ABSTRACT Root canal morphology and its anatomical variations pose a great challenge to endodontists Aim The aim of this in silico study was to perform a qualitative and quantitative analysis of the threedimensional morphological characteristics of the isthmus in the mesial root canals of mandibular molars using microcomputed tomography (micro-CT) Material and Method Six hundred first mandibular molars were selected, including 317 with two mesial canals with isthmuses between the canals, and fully formed root. Isthmus morphology was determined in 3D longitudinal sections using Fan et al. (2010) classification. Root length, and the volume and area of apical and coronal level were measured. Additionally, the structural model index (SMI) of the canals were also assessed Results The prevalence of isthmuses in the mesial root canals was 32% type II, 29% type III, 22% type IV, and 17% type I. The root length was found to be 9.1±0.5 mm, the volume and area, of all root canal system, were 41.8±40.1 mm3 and 63.6±24.2 mm2 respectively. The isthmi volume and area alone were 11.06±9.03 mm3 and 30.02±11.02 mm2. The study confirmed that isthmuses are present in mesial canals of mandibular first molars, being more frequent in the apical third Conclusion The high prevalence of isthmuses with complex morphological features underscores the importance of using intracanal medications to disinfect areas unprepared by instruments.


RESUMO A morfologia do canal radicular e suas variações anatômicas representam um grande desafio para os endodontistas. O objetivo deste estudo ex vivo foi realizar uma análise qualitativa e quantitativa das características morfológicas tridimensionais do istmo nos canais mesiais de molares inferiores por meio de microtomografia computadorizada (micro-CT) Material e Método Foram selecionados 600 primeiros molares inferiores, incluindo 317 com dois canais mesiais com istmos e raiz totalmente formada. A morfologia do istmo foi determinada em cortes longitudinais 3D usando a classificação Fan et al. (2010). Foram mensurados o comprimento da raiz, o volume e a área apical e coronal e da cavidade pulpar. Adicionalmente, também foram avaliados o structure model index (SMI) dos canais Resultados A prevalência de istmos nos canais mesiais foi de 32% tipo II, 29% tipo III, 22% tipo IV e 17% tipo I. O comprimento da raiz foi de 9,1±0,5 mm, o volume e a área de todo o sistema de canais radiculares foram de 41.8±40.1 mm3 e 63.6±24.2 mm2, respectivamente. O volume e área do istmo isoladamente foram 11.06±9,03 mm3 e 30.02±11.02 mm2. O estudo confirmou que os istmos estão presentes em canais mesiais dos primeiros molares inferiores, sendo mais frequentes no terço apical Conclusão A alta prevalência de istmos com características morfológicas complexas ressalta a importância do uso de medicação intracanal para desinfecção de áreas não tocadas por instrumentos.

BACKGROUND: An individual's dietary pattern contributes in different ways to the prevention and control of recurrent cardiovascular events. However, the quality of the diet is influenced by several factors. The present study aimed to evaluate the quality of the diet of individuals with cardiovascular diseases and determine whether there is an association between sociodemographic and lifestyle factors. METHODS: This is a cross-sectional study carried out with individuals with atherosclerosis (coronary artery disease, cerebrovascular disease or peripheral arterial disease) recruited from 35 reference centres for the treatment of cardiovascular disease in Brazil. Diet quality was assessed according to the Modified Alternative Healthy Eating Index (mAHEI) and stratified into tertiles. For comparing two groups, the Mann-Whitney or Pearson's chi-squared tests were used. However, for comparing three or more groups, analysis of variance or Kruskal-Wallis was used. For the confounding analysis, a multinomial regression model was used. p < 0.05 was considered statistically significant. RESULTS: In total, 2360 individuals were evaluated: 58.5% male and 64.2% elderly. The median (interquartile range [IQR]) of the mAHEI was 24.0 (20.0-30.0), ranging from 0.4 to 56.0 points. When comparing the odds ratios (ORs) for the low (first tertile) and medium (second tertile) diet quality groups with the high-quality group (third tertile), it was observed that there was an association between diet quality with a family income of 1.885 (95% confidence intervals [CI] = 1.302-2.729) and 1.566 (95% CI = 1.097-2.235), as well as physical activity of 1.391 (95% CI = 1.107-1.749) and 1.346 (95% CI = 1.086-1.667), respectively. In addition, associations were observed between diet quality and region of residence. CONCLUSIONS: A low-quality diet was associated with family income, sedentarism and geographical area. These data are extremely relevant to assist in coping with cardiovascular disease because they enable an assessment of the distribution of these factors in different regions of the country.

