ABSTRACT

OBJECTIVE To analyze the data of non-invasive prenatal testing based on specific loci of circulating cell-free fetal DNA (cffDNA). METHODS Selected loci of target chromosomes were analyzed by sequence capture and sequencing. Meanwhile, 600 loci were selected from other chromosomes for determining the concentration of cffDNA. RESULTS A total of 768 specific loci were captured on chromosomes 21 and 18, and used to determine whether the two were abnormal. When the minimum concentration of detected cffDNA was set at 3% and the threshold of Z score was set to [-6,6], the specificity of the analysis was 99.37% and the sensitivity was 100%. CONCLUSION A reliable, convenient and low-cost analytical method has been developed. The method requires less sequencing data for non-invasive prenatal testing, and can accurately detect abnormalities of fetal chromosomes 21 and 18, and simultaneously determine the concentration of cffDNA.

ABSTRACT

Objective To establish a noninvasive prenatal testing(NIPT) method based on the chromosome specific site sequencing instead of the conventional whole genome sequencing. Methods Blood plasma samples from 200 pregnant women with known fetus karyotypes were collected.First, the specific loci on chromosome 13,18 and 21 were selected by the database filter. Then, these specific loci were captured by probes, and were performed sequencing. Finally, chromosome aneuploidy was identified by the Z-score.ResultsSeven fetuses with 21-trisomy syndrome,3 with 18-trisomy syndrome and 1 with 13-trisomy syndrome were detected by the chromosome specific site sequencing, and the results were accordant with those of the whole genome sequencing method. Conclusion The NIPT method based on the chromosome specific site sequencing has the advantages of low cost and high throughput, which may replace the NIPT method based on the whole genome sequencing.

ABSTRACT

Objective To investigate the activation and clinical significance of the mammalian target of rapamycin (mTOR) pathway related protein and eukaryotic translation factor 4E-binding protein 1 (4E-BP1) in gastric cancer. Methods The activation of mTOR and 4E-BP1 in gastric cancer tissues of 38 surgical patients were detected by immunohistochemical method. The differences of phosphorylated mTOR and 4E-BP1 expression among cancerous tissues,para-cancerous tissues and normal gastric mucosa tissues and dinicopathological variables were analyzed by Chi square test and Kruskal-Wallis test. Results The positive expression rate of phosphorylated mTOR in the gastric cancerous tissues was significantly higher than that of para-cancerous tissues and normal tissues [71.1% (27/38),50.0 % (19/38) and 44.7 % (17/38),x2 =11.031,P =0.026].The positive expression rate of downstream protein 4E-BP1 in the gastric cancer tissues was also significantly higher than that of paracancerous tissues and normal tissues [68.4%(26/38),57.9%(22/38) and 28.9% (11/38),x2 =13.943,P=0.007].There was no correlation between phosphorylated mTOR and 4E-BP1 expression and tumor Lauren's sub-type,infiltration,differentiation degree, lymph node metastasis and patient's age. There was statistical significant difference between activated 4E-BP1 expression and tumor size in gastric cancer (H=3.86,P<0.05).Conclsions mTOR pathway was over activated in gastric cancer. There was difference between phosphorylation degree of its downstream protein 4E-BP1 and the tumor size.

ABSTRACT

To effectively investigate the spatial variability of heavy metals in soil, produce a higher quality spatial distribution map, and identify the potential pollution sources of heavy metals, geostatistics was employed to evaluate the effect of scale on spatial variability of heavy metals in Beijing agricultural soils. The results revealed that spatial variability of Cr, Ni, Zn, and Hg was dependent on scale. Validation of the optimality of theoretical semivariance and comparative analysis of the estimation accuracy demonstrated that the multi-scale nested model can reveal the spatial structure of heavy metals effectively and improve the estimation accuracy better than the single-scale method, thereby enabling production a higher quality spatial interpolation map. Thus, the multi-scale kriging nested model is a useful tool for revealing spatial variability of heavy metals in soils, while the spatial distribution maps allow the identification of hot spots with high concentrations of heavy metals.

ABSTRACT

Landscape connectivity, as an important indicator of regional landscape functional pattern measured by cost-distances model, could both reveal evidence of, and act as an indicator, for desertification. Using Minqin county as a case study, this study was further to test cost-distance connectivity for indicating desertification, and to analyze temporal changes of connectivity in the county from 1977 to 1997. The results further indicate the connectivity interpreted as lower cost-distance and higher risk of desertification. The temporal change analysis of connectivity provides more detail supplement of desertification processes. Landscape connectivity changes in the county had three distinctive phases, the strongest decline of log cost-distance with overwhelming decrease area and decrease amount in the period 1977--1984, and its slight decline with the relative balance between the high decrease amount and high increase amount in the period 1984--1992, then its the slight decline with the relative balance between the low decrease amount and low increase amount in the period 1992--1997. The frequent temporal and spatial transition in source class, grassland, alkali-saline land and irrigated cultivated land caused negative effects on the oasis environment.

ABSTRACT

This study uses statistical modeling techniques to develop a desertification risk index (RI) for Minqin County, Gansu Province, China. Twenty socio-economic factors were selected and compared with the RI results to explore the spatial and temporal variability of desertification risk in the study area and to identify possible local driving forces behind desertification risk. The explanatory factors were different in 1988, 1992 and 1997, possibly reflecting the role of temporal variation as a contributor to desertification. The average number of sheep per-household was found to be an important indicator of change in desertification risk, while changes in ridge crop planting area explained the distribution of the rate of change in desertification risk in 1988-1992. The results suggests that the RI was useful in expanding the understanding of spatial temporal desertification issues in Minqin County, as well as identifying a current set of agricultural activities related to desertification risk. Further, given the limited nature of consistent data and observations for the area, development of the RI also served to establish a baseline for future investigations into desertification change and the risks such change might pose for the region.
