ABSTRACT

The combined region of eastern Tennessee and western North Carolina has a persistently high risk of pediatric La Crosse virus neuroinvasive disease (LACV-ND). To guide public health intervention in this region, the objectives of this retrospective ecological study were to investigate the geographic clustering and predictors of pediatric LACV-ND risk at the ZIP code tabulation area (ZCTA) level. Data on pediatric cases of LACV-ND reported between 2003 and 2020 were obtained from Tennessee Department of Health and North Carolina Department of Health and Human Services. Purely spatial and space-time scan statistics were used to identify ZCTA-level clusters of confirmed and probable pediatric LACV-ND cases from 2003-2020, and a combination of global and local (i.e., geographically weighted) negative binomial regression models were used to investigate potential predictors of disease risk from 2015-2020. The cluster investigation revealed spatially persistent high-risk and low-risk clusters of LACV-ND, with most cases consistently reported from a few high-risk clusters throughout the entire study period. Temperature and precipitation had positive but antagonistic associations with disease risk from 2015-2020, but the strength of those relationships varied substantially across the study area. Because LACV-ND risk clustering in this region is focally persistent, retroactive case surveillance can be used to guide the implementation of targeted public health intervention to reduce the disease burden in high-risk areas. Additional research on the role of climate in LACV transmission is warranted to support the development of predictive transmission models to guide proactive public health interventions.

ABSTRACT

BACKGROUND: Aedes albopictus is a nuisance pest mosquito of public health importance commonly managed with adulticides and larvicides. We investigated whether adding Gravid Aedes Traps (GATs), Autocidal Gravid Ovitraps (AGOs) or In2Care traps would extend the effectiveness of chemical control methods in Wake County, North Carolina, USA, by combining barrier sprays and larval habitat management (LHM) with each trap type at suburban households. We compared these three treatment groups to untreated controls and to backyards treated only with barrier sprays and LHM. Once a week, for ten weeks, we collected adult mosquitoes at each house using lure-baited surveillance traps and dissected a portion of Ae. albopictus females to determine parity. RESULTS: Barrier sprays and LHM alone or combined with any supplemental autocidal ovitrap significantly reduced female Ae. albopictus through Week 3 post-treatment. GATs significantly extended chemical control effectiveness for the duration of the study. Compared to the untreated control, the AGO and GAT treatment groups had significant overall female Ae. albopictus reductions of 74% and 80.4%, respectively, with populations aging significantly slower at houses treated with AGOs. CONCLUSION: This household-level study, though limited in size, observed significant reductions in nuisance Ae. albopictus when combining AGOs and GATs with chemical controls for an eight-week period. Delayed population aging in AGO-treated yards suggests that traps also could mitigate disease transmission risk. Future studies should test these control methods at the neighborhood level to evaluate large-scale effectiveness as well as assess the effect of autocidal ovitraps without chemical intervention. © 2022 The Authors. ABSTRACT

In 1920, Culex coronator was reported from San Benito, Texas, and later in Arizona, New Mexico, and Oklahoma. In 2005, this species was reported to be spreading across the southeastern USA. Now reported in 14 states, it has been found as far north as northern Oklahoma; Memphis, TN; and Suffolk, VA. The public health significance of Cx. coronator is not firmly established, even though it has been implicated as a potential vector of several arboviral diseases. This study aims to document additional Cx. coronator county-level records, to provide information about its continued expansion across the southern USA, and to provide a short research update into its vector potential. Data acquired through multistate collaborations and author collections resulted in 146 new county records from Alabama, Arkansas, Florida, Georgia, Louisiana, Mississippi, North Carolina, Oklahoma, South Carolina, and Texas. No new county records were presented for Arizona, New Mexico, Tennessee, or Virginia, which had previously reported this species. With these new data, this species has been documented in 386 counties in 14 states of the continental USA.

ABSTRACT

Container Aedes mosquitoes are responsible for the transmission of anthroponotic and zoonotic viruses to people. The surveillance and control of these mosquitoes is an important part of public health protection and prevention of mosquito-borne disease. In this study, we surveyed 327 sites over 2 weeks in late June and early July in 2017 in North Carolina, USA for the presence and abundance of Aedes spp. eggs in an effort to better target potential Ae. aegypti collections. We examined the ability of 2 types of landscape data, Light Detection And Ranging (LIDAR) and National Land Cover Database (NLCD) to explain the presence and abundance of eggs using principal component analysis to deal with collinearity, followed by generalized linear regression. We explained variation of both egg presence and abundance for Aedes albopictus (Skuse) and Aedes triseriatus (Say) using both NLCD and LIDAR data. However, the ability to make robust predictions was limited by variation in the data. Increased sampling time and better landscape data would likely improve the predictive ability of our models, as would a better understanding of oviposition behavior.

