ABSTRACT

The objective of the present study was to evaluate if extradural contact during hemilaminectomy would cause neurological deterioration in the early and/or late postoperative period in dogs with intervertebral disc extrusion. Nineteen dogs with thoracolumbar intervertebral disc extrusion underwent hemilaminectomy for spinal cord decompression and removal of extruded disc material. Meningeal contacts during surgery were quantified. Paraplegia (with nociception) and paraparesis were observed in 11/19 and 8/19 of dogs, respectively, before surgery. At the end of our study, only two (2/19) had paraplegia and one (1/19), paraparesis. There were more extradural contacts when extruded intervertebral disc material was at a ventrolateral position. Extradural contacts during surgery had no influence on neurological progression nor on time to recovery of motor function. Immediately (24 and 48 hours) after surgery, 13/19 dogs had the same neurological stage before surgery. At 7 and 90 days, 13/19 and 17/19 dogs, respectively, showed neurological improvement, compared with their preoperative stage. There was no influence of the number of extradural contacts on neurological recovery. These findings indicate that a careful inspection of the vertebral canal for removal of as much extruded disc material as possible does not cause neurologic deterioration.(AU)


O presente trabalho teve como objetivo avaliar se contatos extradurais durante hemilaminectomia em cães com extrusão de disco intervertebral causariam piora neurológica no pós-operatório imadiato e/ou tardio. Dezenove cães com extrusão toracolombar de disco intervertebral foram submetidos à hemilaminectomia para descompressão medular e remoção do material extruso. Durante o procedimento cirúrgico, os contatos meningomedulares foram quantificados. Antes da cirurgia, 11/19 cães apresentavam paraplegia (com nocicepção) e 8/19 cães, paraparesia. Ao fim do estudo, apenas dois cães (2/19) mostravam paraplegia com dor profunda e um (1/19), paraparesia. Observou-se maior quantidade de contatos extradurais quando o material discal extruso encontrava-se em posição ventrolateral. Os contatos extradurais não mostraram influência estatística na evolução neurológica dos animais, bem como no tempo de recuperação das funções motora. Vinte e quatro e 48 horas após a cirurgia, 13/19 cães apresentavam o mesmo grau neurológico de antes da cirurgia. Após sete e 90 dias de pós-operatório, 13/19 e 17/19 demonstraram melhora neurológica em comparação com o pré-operatório, respectivamente. A quantidade de contatos extradurais não influenciou na recuperação neurológica dos cães. Esses achados indicam que uma inspeção minuciosa do canal vertebral pode ser recomendada, a fim de remover o máximo de material discal extruso, evitando-se piora neurológica por compressão medular.(AU)

ABSTRACT

Objetivou-se avaliar as implicações e os resultados obtidos frente aos tratamentos conservativo e cirúrgico em 37 cães sem distinção de sexo, raça ou idade, que apresentaram fraturas e luxações vertebrais (FLV) toracolombares. Cada paciente passou por anamnese, seguida por exames clínico, neurológico e radiográficos para se obter o diagnóstico neuroanatômico e a severidade da lesão. Os cães foram submetidos ao tratamento conservativo ou cirúrgico de acordo com as características de cada caso. Dos 37 animais deste estudo, 37,7% tinham percepção de dor profunda (PDP) intacta. Vinte animais foram submetidos ao tratamento conservativo, 14 ao tratamento cirúrgico e três animais faleceram antes do tratamento. Dos animais que mantiveram a PDP intacta, a taxa de recuperação foi de 100%, com tempo de recuperação total variando de sete a 75 dias, ao passo que, dos animais que perderam a PDP, nenhum recuperou a deambulação voluntária. Dos 14 animais operados, 42,8% apresentaram alterações vertebrais não visibilizadas ao exame radiográfico convencional, sendo necessária mudança na técnica cirúrgica planejada em 14,2% desses casos. Nos animais que mantiveram a PDP, não houve diferença significante nas taxas nem no tempo de recuperação da deambulação voluntária, independentemente do tipo de tratamento. Entretanto, houve diferença altamente significante entre os graus que mantiveram a PDP intacta com os animais que perderam a percepção de dor profunda em relação à taxa de recuperação. A taxa de eutanásia foi menor que em outros trabalhos descritos na literatura. Dos animais sem PDP submetidos ao tratamento conservativo ou cirúrgico, 31,25% readquiriram a capacidade de caminhar sem recuperar a PDP, sendo essa deambulação involuntária atribuída ao caminhar espinal.


