ABSTRACT

One lucrative method for overcoming challenges in drug discovery or for enhancing undesirable properties of already-approved medications is prodrug design. The goal of this review is to present researchers with a profile of naturally occurring Phytophenols as carriers that would be used for prodrug synthesis as well as their advantages. Phytophenols offer several advantages when used as promoieties as they also possess antioxidant and analgesic properties, they are obtained naturally and their safety profile is well established. Several phytophenols like menthol, thymol, eugenol, guaiacol, sesamol, vanillin, and umbelliferone are some of the phytophenols that have several beneficial properties and are extensively employed in the field of food processing and medicine. In the current review, we have listed all types of promoieties that are used for prodrug synthesis and phytophenols are reviewed in detail, which may help researchers to select phytophenols based on their need and suitability for drug candidates.

ABSTRACT

Cancer, the second leading cause of death worldwide, is a major health problem. Chemotherapy, radiation therapy and surgery are current treatments for cancer. Most anticancer drugs have severe toxic effects and are required to be administered in cycles to reduce toxicity and prevent resistance. Plant-based drugs have shown a potential for treatment of cancer, and various plant secondary metabolites have shown promising antitumor activity against several cancer cell lines, such as leukemia, colon cancer, prostate cancer, breast cancer and lung cancer. Vincristine, etoposide, topotecan and paclitaxel, which are of natural origin, are successfully used in clinical practice, and this has generated interest in natural compounds as anticancer agents. Some phytoconstituents like curcumin, piperine, allicin, quercetin and resveratrol have been extensively researched and reviewed. In the current study, we have reviewed several plants like Athyrium hohenackerianum, Aristolochia baetica, Boswellia serrata, Panax ginseng, Berberis vulgaris, Tanacetum parthenium, Glycine max, Combretum fragrans, Persea americana, Raphanus sativus, Camellia sinensis, and Nigella sativa for their source, key phytoconstituents, and anticancer activity along with their toxicity profile. Few phytoconstituents like boswellic acid, sulforaphane and ginsenoside showed excellent anticancer activity compared to standard drugs and are potential clinical candidates.

ABSTRACT

The mutual prodrugs of aceclofenac with various naturally available antioxidants; menthol, thymol, eugenol, guiacol and vanillin have been synthesized by the DCC coupling method, purified and characterized by spectral data, as well as, partition coefficient, solubility and hydrolytic studies. The title compounds have more lipophilic character as compared to the parent moieties and good stability in acidic environment, which is prerequisite for the oral absorption of the drug. Under gastric as well as intestinal pH conditions these prodrugs showed variable susceptibility towards hydrolysis. The synthesized derivatives were evaluated for antiinflammatory, analgesic activities and ulcerogenic potential. Prodrug showed improved solubility in organic solvents, which implies lipophilic character of ester prodrugs and were also found to be chemically stable in acidic environment. The aceclofenac mutual prodrugs showed improved analgesic and anti-inflammatory activities with reduced ulcerogenicity.
