ABSTRACT

The use of sensor-based measures of rumination time as a parameter for early disease detection has received significant attention in scientific research. This study aimed to assess the accuracy of health alerts triggered by a sensor-based accelerometer system within 2 different management strategies on a commercial dairy farm. Multiparous Holstein cows were enrolled during the dry-off period and randomly allocated to conventional (CON) or sensor-based (SEN) management groups at calving. All cows were monitored for disorders for a minimum of 10 DIM following standardized operating procedures (SOPs). The CON group (n = 199) followed an established monitoring protocol on the farm. The health alerts of this group were not available during the study but were later included in the analysis. The SEN group (n = 197) was only investigated when the sensor system triggered a health alert, and a more intensive monitoring approach according to the SOPs was implemented. To analyze the efficiency of the health alerts in detecting disorders, the sensitivity (SE) and specificity (SP) of health alerts were determined for the CON group. In addition, all cows were divided into 3 subgroups based on the status of the health alerts and their health status, to retrospectively compare the course of rumination time. Most health alerts (87%, n = 217) occurred on DIM 1. For the confirmation of diagnoses, health alerts showed SE and SP levels of 71% and 47% for CON cows. In SEN cows, a SE of 71% and 75% and SP of 48% and 43% were found for the detection of ketosis and hypocalcemia, respectively. The rumination time of the subgroups was affected by DIM and the interaction between DIM and the status of health alert and health condition.

ABSTRACT

Dynamic functional changes in the oviductal microenvironment are the prerequisite for the establishment of pregnancy. The objective of this study was to gain the first insights into oestrous cycle-dependent dynamics of polymorph nuclear neutrophils (PMN) and the mRNA abundance of selected genes and their correlations in the oviduct of living cows. Mini-cytobrush samples were taken from the oviducts of healthy heifers (n = 6) and cows (n = 7) during the follicular (FOL) and luteal phase (LUT) by transvaginal endoscopy. Total RNA was isolated from the samples and subjected to reverse transcription-quantitative PCR for selected pro-inflammatory factors, glycoproteins, and a metabolic marker. The percentage of PMN was determined by cytological examination. The mean PMN percentage was 2.8-fold greater during LUT than FOL. During LUT, significantly greater mRNA abundance of the pro-inflammatory factors IL1B, CXCL1, CXCL3, and CXCL8 was observed. The OVGP1 mRNA abundance was twice as high during FOL than in LUT. Pearson correlation, principal component analysis and heatmap analyses indicated characteristic functional patterns with strong correlations among investigated factors. Using this novel approach, we illustrate complex physiological dynamics and interactions of the mRNA expression of pro-inflammatory factors, mucins, OVGP1, and PMN in the oviduct during the oestrous cycle.

ABSTRACT

The objective of this study was to use a sensor-based accelerometer (ACC) to identify changes in lying, rumination, and activity times in weaned calves during the moving and regrouping process. Overall, 270 healthy Holstein calves (from approximately 16 regrouping events) at the age of approximately 4 months were enrolled and equipped with an ear-attached ACC (SMARTBOW, Smartbow GmbH/ Zoetis LLC). Sensor data were recorded for 5 d before (d -5) until 4 d after moving and regrouping (d 4). The day of regrouping was defined as d 0. Acceleration data were continuously processed by specific algorithms (developed by SMARTBOW) for lying, rumination, and activity. Lying, rumination, and activity times were averaged from d -5 to d -3 to generate a baseline value for each parameter. Parameters on d 0 to d 4 after regrouping were compared to this baseline. All parameters showed significant changes compared with the baseline at d 0. Significant decreases in rumination and inactive times were seen up to d 2. Lying time was significantly lower until d 3. The study results indicate that the ACC can be used to monitor the disruptive effects of regrouping on lying and rumination behaviors. Further research is necessary to elucidate how these changes have an impact on health, performance, and welfare and to evaluate how to reduce these potentially negative effects.