Vector control has been an essential strategy in Brazil to manage vector-borne diseases, and the use of insecticides plays an important role in this effort. Pyriproxyfen (PPF) has become a common insect growth regulator used to control juvenile stages of mosquitoes by disturbing their growth and development. This study assesses the susceptibility and resistance status of Brazilian Ae. aegypti populations that previously showed low resistance levels to PPF. Eggs of Ae. aegypti were collected from six cities located in the northeast states of Ceará (Quixadá, Icó, and Juazeiro do Norte), and Bahia (Itabuna, Brumado, and Serrinha). We used the Ae. aegypti Rockefeller strain as an experimental control and a strain known to be susceptible to insecticides. Inhibition of emergence rates by 50% of Ae. aegypti populations varied from 0.0098-0.046 µg/L. Mosquitoes from Icó, Serrinha, and Brumado showed low resistance levels [resistance ratio (RR50) = 2.33, 4.52, and 4.83, respectively], whereas moderate levels of resistance were detected in populations from Juazeiro do Norte (RR50=5.83) and Itabuna (RR50=7.88). Aedes aegypti collected from the Quixadá population showed a high resistance level to pyriproxyfen (RR50=11). The evolution of resistance in Brazilian Ae. aegypti populations to PPF can compromise vector control efforts. Continuous monitoring of insecticide resistance in Ae. aegypti is essential for making timely management decisions for effective vector control and management.

Introdução: O número de adeptos ao vegetarianismo tem crescido consideravelmente. Observa-se que, entre as vantagens de se adotar uma dieta vegetariana, está o menor risco de desenvolver sobrepeso e obesidade, o que pode estar relacionado com o alto consumo de vegetais e práticas importantes de estilo de vida. Objetivos: Avaliar o estado nutricional e a frequência do consumo alimentar de vegetarianos não estritos e estritos. Métodos: Estudo transversal descritivo, realizado com participantes do grupo "Vegans SLZ", de ambos os sexos e com idade a partir de 18 anos. A coleta de dados ocorreu de janeiro a março de 2020. Foi realizada aplicação de questionários sobre dados de frequência do consumo alimentar, socioeconômico e de estilo de vida e a aferição de medidas de peso corporal, estatura e de percentuais de gordura corporal e massa muscular. Os dados foram tabulados no Excel 2010® e analisados no programa estatístico R Studio versão 3.6.1. Resultados: Dos 43 vegetarianos avaliados, a maioria seguia a dieta vegetariana estrita (55,81%) e era eutrófica (62,79%), 32,56% apresentaram alto percentual de gordura corporal e 53,49% percentual de massa magra corporal normal. Foi frequente o consumo de pães, cereais, tubérculos, leguminosas, oleaginosas, frutas e verduras, e 41% consumiam alimentos industrializados de 4 a 7 vezes por semana. Conclusões: A maioria da amostra tinha estado nutricional eutrófico e consumo regular de todos os grupos alimentares, porém com importante consumo de alimentos industrializados.


Introduction: The number of adherents to vegetarianism has grown considerably. It is observed that, among the advantages of adopting a vegetarian diet, there is a lower risk of developing overweight and obesity, which may be related to the high consumption of vegetables and important lifestyle practices. Objectives: To assess the nutritional status and frequency of food consumption of non-strict and strict vegetarians. Methods: Descriptive cross-sectional study carried out with participants of the "Vegans SLZ" group, of both sexes and aged 18 years and over. Data collection took place from January to March 2020. Questionnaires were applied to data on frequency of food, socioeconomic and lifestyle consumption and measurements of body weight, height and percentages of body fat and muscle mass were measured. Data were tabulated in Excel 2010® and analyzed in the statistical program R Studio version 3.6.1. Results: Of the 43 vegetarians evaluated, the majority followed the strict vegetarian diet (55.81%) and were eutrophic (62.79%), 32.56% had a high percentage of body fat and 53.49% percentage of lean body mass normal. The consumption of breads, cereals, tubers, legumes, oilseeds, fruits and vegetables was frequent, and 41% consumed processed foods 4 to 7 times a week. Conclusions: Most of the sample had a eutrophic nutritional status and regular consumption of all food groups, but with a significant consumption of industrialized foods.

