospital from January 2018 to Juen 2019 were prospectively collected, which were randomly divided into sufentanil and ropivacaine groups (group A, n=50) and fentanyl and ropivacaine (group B, n=50) and the control group without analgesia (group C, n=50); The general conditions of the patients before and after delivery were collected. The visual analogue scale (VAS) score was used to observe and record the delivery process and results. The Apgar score of newborns was performed, and the umbilical artery blood was taken for blood gas analysis. Results:The first and second stage of labor in group A and B were shorter than those in group C ( P<0.05); there was no difference in the third stage of labor and postpartum bleeding between the three groups ( P>0.05). The VAS scores of groups A and B were lower than that of group C ( P<0.05), while there was no difference between group A and group B ( P>0.05). There was no difference in pH value of umbilical artery blood between the three groups ( P>0.05). The umbilical artery blood PO 2 of group A and group B was higher than those of group C ( P<0.05), while there was no difference between group A and group B ( P>0.05). The umbilical artery blood PCO 2 of group A and group B was lower than that of group C( P<0.05), while there was no difference between group A and group B ( P>0.05). The cesarean section rate in group C was higher than that in groups A and B ( P<0.05). There was no difference in fetal heart rate, fetal distress, and neonatal asphyxia among the three groups ( P>0.05). There was significant difference in the overall incidence of adverse conditions in the three groups ( P<0.05). The Apgar scores of the newborns in groups A and B were higher than those in the C group at 1 min postpartum ( P<0.05), while there were no difference in the Apgar scores among the three groups at 5 and 10 min postpartum ( P>0.05). Conclusions:Painless delivery in plateau area can reduce the pain of delivery and the rate of cesarean section, and has high safety and reduce the adverse effects on mothers and infants, which is worthy of clinical promotion.

ABSTRACT

Objective To observe the effects of different doses of Peng-Hui on expression levels of insulin receptor substance 1 (IRS-1) in type 2 diabetes (T2DM) rats. Methods T2DM rat model was established by feeding with high-fat-diet and intraperitoneal injecting streptozotocin (STZ). T2DM model rats were randomized into four groups: T2DM control group, high dose Peng-Hui (150 mg/kg) group, medium dose Peng-Hui (100 mg/kg) group and low dose Peng-Hui (50 mg/kg) group. The T2DM control group was fed with only high-fat-diet. The normal control group was fed with common diet. The expression levels of IRS-1 in pancreas and skeletal muscle tissues were detected by immunohistochemistry method after 8-week treatment, and fasting blood glucose (FBG) and insulin levels were also detected in five groups. Results FBG lev-els were significantly lower in medium dose Peng-Hui group than those of T2DM control group and low dose Peng-Hui group (P<0.05). There were no significant differences in insulin levels between high, medium and low dose Peng-Hui groups and T2DM control group (P>0.05). The expression levels of IRS-1 of pancreas and skeletal muscle tissues were significant-ly lower in high dose Peng-Hui group than that of T2DM control group (P<0.05). Conclusion The expression of IRS-1 was decreased in high dose Peng-Hui group, but no effects on FBG and plasma insulin.