Our aim of this paper was to study the implications and results associated with conservative and surgical treatment of dogs with thoracolumbar Vertebral fractures and luxations (VFL) so that the information obtained can be used to obtain guidelines that lead to greater recovery rates. For such, 37 dogs presenting VFL were examined, without distinction of sex, breed, or age. Each patient had detailed history obtained, followed by clinical, neurologic and radiographic exams for obtaining neuroanatomic location and severity of the nervous and vertebral lesions. The dogs were submitted to conservative or surgical treatment according to each case. Deep pain perception (DPP) was intact in 37.7% of the 37 dogs in this study. Twenty dogs received conservative treatment, 14 underwent surgery and three dogs died before treatment. Of the dogs with intact DPP, 100% recovered, with a recovery time varying between seven and 75 days, while none of the dogs that lost DPP recovered voluntary motion. Of the 14 dogs submitted to surgery, 42.8% had vertebral lesions that weren't detected using conventional radiography, which demonstrates the low sensitivity of this exam, and led to a need for a change in the planned surgical technique in 14.2% of these cases. In dogs with intact DPP there was no significant difference in time or rate of recovery of voluntary motion, regardless of the treatment method. However, there was a highly significant difference between the dogs which retained and lost DPP regarding recovery rates. The percentage of euthanasia was less than in other reports, probably due to the greater awareness and protectiveness of owners nowadays. Of the dogs without DPP treated either surgically or conservatively, 31.25% recovered ambulation without recovery of DPP, characterizing this ambulation as involuntary, attributed to spinal walk.

ABSTRACT

Objetivou-se avaliar viabilidade, eficácia, vantagens e desvantagens da anestesia epidural lombossacral, junto à anestesia geral inalatória, em cirurgias de descompressão da cauda equina, tendo em vista que essas cirurgias são procedimentos longos e dolorosos. Para isso, foram utilizados 11 cães adultos com sinais clínicos de síndrome da cauda equina, que foram submetidos à anestesia geral inalatória e deixados no estágio anestésico mais superficial. Depois, realizou-se a anestesia epidural em seis dos 11 pacientes, por punção espinhal em L7-S1, com bupivacaína a 0,5 por cento. Os parâmetros fisiológicos (cardíacos, vasculares, respiratórios, temperatura corporal e glicemia) foram aferidos antes da medicação pré-anestésica, 10 minutos após esta, 30 minutos depois da epidural, depois da laminectomia, assim como após 60 minutos e 90 minutos da epidural, tanto no grupo com epidural como naquele sem esta (controle). Os animais que possuíam bloqueio epidural apresentaram redução significativa no consumo de anestésico inalatório e no tempo de extubação, não apresentando déficits neurológicos causados pela anestesia epidural, quando comparados com o grupo-controle. Conclui-se que a técnica de anestesia epidural é eficiente e vantajosa na realização de cirurgias descompressivas lombossacrais, pois proporciona menor risco anestésico para o animal...


The objective of this study was to evaluate the feasibility, effectiveness, advantages and disadvantages of lumbosacral epidural anesthesia with inhalational anesthesia in surgical decompression of the cauda equina, since this surgery has a lengthy and painful recovery from anesthesia. For this, 11 adult dogs which showed clinical signs of cauda equina syndrome were used. After the epidural, anesthesia in six of 11 patients, spinal tap in L7-S1, with bupivacaine 0.5 percent anesthetic was used. Physiological parameters (heart, vascular, respiratory and body temperature) were measured before, 10 minutes after, and 30 minutes after the epidural, after the laminectomia and 60 minutes and 90 minutes after the epidural, both in the epidural group and the control group. Animals that had ep showed a significant reduction in consumption of inhaled anesthetic and time of extubation. There were no neurological deficits caused by epidural anesthesia compared with the group without epidural anesthesia. Eriopis connexa (Germar) (Coleoptera: Coccinellidae) occurs in several countries of South America and its mass rearing is important for biological control programmes. This work evaluated biological aspects of E. connexa larva fed on eggs of Anagasta kuehniella (Zeller) (Lepidoptera: Pyralidae) and Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) frozen for one day, fresh eggs of Diatraea saccharalis (Fabricius) (Lepidoptera: Pyralidae), S. frugiperda newly-hatched caterpillars, nymphs of Rhopalosiphum maidis (Fitch) and Schizaphis graminum (Rondani) (Hemiptera: Aphididae). Duration of larva, pupa and larva to adult stages differed among prey offered, whereas the prepupa stage was similar. Larva, pupa, prepupa and larva to adult viabilities were equal or major of 87.5% in all prey, except for larva fed on newly-hatched larvae of S. frugiperda. Eriopis connexa has good adaptation to different prey corroborating its polyphagous feeding habit, which evidences the potential of this natural enemy for controlling corn and sorghum pests.

