ABSTRACT

We investigated the effects of obesity on metabolic, inflammatory, and oxidative stress parameters in the adipose tissue of patients with fatal COVID-19. Postmortem biopsies of subcutaneous adipose tissue were obtained from 25 unvaccinated inpatients who passed from COVID-19, stratified as nonobese (N-OB; body mass index [BMI], 26.5 ± 2.3 kg m-2) or obese (OB BMI 34.2 ± 5.1 kg m-2). Univariate and multivariate analyses revealed that body composition was responsible for most of the variations detected in the metabolome, with greater dispersion observed in the OB group. Fifteen metabolites were major segregation factors. Results from the OB group showed higher levels of creatinine, myo-inositol, O-acetylcholine, and succinate, and lower levels of sarcosine. The N-OB group showed lower levels of glutathione peroxidase activity, as well as higher content of IL-6 and adiponectin. We revealed significant changes in the metabolomic profile of the adipose tissue in fatal COVID-19 cases, with high adiposity playing a key role in these observed variations. These findings highlight the potential involvement of metabolic and inflammatory pathways, possibly dependent on hypoxia, shedding light on the impact of obesity on disease pathogenesis and suggesting avenues for further research and possible therapeutic targets.

ABSTRACT

Mono- and diglycerides play a crucial role in the food industry as multifunctional food additives and emulsifiers. Their importance stems from their unique properties, which allow them to improve the quality, texture, and stability of various food products. Here, results of the kinetic modeling of the mono- and diglycerides synthesis mediated by the lipase Lipozyme® TL 100 L immobilized on the clayey support Spectrogel® type C are reported. The support was characterized by TEM, SEM, and FTIR. Firstly, the influence of pH and lipase load on the immobilization process was analyzed, resulting in an enzymatic activity of 93.2 ± 0.7 U g-1 under optimized conditions (170.9 U g-1 of lipase and pH of 7.1). Afterward, the effects of reaction temperature and concentration of immobilized biocatalyst in the feedstock conversion were evaluated. At optimized parameters, a triglycerides conversion of 97% was obtained at 36.5 °C, 7.9 vol.% of enzyme, a glycerol to feedstock molar ratio of 2:1, and 2 h. The optimized conditions were used to determine the kinetic constants of the elementary reactions involved in the glycerolysis, where a fit superior to 0.99 was achieved between experimental values and predicted data.

ABSTRACT

The topical use of sunscreens is recommended for avoiding the damaging effects of UV radiation. However, improvements are still needed in the existing products to enhance their photoprotection effectiveness and safety. This involves minimizing the use of chemical UV filters while providing enhanced and prolonged photoprotection. This work investigated novel sunscreen formulations and their UV protection effects by encapsulating Uvinul® A, Tinosorb® S, and Uvinul® T150 into nanostructured lipid carriers (NLCs) based on bacuri butter and raspberry seed oil. First, the impact of critical formulation and process parameters on NLCs' particle size was evaluated using a 22 Face Centered Central Composite Design. Then, formulations were evaluated in terms of critical quality factors, in vitro skin permeation, and in vitro and in vivo photoprotection activities. The developed NLCs-containing formulations exhibited appropriate size (122-135 nm), PdI (<0.3), encapsulation efficiency (>90%), and drug content (>80%), which were preserved for at least 90 days under different stability conditions. Moreover, these NLCs-based formulations had equivalent skin permeation to emulsion-based controls, and the addition of NLCs into sunscreen cream bases in the optimum proportion of 20% (w/w) resulted in enhanced UVA and UVB photoprotection levels, despite a 10% reduction in the total filters content. Altogether, these results describe the application of nanoencapsulated organic UV filters in innovative sunscreen formulations to achieve superior photoprotection and cosmeceutical properties.