ABSTRACT

Many studies have been conducted to estimate pregnancy losses between 19 and 34 d after artificial insemination (AI) in dairy cows managed under confinement-based systems, but few studies have examined embryo mortality during this interval in dairy cows managed under gazing systems. The objectives of this prospective cohort study were (1) to assess the diagnostic value of the corpus luteum (CL) blood perfusion (BP) evaluation by Doppler ultrasound (US) to detect nonpregnant cows at 19 to 20 d post-AI, and (2) to assess the rate of potential embryo mortality between 19 to 34 d post-AI. The CL-BP of all cows included in the study (n = 131) was examined on farm by power and color mode of Doppler US and later using an image processing software by a second evaluator. The endometrium thickness and echotexture were evaluated by B-mode US at the same visit to assess if the nonpregnancy diagnosis could be improved at 19 to 20 d post-AI by this additional diagnostic tool. Blood samples were obtained at 19 to 20 d post-AI for progesterone (P4) measurement by chemiluminescence and to determine the mRNA expression of ISG by real-time PCR. Pregnancy diagnosis based on embryo visualization was performed at 33 to 34 d post-AI by US B-mode. In parallel interpretation, ISG15 and MX2 mRNA expression in leukocytes [sensitivity (Se), 100%] were regarded as suitable biomarkers for early pregnancy and were selected for molecular characterization of pregnancy at 19 to 20 d post-AI. At 19 to 20 d post-AI, 61.1% of the cows had positive CL-BP by Doppler US (Se, 98.0%), 62.7% had ISG mRNA expression in leukocytes over the cutoff point (Se, 95.7%), and 50.8% were positive, based on the combination of ISG mRNA expression, CL-BP by Doppler US, and P4 concentration (Se, 100%), and were considered as possible pregnant. At 33 to 34 d, the pregnancy rate was 37.4% diagnosed by the B-mode US. Based on the expression of the selected biomarkers in cows with active CL, we found that 28.1% of the cows could have potentially lost their pregnancy between 19 and 34 d post-AI. The Doppler US color mode showed similar accuracy and a higher negative predictive value than the genes selected as biomarkers. The additional B-mode ultrasound evaluation of the uterine stratum vasculare and the endometrium thickness improved the diagnostic accuracy. Therefore, assessing the CL-BP by Doppler US allowed early detection of nonpregnant cows at 19 to 20 d post-AI. The combination of early CL-BP by Doppler US (d 19 to 20) with early embryo detection by B-mode US (d 33-34) could be used to facilitate earlier rebreeding of dairy cows.

ABSTRACT

The objectives of this study were (1) to characterize the interindividual variation in the relationship between antepartum (ap) backfat thickness (BFT) and subsequent BFT loss during early lactation in a large dairy herd using cluster analysis; (2) to compare the serum concentrations of metabolites (nonesterified fatty acids, ß-hydroxybutyrate), metabolic hormones (leptin and adiponectin), and an inflammatory marker (haptoglobin) among the respective clusters; and (3) to compare lactation performance and uterine health status in the different clusters. An additional objective was (4) to investigate differences in these serum variables and in milk yield of overconditioned (OC) cows that differed in the extent of BFT loss. Using data from a large study of 1,709 multiparous Holstein cows, we first selected those animals from which serum samples and BFT results (mm) were available at d 25 (±10) ap and d 31 (±3 d) postpartum (pp). The remaining 713 cows (parity of 2 to 7) were then subjected to cluster analysis: different approaches based on the BFT of the cows were performed. K-means (unsupervised machine learning algorithm) clustering based on BFT-ap alone identified 5 clusters: lean (5-8 mm BFT, n = 50), normal (9-12 mm, n = 206), slightly fat (SF; 13-16 mm, n = 203), just fat (JF; 16-22 mm, n = 193), and very fat (VF; 23-43 mm, n = 61). Clustering by difference between BFT-ap and BFT-pp (ΔBFT) also revealed 5 clusters: extreme loss (17-23 mm ΔBFT, n = 16), moderate loss (9-15 mm, n = 119), little loss (4-8 mm, n = 326), no loss (0-3 mm, n = 203), and gain (-8 to -1 mm, n = 51). Based on the blood variables measured, our results confirm that cows with greater BFT losses had higher lipid mobilization and ketogenesis than cows with less BFT loss. The serum variables of cows that gained BFT did not differ from normal cows. Milk yield was affected by the BFT-ap cluster, but not by the ΔBFT cluster. Cows categorized as VF had lesser milk yield than other clusters. We further compared the OC cows that had little or no BFT loss (i.e., 2% of VF, 12% of JF, and 31% of SF, OC-no loss, n = 85) with the OC cows that lost BFT (OC-loss, n = 135). Both NEFA and BHB pp concentrations and milk yield were greater in OC-loss cows compared with the OC-no loss cows. The serum concentration of leptin ap was greater in OC-loss than in the OC-no loss cows. Overall, OC cows lost more BFT than normal or lean cows. However, those OC cows with a smaller loss of BFT produced less milk than OC cows with greater losses.

