ABSTRACT

The booting stage and filling stage have been considered as the key stages for arsenic (As) uptake in rice. In this study, a field study was conducted to investigate the influence of the topdressing different amounts of silicon (Si) fertilizer at the key stages on rice As uptake and accumulation. The results showed that topdressing of a low amount of Si fertilizer at the booting stage and filling stage could increase rice yield, promote the formation of iron plaque and the retention of As on iron plaque, and reduce inorganic As content in brown rice. Compared with the control, the rice grain yield was increased by 22.60% with the topdressing of 20 kg·hm-2 Si fertilizer at the grain filling stage. As compared with the control, the Fe and As content in iron plaque under the topdressing of 20 kg·hm-2 Si fertilizer at the booting stage and filling stage was significantly (p < 0.05) increased by 84.34% and 87.78% (Fe content) and 70.96% and 63.80% (As content), respectively. Meanwhile, contents of As in rice roots, stems, and husks at the topdressing of 20 kg·hm-2 Si fertilizer at the booting stage were significantly (p < 0.05) reduced by 45.10%, 33.34%, and 31.23%, respectively, relative to the control. The lowest inorganic As content (0.21 mg·kg-1) in brown rice was obtained at the topdressing of 20 kg·hm-2 Si fertilizer at the booting stage, which was close to the National Food Limit Standard of 0.20 mg·kg-1 (GB 2762-2017). Therefore, topdressing of 20 kg·hm-2 Si fertilizer at the booting stage might be considered as an effective method to reduce inorganic As content in brown rice.

ABSTRACT

In this study, spherical α-Fe2O3 prepared by the hydrothermal method was used as a template for the first time; LaFeO3 perovskite catalysts were successfully synthesized by the molten salt method (M-LF-T), sol-gel method (S-LF-T), and co-precipitation method (C-LF-T), respectively. To determine the optimal synthesis method, X-ray diffraction patterns were obtained and showed that single phase LaFeO3 with good crystallinity was prepared by the molten salt method after calcination at 600 °C for 4 h. SEM and TEM images showed that the M-LF-600 catalyst preserved the spherical structure of α-Fe2O3 template. Compared with the catalysts synthesized by the sol-gel method and co-precipitation method, the M-LF-600 catalyst had the highest BET surface area of 16.73 m2 g-1. X-ray photoelectron spectroscopy analysis showed that the M-LF-600 catalyst had the highest surface Fe3+/Fe2+ molar ratio and the best surface oxygen adsorption capacity. The CO oxidation of the LaFeO3 catalyst demonstrated that the M-LF-600 catalyst had the best catalytic performance.

ABSTRACT

GGC repeat expansions within NOTCH2NLC have been identified as the genetic cause of neuronal intranuclear inclusion disease (NIID). To understand the molecular pathogenesis of NIID, here, we established both a transgenic mouse model and a human neural progenitor cells (hNPCs) model. Expression of the NOTCH2NLC with expanded GGC repeats produced widespread intranuclear and perinuclear polyglycine (polyG), polyalanine (polyA), and polyarginine (polyR) inclusions, leading to behavioral deficits and severe neurodegeneration, which faithfully mimicked the clinical and pathological features associated with NIID. Furthermore, conserved alternative splicing events were identified between the NIID mouse and hNPC models, among which was the enrichment of the binding motifs of hnRNPM, an RNA binding protein known as alternative splicing regulator. Expanded NOTCH2NLC-polyG and NOTCH2NLC-polyA could interact with and sequester hnRNPM, while overexpression of hnRNPM could ameliorate the cellular toxicity. These results together suggested that dysfunction of hnRNPM could play an important role in the molecular pathogenesis of NIID.

