ABSTRACT

Dengue is one of the major diseases causing global public health concerns. Despite technological advances in vaccine production against all its serotypes, it is estimated that the dengue virus is responsible for approximately 390 million infections per year. Laboratory diagnosis has been the key point for the correct treatment and prevention of this disease. Currently, the limiting factor in the manufacture of dengue diagnostic kits is the large-scale production of the non-structural 1 (NS1) antigen used in the capture of the antibody present in the infected patients' serum. In this work, we demonstrate the production of the non-structural 1 protein of dengue virus (DENV) serotypes 1-4 (NS1-DENV1, NS1-DENV2, NS1-DENV3, and NS1-DENV4) in the methylotrophic yeast Pichia pastoris KM71H. Secreted recombinant protein was purified by affinity chromatography and characterized by SDS-PAGE and ELISA. The objectives of this study were achieved, and the results showed that P. pastoris is a good heterologous host and worked well in the production of NS1DENV 1-4 recombinant proteins. Easy to grow and quick to obtain, this yeast secreted ready-to-use proteins, with a final yield estimated at 2.8-4.6 milligrams per liter of culture. We reached 85-91% sensitivity and 91-93% specificity using IgM as a target, and for anti-dengue IgG, 83-87% sensitivity and 81-93% specificity were achieved. In this work, we conclude that the NS1 recombinant proteins are efficiently produced in P. pastoris and have great potential for use in diagnostic kits for dengue virus infections. The transformed yeast obtained can be used for production in industrial-scale bioreactors.

ABSTRACT

Objetivo: analisar as concepções e práticas da equipe de saúde acerca do processo do cuidar em um centro de tratamento oncológico. Metodologia: pesquisa de natureza qualitativa, realizada com profissionais pertencentes à equipe de saúde de uma unidade de tratamento quimioterápico. A coleta de dados ocorreu por meio de um roteiro de entrevistas, cuja análise foi baseada em Lawrence Bardin. Foi aprovada pelo Comitê de Ética em Pesquisa com Seres Humanos da Universidade Federal de Viçosa sob o número 1.000.021. Resultados: os depoimentos indicaram que a equipe interdisciplinar atua como ferramenta estratégica para a implementação do cuidado holístico e que o acolhimento e vínculo terapêutico são interfaces possíveis entre a tríade profissional-família-paciente. Conclusão: esta investigação mostrou que as concepções de cuidado na visão dos participantes da pesquisa vão ao encontro das próprias práticas, ou seja, as mesmas coincidem, o que demonstra que a equipe realiza uma práxis genuinamente coerente àquilo que acreditam ser um cuidado ideal. A equipe interdisciplinar entrevistada mostrou-se capaz de desenvolver um cuidado holístico, ao apresentar habilidades técnicas e humanas para lidar com as demandas do cuidar do paciente oncológico em tratamento quimioterápico.


Objective: to analyse the conceptions and practices of the health team about the care process in a cancer treatment center. Methodology: this is a qualitative research carried out with professionals belonging to the health team of a chemotherapy treatment unit. The data collection took place through a script of interviews, whose analysis was based on Lawrence Bardin. It was approved by the Human Research Ethics Committee of the Federal University of Viçosa under the number 1.000.021. Results: the interviews indicated that the interdisciplinary team acts as a strategic tool for the implementation of holistic care and that the therapeutic link and reception are possible interfaces between the professional-family-patient triad. Conclusion: this research showed that the care conceptions in the participants' view of the research match the practices themselves, that is, they coincide, which demonstrates that the team performs a genuinely coherent praxis to what they believe to be an ideal care. The interdisciplinary team interviewed was able to develop a holistic care, presenting technical and human skills to deal with the demands of the cancer patient care in chemotherapy treatment.