ABSTRACT

One of the most important diseases in calves worldwide is neonatal calf diarrhea (NCD), which impairs calf welfare and leads to economic losses. The aim of this study was to test whether the activity patterns of calves can be used as early indicators to identify animals at risk for suffering from NCD, compared with physical examination. We monitored 310 healthy female Holstein-Friesian calves on a commercial dairy farm immediately after birth, equipped them with an ear tag-based accelerometer (Smartbow, Smartbow GmbH), and conducted daily physical examinations during the first 28 d of life. The Smartbow system captured acceleration data indicative of standing and lying periods and activity levels (active and inactive), shown as minutes per hour. We categorized calves as diarrheic if they showed fecal scores of ≥3 on a 4-point scale on at least 2 consecutive days. Incidence of diarrhea was 50.7% (n = 148). A mixed logistic regression model showed that lying [odds ratio (OR) = 1.19], inactive (OR = 1.14), and active (OR = 0.92) times, 1 d before clinical identification of diarrhea (d -1), were associated with the odds of diarrhea occurring on the subsequent day. Receiver operating characteristics curve showed that lying time at d -1 was a fair predictor for diarrhea on the subsequent day (area under curve = 0.69). Average lying time on d -1 was 64.8 min longer in diarrheic calves compared with their controls. Median lying and inactive times decreased, and active time increased with age over the study period. The 24-h pattern of behavior indices based on the output of the Smartbow system followed periods of resting and active times, and showed that between 2200 h and 0600 h, calves spent the greatest percentage of time lying and inactive. These results showed that the accelerometer system has the potential to detect early indicators associated with NCD. In future studies, additional data for the development and testing of calf- and event-specific algorithms (e.g., for detecting milk intake, playing behavior) should be collected, which might further improve the early detection of diarrhea in calves.

BACKGROUND: The somatic cell count (SCC) of individual cow samples is a useful proxy for monitoring udder health status. AIMS: The present study aimed to provide updated information about udder health in Iranian Holstein dairy cattle, and to quantify the effectiveness of the mastitis control program. METHODS: A total of 17,990 monthly test-day records from 1,663 Holstein dairy cattle in 10 "regular" herds and 2,389 test-day records from 386 Holstein dairy cattle in 2 herds that were assigned to the 10-point mastitis control program ("controlled" herds) were included. Each test-day record comprised the date of recording, daily milk production (kg), fat and protein (%), days in milk, parity, and SCC. RESULTS: Median (Q1-Q3) SCC × 103 for "regular" and "controlled" herds were 136 (52-391) and 64 (24-204) cells/ml, respectively. Also, the percentage of records containing SCC >200,000 cells/ml (elevated SCC) for these groups were 40.3% and 25.5%, respectively. Mixed effects logistic analysis revealed that milk records from cows in the first lactation, early lactation, and with >40 kg daily milk yield had lower odds of elevated SCC. The odds of elevated SCC were lower in summer and autumn than in winter. CONCLUSION: Host and environmental characteristics influence SCC. This should be considered for the interpretation of SCC results. Mastitis control programs can support dairy producers to reach a standard level of udder health.