ABSTRACT

Objective: Although risk factors for excessive daytime sleepiness (EDS) have been reported, there are still few cohort-based predictive models for EDS in Parkinson's disease (PD). This 1-year longitudinal study aimed to develop a predictive model of EDS in patients with PD using a nomogram and machine learning (ML). Materials and methods: A total of 995 patients with PD without EDS were included, and clinical data during the baseline period were recorded, which included basic information as well as motor and non-motor symptoms. One year later, the presence of EDS in this population was re-evaluated. First, the baseline characteristics of patients with PD with or without EDS were analyzed. Furthermore, a Cox proportional risk regression model and XGBoost ML were used to construct a prediction model of EDS in PD. Results: At the 1-year follow-up, EDS occurred in 260 of 995 patients with PD (26.13%). Baseline features analysis showed that EDS correlated significantly with age, age of onset (AOO), hypertension, freezing of gait (FOG). In the Cox proportional risk regression model, we included high body mass index (BMI), late AOO, low motor score on the 39-item Parkinson's Disease Questionnaire (PDQ-39), low orientation score on the Mini-Mental State Examination (MMSE), and absence of FOG. Kaplan-Meier survival curves showed that the survival prognosis of patients with PD in the high-risk group was significantly worse than that in the low-risk group. XGBoost demonstrated that BMI, AOO, PDQ-39 motor score, MMSE orientation score, and FOG contributed to the model to different degrees, in decreasing order of importance, and the overall accuracy of the model was 71.86% after testing. Conclusion: In this study, we showed that risk factors for EDS in patients with PD include high BMI, late AOO, a low motor score of PDQ-39, low orientation score of MMSE, and lack of FOG, and their importance decreased in turn. Our model can predict EDS in PD with relative effectivity and accuracy.

ABSTRACT

Pb accumulation in rice varies significantly at different growth stages. In this study, a hydroponic experiment was conducted to study the effects of exogenous Pb stress on Pb accumulation and transportation in rice plants and determine the key rice growth stages of Pb accumulation and their contribution to the Pb content in brown rice. For the hydroponic experiment, 0.5 mg ·L-1 of exogenous Pb was added to rice at different growth stages, including the tillering stage, jointing stage, booting stage, filling stage, dough stage, maturing stage, and whole growth stage (103 d) of the rice plant. Another treatment of the whole growth stage without Pb stress was established as the control. The results showed that: ① There were no significant effects of Pb stress in any single stage on the plant height, tiller number, and biomass, but the rice plant height and biomass significantly decreased under the Pb stress for the whole growth period. ② Pb contents in different tissues at the maturing stage of the rice plant were in the order of root > stem node 1 > other stem nodes > root rhizome > sti > leaf > ear > husk > brown rice. Pb contents in brown rice ranged from 0.1 to 1.2 mg ·kg-1 for all exogenous Pb treatments, and ranked in the growth stages as booting stage > jointing stage > tillering stage > filling stage > maturing stage > dough stage. ③ The relative contribution rates of Pb accumulation in the whole rice plants were relatively high during the reproductive growth period (filling stage, dough stage, and maturing stage), while those in the above-ground parts of the rice plants were relatively high during the vegetative growth stage (tillering stage, jointing stage, booting stage). ④ The booting stage was the key growth period for Pb accumulation in brown rice. Pb stress at this stage contributed 43.3% of Pb content in brown rice, followed by Pb stress at the jointing stage and the tillering stage, with contribution rates of 24.4% and 21.3%, respectively. ⑤ Water management regimes, application of amendments, or leaf resistance control techniques should be applied appropriately at the booting stage of the rice plants to reduce Pb accumulation in brown rice and to realize the safe use of rice paddies polluted with Pb.

ABSTRACT

The absorption and accumulation of As at different stages of rice growth are significantly different. To study the key growth stages of As accumulation in brown rice and to determine the contribution of As accumulation at different growth stages to As contribution in brown rice, a rice hydroponics experiment was carried out by adding external As during the different rice growth stages: tillering stage (30 d), jointing stage (16 d), booting stage (13 d), filling stage (17 d), dough stage (15 d), maturity stage (13 d), and full growth period (104 d). The results showed that: ① As stress at different growth stages had a significant effect on the biomass of rice plants. In comparison with the control CK, the treatments of As stress with five single-stages decreased the rice biomass, except during the tillering stage. Among these treatments, although the As stress at booting stage had the lowest rice biomass, the biomass of all the rice plants were higher than that for As stress in the full growth period. ② The content of As in brown rice for all the treatments of As stress at six single-stages, ranging from 0.08-0.24 mg ·kg-1, was higher than that of CK. Among these six treatments, the As stress at booting stage had the highest As content in brown rice, accounting for 64.9% of the As content in the full growth period. ③ The accumulation of As in brown rice for all the treatments of As stress at six single-stages was higher than that of CK, and ranged from 1.4-1.5 µg ·plant-1. Among these six treatments, the As stress at booting stage had the highest As accumulation, followed by filling stage. The accumulation of As in brown rice treated with As stress was highest in the full growth period with a value of 5.7 µg ·plant-1. ④ The relative contribution of As stress at the booting stage was the highest for the As accumulation in brown rice and reached to 40.3%, followed by filling stage with 26.0%. Therefore, the rice growth stages of the booting stage and filling stage were the key stages of As accumulation in brown rice.