ABSTRACT

Objetivou-se descrever a ocorrência de lesão iatrogênica meningomedular em um cão da raça Dachshund, macho, oito anos de idade, apresentando tetraparesia flácida há dois meses, que foi submetido à espondilectomia ventral para tratamento de extrusão crônica de disco intervertebral cervical. Durante remoção do material de disco herniado com removedores de tártaro e pinça hemostática, observou-se severa hemorragia dos seios venosos, descolamento meníngeo com laceração medular e imediata queda dos parâmetros cardiovasculorrespiratório, com evolução para a morte ante a ausência de respostas às medidas de ressuscitação cardiopulmonar.


Our objective was to describe the occurrence of an iatrogenic meningo-medullary lesion and the trans-surgical complications in an eight-year-old male Daschund, which presented flacid tetraparesis for two months, and underwent ventral spondylectomy to treat a chronic cervical intervertebral disc extrusion. During the removal of the herniated disc with dental picks and hemostatic forceps, we observed meningeal dislocation with medullary laceration and an immediate reduction in cardiovasculatory and respiratory parameters, which progressed to a cardiorespiratory arrest that was non-responsive to medication and finally led to the death of the patient.

ABSTRACT

Poucos relatos têm descrito avulsões das cabeças lateral ou medial e avulsão bilateral das origens do músculo gastrocnêmio em cães e gatos. Este trabalho descreveu o primeiro relato de avulsão das origens lateral e medial do músculo gastrocnêmio em gata, bem como a técnica de reinserção cirúrgica. Uma gata, sem raça definida, com 10 anos, apresentou claudicação do membro posterior direito com postura plantígrada. Avulsão bilateral das cabeças do músculo gastrocnêmio foi diagnosticada com base nos achados do exame clínico. A técnica de reinserção foi realizada com fio mononáilon, numa sutura de Kessler modificada, ancorada em um orifício criado na porção metafisária distal do fêmur. Após período de imobilização externa complementar seguida de fisioterapia, houve recuperação satisfatória.


Few reports have described avulsions of the medial or lateral heads, as well as bilateral avulsion of the origins of the gastrocnemius muscle in dogs and cats. The objective of this study was to describe the first report of avulsion of the origins of the lateral and medial gastrocnemius muscle in a cat (Felis catus), as well as the surgical reinsertion technique. A mixed breed queen at 10 years of age presented claudication of the right hind limb with plantigrade stance. Bilateral avulsion of the heads of the gastrocnemius muscle was diagnosed based on findings from clinical a examination. The reinsertion technique was performed with monofilament nylon thread in a modified Kessler suture anchored in a hole created in the distal metaphyseal portion of the femur, obtaining satisfactory recovery after a period of additional external immobilization followed by physical therapy.

ABSTRACT

Utilizou-se o eletroneuroestimulador como um meio adequado para confirmação do posicionamento da ponta de agulhas isoladas no espaço epidural lombossal, por meio de pequenas estimulações elétricas que provocam contrações dos músculos inervados, empregando-se agulha de cateter intravenoso em vez de agulha isolada para a eletroneuroestimulação. Foram utilizados 40 cães para procedimentos cirúrgicos ortopédicos, abdominais ou retroumbilicais que necessitassem de bloqueio epidural. Durante a introdução gradativa da agulha no canal vertebral lombossacral, estímulos elétricos de 1,5 a 0,5mA foram aplicados. Diante de contrações musculares em membros pélvicos, cauda e/ou ânus, as quais foram consideradas respostas condizentes com o correto posicionamento da agulha no espaço epidural, estimulando raízes da cauda equina, procedeu-se à administração da associação de lidocaína, bupivacaína e tramadol. Constatou-se a adequada anestesia epidural mediante relaxamento do esfíncter anal, ausência dos reflexos patelares e flexor e a analgesia na área cirúrgica abdominal. Conclui-se que o método foi eficaz para a confirmação do correto posicionamento da agulha no espaço epidural durante a realização dessa anestesia regional em cães.