BACKGROUND: The development of resistance against insecticides in Aedes aegypti can lead to operational failures in control programs. Knowledge of the spatial and temporal trends of this resistance is needed to drive effective monitoring campaigns, which in turn provide data on which vector control decision-making should be based. METHODS: Third-stage larvae (L3) from the F1 and F2 generations of 39 Peruvian field populations of Ae. aegypti mosquitoes from established laboratory colonies were evaluated for resistance against the organophosphate insecticide temephos. The 39 populations were originally established from eggs collected in the field with ovitraps in eight departments of Peru during 2018 and 2019. Dose-response bioassays, at 11 concentrations of the insecticide, were performed following WHO recommendations. RESULTS: Of the 39 field populations of Ae. aegypti tested for resistance to temephos , 11 showed high levels of resistance (resistance ratio [RR] > 10), 16 showed moderate levels of resistance (defined as RR values between 5 and 10) and only 12 were susceptible (RR < 5). The results segregated the study populations into two geographic groups. Most of the populations in the first geographic group, the coastal region, were resistant to temephos, with three populations (AG, CR and LO) showing RR values > 20 (AG 21.5, CR 23.1, LO 39.4). The populations in the second geographic group, the Amazon jungle and the high jungle, showed moderate levels of resistance, with values ranging between 5.1 (JN) and 7.1 (PU). The exception in this geographic group was the population from PM, which showed a RR value of 28.8 to this insecticide. CONCLUSIONS: The results of this study demonstrate that Ae. aegypti populations in Peru present different resistance intensities to temephos, 3 years after temephos use was discontinued. Resistance to this larvicide should continue to be monitored because it is possible that resistance to temephos could decrease in the absence of routine selection pressures.

ABSTRACT

O presente estudo tem como objetivo investigar as características neuropsicológicas do desenvolvimento no primeiro ano de bebês prematuros comparados com bebês a termo, através de uma metodologia de revisão de literatura. A busca foi realizada nas bases MEDLINE, APAPsycNet, Science Direct e SciELO, no período de 2004 a 2021, utilizando a palavra-chave "neuropsychology" combinada com "infant premature" e "premature birth". Apenas quatro estudos atenderam aos critérios de inclusão, que foram: amostras de no mínimo dez prematuros, comparados com bebês a termo de até um ano de idade; estudos com, no mínimo, uma testagem cognitiva ou de linguagem, motora e socioemocional. Os estudos encontrados relataram os seguintes resultados: foi observada uma maior reatividade nos prematuros para raiva, mas menores reatividades ao medo e a brincadeiras; foram relatados também prejuízos motores, diferenças em relação ao índice de desenvolvimento cognitivo (considerando-se a idade cronológica), assim como escores menores no desempenho audiovisual, cognitivo e de linguagem. Neste sentido, os estudos sugerem que bebês prematuros apresentam comprometimentos distintos quando comparados aos bebês nascidos a termo. Com poucos estudos longitudinais, argumenta-se que mais estudos com enfoque no desenvolvimento integral são necessários.


This study aimed to investigate the neuropsychological characteristics of development in the first year of premature infants compared to full term infants through a literature review methodology. The search was conducted in the MEDLINE, APAPsycNet, Science Direct and SciELO databases, from 2004 to 2021, with the keyword "neuropsychology" combined with "infant premature" and "premature birth". Only four studies met the inclusion criteria, which were: samples of at least ten premature infants, compared to full-term infants up to one year of age; studies with at least a cognitive or language, motor and socio-emotional testing. The studies found reported the following results: a higher reactivity for anger in the premature, but lower reactivity for fear and play; motor impairment, difference in cognitive development index (considering chronological age), as well as lower scores in audiovisual, cognitive and language performance. In this sense, the studies suggest that preterm infants present distinct impairments when compared to full-term infants. With few longitudinal studies, it is argued that further studies focusing on integral development are needed.