ABSTRACT

Alzheimer's disease (AD) is a neurodegenerative disorder caused by accumulation of amyloid-ß oligomers (AßO) in the brain, neuroinflammation, oxidative stress, and cognitive decline. Grandisin, a tetrahydrofuran neolignan, exhibits relevant anti-inflammatory and antioxidant properties. Interestingly, grandisin-based compounds were shown to prevent AßO-induced neuronal death in vitro. However, no study has assessed the effect of these compounds on the AD animal model. This study focuses on a triazole grandisin analogue (TGA) synthesized using simplification and bioisosteric drug design, which resulted in improved potency and solubility compared with the parent compound. This study aimed to investigate the possible in vivo effects of TGA against AßO-induced AD. Male C57/Bl6 mice underwent stereotaxic intracerebroventricular AßO (90 µM) or vehicle injections. 24 h after surgery, animals received intraperitoneal treatment with TGA (1 mg/kg) or vehicle, administered on a 14 day schedule. One day after treatment completion, a novel object recognition task (NORT) was performed. Memantine (10 mg/kg) was administered as a positive control. NORT retention sessions were performed on days 8 and 16 after AßO injection. Immediately after retention sessions, animals were euthanized for cortex and hippocampus collection. Specimens were subjected to oxidative stress and cytokine analyses. TGA reduced the level of cortex/hippocampus lipoperoxidation and prevented cognitive impairment in AßO-injected mice. Additionally, TGA reduced tumor necrosis factor (TNF) and interferon-γ (IFN-γ) levels in the hippocampus. By contrast, memantine failed to prevent cortex/hippocampus lipid peroxidation, recognition memory decline, and AßO-induced increases in TNF and IFN-γ levels in the hippocampus. Thus, memantine was unable to avoid the AßO-induced persistent cognitive impairment. The results showed that TGA may prevent memory impairment by exerting antioxidant and anti-inflammatory effects in AßO-injected mice. Moreover, TGA exhibited a persistent neuroprotective effect compared to memantine, reflecting an innovative profile of this promising agent against neurodegenerative diseases, such as AD.

ABSTRACT

Corals are colonial animals within the Phylum Cnidaria that form coral reefs, playing a significant role in marine environments by providing habitat for fish, mollusks, crustaceans, sponges, algae, and other organisms. Global climate changes are causing more intense and frequent thermal stress events, leading to corals losing their color due to the disruption of a symbiotic relationship with photosynthetic endosymbionts. Given the importance of corals to the marine environment, monitoring coral reefs is critical to understanding their response to anthropogenic impacts. Most coral monitoring activities involve underwater photographs, which can be costly to generate on large spatial scales and require processing and analysis that may be time-consuming. The Marine Ecology Laboratory (LECOM) at the Federal University of Rio Grande do Norte (UFRN) developed the project "#DeOlhoNosCorais" which encourages users to post photos of coral reefs on their social media (Instagram) using this hashtag, enabling people without previous scientific training to contribute to coral monitoring. The laboratory team identifies the species and gathers information on coral health along the Brazilian coast by analyzing each picture posted on social media. To optimize this process, we conducted baseline experiments for image classification and semantic segmentation. We analyzed the classification results of three different machine learning models using the Local Interpretable Model-agnostic Explanations (LIME) algorithm. The best results were achieved by combining EfficientNet for feature extraction and Logistic Regression for classification. Regarding semantic segmentation, the U-Net Pix2Pix model produced a pixel-level accuracy of 86%. Our results indicate that this tool can enhance image selection for coral monitoring purposes and open several perspectives for improving classification performance. Furthermore, our findings can be expanded by incorporating other datasets to create a tool that streamlines the time and cost associated with analyzing coral reef images across various regions.

ABSTRACT

To investigate the impact of different exercise training schedules (following a fixed schedule or at random times of the day) on clock genes and myokine expression patterns in the skeletal muscle of tumor-bearing mice. Mice were divided into three groups: tumor (LLC), tumor + exercise training (LLC + T) always performed at the same time of the day (ZT2) and exercise training at random times of the day (ZTAlt). Mice were inoculated subcutaneously with Lewis lung carcinoma cells. The gastrocnemius muscle was dissected and the clock gene expression (Clock/Per1/Per2/Per3/Rev-Erbα/GAPDH) was investigated by quantitative reverse transcription polymerase chain reaction with SYBR® Green. Myokine content in muscle (tumour necrosis factor alpha/IL-10/IL-4) was assessed by enzyme-linked immunosorbent assay. At the end of the protocol, the trained groups showed a reduction in total weight, when compared to Lewis lung carcinoma. Tumor weight was lower in the LLC + T (ZTAlt), when compared to LLC. Clock gene mRNA expression showed a significant increase for ZT20 in the groups that performed physical exercise at LLC + T (ZTAlt), when compared with LLC. The Per family showed increased mRNA expression in ZT4 in both trained mice groups, when compared with LLC. LLC + T (ZTAlt) presented reduction of the expression of anti-inflammatory myokines (Il-10/IL-4) during the night, compared with LLC + T(ZT2). Exercise training is able to induce marked modification of clock gene expression and of the production of myokines, in a way that is dependent on schedule exercise training strategy. Taken together, the results show that exercise is a potent Zeitgeber and may thus contribute to change clock genes expression and myokines that are able to reduce the tumor weight.

