ABSTRACT

@#For the quality control of cetomacrogol 1000, a gas chromatographic method for the determination of residual impurities in cetomacrogol 1000, such as ethylene oxide, 1, 4-dioxane, ethylene glycol, diethylene glycol and triethylene glycol, was established and validated.The DB-1 column with headspace injection was used to detect ethylene oxide and 1, 4-dioxane with the inlet temperature of 150 °C, the FID temperature of 250 °C, the headspace equilibration temperature of 70 °C and the equilibration time of 45 min.The VF-17MS column with liquid injection was used to detect ethylene glycol, diethylene glycol and triethylene glycol with the inlet temperature of 270 °C, and the FID temperature of 290 °C.The results showed that ethylene oxide and 1,4-dioxane have a good linearity within their specified addition amount ranges (r > 0.999), with the RSD of precision of below 8.0% and the average recovery rates of 90.6% and 101.2%; and that ethylene glycol, diethylene glycol and triethylene glycol also have a good linearity between 3 ? 60 μg/mL (r > 0.999), with the RSD of precision of below 3.0%, and the recovery rates of 96% ~ 103%.The method established in this study has good specificity, linearity, precision and recovery rate, which can effectively detect the multi-component and trace impurities.

ABSTRACT

Objective:To explore the biological functions of differentially expressed miRNAs and their target genes, and their regulatory relations in growth hormone (GH)-secreting pituitary adenoma.Methods:Four databases (miRDB, miRwalk, Targetscan7.2 and Starbase) were used to predict the target genes of differentially expressed miRNAs in GH-secreting pituitary adenoma, and GO, KEGG and protein-protein interaction network (PPI) analyses were performed on the target genes. Meaningful hub target genes were screened out and compared with mRNA sequencing results from the database; and then, a visual miRNA-target gene interaction network was constructed.Results:Totally, 113 up-regulated miRNAs target genes and 128 down-regulated miRNAs target genes were obtained with the protein interaction degree≥20. Further intersection was conducted to obtain 13 hub target genes; these genes were mainly involved in ubiquitin mediated proteolysis pathway. The regulatory miRNAs of the hub target genes were miR-15b, miR-365, miR-32-3p, and miR-486-5p.Conclusion:A miRNAs-hub target gene mutual regulation network closely related to GH-secreting pituitary adenoma is constructed with bioinformatics analysis tools, and some potential proteins and pathways related to the pathogenesis and pathological process of GH-secreting pituitary adenoma are discovered.

ABSTRACT

@#The gas chromatography method was developed for the determination of ethylene glycol, diethylene glycol and triethylene glycol in poloxamer 188 to provide scientific basis for the quality control. The samples was separated on column VF-17ms(30 m×0. 53 mm, 1. 0 μm)with temperature programming, inlet temperature was 270 °C, detector temperature was 290 °C and the split ratio was 10 ∶1. The method showed great linearity over the range of 6-15 μg/mL(r≥0. 999). The injection precision(n=8)of the three residual impurities were 3. 3%, 3. 0%, 2. 3% and the average recoveries were 99. 05%(RSD=2. 9%, n=9), 102. 20%(RSD=4. 0%, n=9), 101. 91%(RSD=3. 1%, n=9), respectively. The analytical method is specific, accurate and sensitive, which is suitable for the determination of ethylene glycol, diethylene glycol and triethylene glycol in poloxamer 188, providing reference and guidance for the production and quality control of poloxamer 188.

ABSTRACT

Objective To compare the effect and safety of arthroscopic debridement enmbined with intercondylar fossa angioplasty in the elder with knee osteoarthritis.Methods Six-eight elderly with knee osteoarthritis were selected from June 2012 to December 2014 in this hospital and they were averagely and randomly divided into group A and B.Patients in group A were given arthroscopic debridement,while in group B were given and arthroscopic debridement,intercondylar fossa angioplasty.The levels of VAS scores,Lysholm scores,WOMAC scores and joint range of motion were measured and compared between the two groups after 3 months,6 months and 12 months of operation.The adverse events of the two groups were supervised and compared as well.Results All elder in the two groups had apparently decreasing in VAS scores and WOMAC scores (P<0.05) after operation.Compared with group A,patients in group B had lower levels of VAS scores and WOMAC scores(P<0.05) at 12 months after operation.The level of Lysholm score was significantly increased in the two groups after treatment(P<0.05),while group B had a higher level at 12 months after treatment than group A (P<0.05).The joint range of motion in the two groups were improved after treatment(P<0.05).The elder patients in group B had better joint range of motion than group A at 6 and 12 months after operation (P<0.05).Early postoperative swelling in the joints was the adverse event after operation,but there was no statistically difference between two groups.Conclusion The combination of arthroscopic debridement and intercondylar fossa angioplasty is effective to release knee pain,increase the clinical efficacy and improve the knee function.