Este estudio tuvo como objetivo investigar las características neuropsicológicas del desarrollo del primer año de los recién nacidos prematuros en comparación con los recién nacidos a término. Se realizaron búsquedas en las bases de datos MEDLINE, APAPsycNet, Science Direct y SciELO, de 2004 a 2021, con las palabras clave: "neuropsicología" combinada con "lactante prematuro" y "nacimiento prematuro". Solo cuatro estudios cumplieron con los criterios de inclusión: muestras de al menos diez recién nacidos prematuros, en comparación con recién nacidos a término hasta un año de edad, estudios con al menos una prueba cognitiva o de lenguaje, motora y socioemocional. Se encontró: mayor reactividad en prematuros ante la ira y menor en el miedo y los juegos, deterioro motor, diferencia en relación al índice de desarrollo cognitivo, considerando la edad cronológica, así como menores puntuaciones en desempeño audiovisual, cognitivo y lingüístico. Los bebés prematuros parecen tener diferentes discapacidades en comparación con los bebés nacidos a término. Con pocos estudios longitudinales, se argumenta que se necesitan más estudios con un mayor enfoque en el desarrollo integral.

RESUMEN Introducción. Paraguay es un país endémico para el dengue y la intervención primaria para esta arbovirosis es el control químico de su vector Aedes aegypti, siendo necesario estudiar su resistencia frente a insecticidas utilizados para el control y conocer su sostenibilidad en espacio y tiempo. Objetivo. Determinar la susceptibilidad de poblaciones silvestres de Ae. aegypti procedentes de Villa Elisa-Central, frente al adulticida deltametrina y conocer el área potencial de distribución de estas poblacionesresistentes/susceptibles. Metodología. Se utilizaron mosquitos Ae. aegypti adultos de la Filial 1 obtenidos de ovitrampas colocadas en viviendas geo-localizadas entre diciembre 2017-marzo 2018. Las hembras resultantes de la cría en condiciones ad hoc en laboratorio fueron expuestas a concentraciones crecientes del insecticida. La distribución real de las poblaciones de mosquitos y su área de influencia, fueron modeladas mediante DIVA-GIS. Resultados. La mortalidad promedio observada para cada dosis de deltametrina fue: 5,3% para (0,03%); 47,4% para (0,15%) y 61,2% para (0,3). Se observó áreas de influencia para la presencia de Ae. aegypti. Conclusión. los valores correspondientes a la mortalidad resultaron inferiores al 98%, lo cual fue indicativo de resistencia a dosis diagnóstica. El modelo de distribución geográfica de la población evaluada demostró la existencia de áreas idóneas con condiciones bioclimáticas óptimas (niveles de precipitación, temperatura y humedad), propicias para la aparición de reservorios de agua y presencia del vector, tanto en Villa Elisa, como en los distritos localizados en la periferia tales como San Lorenzo, Lambaré, Ñemby, San Antonio y Fernando de la Mora.


ABSTRACT Introduction. Methodology. Mosquitoes Ae. aegypti adults from Filial 1 obtained from ovitraps placed in geo-located dwellings between December 2017 and March 2018. The females resulting from rearing under ad hoc conditions in the laboratory were exposed to increasing concentrations of the insecticide. The actual distribution of mosquito populations and their area of ​​influence were modeled using DIVA-GIS. Results. The average mortality observed for each dose of deltamethrin was: 5.3% for (0.03%); 47.4% for (0.15%) and 61.2% for (0.3). Areas of influence were observed for the presence of Ae. aegypti. Conclusion. The values ​​corresponding to mortality were lower than 98%, which was indicative of resistance to diagnostic dose. The geographic distribution model of the evaluated population demonstrated the existence of suitable areas with optimal bioclimatic conditions (levels of precipitation, temperature and humidity), conducive to the appearance of water reservoirs and the presence of the vector, both in Villa Elisa and in the districts located on the outskirts such as San Lorenzo, Lambaré, Ñemby, San Antonio and Fernando de la Mora.

ABSTRACT

White light during mycelial growth influences high conidial stress tolerance of the insect-pathogenic fungus Metarhizium robertsii, but little is known if low- or high-white light irradiances induce different stress tolerances. The fungus was grown either in the dark using two culture media: on minimal medium (Czapek medium without sucrose = MM) or on potato dextrose agar (PDA) or PDA medium under five different continuous white light irradiances. The stress tolerances of conidia produced on all treatments were evaluated by conidial germination on PDA supplemented with KCl for osmotic stress or on PDA supplemented with menadione for oxidative stress. Conidia produced on MM in the dark were more tolerant to osmotic and oxidative stress than conidia produced on PDA in the dark or under the light. For osmotic stress, growth under the lower to higher irradiances produced conidia with similar tolerances but more tolerant than conidia produced in the dark. For oxidative stress, conidia produced under the white light irradiances were generally more tolerant to menadione than conidia produced in the dark. Moreover, conidia produced in the dark germinated at the same speed when incubated in the dark or under lower irradiance treatment. However, at higher irradiance, conidial germination was delayed compared to germination in the dark, which germinated faster. Therefore, growth under light from low to high irradiances induces similar conidial higher stress tolerances; however, higher white light irradiances cause a delay in germination speed.