ABSTRACT

Automated sensor-based monitoring of cows has become an important tool in herd management to improve or maintain animal health and welfare. Location systems offer the ability to locate animals within the barn for, for example, artificial insemination. Furthermore, they have the potential to measure the time cows spend in important areas of the barn, which might indicate need for improvement in the management of the herd or individuals. In this study, we tested the sensor-based real-time location system (RTLS) Smartbow (SB, Smartbow GmbH) under field conditions. The objectives of this study were (1) to determine the accuracy of the system to predict the location of the cow and the agreement between visual observations and RTLS observations for the total time spent by cows in relevant areas of the barn and (2) to compare the performance of 2 different algorithms (Alg1 and Alg2) for cow location. The study was conducted on a commercial Austrian dairy farm. In total, 35 lactating cows were video recorded for 3 consecutive days. From these recordings, approximately 1 h was selected randomly each day for every cow (3 d × 35 cows). Simultaneously, location data were collected and classified by the RTLS system as dedicated to the alley, feed bunk, or cubicle on a 1-min resolution. A total of 6,030 paired observations were derived from visual observations (VO) and the RTLS and used for the final data analysis. Substantial agreement of categorical data between VO and SB was obtained by Cohen's kappa for both algorithms (Alg1 = 0.76 and Alg2 = 0.78). Similar results were achieved by both algorithms throughout the study, with a slight improvement for Alg2. The ability of the system to locate the cows in the predefined areas was assessed, and the results from Alg2 showed sensitivity, specificity, and positive predictive value of alley (74.0, 91.2, and 76.9%), feed bunk (93.5, 86.2, and 89.1%), and cubicle (90.5, 83.3, and 95.4%) and an overall accuracy of 87.6%.The correlation coefficient (r) between VO and SB for the total time cows spent (within 1 h) in the predefined areas was good to strong (r = 0.82, 0.98, and 0.92 for alley, feed bunk, and cubicle, respectively). These results show the potential of the system to automatically assess total time spent by cows in important areas of the barn for indoor settings. Future studies should focus on evaluating 24-h periods to assess time budgets and to combine technologies such as accelerometers and location systems to improve the performance of behavior prediction in dairy cows.

Dairy farms face many challenges and changes. With increasing herd sizes and fewer farmers or employees per cow, new strategies to maintain or improve reproductive management are required. One of the major challenges is to detect cows in estrus and to estimate the perfect time for artificial insemination (AI). Several estrus and ovulation synchronization programs with timed AI as well as estrus detection aids, e.g., tail-paint, pedometer, accelerometer, and others are available. A combination of ovulation synchronization programs and technical solutions, however, has rarely been tested. This study was designed to gain insights into behavioral patterns of cows subjected to an Ovsynch program and to test if behavioral data could be used to optimize the timing of insemination within an Ovsynch program. In this study, we used an ear-tag based 3D-accelerometer system (SMARTBOW, Smartbow GmbH, Weibern, Austria) to generate data of behavioral patterns, i.e., rumination and activity. In Part 1 of this study, behavioral patterns during the peri-estrus period were compared between cows with physiological estrus and cows subjected to an Ovsynch protocol. On the day before estrus and on the day of estrus/AI, cows with natural estrus showed a clear drop in rumination and "inactivity" and an increase in "high activity", based on an algorithm of the accelerometer system, whereas, cows in the Ovsynch protocol showed only minor changes in behavioral patterns. In Part 2, we analyzed behavioral patterns between synchronized cows that became pregnant after AI and synchronized cows that remained open. As a result, no differences were detected between these two Ovsynch groups before AI. Thus, in this study we found no evidence that behavioral patterns can be used to improve conception rates within an Ovsynch protocol.

ABSTRACT

The aim of the study was to determine whether two postpartum intramuscular treatments with 200 mg of beta-(ß-)carotene (Carofertin; Alvetra u. Werfft, Vienna, Austria) in a 14-day interval increases ß-carotene concentrations in blood, particularly around the time of the first artificial insemination (AI), and to test the effect of the treatment on fertility parameters, luteal size, and progesterone blood levels of dairy cows. A total of 297 Holstein dairy cows were enrolled in the study. Between 28 and 34 days postpartum (dpp) ß-carotene concentrations were measured in blood samples using an on-site test (iCheck carotene; BioAnalyt, Teltow, Germany). Cows with a ß-carotene concentration <3.5 mg/L, indicating a deficiency of ß-carotene, were allocated either to the ß-carotene treatment group BCT (n = 123) or to the control group CON (n = 121). Cows with concentrations ≥3.5 mg/L were assigned to an optimally supplied reference group (REF; n = 53). Cows in the BCT group received 200 mg of ß-carotene intramuscularly at 28-34 dpp and at 42-48 dpp. Further blood samples were collected at 35-41 dpp, 42-48 dpp, 49-55 dpp, and in the week after the first AI and their ß-carotene concentrations were analyzed. Between day 10 and 14 after the first AI, the blood progesterone concentration was measured and the size of the corpus luteum (CL) was determined by ultrasound. Blood ß-carotene concentrations increased in the BCT cows in the week after the treatment with a peak at 49-55 dpp and were significantly higher than in the CON group at each time point after the first treatment. Logistic regression models, however, revealed that the treatment with ß-carotene had no effect on first service conception rate, days to first service, time to pregnancy, or percentage of pregnant cows within 150 dpp. Furthermore, there was no effect on progesterone concentration or the size of the CL between the groups. In conclusion, two treatments with Carofertin postpartum increased ß-carotene blood concentrations but had no effect on the fertility parameters in this study.

