ABSTRACT

Our objectives for this study were to understand the biological basis of meat tenderness and to provide an overview of the gene expression profiles related to meat quality as a tool for selection. Through deep mRNA sequencing, we analyzed gene expression in muscle tissues of two Italian cattle breeds: Maremmana and Chianina. We uncovered several differentially expressed genes that encode for proteins belonging to a family of tripartite motif proteins, which are involved in growth, cell differentiation and apoptosis, such as TRIM45, or play an essential role in regulating skeletal muscle differentiation and the regeneration of adult skeletal muscle, such as TRIM32. Other differentially expressed genes (SCN2B, SLC9A7 and KCNK3) emphasize the involvement of potassium-sodium pumps in tender meat. By mapping splice junctions in RNA-Seq reads, we found significant differences in gene isoform expression levels. The PRKAG3 gene, which is involved in the regulation of energy metabolism, showed four isoforms that were differentially expressed. This distinct pattern of PRKAG3 gene expression could indicate impaired glycogen storage in skeletal muscle, and consequently, this gene very likely has a role in the tenderization process. Furthermore, with this deep RNA-sequencing, we captured a high number of expressed SNPs, for example, we found 1462 homozygous SNPs showing the alternative allele with a 100% frequency when comparing tender and tough meat. SNPs were then classified into categories by their position and also by their effect on gene coding (174 non-synonymous polymorphisms) based on the available UMD_3.1 annotations.

ABSTRACT

Chianina and Maremmana breeds play an important role in the Italian cattle meat market. The Chianina breed is an ancient breed principally raised for draught. Now this breed is the worldwide recognized producer of top quality beef, tasteful and tender, specifically the famous "Florentine steak". The Maremmana characterized by a massive skeletal structure, is a rustic cattle breed selected for adaptability to the marshy land of the Maremma region. We used a high throughput mRNA sequencing to analyze gene expression in muscle tissues of two Italian cattle breeds, Maremmana (MM) and Chianina (CN) with different selection history. We aim to examine the specific genetic contribution of each breed to meat production and quality, comparing the skeletal muscle tissue from Maremmana and Chianina. Most of the differentially expressed genes were grouped in the Glycolysis/Gluconeogenesis pathways. The rate and the extent of post-mortem energy metabolism have a critical effect on the conversion of muscle to meat. Furthermore, we aim at discovering the differences in nucleotide variation between the two breeds which might be attributable to the different history of selection/divergence. In this work we could emphasize the involvement of pathways of post-mortem energy metabolism. Moreover, we detected a collection of coding SNPs which could offer new genomic resources to improve phenotypic selection in livestock breeding program.

ABSTRACT

The antibacterial activity of an ethylacetate neem cake extract (NCE) against bacteria that affect meat quality, namely Campylobacter jejuni, Carnobacterium spp., Lactobacillus curvatus, Lactobacillus sakei and Leuconostoc sp., is reported. The antibacterial activity was detected using standardised disc diffusion and macrodilution methods. The bacterial growth inhibition zone ranged from 11.33 ± 0.58 to 22.67 ± 0.58 mm (100 µL NCE). There is significant difference between the growth inhibition zone of NCE and the control (ciprofloxacin 100 µg). The percent of bacterial growth reduction range was 79.75 ± 1.53 to 90.73 ± 1.53 (100 µg NCE) as compared with control (without NCE). NCE in different amounts counteracted the growth of all tested bacteria.

ABSTRACT

Four different extraction-derivatization methods commonly used for fatty acid analysis in meat (in situ or one-step method, saponification method, classic method and a combination of classic extraction and saponification derivatization) were tested. The in situ method had low recovery and variation. The saponification method showed the best balance between recovery, precision, repeatability and reproducibility. The classic method had high recovery and acceptable variation values, except for the polyunsaturated fatty acids, showing higher variation than the former methods. The combination of extraction and methylation steps had great recovery values, but the precision, repeatability and reproducibility were not acceptable. Therefore the saponification method would be more convenient for polyunsaturated fatty acid analysis, whereas the in situ method would be an alternative for fast analysis. However the classic method would be the method of choice for the determination of the different lipid classes.

