ABSTRACT

A recent study reported the oncogenic function of lncRNA NR2F1-AS1 in liver cancer. Interestingly, by analyzing TCGA data set, downregulation of NR2F1-AS1 in colorectal cancer (CRC) was observed. This observation triggered interest to analyze the functions of NR2F1-AS1 in CRC. It was observed that NR2F1-AS1 was downregulated in CRC and predicted poor survival. NR2F1-AS1 can directly interact with miR-371a-3p but their overexpression failed to affect the expression of each other. However, NR2F1-AS1 overexpression led to the upregulation of TOB1, a target of miR-371a-3p. Cell proliferation analysis revealed reduced proliferation rate of CRC cells after NR2F1-AS1 and TOB1 overexpression. MiR-371a-3p overexpression played an opposite role and reduced the effects of NR2F1-AS1 and TOB1 overexpression. In conclusion, NR2F1-AS1 regulates miR-371a-3p/TOB1 axis to suppress proliferation of CRC cells.

ABSTRACT

PURPOSE: Recently, dysregulated circular RNAs (circRNAs) have been associated with the progression of numerous malignant tumors. However, the mechanism through which circRNAs participate in breast cancer (BC) remains unclear. This study was designed to illustrate the role of hsa_circ_0068033 in BC. METHODS: We detected the expression levels of hsa_circ_0068033 in BC tissues using quantitative real-time PCR (qRT-PCR) and in situ hybridization (ISH). A series of functional experiments were conducted to assess the function of hsa_circ_0068033 in BC development and the underlying mechanisms. RESULTS: The results suggested that the expression of hsa_circ_0068033 was downregulated in BC tissues, and its expression was markedly correlated with tumor size (P=0.021), and the Tumor, Node, and Metastasis stage (P=0.023). Receiver operating characteristic analysis showed that hsa_circ_0068033 testing yielded an area under the curve value of 0.8480 in discriminating BC from non-tumor controls. Functionally, in-vitro experiments demonstrated that overexpression of hsa_circ_0068033 could inhibit the growths, clone formation, invasion and migration of MCF-7 and MDA-MB-231 cells while activating the intrinsic apoptotic pathway to induce apoptosis. The xenograft experiment revealed that exogenous hsa_circ_0068033 is able to evidently reduce the tumorigenic ability of MDA-MB-231 cells in nude mice. Rescue assays further proved that hsa_circ_0068033 exerts biological functions by sponging miR-659. CONCLUSION: Collectively, this study revealed for the first time that hsa_circ_0068033 acts as a tumor suppressor gene in BC, and the hsa_circ_0068033/miR-659 axis participates in the progression of BC.

ABSTRACT

Long noncoding RNAs (lncRNAs) have emerged as important regulators of human cancers. LncRNA GAS5 (GAS5) is identified as a tumor suppressor involved in several cancers. However, the roles of GAS5 and the mechanisms responsible for its functions in gastric cancer (GC) have not been well documented. Herein, the decreased GAS5 and increased miRNA-106a-5p levels were observed in GC and cell lines. GAS5 level was significantly inversely correlated with miRNA-106a-5p level in GC tissues. Moreover, dual-luciferase reporter and qRT-PCR assays showed that GAS5 bound to miRNA-106a-5p and negatively regulated its expression in GC cells. Functional experiments showed that GAS5 overexpression suppressed GC cell proliferation, migration and invasion capabilities, and promoted apoptosis, while miRNA-106a-5p overexpression inverted the functional effects induced by GAS5 overexpression. In vivo, GAS5 overexpression inhibited tumor growth by negatively regulating miRNA-106a-5p expression. Mechanistic investigations revealed that GAS5 overexpression inactivated the Akt/mTOR pathway by suppressing miRNA-106a-5p expression in vitro and in vivo Taken together, our findings conclude the GAS5 overexpression suppresses tumorigenesis and development of gastric cancer by sponging miR-106a-5p through the Akt/mTOR pathway.

ABSTRACT

Coal fires are a serious threat to the workers' security and safe production in open pit mines. The coal fire source is hidden and innumerable, and the large-area cavity is prevalent in the coal seam after the coal burned, causing the conventional extinguishment technology difficult to work. Foams are considered as an efficient means of fire extinguishment in these large-scale workplaces. A noble foam preparation method is introduced, and an original design of cavitation jet device is proposed to add foaming agent stably. The jet cavitation occurs when the water flow rate and pressure ratio reach specified values. Through self-building foaming system, the high performance foams are produced and then infused into the blast drilling holes at a large flow. Without complicated operation, this system is found to be very suitable for extinguishing large-scale coal fires. Field application shows that foam generation adopting the proposed key technology makes a good fire extinguishment effect. The temperature reduction using foams is 6-7 times higher than water, and CO concentration is reduced from 9.43 to 0.092‰ in the drilling hole. The coal fires are controlled successfully in open pit mines, ensuring the normal production as well as the security of personnel and equipment.