During stage II of parturition, the bovine fetus is at risk of oxygen deficiency caused by insufficient gas exchange between the dam and the fetus. The early detection of this critical condition, followed by assistance at calving, can help to improve the vitality of the newborn calf, or even prevent it from being born dead. By using pulse oximetry, the arterial oxygen saturation, as well as the pulse rate, can be continuously and non-invasively measured. The aim of our study was to identify critical thresholds for the parameters 'arterial oxygen saturation (FSpO2)' and 'pulse rate (PR)' that indicate a severe postnatal risk for calves to suffer from acidosis. FSpO2 and PR from 40 bovine fetuses were recorded during the last 25 min of calving with a commercially available pulse oximeter (Radius-7, Masimo Corporation, Irvine, USA). The calves were tested immediately after birth for acidosis by analyzing their blood with a portable blood gas analyzer (VetScan iStat1, Abaxis Inc., Union City, USA). Retrospectively, the pulsoximetric data were scanned for predefined patterns. The validity of these patterns to predict acidosis in newborn calves was analyzed by using receiver operating characteristics (ROC) curve analyses. In general, PR was a stronger predictive parameter for acidosis than FSpO2, with the greatest area under the curve (AUC) for the PR criteria 'Pulse rate > 120 beats per minute (bpm) for at least 2 min', with an AUC of 0.764, in contrast to an AUC of 0.613 for the best FSpO2 criteria 'FSpO2 < 40% for at least 50% of the measurement'. Further studies should investigate whether vitality after calving can be improved and fetal death rate can be reduced when obstetric assistance is performed as soon as one of these criteria apply to the bovine fetus. For more practical implementation in the field, improvement of the device's hardware would be necessary.

ABSTRACT

Several factors have been reported to influence colostrum quality (immunoglobulin concentration). To date, knowledge of the influence of climatic factors in association with other potential influencing factors on colostrum quality is scarce. Associated influential factors are parity, body condition score, length of dry period, ration fed ante partum (AP), ß-hydroxybutyrate postpartum (PP), milk yield, milk fat and protein, as well as somatic cell counts from previous and current lactation. The objective of the present study was to examine the effect of barn climate and the aforementioned factors on colostrum quality. Data were collected from 1,381 multiparous Holstein Friesian cows kept on one dairy farm over a period of one year (August 2014 to August 2015). Colostrum was harvested on farm within 1 h PP. The quantity and quality of first colostrum (estimated by Brix refractometry) were recorded for each cow. Additional data recorded were parity, body condition score at drying off, length of dry period, ration fed AP, milk yield data from previous and current lactation, milk somatic cell counts, and ß-hydroxybutyrate PP. During the study period, temperature and humidity were recorded in the barn every hour, and temperature-humidity-index (THI) was calculated. Linear regression was performed with colostrum quality as the dependent variable. In the final model, colostrum quantity (L), length of dry period, parity, and climatic factors (specifically, median humidity in the 3rd week AP and hours with THI ≥72 in the last 14 and 21 d AP, respectively) were significant. Colostrum quality improved with parity and length of dry period and decreased with colostrum quantity, humidity, and hours with a THI ≥72. A classification and regression tree analysis revealed that colostrum quantity was the most important factor in this model [normalized importance (NI) 100%]. Parity (NI 42.7%), length of dry period (NI 37.1%), and climatic factors (NI 0.4 to 1.9%) followed with decreasing importance. These results indicate that the most important factors for colostrum quality (i.e., colostrum quantity and parity) may not be influenced by management. The 2 factors that can be influenced by management [i.e., length of dry period and THI (e.g., by cooling)], were quantitatively of minor importance compared with the other 2 factors. Further studies are necessary to determine whether changing these factors can improve colostrum quality significantly.

