ABSTRACT

Current climate projections for mid-latitude regions globally indicate an intensification of wind-driven coastal upwelling due to warming conditions. The dynamics of mid-latitude coastal upwelling are marked by environmental variability across temporal scales, which affect key physiological processes in marine calcifying organisms and can impact their large-scale distribution patterns. In this context, marine invertebrates often exhibit phenotypic plasticity, enabling them to adapt to environmental change. In this study, we examined the physiological performance (i.e., metabolism, Thermal Performance Curves, and biomass and calcification rates) of individuals of the intertidal mollusk Chiton granosus, a chiton found from northern Peru to Cape Horn (5° to 55°S). Our spatial study design indicated a pattern of contrasting conditions among locations. The Talcaruca site, characterized by persistent upwelling and serving as a biogeographic break, exhibited lower pH and carbonate saturation states, along with higher pCO2, compared to the sites located to the north and south of this location (Huasco and Los Molles, respectively). In agreement with the spatial pattern in carbonate system parameters, long-term temperature records showed lower temperatures that changed faster over synoptic scales (1-15 days) at Talcaruca, in contrast to the more stable conditions at the sites outside the break. Physiological performance traits from individuals from the Talcaruca population exhibited higher values and more significant variability, along with significantly broader and greater warming tolerance than chitons from the Huasco and Los Molles populations. Moreover, marked changes in local abundance patterns over three years suggested population-level responses to the challenging environmental conditions at the biogeographic break. Thus, C. granosus from the Talcaruca upwelling zone represents a local population with wide tolerance ranges that may be capable of withstanding future upwelling intensification on the Southern Eastern Pacific coast and likely serving as a source of propagules for less adapted populations.

ABSTRACT

Environmental contamination by chromium represents a serious public health problem. Therefore, it is crucial to develop and optimize remediation technologies to reduce its concentration in the environment. The aims of this study were to evaluate the uptake of chromium by live and complete microbial mats in experimental mesocosms under different pH and salinity conditions to understand how these factors affect the microphytobenthic community and, consequently, how chromium removal process is influenced. Microbial mats from the estuarine environment were exposed to 15 mg Cr/L under different pH (2, 4, and 8) and salinity (2, 15, and 33) conditions. Salinity, redox potential, and pH were measured throughout the trial in solutions and in microbial mats, while total Cr determinations were performed at the end of the assay. The results demonstrated that the removal efficiency of Cr by microbial mats was significantly improved in solutions at pH 2, remaining unaffected by variations in salinity. Notably, both cyanobacteria and diatoms showed remarkable resistance to Cr exposure under all conditions tested, highlighting their exceptional adaptability. Microbial mats have proved to be effective filters for reducing the concentration of chromium in aqueous solutions with varying pH and salinity levels.

ABSTRACT

This study addresses the limited understanding of chromium-microbial mat interactions in estuarine tidal flats. The aims were to evaluate (1) the efficiency of the microbial consortium in Cr(III) removal from seawater; (2) the elemental and mineralogical composition of the microbial mat as a natural system in the Cr removal, (3) the effects of metal on microphytobenthos, and (4) possible interactions of Cr with other metals present in the consortium. Microbial mats were exposed to Cr(III) solutions at different concentrations (2-30 mg Cr/L). Analysis such as metal concentration, organic matter content, chlorophyll a and phaeopigment concentrations, abundance of diatoms and cyanobacteria, SEM-EDS, and XRD were performed. Most of the Cr(III) was deposited, as chromium oxide/hydroxide, on the surface of all microbial mat components. The complete microbial mat, comprising sediments, detritus, EPS, and diverse microorganism communities, exhibited a remarkable capacity to accumulate Cr(III), retaining over 87% in the solution.

ABSTRACT

Abstract The objective of this work was to compare the performance of treponemal and non-treponemal screening in two periods close in time in blood donors and to analyse the association of the signal strength (signal-to-cutoff, S/ CO) of the chemiluminescent immunoassay (CIA) with the reactivity of the enzymeimmunoassay (EIA) and the rapid plasma reagin (RPR). Donors were screened with treponemal tests. The distribution of the S/CO values obtained by CIA was analysed based on EIA and RPR, and its association was evaluated. Group 1: ELISA+/RPR+ donors and Group 2: ELISA+/RPR-. A total of 76,794 voluntary donations were processed. Comparing the means between groups, it was found that the CIA S/CO ratio was significantly higher for Group 1 donors (19.5 vs. 8.10; p<0.001). In conclusion, the signal strength of the qualitative CIA test would be associated with the reactivity of the RPR and would be related to the course of the infection.