The electric neurostimulator was used as an adequate mean for the confirmation of the correct placement of the tip of the insulated needles in the lumbosacral epidural space through small electric stimulations causing muscle contractions by using an intravenous catheter needle instead of an electric neurostimulation insulated needle. Forty dogs were selected based on the need for an epidural block for orthopedic, abdominal or retroumbilical surgical procedures. Electrical stimuli were applied from 1.5 to 0.5mA during a gradual introduction of the needle into the lumbosacral vertebral canal. Responses that were considered as indicative of the correct placement of the needle and stimulation of the cauda equina were muscle contractions in the pelvic limbs, tail or anus. An association of lidocaine, bupivacaine and tramadol was then administered. Adequate epidural anesthesia was confirmed through relaxation of the anal sphincter, absence of patellar and flexor reflexes and analgesia of the abdominal surgical area. The authors were able to conclude that the method used was effective to confirm the correct needle placement in the epidural space during regional anesthesia in dogs.

ABSTRACT

A luxação da articulação atlanto-occipital é considerada uma afecção incomum no homem e nos animais. Radiografias laterais são recomendadas para o diagnóstico. No entanto, estão sujeitas a erros relacionados ao ângulo de radiação, ao alvo da imagem, à distância e à sobreposição óssea. Objetivou-se neste relato descrever os achados clínicos e radiográficos de dois cães com luxação traumática da articulação atlanto-occipital que apresentavam tetraparesia, dor cervical cranial, incapacidade de elevar a cabeça e déficits de nervos cranianos. No primeiro animal, observaram-se deslocamento craniodorsal do processo articular do atlas em relação a um dos côndilos do occipital, ausência de sobreposição dos forames vertebrais laterais e sobreposição do côndilo do occipital ao processo articular do atlas, no lado direito, caracterizando uma luxação unilateral. No segundo animal, observou-se deslocamento craniodorsal dos processos articulares do atlas em relação aos côndilos do occipital, com sobreposição dos forames vertebrais laterais e ausência de visibilização dos côndilos do occipital em virtude da projeção cranial dos processos articulares do atlas em direção ao crânio, caracterizando luxação bilateral. Conclui-se que o exame radiográfico simples, nas projeções laterolateral e ventrodorsal, apesar da dificuldade de ser interpretado, é eficiente para confirmar o diagnóstico da luxação atlanto-occipital traumática, tanto a simétrica quanto a assimétrica.


Atlanto-occipital luxation is considered rare in both humans and animals. Lateral radiographs are recommended for diagnosis, however, errors may occur related to the angle of radiation, image target, distance and overlapping of bone. Our objective is to report the clinical and radiographic findings in two dogs with traumatic atlanto-occipital luxation, which had tetraparesis, cranial neck pain, and inability to raise the head and cranial nerve deficits. The first animal had a cranio-dorsal dislocation of the articular process of the atlas in relation to one of the occipital condoles, with no overlapping of the transverse foramens or overlapping of the occipital condile in relation to the articular process of the atlas, on the right side, which characterizes a unilateral luxation. The second animal presented with a cranio-caudal dislocation of the articular processes of the atlas regarding the occipital condoles, with overlapping of the transverse foramens and inability to visualize the occipital condoles due to the cranial advancement of the articular processes of the atlas towards the skull, which characterizes a bilateral luxation. We conclude that a simple radiographic exam, in lateral and ventrodorsal projections, though difficult to interpret, is efficient in confirming a diagnosis of traumatic atlanto-occipital luxation, both symmetric and asymmetric.

ABSTRACT

Despite the increasing concern given to air quality in urban and industrial areas in recent years, particular emphasis on regulation, control, and reduction of air pollutant emissions is still necessary to fully characterize the chain emissions-air quality-exposure-dose-health effects, for specific sources. The Estarreja region was selected as a case study because it has one of the largest chemical industrial complexes in Portugal that has been recently expanded, together with a growing urban area with an interesting location in the Portuguese coastland and crossed by important road traffic and rail national networks. This work presents the first air quality assessment for the region concerning pollutant emissions and meteorological and air quality monitoring data analysis, over the period 2000-2009. This assessment also includes a detailed investigation and characterization of past air pollution episodes for the most problematic pollutants: ozone and PM10. The contribution of different emission sources and meteorological conditions to these episodes is investigated. The stagnant meteorological conditions associated with local emissions, namely industrial activity and road traffic, are the major contributors to the air quality degradation over the study region. A set of measures to improve air quality--regarding ozone and PM10 levels--is proposed as an air quality management strategy for the study region.