ABSTRACT

Precision dairy farming technologies have tremendous potential to improve and support farmers in herd management decisions, particularly in reproductive management. Nowadays, estrus detection in cows is challenging and several supporting tools are available. In this study, a 3D-accelerometer integrated into an ear-tag (SMARTBOW, Smartbow GmbH, Weibern, Austria) was used for the detection of cows in estrus. Movement pattern based on accelerometer data were analyzed and processed by algorithms and machine learning, resulting in estrus alerts. For the evaluation of the system, reproductive performance data of 579 estrus events of multiparous cows were used to retrospectively evaluate the accuracy of estrus alerts generated by the accelerometer-based system and the overall performance of the system. Estrus events were classified as 'gold standard' events, if an estrus followed by AI resulted in pregnancy, and as 'recorded estrus' events, if two estrus events with an interval of 18-25 d were in the herd records, independent of whether estrus was followed by AI or pregnancy. In total, 316 'gold standard' events were matched with estrus alerts generated by the accelerometer-based system, resulting in a sensitivity of 97%. Furthermore, 263 'recorded estrus' events were compared with correct or incorrect estrus alerts by the system. Sensitivity, specificity, positive and negative predictive values, accuracy, and error rate for 'recorded estrus' events were 97%, 98%, 96%, 94%, 96%, and 2%, respectively. In summary, the SMARTBOW system is suitable for an automated detection of estrus events of multiparous cows in indoor housed dairy cows.

ABSTRACT

Pulse oximetry is a well-established technique in human and veterinary medicine. In farm animals, it could also be a useful tool for the detection of critical conditions relating to oxygen supply and the cardiovascular system. Among other uses, an innovative application could be the monitoring of fetuses during birth. This could help in the early identification of critical situations and support farmers and veterinarians in their decision to start obstetric or life-support interventions. Until now, however, its use in ruminant medicine was still limited to experimental applications. The objective of this study was to evaluate the accuracy of the Radius-7 Wearable Pulse CO-Oximeter (Masimo Corporation, Irvine, CA) for monitoring vital parameters in newborn calves. All measurements were conducted on animals in the lying down position. The sensor of the pulse oximeter was placed in the interdigital space of the calves' front legs and fixed with a homemade latex hoof cover. The pulsoximetric measurements of arterial oxygen saturation (SpO2) in 40 newborn calves were compared with the corresponding results (SaO2) from a portable blood gas analyzer (VetScan iStat1, Abaxis Inc., Union City, CA), which served as the reference. For this, an arterial blood sample was taken from the medial intermediate branch of the caudal auricular artery. In addition, the pulse rate was measured in 10 calves aged between 0 and 7 d with the pulse oximeter and simultaneously with a heart rate belt (Polar Equine Belt, Polar Electro Oy, Kempele, Finland) to determine their level of agreement. Spearman correlation coefficient for oxygen saturation was 93.8% for the pulse oximeter and the blood gas analyzer, and 97.7% for the pulse rate measured with the pulse oximeter and the heart rate belt. Bland-Altman plots revealed an overestimation of SaO2 by 2.95 ± 6.39% and an underestimation of the pulse rate by -0.41 ± 3.18 beats per minute compared with the corresponding reference methods. In summary, the pulse oximeter is suitable for continuous monitoring of arterial oxygen saturation and pulse in newborn Holstein Friesian calves. For practical use, purpose-built technical equipment is required to attach the sensor to the calves' legs.