ABSTRACT

Microbial mats have a special role in the removal of metals in marine environment. This study aimed to experimentally determine the efficiency of Cr removal from seawater by microbial mats. The effect of Cr on the microphytobenthic community and the influence of an aerated condition on the removal of metal and microorganisms were also considered. Hence, microbial mat samples were subdivided in 4 groups: submerged in Cr (chromium 2 mg/L without aeration), Cr + O2 (chromium 2 mg/L with aeration), SW + O2 (filtered seawater with aeration), and a control: SW (filtered seawater with neither chromium nor aeration). Water and microbial mat subsamples were used to determine Cr concentrations, organic matter content, granulometry, physicochemical parameters, chlorophyll a, phaeopigments, and quantitative analysis of the microphytobenthic community. Cr removal efficiency from seawater was 95% for the Cr treatment and 99% for the Cr + O2 treatment. The abundance of cyanobacteria tended to decrease between the initial and final days of the assay, whereas the opposite trend was observed for diatoms. Two aspects are worth noting in the paper: microbial mats efficiently removed Cr from seawater at a concentration of 2 mg Cr/L and the removal of Cr by microbial mats was more efficient with water aeration.

ABSTRACT

Several vaccines have been developed to control the COVID-19 pandemic. CoronaVac®, an inactivated SARS-CoV-2 vaccine, has demonstrated safety and immunogenicity, preventing severe COVID-19 cases. We investigate the safety and non-inferiority of two immunization schedules of CoronaVac® in a non-inferiority trial in healthy adults. A total of 2302 healthy adults were enrolled at 8 centers in Chile and randomly assigned to two vaccination schedules, receiving two doses with either 14 or 28 days between each. The primary safety and efficacy endpoints were solicited adverse events (AEs) within 7 days of each dose, and comparing the number of cases of SARS-CoV-2 infection 14 days after the second dose between the schedules, respectively. The most frequent local AE was pain at the injection site, which was less frequent in participants aged ≥60 years. Other local AEs were reported in less than 5% of participants. The most frequent systemic AEs were headache, fatigue, and myalgia. Most AEs were mild and transient. There were no significant differences for local and systemic AEs between schedules. A total of 58 COVID-19 cases were confirmed, and all but 2 of them were mild. No differences were observed in the proportion of COVID-19 cases between schedules. CoronaVac® is safe, especially in ≥60-year-old participants. Both schedules protected against COVID-19 hospitalization.

ABSTRACT

Every year the concern from environmental pollution increases as it affects human health. In the current paper, it is analyzed the evolution of the air quality in different cities on international level related to the reduction in mobility trend due to the emergence of the pandemic. The air pollution is studied in order to achieve healthier cities in the future and create an awareness for the citizens. The COVID-19 pandemic situation was declared during the first months of 2020 and has shown (indirectly) the effects that restrictions on human activity and road traffic have on atmospheric pollutant values. This pandemic condition derived, in most cases, in mobility restrictions in order to decrease the spread of the virus through the density of population in different places and, as a result of these restrictions, pollutant presence decreased equally. This article analyzes some of the largest cities around the world, such as Tsuen Wan district in Hong Kong (China), Los Angeles (USA), London (UK), São Paulo (Brazil), Bangalore (India), Johannesburg (South Africa), and Sydney (Australia) in order to check the different mobility restriction policies established in each location, and how those restrictions led to decrease the levels of some pollutant particles such as NO2 in the atmosphere. This should serve as a focus of awareness of the city and political interests of how it affects human activity, and those measures should be taken to reduce pollutants.