ABSTRACT

There is a clear need for improved vector surveillance approaches that are affordable, labor efficient, and safer than traditional methods. The BG-Counter (Biogents USA, Moorefield, WV) is a device for remotely monitoring mosquito activity in combination with the BG-Sentinel (Biogents USA), a widely used trap for the collection of host-seeking mosquitoes. The BG-Counter uses a wireless connection to provide real-time counts of mosquitoes captured by the BG-Sentinel, allowing users to remotely monitor mosquito populations. This study tested the effectiveness of the BG-Counter in 5 North Carolina counties. A total of 96 trap-days resulted in the collection of >45,000 individual mosquitoes representing 35 species. Aedes albopictus was the most common species collected in all counties, except for New Hanover County where Culex nigripalpus was the most common. The mean daily accuracy ranged from 80.1% (New Hanover County) to 9.4% (Jackson County). There was a significant linear relationship between the actual number of mosquitoes collected and the device counts at all sites except Jackson County, the site with the lowest relative mosquito abundance compared with nontarget organisms. A linear regression of daily BG-Counter accuracy and the daily proportion of mosquitoes to the total number of arthropods collected revealed a significant positive linear relationship, supporting the premise that the BG-Counter is less effective when the relative abundance of mosquitoes is low. Mosquito surveillance programs using the BG-Counter should recognize its context-dependent accuracy and routinely evaluate the accuracy of the device based on local conditions.

ABSTRACT

Mosquitoes may develop resistance to insecticide active ingredients (AIs) found in formulated products (FPs) due to environmental exposure from insecticides in mosquito control and/or unrelated to mosquito control, e.g., agricultural, household pest control. Mosquito control programs should implement resistance management strategies by assessing resistance in targeted populations, rotating different classes of insecticides based on resistance testing, and/or increasing insecticide concentration (i.e., saturation, using maximum labeled rate) to overcome emerging resistance. Resistance testing is often done solely on AIs, but should, in some cases, include both AIs and FPs at the concentrations mosquitoes may encounter in the field. The resistance/susceptibility status was determined for adulticides used in mosquito control. Centers for Disease Control and Prevention (CDC) bottle bioassays were used to assess resistance/susceptibility status for eight AIs (i.e., bifenthrin, permethrin, sumethrin/prallethrin, deltamethrin, tau-fluvalinate, chlorpyrifos, malathion, and naled) and eight FPs (TalStar, Biomist 3 + 15, Duet, Suspend Polyzone, Mavrik, MosquitoMist, Fyfanon, and Dibrom) that respectively contain the AIs. Current CDC guidelines were utilized: susceptible (97-100% mortality at diagnostic time [DT]), developing resistance (90-96% mortality at DT), or resistant (<90% mortality at DT). Significant differences were observed in mosquito susceptibility/resistance among and between AIs and FPs.

ABSTRACT

After a dengue outbreak in Key West, Florida, during 2009-2010, authorities, considered conducting the first US release of male Aedes aegypti mosquitoes genetically modified to prevent reproduction. Despite outreach and media attention, only half of the community was aware of the proposal; half of those were supportive. Novel public health strategies require community engagement.

BACKGROUND: In 2009-2010, 93 cases of dengue were identified in Key West, Florida. This was the first outbreak of autochthonous transmission of dengue in Florida since 1934. In response to this outbreak, a multifaceted public education outreach campaign was launched. The aim of this study is to compare dengue prevention knowledge, attitudes, perceptions, and prevention practices among residents of subsidized public housing to the general population in Key West and to assess whether there were barriers preventing effective outreach from reaching specific vulnerable populations. METHODS: A randomized population-based evaluation of knowledge, attitudes, and behaviors toward dengue prevention consisting of 521 separate household interviews was undertaken in July of 2011. A subset analysis was performed on interviews collected from 28 public housing units within four subsidized public housing complexes. Analysis was performed to determine whether knowledge, attitudes, and behaviors exhibited by public housing residents differed from the non-public housing study population. RESULTS: Public housing residents recalled fewer outreach materials (p=0.01) and were 3.4 times (95% confidence interval [CI] 1.4-8.3) more likely not to recall any outreach materials. Public housing residents were less likely to correctly identify how dengue transmission occurs (61% vs. 89%), where mosquitoes lay their eggs (54% vs. 85%), or to identify any signs or symptoms related to dengue (36% vs. 64%). Public housing residents were less likely to perform dengue prevention practices such as removing standing water or always using air conditioning. CONCLUSIONS: Examination of public housing residents identified an at-risk population that recalled less exposure to outreach materials and had less knowledge about dengue infection and prevention than the randomized study population. This provides public health systems the opportunity to target or modify future health messages and interventions to this group. Differences identified in the demographics of this population suggest that alternative methods or non-English materials may be required to reach desired outcomes.