ABSTRACT

The objective of this study was to evaluate the ear-tag-based accelerometer system Smartbow (Smartbow GmbH, Weibern, Austria) for detecting rumination time, chewing cycles, and rumination bouts in indoor-housed dairy cows. For this, the parameters were determined by analyses of video recordings as reference and compared with the results of the accelerometer system. Additionally, we tested the intra- and inter-observer reliability as well as the agreement of direct cow observations and video recordings. Ten Simmental dairy cows in early lactation were equipped with 10-Hz accelerometer ear tags and kept in a pen separated from herd mates. A total mixed ration was fed twice a day via a roughage intake control system. During the study, cows' rumination and other activities were directly observed for 20 h by 2 trained observers. Additionally, cows were video recorded for 19 d, 24 h a day. After exclusion of unsuitable videos, 2,490 h of cow individual 1-h video sequences were eligible for further analyses. Out of this, one hundred 1-h video sequences were randomly selected and visually and manually classified by a trained observer using professional video analyses software. Based on these analyses, half of the data was used for development (based on data of 50-h video analyses) and testing (based on data of additional 50-h video analyses) of the Smartbow algorithms, respectively. Inter- and intra-observer reliability as well as the comparison of direct against video observations revealed in high agreements for rumination time and chewing cycles with Pearson correlation coefficients >0.99. The rumination time, chewing cycles, as well as rumination bouts detected by Smartbow were highly associated (r > 0.99) with the analyses of video recordings. Algorithm testing revealed in an underestimation of the average ± standard deviation rumination time per 1-h period by the Smartbow system of 17.0 ± 35.3 s (i.e., -1.2%), compared with visual observations. The average number ± standard deviation of chewing cycles and rumination bouts was overestimated by Smartbow by 59.8 ± 79.6 (i.e., 3.7%) and by 0.5 ± 0.9 (i.e., 1.6%), respectively, compared with the video analyses. In summary, the agreement between the Smartbow system with video analyses was excellent. From a practical and clinical point of view, the detected differences were negligible. However, further research is necessary to test the system under various field conditions and to evaluate the benefit of incorporating rumination data into herd management decisions.

ABSTRACT

The bovine oviduct provides the site for fertilization and early embryonic development. Modifications to this physiological environment, for instance the presence of pathogenic bacterial species, could diminish reproductive success at early stages of pregnancy. The aim of this study was to elucidate the inflammatory responses of bovine oviductal epithelial cells (BOEC) to a pathogenic bacterial species (Trueperella pyogenes) and a potentially pathogenic bacterium (Bacillus pumilus). BOEC from four healthy animals were isolated, cultured in passage 0 (P0) and passaged until P3. Trypan blue staining determined BOEC viability during 24 h co-culture with different multiplicities of infection (MOI) of T. pyogenes (MOI 0.01, 0.05, 0.1 and 1) or B. pumilus (MOI 1 and 10). BOEC remained viable when co-cultured with T. pyogenes at MOI 0.01 and with B. pumilus at MOI 1 and 10. Extracted total RNA from control and bacteria co-cultured samples was subjected to reverse transcription-quantitative polymerase chain reaction (RTq-PCR) to determine mRNA expression of various studied genes. The rate of release of interleukin 8 (IL8) and prostaglandin E2 (PGE2) from BOEC was measured by ELISA after 24 h co-culture with bacteria. RT-qPCR of various selected pro-inflammatory factors revealed similar mRNA expression of pro-inflammatory factors in BOEC co-cultured with T. pyogenes and in the controls. Higher mRNA expression of IL 1A, -1B, tumor necrosis factor alpha and CXC ligand (CXCL) 1/2, -3, -5 and IL8 and PG synthesis enzymes in BOEC co-cultured with B. pumilus was observed. In the presence of B. pumilus a higher amount of IL8 and PGE2 was released from BOEC than from controls. The viability and pro-inflammatory response of P3 BOEC incubated with bacteria was lower than in P0 BOEC. These findings illustrate the pathogenicity of T. pyogenes towards BOEC in detail and the potential role of B. pumilus in generating inflammation in oviductal cells. Culturing conditions influenced the pro-inflammatory responses of BOEC towards bacteria. Therefore, researchers conducting epithelial-bacterial in vitro co-culture should not underestimate the effects of these parameters.

