ABSTRACT

The long axis of epidermal cambial cells determining the direction of their division is determined before the beginning of division and is oriented perpendicularly, but not parallel, to the basal membrane, as in other basal cells. As a result, only one of the two newly formed cells adheres to the basal membrane and at the expense of traction forces detaching one cell from the other and elastic force of the basal membrane is formed as a maternal cell and stays in the focus of multiplication. The other cell, turning around under the effect of traction forces perpendicularly to the maternal cell, cardinally changes the direction of its long axis and is polarized parallel to the basal membrane like other cells. This cell becomes the daughter cell, its shape with the "head" and "tail" allows its rapid migration into other rosettes for differentiation.

ABSTRACT

The role of pancreatic beta-cell antigenic structures in modulation of insulin secretion in vitro was recently demonstrated by others. Here we report generation of a monoclonal antibody (mAB) ICA-1 to non-species specific beta-cell antigen(s) 64, 67 and 69 kDa. The mAB inhibits glucose stimulated insulin secretion in islet cell cultures. The ability of mAB ICA-1 to immunoprecipitate active glutamic acid decarboxylase from high speed supernatants of pancreatic and brain crude extracts was demonstrated. The 64, 67 and 69 kDa antigenic material was affinity purified from pancreatic islet cell high speed supernatants, active glutamic acid decarboxylase was found in the material. Immunoaffinity purification with mAB ICA-1 of GAD-like pancreatic beta-cell antigenic material has provided evidence of possible involvement of glutamic acid decarboxylase in modulation of insulin secretion.

ABSTRACT

Outbred laboratory mice, pretreated with an ascites-forming stimulator and an irradiation dose of 6 Gy, can be used for the production of monoclonal antibodies. However, the passage of hybridomas in irradiated outbred mice rapidly leads to the death of the cells. The properties of antibodies obtained from ascitic fluid of outbred mice are no different from those of antibodies obtained from BALB/c mice.

ABSTRACT

Monoclonal antibodies (MoAB) ICO-45 discovered the antigen with 45 kD molecular mass, expressed on the surface of 77% thymocytes, 68% monocytes, 95% granulocytes, 46% T-lymphocytes, 59% non-T-lymphocytes. MoAB ICO-45 blocked T-lymphocyte blast transformation and inhibited the respiratory burst of monocytes and granulocytes of the peripheral blood.

ABSTRACT

The panel of monoclonal antibodies (MCA) ICO-16, ICO-17, ICO-18, ICO-19, ICO-20, ICO-27, ICO-28 IgG2a isotypes to CD38 antigen was obtained. MCA discovered the antigen with 45 kD molecular mass, expressed on the surface of 100% thymocytes, 43-53% lymphocytes, 32-46% monocytes. All obtained MCA blocked the binding each other with thymocyte of man. MCA reacts in complement-dependent cytotoxic test. The antigen CD38 is expressed on blast cells of patients with T-cells subset of ALL.

ABSTRACT

Monoclonal antibodies (Mab) Ig G2a isotypes reacting in indirect immunofluorescence assay with 68.7 +/- 4.1% of thymocytes, 7% of T-cells and not determining the antigen on other blood cells were obtained. Mab ICO-20 reacted in complement-dependent cytotoxic test. The antigen was expressed on colony-forming cells of granulocyte-macrophage row. Mab ICO-20 reaction with 100% of thymocytes was defined by flow cytometry. Antigen molecular mass is 45000 Dalton. The antigen was expressed on blast cells of patients with ALL and AML. Mab ICO-20 reaction was more more often with T-cell ALL.

ABSTRACT

Monoclonal antibodies (Mab) ICO-13 of IgM isotype reacted in indirect surface immunofluorescence test with 88.5 +/- 3.4% of thymocytes and 7.1 +/- 2.4% of T cells, but failed to react with other peripheral blood cells from healthy donors. The antigen disappeared in cells stored at 4 degrees C overnight or at -196 degrees C in liquid nitrogen. The antigen detected by Mab ICO-13 was expressed on blast cells of acute lymphoblast leukemias and lymphomas. The antigen was absent in chronic lymphoblast leukemia and blast crisis of chronic myeloid leukemia.

ABSTRACT

The goal of the investigation was to further characterize ICO-II monoclonal antibodies. ICO-II have been shown to block NK activity of mononuclear cells from the blood of healthy donors against K-562 and Molt-4 target cells, cytotoxic T lymphocytes induced in a 7-day mixed lymphocyte culture, the reaction of lymphocyte blast cell transformation to phytohemagglutinin and the formation of En-rosettes. The molecular weight of the antigen detected by ICO-II is 180 KD. ICO-II are shown to detect alpha-subunit of human lymphocyte function-associated antigen-I (LFA-I).