Resumo Introdução Considerando-se a magnitude da hipertensão arterial sistêmica e suas complicações, assim como a importância da atividade física regular na sua prevenção e/ou tratamento, é necessário identificar os fatores associados à prática de atividade física insuficiente. Objetivo Analisar os fatores associados à atividade física insuficiente em hipertensos atendidos na atenção primária à saúde. Método Estudo transversal realizado com amostra aleatória de 305 hipertensos em três unidades de saúde de São Luís (MA). Foi realizada análise bivariada, com estimativas de razões de prevalência, intervalos de confiança e análise multivariada por regressão de Poisson. Resultados A atividade física insuficiente foi observada em 38,7% da amostra. Atividade física insuficiente foi associada a menos de 8 anos de educação formal (razão de prevalência - RP = 1,58; intervalo de confiança de 95% - IC = 1,04-2,39), colesterol total ≥200 mg/dL (RP = 0,78; IC 95% = 0,58 -1,04), lipoproteína de baixa densidade colesterol-LDLc ≥100 mg/dL (RP = 0,79; IC 95% = 0,53-0,95) e lipoproteína de alta densidade colesterol-HDLc <40 mg / dL entre homens e <50 mg/dL entre mulheres (RP = 1,21; IC 95% = 0,90-1,64). A escolaridade <8 anos (RP = 1,50; IC 95% = 0,99-2,29) e LDLc ≥100 mg/dL (RP = 0,72; IC 95% = 0,54-0,96) também estiveram associados na regressão multivariada. Conclusão Observou-se alta prevalência de atividade física insuficiente e sua associação com baixa escolaridade e LDLc alterado em hipertensos.


Abstract Background Considering the magnitude of systemic hypertension (SH) and its complications, as well as the importance of regular physical activity in its prevention and/or treatment, there is a necessity to identify the factors associated with insufficient practice of physical activity. Objective To analyze factors associated with insufficient physical activity in hypertensive patients treated at Primary Health Care (PHC). Method This cross-sectional study was performed with a random sample of 305 hypertensive patients in three PHC units in São Luis, state of Maranhão, Brazil. A bivariate analysis was performed with estimates of prevalence ratios and confidence intervals, and multivariate analysis using Poisson regression. Results Insufficient physical activity was observed in 38.7% of the sample. Insufficient physical activity was associated with having <8 years of formal education (prevalence ratio-PR=1.58; 95% confidence interval-CI=1.04-2.39), total cholesterol ≥200 mg/dL (PR=0.78; 95% CI=0.58-1.04), low-density lipoprotein cholesterol-LDL ≥100 mg/dL (PR=0.79; 95% CI=0.53-0.95), and high-density lipoprotein cholesterol-HDL <40 mg/dL among men and <50 mg/dL among women (PR=1.21; 95% CI=0.90-1.64). Length of education <8 years (PR=1.50; 95% CI=0.99-2.29) and LDL ≥100 mg/dL (PR=0.72;95% CI=0.54-0.96) were also associated in the multivariate regression. Conclusion High prevalence of insufficient physical activity and its association with low education level and altered LDL serum levels were observed in hypertensive patients.