ABSTRACT

Rifampicin is one of the most commonly used antibiotics for treating tuberculosis, but shows low bioavailability and requires long-term administration, and hence its use may result in severe side effects. Encapsulation of rifampicin in polymeric reservoirs allows it to be administered locally and improves its pharmacological action. High rifampicin loading is crucial for obtaining an adequate therapeutic effect. Generally, the drug loading is a complex function of reservoir fabrication parameters. In the current work, we systematically varied the drug (rifampicin), polymer (PLGA) and dispersed phase contents as well as the solvent evaporation rate, particle size and number of particle washing cycles to characterize the challenges involved in encapsulating rifampicin. Physical insight into the low encapsulation efficiencies was provided, as well as an optimization of fabrication conditions to achieve higher drug loading levels. The particle solidification stage was found in the current work to be the most crucial step, where a significant amount of rifampicin was lost enhanced by its solubility in the aqueous medium. Increases in polymer concentration, solvent evaporation rate and particle size each significantly improved the drug loading by hindering of solvent-assisted escape of the drug. Based on our observation of the drug loading being extremely sensitive to the particle recovery and washing procedure after the solvent evaporation, most of the encapsulated rifampicin was concluded to be located on or very near the reservoir surface. Encapsulation could be significantly improved by fabricating multiple emulsions, especially double w/o/w emulsions, but the resultant particles were relatively large and porous, which might be a drawback for drug administration.

ABSTRACT

The process of population aging will lead to an increase in health problems in older people, mainly related to their functionality. Accordingly, the countries of the Region of the Americas must begin to act to meet this challenge. One of the fundamental tasks involves the ability to measure and monitor functionality in the population. The tools used to evaluate functionality focus mainly on the basic activities of daily living, which limits their preventive capacity, since instrumental activities provide a better prognosis. Using the case of Chile, challenges for the Region were identified, mainly related to the advantages of adopting new methodologies that not only evaluate functional capacity, as current strategies do, but also detect its deterioration early and monitor its stages throughout the dependence stage in older people. This will also make it possible to evaluate initiatives for the prevention and management of loss of functionality.

ABSTRACT

Ocean acidification (OA) is expected to rise towards the end of the 21st century altering the life history traits in marine organisms. Upwelling systems will not escape OA, but unlike other areas of the ocean, cooling effects are expected to intensify in these systems. Regardless, studies evaluating the combined effects of OA and cooling remain scarce. We addressed this gap using a mesocosm system, where we exposed juveniles of the intertidal muricid snail Acanthina monodon to current and projected pCO2 (500 vs. 1500 ppm) and temperature (15 vs. 10 °C) from the southeast Pacific upwelling system. After 9 weeks of experimental exposure to those conditions, we conducted three estimations of growth (wet weight, shell length and shell peristomal length), in addition to measuring calcification, metabolic and feeding rates and the ability of these organisms to return to the normal upright position after being overturned (self-righting). Growth, feeding and calcification rates increased in projected cooling conditions (10 °C) but were unaffected by pCO2 or the interaction between pCO2 and temperature. Instead, metabolic rates were driven by pCO2, but a significant interaction with temperature suggests that in cooler conditions, metabolic rates will increase when associated with high pCO2 levels. Snail self-righting times were not affected across treatments. These results suggest that colder temperatures projected for this area would drive this species growth, feeding and calcification, and consequently, some of its population biology and productivity. However, the snails may need to compensate for the increase in metabolic rates under the effects of ocean acidification. Although A. monodon ability to adjust to individual or combined stressors will likely account for some of the changes described here, our results point to a complex dynamic to take place in intertidal habitats associated with upwelling systems.

ABSTRACT

Widespread intertidal mussels are exposed to a variety of natural and anthropogenic stressors. Even so, our understanding of the combined influence of stressors such as predation risk and ocean acidification (OA) on these species remains limited. This study examined the response of the purple mussel (Perumytilus purpuratus), a species distributed along Pacific southeastern rocky shores, to the effects of predation risk and OA. Using a laboratory 2 × 2 cross design, purple mussels were either devoid or exposed to predator cues from the muricid snail Acanthina monodon, while simultaneously exposing them to current (500 ppm) or projected OA conditions (1500 ppm). The response of purple mussels to these factors was assessed using growth, calcification, clearance, and metabolic rates, in addition to byssus production. After 60 d, the presence of predator cues reduced mussel growth in width and length, and in the latter case, OA enhanced this response making the effects of predator cues more severe. Calcification rates were driven by the interaction between the two stressors, whereas clearance rates increased only in response to OA, likely explaining some of the growth results. Mussel byssus production also increased with pCO2 but interacted with predation risk: in the absence of predator cues, byssus production increased with OA. These results suggest that projected levels of OA may alter and in some cases prevail over the natural response of purple mussels to predation risk. Considering the role played by this mussel as a dominant competitor and ecosystem engineer in rocky shores, these results have community-wide implications.