ABSTRACT

The primary objective of this study was to evaluate the repeatability of measurements of ß-hydroxybutyrate (BHB) concentrations in capillary blood obtained from minimal-invasive puncturing of different sites at the external vulva skin of dairy cows. A total of 240 multiparous cows sampled once within 21 d in milk were enrolled in the study. Capillary blood samples were obtained from the right and left labium of the external vulva skin and additionally from a site close to the first puncture. Samples were analyzed by using an electronic hand-held device [FreeStyle Precison Neo (FSP-Neo), Abbott GmbH & Co., Wiesbaden, Germany)]. Additionally, samples from a coccygeal vessel were analyzed as reference in a laboratory. Mean deviations (± standard deviation) between the 3 capillary sampling sites ranged from 0.01 ± 0.14 (left vs. right labium) to 0.02 ± 0.13 mmol/L (left or right labium vs. additional site). The BHB concentrations in capillary blood compared with the reference showed a mean deviation of 0.14 ± 0.18 mmol/L. The Spearman correlation coefficient between BHB concentrations in capillary blood and coccygeal blood was 85.4%. Mixed model analyses showed no significant effect of sampling site or squeezing the skin during sampling procedures on the differences toward the reference. Based on laboratory BHB concentrations of ≥1.2 and ≥1.4 mmol/L, respectively, the prevalence of hyperketonemia was 10.4 (n = 25) and 5.0% (n = 12). Thresholds to detect hyperketonemia with the FSP-Neo were optimized by receiver operating characteristic analyses. Based on a laboratory BHB concentration of ≥1.2 mmol/L thresholds were determined as 1.1 (left labium, 2nd puncture) and 1.0 mmol/L (right labium), with sensitivities between 94 and 99% and specificities between 89 and 97%. Based on a serum BHB concentration of 1.4 mmol/L, receiver operating characteristic analyses resulted in optimized cutoffs of 1.4 mmol/L for all sampling sites at the vulva, with sensitivities of 100% and specificities between 96 and 99%. In conclusion, vulvar capillary blood is suitable for accurate measuring of BHB concentrations by using the FSP-Neo. After adjusting the thresholds, the FSP-Neo provides very good test characteristics independent of the capillary sampling site or squeezing the skin and allows a practical and reliable screening tool for dairy cows.

ABSTRACT

The objective of the present study was to test the association between the presence of fluid in uterine lumen and the fertility of repeat breeder cows (RBC). Grazing Holstein cows that were clinically normal and were not pregnant after at least 3 consecutive inseminations were defined as RBC and included in the study (n = 358). A cut-point ≥2 mm of uterine lumen visible by ultrasonography was used as indicative of the presence of fluid in the uterus. The effect of this fluid on the fertility of RBC was assessed by mixed logistic regression. Twenty percent of the RBC had ≥2 mm of fluid in their uterus, and this finding was associated with a marked reduction in the odds of conception (odds ratio = 0.46). Additional intrauterine sampling revealed that 24.0% of RBC showed signs of subclinical endometritis [≥5% neutrophils in endometrial cytology (SCE)]. However, we found no agreement between the finding of uterine fluid and the diagnosis of SCE (kappa coefficient = 0.03, 95% confidence interval = -0.07 to 0.14). In conclusion, the presence of fluid (≥2 mm) in uterine lumen, detected by ultrasonography, was associated with lower fertility in RBC, but was not related to the diagnosis of SCE. Thus, ultrasonographic evaluation is an important tool to assist in assessment of RBC.

ABSTRACT

The objective of this study was to assess the effect of a selective antibiotic treatment strategy based on a quick bacteriological on-farm test (Petrifilm, 3M Corp., St. Paul, MN) compared with the conventional antibiotic treatment of all cows having clinical endometritis (CE) defined by the presence of purulent vaginal discharge on both clinical cure rate and reproductive performance. The study was simultaneously conducted with dairy cows reared under a highly supplemented rotational grazing system in Argentina and in a freestall system in Slovakia. Cows having an abnormal vaginal discharge (VD, indicative of clinical endometritis) on 21 to 35 d in milk (DIM) were randomly allocated to 1 of 2 study groups: selective treatment (ST) or conventional treatment (CT). All cows in the CT group (n = 174) received a single intrauterine administration of 500 mg of cephapirin. In the ST group (n = 178), treatment decision was made according to the results of the bacteriological on-farm test. For this test, we collected intrauterine samples with the cytobrush technique and stroke the brushes onto 2 different Petrifilm plates, one for aerobic count and another for Enterobacteriaceae count, incubated the plates, and counted the number of colonies after 24 h. Positive cows (≥5 colonies in one or both plates) received a single intrauterine treatment with 500 mg of cephapirin, whereas negative cows (<5 colonies) remained untreated. Clinical cure rate was assessed by direct vaginal inspection at 14 d after treatment (VD-0). The odds for conception at first artificial insemination, artificial insemination by 80 DIM, pregnancy by 100 DIM, and for nonpregnancy by 200 DIM were estimated with mixed logistic regression models. The hazard of conception was also assessed with proportional hazard regression model. The selective antibiotic treatment strategy based on the outcome of Petrifilm test reduced the number of required treatments (57%) and maintained similar efficacy in terms of clinical cure and reproductive performance as the conventional antibiotic treatment of all endometritic cows.