ABSTRACT

Mosquitoes such as those in the Culex pipiens complex are important vectors of disease. This study was conducted to genetically characterize Cx. pipiens complex populations in the state of Colorado, USA, and to determine the number of genetic clusters represented by the data. Thirteen populations located among four major river basins were sampled (n = 597 individuals) using a panel of 14 microsatellites. The lowest-elevation sites had the highest Expected Heterozygosity (H(E)) values (range 0.54-0.65). AMOVA results indicated the presence of statistically significant amounts of variation within each level when populations were analyzed as one group or when they were grouped either by river basin or by their position on the east or west side of the Rocky Mountains. Most pairwise F(ST) values were significant via permutation test (range 0-0.10), with the highest values from comparisons with Lamar, in southeast CO. A neighbor joining tree based on Cavalli-Sforza and Edwards's chord distances was consistent with the geographic locations of populations, as well as with the AMOVA results. There was a significant isolation by distance effect, and the cluster analysis resolved five groups. Individuals were also assayed with an additional microsatellite marker, Cxpq78, proposed to be monomorphic in Cx. pipiens but polymorphic in the closely related but biologically distinct species Cx. quinquefasciatus. Low frequencies (≤3%) of Cx. quinquefasciatus alleles for this marker were noted, and mostly confined to populations along the Interstate 25 corridor. Pueblo was distinct in that it had 10% Cx. quinquefasciatus alleles, mostly of one allele size. The degree of population genetic structure observed in this study is in contrast with that of Cx. tarsalis, the other major vector of WNV in the western U.S., and likely reflects the two species' different dispersal strategies.

ABSTRACT

Early identification of increasing mosquito activity is critical to effective mosquito control, particularly when increasing host-seeking behavior may be associated with increased risk of mosquito-borne disease. In this paper, we analyzed the temporal abundance pattern of the West Nile Virus vector, Culex tarsalis, in Fort Collins, CO, using an autoregressive integrated moving average model. We determined that an autoregressive model order 5 with lagged minimum temperatures was best at describing the seasonal abundance of Cx. tarsalis. We then tested the effect of using both temporal and spatial subsets of the data to determine the effect of reduced sampling effort on abundance predictions. We found that, if reduced trapping is necessary due to limited resources, removal of the least productive 1/3 or 1/4 of the traps produced the least erroneous predictions of seasonality represented in the observed data. We show that this productivity-based subset scheme performs better than other sampling effort reductions in generating the best estimate of Cx. tarsalis abundance per trap-night.

ABSTRACT

In 2006-2007, stable flies, Stomoxys calcitrans (L.) (Diptera: Muscidae), were suspected of being enzootic vectors of West Nile virus (family Flaviviridae, genus Flavivirus, WNV) during a die-off of American white pelicans (Pelecanus erythrorhynchos Gmelin) (Pelecanidae) in Montana, USA. WNV-positive stable flies were observed feeding en masse on incapacitated, WNV-positive pelicans, arousing suspicions that the flies could have been involved in WNV transmission among pelicans, and perhaps to livestock and humans. We assessed biological transmission by infecting stable flies intrathoracically with WNV and testing them at 2-d intervals over 20 d. Infectious WNV was detected in fly bodies in decreasing amounts over time for only the first 6 d postinfection, an indication that WNV did not replicate within fly tissues and that stable flies cannot biologically transmit WNV. We assessed mechanical transmission using a novel technique. Specifically, we fed WNV-infected blood to individual flies by using a cotton swab (i.e., artificial donor), and at intervals of 1 min-24 h, we allowed flies to refeed on a different swab saturated with WNV-negative blood (i.e., artificial recipient). Flies mechanically transmitted viable WNV from donor to recipient swabs for up to 6 h postinfection, with the majority of the transmission events occurring within the first hour. Flies mechanically transmitted WNV RNA to recipient swabs for up to 24 h, mostly within the first 6 h. Given its predilection to feed multiple times when disturbed, these findings support the possibility that the stable fly could mechanically transmit WNV.

ABSTRACT

Resistance to permethrin and the activity of metabolic enzymes were monitored in field-collected Culex tarsalis from northeastern Colorado during 2005 and 2006 and compared with a lab strain from Bakersfield, CA. Collections were made from 13 different sites within and outside of the mosquito abatement area. Resistance levels by year and early vs. late season were examined and median lethal time values and 95% confidence intervals were compared with the lab strain. Mosquitoes collected in 2005 were more resistant to permethrin than either the lab strain or mosquitoes collected in 2006. Glutathione S-transferase levels recorded in 2005 were 9-12x higher than either the lab strain or 2006 mosquitoes; activity of mixed-function oxidases was also greater. Both mechanisms may play a role in permethrin resistance or reflect nontarget exposure to other pesticides in Cx. tarsalis in northeastern Colorado.