ABSTRACT

Myxozoans of Ceratomyxidae Doflein, 1899 are common coelozoic parasites of marine life, and are also found less frequently in freshwater fish. The present study describes Ceratomyxa ranunculiformis n. sp. as a new freshwater myxosporean species infecting the gall bladder of the Amazonian sciaenid Plagioscion squamosissimus. The new Ceratomyxa was described based on its host, myxospore morphology, ribosomal rDNA gene sequencing, parasite distribution, and phylogenetic analysis. Immature and mature plasmodia were tadpole-shaped or pyriform, and exhibited slow undulatory motility. The myxospores were elongated and crescent-shaped in the frontal view, with a sutural line between two valves, which had rounded ends. The measurements of the formalin-fixed myxospores were: average length 4.9 (4.0-6.6) µm, average thickness 37.6 (32.4-43.9) µm, average posterior angle 165° (154°-173°). Two ovoid polar capsules of equal size, average length 2.0 (1.4-3.0) µm and average width 1.9 (1.4-2.4) µm, were located adjacent to the suture and contained polar filaments with 2-3 coils. The integrated comparative analysis of the morphological characteristics and molecular analyses of the ribosomal rDNA genes supported the identification of a new species of coelozoic Ceratomyxa. Maximum likelihood analyses showed the new species clustering within a well-supported clade, together with all the other Amazonian freshwater ceratomyxids.

ABSTRACT

Chagas disease is a neglected tropical disease with only two drugs available for treatment and the plant Cecropia pachystachya has several compounds with antimicrobial and anti-inflammatory activities. This study aimed to evaluate a supercritical extract from C. pachystachya leaves in vitro and in vivo against Trypanosoma cruzi. A supercritical CO2 extraction was used to obtain the extract (CPE). Cytotoxicity and immunostimulation ability were evaluated in macrophages, and the in vitro trypanocidal activity was evaluated against epimastigotes and trypomastigotes forms. In vivo tests were done by infecting BALB/c mice with blood trypomastigotes forms and treating animals orally with CPE for 10 days. The parasitemia, survival rate, weight, cytokines and nitric oxide dosage were evaluated. CPE demonstrated an effect on the epi and trypomastigotes forms of the parasite (IC50 17.90 ± 1.2 µg/mL; LC50 26.73 ± 1.2 µg/mL) and no changes in macrophages viability, resulting in a selectivity index similar to the reference drug. CPE-treated animals had a worsening compared to non-treated, demonstrated by higher parasitemia and lower survival rate. This result was attributed to the anti-inflammatory effect of CPE, demonstrated by the higher IL-10 and IL-4 values observed in the treated mice compared to the control ones. CPE demonstrated a trypanocidal effect in vitro and a worsening in the in vivo infection due to its anti-inflammatory activity.

ABSTRACT

This paper presents novel current sensorless maximum-power point-tracking (MPPT) algorithms based on compensators/controllers and a single-input voltage sensor. The proposed MPPTs eliminate the expensive and noisy current sensor, which can significantly reduce the system cost and retain the advantages of the widely used MPPT algorithms, such as Incremental Conductance (IC) and Perturb and Observe (P&O) algorithms. Additionally, it is verified that the proposed algorithms, especially the proposed Current Sensorless V based on PI, can reach outstanding tracking factors (TFs) such as the IC and P&O based on PI algorithms. In this sense, the insertion of controllers inside the MPPT gives them adaptive characteristics, and the experimental TFs are in the remarkable range of more than 99%, with an average yield of 99.51% and a peak of 99.80%.

ABSTRACT

Failure by fatigue can be sudden and catastrophic. Therefore, ensuring that dental implants, which are under constant cyclic loading, do not fail to fatigue is imperative. The majority of the studies about the topic only performed in vitro tests, which are expensive and time-consuming. The Finite Element (FE) method is less costly and it allows the simulation of several different loading scenarios. Nonetheless, there are only a few studies analysing fatigue in dental prostheses using FE models, and the few available did not include all the relevant parameters, such as geometry effect, surface finishing, etc. Therefore, this study aimed to analyse the fatigue behaviour of a single-unit dental implant with two screws using a combination of the numerical results and the traditional fatigue criteria - a combination that was not yet fully and correctly explored. A finite element model comprising a single implant, one abutment, one abutment screw, one fixation screw and one prosthetic crown was developed. Material properties were assigned based on literature data. A 100 N load was applied to mimic the mastication forces and fatigue analysis was conducted using the Gerber, Goodman and Soderberg fatigue criteria. The fatigue analysis demonstrated that the abutment screw could fail in less than 1 year, depending on the criteria, while the fixation screw exhibits an infinite life. The results illustrated the importance of analysing the fatigue behaviour of dental implants and highlighted the potential of finite element models to simulate the biomechanical behaviour of dental implants.

