ABSTRACT

The breeding of remontant rose cultivars that are resistant to diseases and adverse conditions, with high decorative value and continuous flowering is the most important task during work with the gene pool of garden roses. Currently, intercultivar hybridization within a single garden group has largely outlived its usefulness. It is necessary to breed for highly decorative forms or cultivars that have outstanding resistance, morphological characters and patterns of seasonal rhythms, and use these plants as parental forms in further breeding. This study represents a comparative analysis of rose cultivars from two garden groups, Grandiflora (Gurzuf, Lezginka, Korallovy Syurpriz, Queen Elizabeth, Komsomolsky Ogonyok, Love) and Rosa Kordesii (Letniye Zvyozdy, Dortmund, Gutsulochka). These cultivars proved themselves during many years of testing in harsh climatic conditions. The objectives of the study were to determine the genetic relationship within the groups and to assign phenotypically different cultivars to one or another garden group. The analysis was carried out by morphological, phenological and ISSR markers. According to the phenological observations on the Grandiflora cultivars, Komsomolsky Ogonyok had later budding and flowering stages. Polymorphic data generated from the ISSR markers showed that this cultivar was the most distant from the others and formed a separate cluster on the dendrogram. A comparison of the morphological characters (flower diameter, number of petals, peduncle length, bush height) showed a significant difference ( p < 0.05) between Komsomolsky Ogonyok and the other Grandiflora cultivars. A dendrogram based on a molecular analysis showed a lack of close relationships between Komsomolsky Ogonyok and the Kordesii group, which formed a separate cluster. A pairwise comparison of the morphological characters in Komsomolsky Ogonyok with the Kordesii group revealed a significant ( p <0.05) difference in three of the four characters studied. The exceptions were flower diameter when comparing with Dortmund and Letniye Zvyozdy and peduncle length when comparing with Gutsulochka. Although Komsomolsky Ogonyok has a pattern of seasonal development similar to Dortmund in the Kordesii group, the molecular analysis did not assign the former to this group of roses. The cultivars that have valuable characters that no average rose does and that are phenotypically different from such roses represent the most valuable breeding material.

ABSTRACT

A hybridological study of biotypes of species close to Elymus caninus: E. prokudinii, E. viridiglumis, E. goloskokovii, as well as a number of morphologically deviant biotypes in Russia and Kazakhstan, was carried out. The objectives were to study the levels of reproductive relationships and the degree of integration of the species E. goloskokovii, E. prokudinii, and E. viridiglumis into the E. caninus complex. Our estimates of the seed fertility of natural parental biotypes were within 60-90 %. Among the combinations of crossing in F1, the highest seed setting was found in the hybrids formed by parental pairs from close habitats, regardless of the taxonomic rank of biotypes. The highest fertility values (55.6 and 46.1 %) were found in combinations involving E. caninus, E. viridiglumis and E. goloskokovii. It has been concluded that the biotypes of these species included in sexual hybridization form a single recombination gene pool, within which slight differences in reproductive compatibility are observed. The nature of the inheritance of the diagnostic features of lemmas "presence of trichomes" and "length of awns", according to the digenic and monogenic type, respectively, is shown. The high seed fertility of the created hybrids and the presence of intermediate forms in the F2 generation according to distinctive features indicate the possibility of interspecific introgression when species grow together in natural populations. Thus, the assessment of the inheritance of diagnostic characters makes it possible to classify E. goloskokovii, E. prokudinii, and E. viridiglumis as intraspecific taxa of E. caninus s. l. Data were obtained on the morphological and reproductive properties of interspecific hybrids with the participation of the species E. mutabilis as a possible donor in the speciation of taxa close to E. caninus. In cross combinations of E. caninus × E. mutabilis and E. mutabilis × E. caninus, lower values of seed fertility of hybrids in the F1 and F2 generations were noted compared to hybrids between the species E. caninus, E. goloskokovii, E. prokudinii and E. viridiglumis. Nevertheless, on the basis of chorological and morphological criteria, we concluded that E. caninus and E. mutabilis are independent species.