ABSTRACT

Among various therapeutic approaches for stroke, treatment with human umbilical cord mesenchymal stem cells (hUC-MSCs) has acquired some promising results. However, the underlying mechanisms remain unclear. We analyzed the protein expression spectrum of the cortical peri-infarction region after ischemic stroke followed by treatment with hUC-MSCs, and found 16 proteins expressed differentially between groups treated with or without hUC-MSCs. These proteins were further determined by Gene Ontology term analysis and network with CD200-CD200R1, CCL21-CXCR3 and transcription factors. Three of them: Abca13, Grb2 and Ptgds were verified by qPCR and ELISA. We found the protein level of Abca13 and the mRNA level of Grb2 consistent with results from the proteomic analysis. Finally, the function of these proteins was described and the potential proteins that deserve to be further studied was also highlighted. Our data may provide possible underlying mechanisms for the treatment of stroke using hUC-MSCs.

ABSTRACT

Objective To observe the hemostatic efficacy of absorbable Surgiflo haemosatic matrix (trade name:Surgiflo) in endoscopic transsphenoidal pituitary adenomectomy.Methods A total of 40 patients performed endoscopic transsphenoidal pituitary adenomectomy in our hospital from January 2016 to June 2016,were recruited in our study;16 had tumor cavity bleeding and 24 had cavernous sinus/intercavernous sinus bleeding;10 patients had Grade 2 bleeding,21 Grade 3 bleeding,9 Grade 4 bleeding and none had Grade 5 bleeding.Surgiflo was used during the surgery.The clinical data and hemostatic efficacy of these patients were retrospectively analyzed.Results In 40 patients,36 patients achieved haemostasis only by Surgiflo and 4 by Surgiflo with electrocoaulation,bone wax,and hemostatic gauze.Patients with Grade 2,3 and 4 bleeding had 1,1.5 and 2.5 times of spray with Surgiflo.Postoperative CT revealed no bleeding.No advance reactions were recorded after one month of follow-up.Conclusion Surgiflo enjoys good,fast and safe haemostasis efficacy in endoscopic transsphenoidal pituitary adenomectomy,which is appropriate to tumor cavity bleeding,cavernous sinus/ intercavemous sinus bleeding in transsphenoidal pituitary adenomectomy.

ABSTRACT

Objective To discuss the preoperative diagnosis and the effect of microsurgical transsphenoidal surgery of pituitary Rathke's cleft cysts.Methods Retrospective analysis was performed from January,2011 to May,2015 on 62 cases of Rathke's cleft cyst which confirmed by surgery and pathology at the First Affiliated Hospital of Sun Yat-sen University.Sixty-two cases were performed by the surgery of transsphenoidal approach.Results There were 50 cases with a correct preoperative diagnosis of and consider Rathke's cleft cyst,12 cases of misdiagnosis.Postoperative follow-up was performed within 6-12 months,and the patient's clinical symptons were improved in different degrees,1 case of recurrence,no deaths and serious complications.Conclusion The diagnosis of Rathke's cleft cyst need for comprehensive considerations from many aspects,and the improvement of diagnosis rate base on the clinical features,endocrine examination,and imaging data.The microsurgical transsphenoidal surgery is safe and effective treatment for Rathke's cleft cyst.

ABSTRACT

In this study, a highly pathogenic porcine reproductive and respiratory syndrome virus (HP-PRRSV) strain, PRRSV GD07 was continuously propagated in MARC-145 cell cultures primed with swIFN-ß for 50 passages to develop the PRRSV GDßfn strains. And a control strain PRRSV GDfn was passaged without swIFN-ß. The sequencing analysis indicated that under swIFN-ß immune pressure, molecular variation of PRRSV GP5 was accelerated in gene (NS/S>2.50), and the acceleration of GP3 was not significant (NS/S<2.50). swIFN-ß mRNA level induced by Poly(I:C) is lower in cells primed with PRRSV GDßfn than in cells without PRRSV GDfn, although both of them are much less than the control group. Effect of GP5 on IRF3 was analyzed by SDS-PAGE and western-blot. Our results indicated that GP5 protein prevents IRF3 phosphorylation. Therefore, we conclude that swIFN can promote viral mutation in GP5, and, in turn GP5 inhibits IRF3 activation to escape from swIFN-ß.

Objective To study the criteria and technique of transsphenoidal microsurgery on pituitary adenoma invaded cavernous sinus Methods 42 patients with pituitary adenoma underwent 45 transsphenoidal operations Sellar base was opened as large as possible under C arm X ray fluoroscopy or neuronavigation monition Dura matter was cut open under microscope Tumors were removed through the direction and tunnel they in invade Bromocriptine was administered to those with little remains Partial remains and those have no response to bromocriptine were given radiotherapy Results No modality 21 tumors were nearly totally removed, 18 tumors were subtotally resected, 3 were partially resected MRI scanned in 2~3 months after the operation showed that 19 tumors disappeared, 20 tumors had little residul, 3 tumors partially remained Conclusion Pituitary adenomas invading the cavernous sinus with enlarged sella turcica and shown soft in MRI can be submitted to transsphenoidal operation Microsurgical technique can ease tumor removal and protection of the sellar and internal carotid artery