This study presents a methodology that combines experimental tests and the finite element method, which is able to analyse the influence of the geometry on the mechanical behaviour of stents made of bioabsorbable polymer PLA (PolyLactic Acid) during their expansion in the treatment of coarctation of the aorta (CoA). Tensile tests with standardized specimen samples were conducted to determine the properties of a 3D-printed PLA. A finite element model of a new stent prototype was generated from CAD files. A rigid cylinder simulating the expansion balloon was also created to simulate the stent opening performance. A tensile test with 3D-printed customized stent specimens was performed to validate the FE stent model. Stent performance was evaluated in terms of elastic return, recoil, and stress levels. The 3D-printed PLA presented an elastic modulus of 1.5 GPa and a yield strength of 30.6 MPa, lower than non-3D-printed PLA. It can also be inferred that crimping had little effect on stent circular recoil performance, as the difference between the two scenarios was on average 1.81%. For an expansion of diameters ranging from 12 mm to 15 mm, as the maximum opening diameter increases, the recoil levels decrease, ranging from 10 to 16.75% within the reported range. These results point out the importance of testing the 3D-printed PLA under the conditions of using it to access its material properties; the results also indicate that the crimping process could be disregarded in simulations to obtain fast results with lower computational cost and that new proposed stent geometry made of PLA might be suitable for use in CoA treatments-the approach that has not been applied before. The next steps will be to simulate the opening of an aorta vessel using this geometry.

PURPOSE: To evaluate the influence of different surface treatments on the clinical behavior of non-carious cervical sclerotic lesions (NCCLs) over an 18-month follow-up period. METHODS: 128 NCCLs from 32 volunteers were randomized into four groups (n=32): G1-control, without preoperative treatment of the dentin surface; G2, dentin conditioning with 17% ethylenediaminetetracetic acid (EDTA) for 2 minutes; G3, increase in dentin surface roughness with diamond bur and G4, increase in dentin surface roughness with diamond bur + dentin conditioning with 17% EDTA for 2 minutes. RESULTS: Differences between groups were tested using the Friedman test (α= 0.05). A questionnaire was administered to volunteers about risk factors related to NCCLs. The relationship between the questionnaire data and the clinical performance of the restorations was analyzed using the multiple logistic regression test (α= 0.05). The variables related to parafunctional habits, anxiety and/or depression were significantly related to the manifestation of postoperative sensitivity. Roughening the sclerotic dentin with a diamond bur increased postoperative sensitivity within 12 months. The presence of parafunctional habits and anxiety/depression may lead to postoperative sensitivity. CLINICAL SIGNIFICANCE: Roughening the sclerotic dentin with a diamond bur increased postoperative sensitivity within 12 months.

ABSTRACT

The present study describes a new species of Henneguya infecting the ornamental fish Caquetaia spectabilis from the Brazilian Amazon. Fish specimens were collected where the Tapajós and Amazon rivers merge, municipality of Santarém in the State of Pará, Brazil. Infections were intense, with several plasmodia spread on the opercula, fins and eye. Phylogenetic characterization and host-parasite relationship studies of the new Henneguya species used a combination of small subunit ribosomal DNA (ssrDNA) and morphological (photonic and transmission electron microscopy) analyses. Plasmodia were white round to ellipsoidal measuring up to 1.8 mm. The myxospores body measured 20.5 ± 3.9 (15-27) in length, 7.9 µm (6.2-10.8) in width, 6.7 µm (6.0-7.6) in thickness, 20.5 µm (14.4-32.3) in caudal appendages length, and 40.6 µm (34.2-54.6) in total length. The two polar capsules were elongated and equal in size, measuring 4.3 µm (3.3-5.4) in length and 2.1 µm (1.3-2.8) in width. Histological analysis revealed the parasite development in connective tissues of the fins, eyes and opercula. The skin of the fins and opercula presented detachment of the epidermis, however, no inflamatory infiltrate was observed. In the eye were observed inflammatory infiltratate in the epithelium and stroma of the cornea. Ultrastructure analysis showed the connective tissue capsule composed by an inner cellular layer with fibroblasts and outer layer where collagen fibers arranged transversely yet interspersed by layers of fibers arranged longitudinally. Numerous invaginations and extensive pinocytotic channels were observed in the plasmodial membrane. A layer of microfilament-like microfilament-like material was observed in the ectoplasm area and along to the internal surface of the plasmodial membrane. Generative cells and early stages of sporogenesis were seen more internally. The ssrDNA based phylogeny showed the South American species grouped in two lineages and the new species arises in a well-sustained subclade as sister branch of the clade composed by Henneguya spp. parasites of cichlids fish.

