ABSTRACT

We propose a histogram clustering (HC) method to accelerate fluorescence lifetime imaging (FLIM) analysis in pixel-wise and global fitting modes. The proposed method's principle was demonstrated, and the combinations of HC with traditional FLIM analysis were explained. We assessed HC methods with both simulated and experimental datasets. The results reveal that HC not only increases analysis speed (up to 106 times) but also enhances lifetime estimation accuracy. Fast lifetime analysis strategies were suggested with execution times around or below 30 µs per histograms on MATLAB R2016a, 64-bit with the Intel Celeron CPU (2950M @ 2GHz).

ABSTRACT

Single-photon avalanche diodes (SPAD) are powerful sensors for 3D light detection and ranging (LiDAR) in low light scenarios due to their single-photon sensitivity. However, accurately retrieving ranging information from noisy time-of-arrival (ToA) point clouds remains a challenge. This paper proposes a photon-efficient, non-fusion neural network architecture that can directly reconstruct high-fidelity depth images from ToA data without relying on other guiding images. Besides, the neural network architecture was compressed via a low-bit quantization scheme so that it is suitable to be implemented on embedded hardware platforms. The proposed quantized neural network architecture achieves superior reconstruction accuracy and fewer parameters than previously reported networks.

ABSTRACT

Estimation of signal parameters via rotational invariance techniques is a classical algorithm widely used in array signal processing for direction-of-arrival estimation of emitters. Inspired by this method, a new signal model and new fluorescence lifetime estimation via rotational invariance techniques (FLERIT) were developed for multiexponential fluorescence lifetime imaging (FLIM) experiments. The FLERIT only requires a few time bins of a histogram generated by a time-correlated single-photon counting FLIM system, greatly reducing the data throughput from the imager to the signal processing units. As a noniterative method, the FLERIT does not require initial conditions, prior information nor model selection that are usually required by widely used traditional fitting methods, including nonlinear least square methods or maximum-likelihood methods. Moreover, its simplicity means it is suitable for implementations in embedded systems for real-time applications. FLERIT was tested on synthesized and experimental fluorescent cell data showing the potentials to be widely applied in FLIM data analysis.