ABSTRACT

The generic complex Miscanthus Anderss. (Poaceae) is a unique example among herbaceous plants characterized by high values of growth of aboveground vegetative mass and practical use as a valuable source of alternative energy. Miscanthus is one of the most eff icient solar energy accumulators, and since phytomeliorative use implies the cultivation of these resource plants in inconvenient and semi-shady areas, the question about the effect of insuff icient lighting on the productivity of Miscanthus arises. As a result of a long-lasing introduction effort, the Central Siberian Botanical Garden SB RAS created a population of Miscanthus saccharif lorus (Maxim.) Benth., which has good prospects for growing under the conditions of the forest-steppe area in Western Siberia. The goals of our study were: (1) to determine the peculiarities of shoot formation, (2) to assess the cellulose and lignin accumulation in M. saccharif lorus populations under different lighting conditions and (3) to perform a DNA passportization of the Miscanthus population by ISSR marking. Evaluation of shoot formation and the amount of accumulated cellulose and lignin in plants was carried out under different degrees of illumination: one variant was grown in a sunny area, and the other, in partial shade. As a result of analysis of variance, it was found that the number of shoots does not depend on environmental conditions, but on the age of the plant, while environmental conditions have a signif icant effect on plant height. Although the samples of both M. saccharif lorus variants were characterized by different rates of creation of a continuous projective cover, plants in semi-shaded areas formed up to 89.34 % of shoots compared to their peers in illuminated areas, which did not affect signif icantly the size of the aboveground mass and the cellulose content in it. As a result of ISSR-analysis of genomic DNA in the M. saccharif lorus population, unique molecular polymorphic fragments were identif ied, which can be used for identif ication and DNA passportization at the inter-population level. Thus, the complex use of M. saccharif lorus as a valuable meliorative and bioenergetic culture is due to the high adaptive potential of this species. It was found that the illumination factor has virtually no effect on the amount of the cellulose content in the shoot, and a reduced content of the technologically undesirable lignin was observed in plants growing in the partial shade conditions.

ABSTRACT

In the present study, a protocol was developed for the in vitro propagation of a rare medicinal plant, Hedysarum theinum (tea sweetvetch), from axillary buds, and identification of the regenerants was performed with the use of ISSR markers. It was demonstrated that Gamborg and Eveleigh medium supplemented with 5 µM 6-benzylaminopurine was the best for H. theinum for initial multiplication. On the other hand, half-strength Murashige and Skoog (MS) basal medium supplemented with 7 µM α-naphthaleneacetic acid proved to be the best for explant rooting. Molecular genetic analysis of the H. theinum mother plants and the obtained regenerants was performed with six ISSR markers. Depending on the primer, four to ten amplified fragments with sizes ranging from 250 to 3000 bp were identified. Our results confirmed the genetic stability of regenerants obtained in five passages and their identity to the mother plant.

ABSTRACT

The objective of the research described in this paper was to evaluate the level of genetic variability and differentiation between the populations of Hedysarum theinum Krasnob., endemic species of Altai-Sayan Mountains. In this study, six inter-simple sequence repeat (ISSR) primers were used and generated a total of 132 molecular markers, 126 of which were polymorphic (95.5%). The large amount ofDNA polymorphism at the intrapopulation level (H(pop)/H(sP) = 70.5) was evinced by the analysis of a molecular variance (AMOVA) of 88.2%. The high genetic similarity (I = 0.875) detected between the populations of H. theinum reflects the strongly restricted endemic range of H. theinum, which facilitates the gene flow among populations and out-crossing. Due to the significant genetic diversity revealed among individuals against a background of moderate population differentiation collecting samples for ex situ H. theinum conservation from a small number of populations is acceptable. Finally, selected highly polymorphic ISSR markers produced consistently repeatable patterns are discussed as a powerful tool for genotype identification and the qualification of root feedstock.

ABSTRACT

Cloning of the Arabidopsis thaliana genomic DNA fragment presumably corresponding to the promoter region of the ornithine-delta-aminotransferase (OAT) gene is reported. The reporter-gene construct, containing the Escherichia coli beta-glucouronidase gene under control of the OAT gene promoter was generated. The Nicotian tabacum SR1 transformants carrying this construct were obtained. It was demonstrated that in normal conditions, expression of the reporter gene was associated with the meristems and the zones of intensive shoot growth. Possible role of the OAT gene in nitrogen metabolism and shoot development is discussed.

Legumes are one of the cheapest protein sources. But most of them contains inhibitors tripsin and needs in long processing. Micronization and cooking of legumes only partly destructs the protease inhibitors. The using this products as fast processing food is unwished.