ABSTRACT

In this study, we investigated the effects of exogenous phosphorus on the accumulation and transportation of cadmium in rice plants through a hydroponic experiment. In the experiment, the rice variety was Huanghuazhan, P solution concentrations were 10.0-45.0 mg ·L-1 that was made using NaH2PO4, and Cd solution concentrations were 0.1 mg ·L-1 and 0.2 mg ·L-1. The results showed that: ① the biomass in all parts of rice plants and contents of photosynthetic pigment (chlorophyll a, chlorophyll b, and carotenoid) increased gradually with an increase in exogenous P. ② Content of Cd in rice stems, leaves, husk, and brown rice increased gradually with an increase in the amounts of exogenous P. The content of Cd in brown rice increased by 2.8%-22.8% and 40.9%-61.8% when treated with Cd concentrations of 0.1 mg ·L-1 and 0.2 mg ·L-1 in hydroponic solutions, respectively. ③ Cd accumulation in rice plants was accelerated due to the application of exogenous P. Cd accumulating amounts increased from 395.1 µg ·plant-1 to 542.6 µg ·plant-1 and 639.6 µg ·plant-1 to 1082.0 µg ·plant-1 when treated with Cd concentrations of 0.1 and 0.2 mg ·L-1 in hydroponic solutions, respectively. ④ With an increase in the applied amounts of exogenous P, the P/Cd quality ratio in rice roots increased, while those in rice stems, leaves, husks, and brown rice decreased; meanwhile, the Cd transfer coefficients from root to stem (TFroot-stem) and stem to leaf (TFstem-leaf) increased. This showed that there was a certain synergistic effect between P and Cd in the rice parts. Finally, the application of exogenous P promoted the transfer of Cd from the rice root to other rice tissues, resulting in a synergistic effect on Cd accumulation and transportation in various rice tissues and increased Cd contents in brown rice.

ABSTRACT

This study aimed to determine the mutational spectrum of familial Parkinson's disease and sporadic early-onset Parkinson's disease (sEOPD) in a mainland Chinese population and the clinical features of mutation carriers. We performed multiplex ligation-dependent probe amplification assays and whole-exome sequencing for 1676 unrelated patients with Parkinson's disease in a mainland Chinese population, including 192 probands from families with autosomal-recessive Parkinson's disease, 242 probands from families with autosomal-dominant Parkinson's disease, and 1242 sEOPD patients (age at onset ≤ 50). According to standards and guidelines from the American College of Medical Genetics and Genomics, pathogenic/likely pathogenic variants in 23 known Parkinson's disease-associated genes occurred more frequently in the autosomal-recessive Parkinson's disease cohort (65 of 192, 33.85%) than in the autosomal-dominant Parkinson's disease cohort (10 of 242, 4.13%) and the sEOPD cohort (57 of 1242, 4.59%), which leads to an overall molecular diagnostic yield of 7.88% (132 of 1676). We found that PRKN was the most frequently mutated gene (n = 83, 4.95%) and present the first evidence of an SNCA duplication and LRRK2 p.N1437D variant in mainland China. In addition, several novel pathogenic/likely pathogenic variants including LRRK2 (p.V1447M and p.Y1645S), ATP13A2 (p.R735X and p.A819D), FBXO7 (p.G67E), LRP10 (c.322dupC/p.G109Rfs*51) and TMEM230 (c.429delT/p.P144Qfs*2) were identified in our cohort. Furthermore, the age at onset of the 132 probands with genetic diagnoses (median, 31.5 years) was about 14.5 years earlier than that of patients without molecular diagnoses (i.e. non-carriers, median 46.0 years). Specifically, the age at onset of Parkinson's disease patients with pathogenic/likely pathogenic variants in ATP13A2, PLA2G6, PRKN, or PINK1 was significantly lower than that of non-carriers, while the age at onset of carriers with other gene pathogenic/likely pathogenic variants was similar to that of non-carriers. The clinical spectrum of Parkinson's disease-associated gene carriers in this mainland Chinese population was similar to that of other populations. We also detected 61 probands with GBA possibly pathogenic variants (3.64%) and 59 probands with GBA p.L444P (3.52%). These results shed insight into the genetic spectrum and clinical manifestations of Parkinson's disease in mainland China and expand the existing repertoire of pathogenic or likely pathogenic variants involved in known Parkinson's disease-associated genes. Our data highlight the importance of genetic testing in Parkinson's disease patients with age at onset < 40 years, especially in those from families with a recessive inheritance pattern, who may benefit from early diagnosis and treatment.