ABSTRACT

Light is an important signal for fungi in the environment and induces many genes with roles in stress and virulence responses. Conidia of the entomopathogenic fungi Aschersonia aleyrodis, Beauveria bassiana, Cordyceps fumosorosea, Lecanicillium aphanocladii, Metarhizium anisopliae, Metarhizium brunneum, Metarhizium robertsii, Simplicillium lanosoniveum, Tolypocladium cylindrosporum, and Tolypocladium inflatum were produced on potato dextrose agar (PDA) medium under continuous white light, on PDA medium in the dark, or under nutritional stress (= Czapek medium without sucrose = MM) in the dark. The conidial tolerance of these species produced under these different conditions were evaluated in relation to heat stress, oxidative stress (menadione), osmotic stress (KCl), UV radiation, and genotoxic stress caused by 4-nitroquinoline 1-oxide (4-NQO). Several fungal species demonstrated greater stress tolerance when conidia were produced under white light than in the dark; for instance white light induced higher tolerance of A. aleyrodis to KCl and 4-NQO; B. bassiana to KCl and 4-NQO; C. fumosorosea to UV radiation; M. anisopliae to heat and menadione; M. brunneum to menadione, KCl, UV radiation, and 4-NQO; M. robertsii to heat, menadione, KCl, and UV radiation; and T. cylindrosporum to menadione and KCl. However, conidia of L. aphanocladii, S. lanosoniveum, and T. inflatum produced under white light exhibited similar tolerance as conidia produced in the dark. When conidia were produced on MM, a much stronger stress tolerance was found for B. bassiana to menadione, KCl, UV radiation, and 4-NQO; C. fumosorosea to KCl and 4-NQO; Metarhizium species to heat, menadione, KCl, and UV radiation; T. cylindrosporum to menadione and UV radiation; and T. inflatum to heat and UV radiation. Again, conidia of L. aphanocladii and S. lanosoniveum produced on MM had similar tolerance to conidia produced on PDA medium in the dark. Therefore, white light is an important factor that induces higher stress tolerance in some insect-pathogenic fungi, but growth in nutritional stress always provides in conidia with stronger stress tolerance than conidia produced under white light.

ABSTRACT

BACKGROUND: Nut consumption has been related to improvements on cardiometabolic parameters and reduction in the severity of atherosclerosis mainly in primary cardiovascular prevention. The objective of this trial is to evaluate the effects of the Brazilian Cardioprotective Diet (DIeta CArdioprotetora Brasileira, DICA Br) based on consumption of inexpensive locally accessible foods supplemented or not with mixed nuts on cardiometabolic features in patients with previous myocardial infarction (MI). METHODS: DICA-NUTS study is a national, multicenter, randomized 16-week follow-up clinical trial. Patients over 40 years old with diagnosis of previous MI in the last 2 to 6 months will be recruited (n = 388). A standardized questionnaire will be applied to data collection and blood samples will be obtained. Patients will be allocated in two groups: Group 1: DICA Br supplemented with 30 g/day of mixed nuts (10 g of peanuts, 10 g of cashew, 10 g of Brazil nuts); and Group 2: only DICA Br. The primary outcome will consist of LDL cholesterol means (in mg/dL) after 16 weeks of intervention. Secondary outcomes will consist of other markers of lipid profile, glycemic profile, and anthropometric data. DISCUSSION: It is expected that DICA Br supplemented with mixed nuts have superior beneficial effects on cardiometabolic parameters in patients after a MI, when compared to DICA Br. ABSTRACT

This work is a Brazilian-Indian collaboration. It aims at investigating the structural properties of Lenticular galaxies in the Stripe 82 using a combination of S-PLUS (Southern Photometric Local Universe Survey) and SDSS data. S-PLUS is a novel optical multi-wavelength survey which will cover nearly 8000 square degrees of the Southern hemisphere in the next years and the first data release covers the Stripe 82 area. The morphological classification and study of the galaxies' stellar population will be performed combining the Bayesian Spectral type (from BPZ) and Morfometryka (MFMTK) parameters. BPZ and MFMTK are two complementary techniques, since the first one determines the most likely stellar population of a galaxy, in order to obtain its photometric redshift (phot-z), and the second one recovers non-parametric morphological quantities, such as asymmetries and concentration. The combination of the two methods allows us to explore the correlation between galaxies shapes (smooth, with spiral arms, etc.) and their stellar contents (old or young population). The preliminary results, presented in this work, show how this novel data set opens a new window on our understanding of the nearby universe.