ABSTRACT

Calpain system (µ- and m-calpain, calpastatin) and myofibril fragmentation index (MFI) were analysed on muscle longissimus thoracis et lumborum of 16 male fallow deer, half 18- and half 30-month-old, slaughtered after a 4-month feeding trial on pasture alone or with concentrate supplement. Diet did not produce any difference on the measured variables. The muscle of the elder deer had a higher calpastatin level 6 h post-mortem (136.0 vs. 99.7 units/50 g, for the 30- and 18-months-old, respectively), and a lower MFI value, particularly 24 h post-mortem (71.1 vs. 89.1). This is in agreement with previous data showing tougher meat from 30-month-old animals.

ABSTRACT

We present the new host molecule 1 which binds basic amino acid esters in water. It recognizes both positively charged groups of the amino acid esters by electrostatic and hydrogen bond interactions with its four strategically placed phosphonate anions. Selectivity for lysine is achieved by the correct distance between both bisphosphonate pairs. By contrast, the smaller amino acid esters arginine, ornithine, and histidine form 2:1 complexes with 1. In methanol, a double chelate assembly enforced by pi-cation interactions with the imidazolium cation leads to a very high association constant for the 1:histidine complex of 3 x 10(4) M(-)(1).

ABSTRACT

[structure: see text] We provide access to a new class of C1- or C2-symmetrical host molecules 1 and 2 based on a spirobisindane skeleton. Whereas 1 is selective for short, rigid diamines, 2 prefers longer alpha,omega-dications. Of all the amino acid methyl esters, only those of lysine and arginine with the correct distance between their cationic groups form strong 1:1 complexes in DMSO with 2. NMR titrations reveal high association constants as well as discrimination between the enantiomers of lysine and arginine.

Systemic lupus erythematosus (SLE) is a chronic illness that has no known cause. As an autoimmune, inflammatory disease, it can affect various organs in different ways. Because of its fluctuating, unpredictable qualities, it requires the implementation of physiologic and psychosocial interventions that may change daily. A plan of care unique to each individual with SLE is essential to its management.

ABSTRACT

Approximately 58% of the RN nursing programs in the United States are associate degree programs (ADN). The curricula of these programs are designed to prepare graduates to competently provide direct patient care. The purpose of this descriptive comparative study was to evaluate the nursing performance of ADN graduates as perceived by graduates, faculty, and employers. In the first phase of the study, graduates and faculty completed the Six-Dimension Scale of Nursing Performance (6-DSNP) at the time of graduation. Six months later, the graduates again were asked to complete the same questionnaire along with their employers. Findings indicated a significant difference in the planning and evaluation performance of graduates at graduation and 6 months later as perceived by the graduates. Graduates rated themselves significantly higher in all areas of performance when compared to faculty and employers. There was no significant difference in the perception of graduate performance between faculty and employers. This outcome assessment provides needed data on perceptions of ADN programs.

ABSTRACT

Children with special health care needs pose many challenges for community-based providers. Programs such as school, child care, and Head Start provide services to children that previously would have been delivered in hospitals or at home. The need for a system of care to assure safe, quality services for these children is paramount. While health procedure manuals for schools have been developed, few recommendations address the actual training of providers or the trainers of providers. This article describes a system of training which includes steps in managing the care of children with special health care needs, levels of training, and expected competencies for the care providers of children with special health care needs.

ABSTRACT

The intraocular pressure (IOP) of nine cadaver eyes was set by a manometer at 10 mm Hg increments from 10 to 50 mm Hg. Pressure measurements at each of these settings were taken using the Digilab Pneumatonometer and Tono-Pen to determine the accuracy of these instruments. In addition, four brands of therapeutic contact lenses were placed on the eyes, and IOPs were measured through them to determine whether or not comparable IOPs could be obtained through a range of bandage contact lenses. We found a significant difference (p < or = 0.0000) between the measurements obtained by the two instruments at a given manometric setting. Digilab Pneumatonometer's pressures correlated well with manometric pressures. Tono-Pen consistently underestimated manometric pressures. At 50 mm Hg, Digilab Pneumatonometer's mean measurement was 47.5 mm Hg, whereas Tono-Pen's mean was only 38.9 mm Hg. IOPs assessed through all contact lenses were comparable to the measurements without lenses. Multiple regression of score showed that the measured pressure was explained by manometric pressure (81%), the choice of instrument used (5.5%), and the lens thickness (0.09%). The Pierson correlation coefficient was 0.93. This study showed that Pneumatonometer could accurately measure IOP with and without a therapeutic contact lens, but Tono-Pen was equally inaccurate with and without lenses, giving false low measurements.