ABSTRACT The process of population aging will lead to an increase in health problems in older people, mainly related to their functionality.

ABSTRACT

The toxic effects of the insecticides chlorpyrifos and cypermethrin, and the herbicide glyphosate on the growth, biovolume and ultrastructure of the green microalgae Selenastrum capricornutum were evaluated. Concentrations between 9.37-150 mg L-1 of chlorpyrifos, 3.12-100 mg L-1 of cypermethrin and 4.7-60 mg L-1 of glyphosate were assayed along with a control culture. The assayed concentrations were prepared using commercial formulations. After 48 h all tested concentrations of the three pesticides reduced significantly the population growth. The 96 h effective concentration 50 (EC50) was 14.45 mg L-1 for chlorpyrifos, 12.37 mg L-1 for cypermethrin and 15.60 mg L-1 for glyphosate. Cells exposed to the three pesticides showed an increase in the cellular size related to the increase in pesticide concentration and exposure time. The most significant damages observed on the ultrastructure of cells exposed to the three pesticides included thylakoids and mitochondria disruption, formation of electrodense bodies, accumulation of lipids and increase in the size and number of starch granules. The present study demonstrates that the effects of pesticides also extend to non-target organisms having significant ecological implications.

ABSTRACT

Cardiac hypertrophy is the result of responses to various physiological or pathological stimuli. Recently, we showed that polycystin-1 participates in cardiomyocyte hypertrophy elicited by pressure overload and mechanical stress. Interestingly, polycystin-1 knockdown does not affect phenylephrine-induced cardiomyocyte hypertrophy, suggesting that the effects of polycystin-1 are stimulus-dependent. In this study, we aimed to identify the role of polycystin-1 in insulin-like growth factor-1 (IGF-1) signaling in cardiomyocytes. Polycystin-1 knockdown completely blunted IGF-1-induced cardiomyocyte hypertrophy. We then investigated the molecular mechanism underlying this result. We found that polycystin-1 silencing impaired the activation of the IGF-1 receptor, Akt, and ERK1/2 elicited by IGF-1. Remarkably, IGF-1-induced IGF-1 receptor, Akt, and ERK1/2 phosphorylations were restored when protein tyrosine phosphatase 1B was inhibited, suggesting that polycystin-1 knockdown deregulates this phosphatase in cardiomyocytes. Moreover, protein tyrosine phosphatase 1B inhibition also restored IGF-1-dependent cardiomyocyte hypertrophy in polycystin-1-deficient cells. Our findings provide the first evidence that polycystin-1 regulates IGF-1-induced cardiomyocyte hypertrophy through a mechanism involving protein tyrosine phosphatase 1B.

ABSTRACT

This study examines the distribution of primary and secondary space holders integrated in morphological and feeder functional groups as well as the species richness over the seasons on northern Patagonian rocky shores. We also evaluated whether the temporal changes in species richness, assemblage structure and species turnover were related to seasonal changes. These patterns were studied by non-destructive samplings between December 2013 and March 2015. Thirty taxa were identified, including six algal morphological functional and three invertebrate feeder groups. The cover of seaweeds and invertebrates was positively correlated with changes in the environmental factors. The marked seasonality in this Patagonian zone changed between two contrasting periods. Species richness and assemblage structure were associated with seasonal changes and were influenced by the more abundant morphological and feeder functional groups. We suggest that habitat-formers, such as articulated calcareous and suspensor feeder groups, can expand and retract over time-scales of months to seasons, generating available space for the colonization and growth of other organisms. We conclude that seasonal changes and the dynamics of articulated calcareous and suspensor feeder groups drive changes in the algal and invertebrate abundances, in turn driving changes in species richness and assemblage structure.