ABSTRACT

Background: Diabetes mellitus (DM) is associated with a higher prevalence of metabolic syndrome (MS) and cardiovascular mortality. However, few Brazilian studies evaluated MS in diabetic individuals with cardiovascular disease (CVD). Nevertheless, the objective of this study was to compare the prevalence of MS in cardiac patients with and without DM. Methods: Cross-sectional study of BALANCE Program Trial with patients with CVD, ≥45 years old, who had been attending specialized ambulatories on cardiovascular health, in eight states in the Northeast of Brazil. The components of MS were evaluated by following the criteria of National Cholesterol Education Program's Adult Treatment Panel III (NCEP ATP III) and Joint Interim Statement (JIS). In addition, there were investigated different indexes of abdominal obesity, variables related to lifestyle, and food intake. The statistical analysis included descriptive statistics and the Student's t-test, Mann-Whitney, and chi-squared tests for the comparison of groups. There were significant values of P < 0.05. Results: Six hundred forty-seven individuals were evaluated with average (standard deviation) age of 63.1 (9.3) years, being 50.5% females and 40.3% diabetic patients. When the groups of patients who were diabetic and the nondiabetic ones, the first showed higher percentage of obesity (38.5% vs. 23.2%, P < 0.001), of high waist circumference (84.8% vs. 71.9%; P < 0.001), higher waist-height ratio [0.6 (0.6-0.7) vs. 0.6 (0.5-0.6); P < 0.001], conicity index [1.35 (1.29-1.39) vs. 1.32 (1.27-1.38); P = 0.004], and prevalence of MS, because of the criteria of NCEP ATP III (98.8% vs. 80.4%; P < 0.001), as well as the criteria of JIS (99.2% vs. 89.3%; P < 0.001). The component of higher frequency was high blood pressure and/or hypertension (95.0%), followed by hypertriglyceridemia (93.0%). There were no differences regarding age, lifestyle, and food intake. Conclusions: Diabetic individuals with CVD showed more prevalence of MS and more abdominal obesity than nondiabetic individuals. ClinicalTrials.gov ID: NCT01620398.

ABSTRACT

Aedes (Stegomyia) aegypti (Linnaeus, 1762) is a mosquito species of significant medical importance. The use of this vector in research studies usually requires a large number of mosquitoes as well as rearing and maintenance in a laboratory-controlled environment. However, laboratory conditions may be different from field environments, presenting stressful challenges such as low food concentration, especially during larval stages, which may, in turn, impair vector biology. Therefore, we tested herein if larval food availability (0.004, 0.009, 0.020, and 0.070% diets) would affect overall adult insect fitness. We observed slower development in mosquitoes fed a 0.004% diet 15 d post-eclosion (DPE) and shorter mean time in mosquitoes fed a 0.020% diet (7 DPE). Larval diet and adult mosquito weight were positively correlated, and heavier females fed higher larval diets exhibited greater blood feeding capacity and oviposition. In addition, larval diet concentrations led to median adult lifespan variations (male/female in days-0.004%: 30 ± 1.41, 45 ± 1.3; 0.009%: 31.5 ± 1.33, 41 ± 1.43; 0.020%: 26 ± 1.18, 41 ± 1.45; 0.070%: 29 ± 1.07, 44 ± 1.34), reduced tolerance to deltamethrin (1 mg/m2) and changes in detoxification enzyme activities. Moreover, in the larval 0.070% diet, females presented higher Zika susceptibility (plaque-forming unit [PFU]: 1.218 × 106) compared with other diets (0.004%: 1.31 × 105; 0.009%: 2.0 × 105; 0.020%: 1.25 × 105 PFU). Altogether, our study demonstrates that larval diet restriction results not only in larval developmental arrest but also in adult fitness impairment, which must be considered in future assessments.

BACKGROUND: Chemical mosquito control using malathion has been applied in Brazil since 1985. To obtain chemical control effectiveness, vector susceptibility insecticide monitoring is required. This study aimed to describe bioassay standardizations and determine the susceptibility profile of Ae. aegypti populations to malathion and pyriproxyfen, used on a national scale in Brazil between 2017 and 2018, and discuss the observed impacts in arbovirus control. METHODS: The diagnostic-doses (DD) of pyriproxyfen and malathion were determined as the double of adult emergence inhibition (EI) and lethal doses for 99% of the Rockefeller reference strain, respectively. To monitor natural populations, sampling was performed in 132 Brazilian cities, using egg traps. Colonies were raised in the laboratory for one or two generations (F1 or F2) and submitted to susceptibility tests, where larvae were exposed to the pyriproxyfen DD (0.03 µg/l) and adults, to the malathion DD determined in the present study (20 µg), in addition to the one established by the World Health Organization (WHO) DD (50 µg) in a bottle assay. Dose-response (DR) bioassays with pyriproxyfen were performed on populations that did not achieve 98% EI in the DD assays. RESULTS: Susceptibility alterations to pyriproxyfen were recorded in six (4.5%) Ae. aegypti populations from the states of Bahia and Ceará, with Resistance Ratios (RR95) ranging from 1.51 to 3.58. Concerning malathion, 73 (55.3%) populations distributed throughout the country were resistant when exposed to the local DD 20 µg/bottle. On the other hand, no population was resistant, and only 10 (7.6%) populations in eight states were considered as exhibiting decreased susceptibility (mortality ratios between 90 and 98%) when exposed to the WHO DD (50 µg/bottle). CONCLUSIONS: The feasibility of conducting an insecticide resistance monitoring action on a nation-wide scale was confirmed herein, employing standardized and strongly coordinated sampling methods and laboratory bioassays. Brazilian Ae. aegypti populations exhibiting decreased susceptibility to pyriproxyfen were identified. The local DD for malathion was more sensitive than the WHO DD for early decreased susceptibility detection.