This paper presents the development, analysis, and application of chirped fiber Bragg gratings (CFBGs) for dynamic and static measurements of beams of different materials in the single-cantilever configuration. In this case, the beams were numerically analyzed using the finite-element method (FEM) for the assessment of the natural frequencies and vibration modes of the beam for the dynamic analysis of the structural element. Furthermore, the static numerical analysis was performed using a load at the free end of the beam, where the maximum strain and its distribution along the beam were analyzed, especially in the region at which the FBG was positioned. The experimental evaluation of the proposed CFBG sensor was performed in static conditions for forces from 0 to 50 N (in 10 N steps) applied at the free end of the beam, whereas the dynamic evaluation was performed by means of positioning an unbalanced motor at the end of the beam, which was excited at 16 Hz, 65 Hz, 100 Hz, and 131 Hz. The results showed the feasibility of the proposed device for the simultaneous assessment of the force and strain distribution along the CFBG region using the wavelength shift and the full-width at half-maximum (FWHM), respectively. In these cases, the determination coefficients of the spectral features as a function of the force and strain distribution were higher than 0.99 in all analyzed cases, where a potential resolution of 0.25 N was obtained on the force assessment. In the dynamic tests, the frequency spectrum of the sensor responses indicated a frequency peak at the excited frequency in all analyzed cases. Therefore, the proposed sensor device is a suitable option to extend the performance of sensors for structural health assessment, since it is possible to simultaneously measure different parameters in dynamic and static conditions using only one sensor device, which, due to its multiplexing capabilities, can be integrated with additional optical fiber sensors for the complete shape reconstruction with millimeter-range spatial resolution.

ABSTRACT

Due to their high specificity, monogenoids from fish provide an interesting model to study historical associations of hosts and parasites. High agreement between host and parasite phylogeny is often interpreted as evidence of cospeciation. However, cophylogenetic signal may also arise from other, either adaptive or non-adaptive, processes. We applied the recently developed Cophylospace Framework to better understand the evolutionary relationship between monogenoids and marine catfish from the Atlantic coast of South America. The associations between 12 marine catfish and 10 monogenoid species were assessed. Molecular data of host and parasite species were used for phylogenetic reconstruction. We used anchor morphology based on Procrustes coordinates to evaluate whether closely related hosts are associated with morphologically similar parasites. To assess the association between parasite phylogeny and host morphology, we produced a distance matrix based on morphological characters of catfishes. Agreement between phylogenies and between phylogeny and morphology was measured using Procrustes R2 computed with PACo. The parasite phylogeny obtained in this study represents the first complete phylogenetic hypothesis of monogenoids parasitizing ariids from South America. The Cophylospace analysis suggested that phylogenetic and morphological distance of monogenoids contributes similarly to explain the pattern of host­parasite associations, whereas parasite phylogeny is more strongly associated with the morphological traits of the hosts than with host phylogeny. This evidence suggests that cospeciation is not a major force accounting for diversification in the monogenoids studied. Rather host morphological traits seem to be a more important driver, which conforms with evidence from other host‒monogenoid systems.

ABSTRACT

The present study investigated by palynological and chemical analysis (Flame Atomic Absortion Spectrometry) about the botanical origin and the heavy metals content (arsenic, cadmium, chromium, lead and mercury) of monthly honey samples of Apis mellifera L. over two years. The pollen types Apiaceae, Mimosa caesalpiniifolia, M. tenuiflora and Myrcia indicated the main floristic sources used by bees. M. tenuiflora was the most frequent of the pollen types, and because it predominates in different months in each year, which may indicate more than one species of the genus being foraged by the beehive. The climatic influence (rainfall and temperature) on the pollen diversity was investigated and was not statistically supported. The chemical analysis showed that the heavy metal content of the samples were below their respective limits of quantification, and, therefore, the samples can be considered safe for human consumption.

