ABSTRACT

Biofluorescence in echinoderms is largely unexplored, and even though the green sea urchinStrongylocentrotus droebachiensisis a well-studied species, the presence and/or function of fluorescence remains very poorly understood. Hyperspectral imaging was conducted on adult sea urchins (N = 380) while fluorospectrometric analysis was conducted on sea urchin coelomic fluid (N = 30). Fluorescence was documented in both the spines and coelomic fluid ofS. droebachiensis. Intact spines exhibited a low intensity green emission (∼550-600 nm), while broken spines averaged a high emission peak in the green spectrum (∼580 nm). Sea urchins produce a red exudate with a pronounced emission peak (∼680 nm) with a shoulder peak (∼730 nm). The sampled coelomic fluid exhibited high variability, with a majority exhibiting a low-level green fluorescence while pronounced emission peaks (N = 5) were found in the red spectrum (∼680 nm). The complex fluorescence produced byS. droebachiensiswarrants further investigation on its applicability for monitoring welfare of sea urchins in aquaculture facilities.

ABSTRACT

The advanced development of sensor technologies has led to the emergence of fish biosensors that are currently used for research and commercial purposes. AEFishBIT is a miniaturized biosensor attached to fish operculum that measures physical activity and respiration frequencies. In this study, we determined the effect of the tagging method and evaluated the use of this biosensor to monitor post-smolt Atlantic salmon in a tank-based system. The use of piercing fish tag had a negative impact on the gills and operculum, unlike the identical protocols used in gilthead sea bream and European sea bass. In contrast, a surgical thread did not show any apparent tissue damage. Two data recording schedules were considered to monitor immediate early reaction to fish handling and light regime changes (records every 15 min over 2 days) or adaptation to new light conditions (records every 30 min over 4 days). Data showed stabilization of physical activity 8 h post-tagging, with different steady states for the activity/respiratory ratio after changes in light intensity that reflected a different time course adaptation to new light conditions. High correlations were observed between AEFishBIT and video recording data. These findings supported the use of AEFishBIT as a promising tool for smart sensing of Atlantic salmon.

ABSTRACT

Individuals in a fish population differ in key life-history traits such as growth rate and body size. This raises the question of whether such traits cluster along a fast-slow growth continuum according to a pace-of-life syndrome (POLS). Fish species like salmonids may develop a bimodal size distribution, providing an opportunity to study the relationships between individual growth and behavioural responsiveness. Here we test whether proactive characteristics (bold behaviour coupled with low post-stress cortisol production) are related to fast growth and developmental rate in Atlantic salmon, Salmo salar. Boldness was tested in a highly controlled two-tank hypoxia test were oxygen levels were gradually decreased in one of the tanks. All fish became inactive close to the bottom at 70% oxygen saturation. At 40% oxygen saturation level a fraction of the fish actively sought to avoid hypoxia. A proactive stress coping style was verified by lower cortisol response to a standardized stressor. Two distinct clusters of bimodal growth trajectories were identified, with fast growth and early smoltification in 80% of the total population. There was a higher frequency of proactive than reactive individuals in this fast-developing fraction of fish. The smolts were associated with higher post-stress plasma cortisol than parr, and the proactive smolts leaving hypoxia had significant lower post-stress cortisol than the stayers. The study demonstrated a link between a proactive coping and fast growth and developmental ratio and suggests that selection for domestic production traits promotes this trait cluster.