BACKGROUND: Aedes aegypti is a vector of high relevance, since it transmits several arboviruses, including dengue, chikungunya and Zika. Studies on vector biology are usually conducted with laboratory strains presenting a divergent genetic composition from field populations. This may impair vector control policies that were based on laboratory observations employing only long maintained laboratory strains. In the present study we characterized a laboratory strain interbreed with Ae. aegypti collected from five different localities in Rio de Janeiro (Aedes Rio), for insecticide resistance (IR), IR mechanisms, fitness and Zika virus infection. METHODS: We compared the recently established Aedes Rio with the laboratory reference strain Rockefeller. Insecticide resistance (deltamethrin, malathion and temephos), activity of metabolic resistance enzymes and kdr mutation frequency were determined. Some life table parameters (longevity, blood-feeding, number and egg viability) and Zika virus susceptibility was also determined. RESULTS: Aedes Rio showed resistance to deltamethrin (resistance ratio, RR50 = 32.6) and temephos (RR50 = 7.0) and elevated activity of glutathione S-transferase (GST) and esterases (α-EST and pNPA-EST), but not acetylcholinesterase (AChE). In total, 92.1% of males genotyped for kdr presented a "resistant" genotype. Weekly blood-fed females from both strains, presented reduced mortality compared to sucrose-fed mosquitoes; however, Aedes Rio blood-fed females did not live as long (mean lifespan: Rockefeller = 70 ± 3.07; Aedes Rio = 53.5 ± 2.16 days). There were no differences between strains in relation to blood-feeding and number of eggs, but Aedes Rio eggs presented reduced viability (mean hatch: Rockefeller = 77.79 ± 1.4%; Aedes Rio = 58.57 ± 1.77%). Zika virus infection (plaque-forming unit, PFU) was similar in both strains (mean PFU ± SE: Aedes Rio: 4.53 × 104 ± 1.14 × 104 PFU; Rockefeller: 2.02 × 104 ± 0.71 × 104 PFU). CONCLUSION: Selected conditions in the field, such as IR mechanisms, may result in pleiotropic effects that interfere in general physiology of the insect. Therefore, it is important to well characterize field populations to be tested in parallel with laboratory reference strains. This practice would improve the significance of laboratory tests for vector control methods.

ABSTRACT

Fungi sense light and utilize it as a source of environmental information to prepare against many stressful conditions in nature. In this study, Metarhizium robertsii was grown on: 1) potato dextrose agar medium (PDA) in the dark (control); 2) under nutritive stress in the dark; and 3) PDA under continuous (A) white light; (B) blue light lower irradiance = LI; (C) blue light higher irradiance = HI; (D) green light; and (E) red light. Conidia produced under these treatments were tested against osmotic stress and UV radiation. In addition, a suite of genes usually involved in different stress responses were selected to study their expression patterns. Conidia produced under nutritive stress in the dark were the most tolerant to both osmotic stress and UV radiation, and the majority of their stress- and virulence-related genes were up-regulated. For osmotic stress tolerance, conidia produced under white, blue LI, and blue HI lights were the second most tolerant, followed by conidia produced under green light. Conidia produced under red light were the least tolerant to osmotic stress and less tolerant than conidia produced on PDA medium in the dark. For UV tolerance, conidia produced under blue light LI were the second most tolerant to UV radiation, followed by the UV tolerances of conidia produced under white light. Conidia produced under blue HI, green, and red lights were the least UV tolerant and less tolerant than conidia produced in the dark. The superoxide dismutases (sod1 and sod2), photolyases (6-4phr and CPDphr), trehalose-phosphate synthase (tps), and protease (pr1) genes were highly up-regulated under white light condition, suggesting a potential role of these proteins in stress protection as well as virulence after fungal exposure to visible spectrum components.