ABSTRACT

PURPOSE: Henneguya Thélohan, 1892 is one of the most species-rich genera of myxosporean parasites and infects fish around the world. The present study describes a new species infecting the gill filaments, fins, and kidneys of Plagioscion squamosissimus (Heckel, 1840), an economically important freshwater fish distributed in watersheds in the north of South America. METHODS: A total of 108 P. squamosissimus specimens were examined from three geographic localities in the Amazon basin: the Lago Grande do Curuai, a marginal lake of the Amazon River; the Tapajós River, in the state of Pará; and the Solimões River, in the state of Amazonas, Brazil. The analyses were based on the myxospore morphology, ribosomal DNA sequencing, phylogeny, prevalence, and geographic distribution of the host and its parasite. RESULTS: Parasite prevalences were 50% in both the Tapajós and Solimões rivers, and 35.4% in the Lago Grande do Curuai. In terms of the site of infection, the prevalence total was 23.1% in the gill filament, 29.6% in the fins, and 1.8% in the kidney. Regarding gender, the prevalence was 59.5% for males, 32.5% for females, and 21.7% for undetermined sex. The specimens found here were both morphologically and molecularly identical regardless of the infected organ and geographic locality, but distinct from all other Henneguya species, revealing that the parasite reported represents a novel species named Henneguya longisporoplasma n. sp. Despite the sampling being carried out in three different geographic localities of the Amazon basin, no population-level genetic variation was observed, even in the typically more variable ITS-1 region, revealing a panmictic population of H. longisporoplasma n. sp. in this large watershed. Maximum likelihood and Bayesian analyses showed the novel Henneguya clustered as a sister branch of the subclade formed of Henneguya that infect fish belonging to the family Cichlidae. CONCLUSIONS: A novel Henneguya species was identified parasitizing P. squamosissimus. The parasite presented wide geographic distribution in the Amazon basin and genetic analyses showed it as revealing a panmictic population.

ABSTRACT

This paper presents the development, analysis and application of a fiber Bragg grating (FBG) array for two-dimensional (2D) shape reconstruction in a cantilever beam. The structural elements made of Pinus wood and Nylon 6.0 were numerically analyzed using the finite element method for the strain distribution when constant loading is applied at the free end of the beam. In addition, the temperature compensation method is proposed to decouple the temperature cross-sensitivity in the deflection analysis. In this case, the temperature sensitivities of all sensing elements of the 5-FBG array were obtained. An additional FBG was encapsulated in a silicone mold for increased sensitivity and positioned in the clamping point in which deflection was negligible. Temperature compensation was achieved considering the temperature measured by the silicone-embedded FBG (sensitivity of 27.78 pm/°C) and the sensitivity of all five FBGs of the deflection-sensing array (9.14 pm/°C ± 0.33 pm/°C). In the deflection experiments, the sensors presented a high linearity, in which a determination coefficient (R2) higher than 0.995 was obtained in all of the analyzed cases. Furthermore, the 2D shape construction using the proposed sensor approach resulted in the elastic line estimation for all analyzed beams, where the experimental results were in agreement with the theoretical and numerical analysis with a R2 higher than 0.99 in all of the analyzed cases. Therefore, the proposed sensor array is a feasible approach for real-time shape reconstruction of structural elements with the advantages related to the possibility of direct embedment in the measured structure.

While around world, species of the genus Ceratomyxa parasite majority marine hosts, growing diversity has been reported in South American freshwater fish. The present study reports Ceratomyxa barbata n. sp. parasitizing the gallbladder of the Rhaphiodon vulpinus fish from the Amazon and La Plata basins. Morphological (light and transmission electron microscopy), molecular (sequencing of small subunit ribosomal DNA - SSU rDNA), and phylogenetic analyses were used to characterize the new species. Worm-like plasmodia endowed with motility were found swimming freely in the bile. The myxospores were elongated, lightly arcuate, with rounded ends and had polar tubules with 3 coils in the polar capsules. Ultrastructural analysis revealed plasmodia composed of an outer cytoplasmic region, where elongated tubular mitochondria, a rough endoplasmic reticulum, sporogonic stages, and a large vacuole occupying the internal area were observed. Phylogenetic analysis, based on SSU rDNA, found that among all South America freshwater Ceratomyxa species, C. barbata n. sp. arises as an earlier divergent species. The present study reveals the occurrence of this host-parasite system (R. vulpinus/C. barbata n. sp.) in the two largest watersheds on the continent.