ABSTRACT

Compararam-se os achados musculotendinosos e osteoarticulares encontrados em cães com luxação patelar medial, por meio do exame ortopédico realizado com e sem anestesia geral. Para tal, foram utilizados 11 joelhos de 10 cães, sem distinção de sexo, idade e raça, os quais apresentaram luxação patelar medial dos graus II, III e IV, diagnosticada clinicamente. O exame ortopédico específico da articulação do joelho foi realizado previamente à cirurgia, com o animal sem anestesia, e, no dia da cirurgia, com o cão já anestesiado. Verificou-se, quanto aos achados osteoarticulares, que não houve diferenças. Já em relação aos achados musculotendinosos, houve diferença entre as avaliações do movimento de gaveta e dos músculos sartório e retofemoral.


This study aimed to compare the osteoarticular and musculotendineal findings in dogs with medial patellar luxation by orthopedic examination performed with and without general anesthesia. For this we used 11 knees of 10 dogs without distinction of sex, age and race, which had medial patellar luxation in grades II, III and IV, diagnosed clinically. The specific orthopedic examination of the knee joint was performed prior to surgery with the non-anaesthetised dog, and on the day of surgery with the same patient already anesthetized. Regarding the osteoarticular results, no differences were found. In relation to the musculotendineal results, differences were observed between the assessments of the drawer movement and sartorius and rectum femoral muscle tension.

Surveys on Coccinellidae (Coleoptera) in Sete Lagoas city, Minas Gerais state, Brazil, indicated the parasitism of adults of the species Coleomegilla maculata De Geer, 1775, Eriopis connexa (Germar, 1824) and Olla v-nigrum (Mulsant, 1866), by Dinocampus coccinellae (Schrank, 1802) (Hymenoptera; Braconidae: Euphorinae). Since then, the parasitoid have been maintained in its original hosts at the Insect Rearing Laboratory - LACRI of the Brazilian Agricultural Research Institution - Embrapa Milho e Sorgo. Besides the citation of occurrence in Brazil, this work also indicates the parasitoid preference for C. maculata (70% of parasitism), followed by O. v-nigrum (43.3% of parasitism) and E. connexa (36.7% of parasitism). Total life cycle of D. coccinellae was longer on C. maculata (32.4 ± 0.48 days), compared to O. v-nigrum (29.5 ± 0.49 days) and E. connexa (27.8 ± 0.4 days). Due to the relatively high percentage of field parasitism, D. coccinellae can reduce the efficiency of biological pest control by Coccinellidae predators especially in the case of C. maculata.

ABSTRACT

Six surgical corrections were performed with techniques of bone and soft tissue reconstruction, according to the individual presentation of each patient, in four Toy breed dogs, which had grades III and IV luxation, two of them bilaterally operated. During the recovery excessive medial tension associated to the Sartorius muscle was detected, which motivated its release at the iliac insertion. The animals had a functional recovery of the limb after 15 days, and complications or loss of function related to the release of the Sartorius muscle were not observed even after six months. It is believed that the proximal disinsertion of this muscle can be necessary and effective in some patients as a complementary technique to release medial tension in dogs with grades III and IV medial patellar luxation.

ABSTRACT

Elevation in the intracellular Ca(2+) concentration stimulates glucagon secretion from pancreatic α-cells. The Transient Receptor Potential Melastatin 4 channel (TRPM4) is critical for Ca(2+) signaling. However, its role in glucagon secreting α-cells has not been investigated. We identified TRPM4 gene expression and protein in the αTC1-6 cell line using RT-PCR and immunocytochemistry. Furthermore, we performed a detailed biophysical characterization of the channel using the patch-clamp technique to confirm that currents typical for TRPM4 were present in αTC1-6 cells. To investigate TRPM4 function, we generated a stable knockdown clone using shRNA and a lentiviral vector. Inhibition of TRPM4 significantly reduced the responses to different agonists during Ca(2+) imaging analysis with Fura-2AM. The reduction in the magnitude of Ca(2+) signals resulted in decreased glucagon secretion. These results suggested that depolarization by TRPM4 may play an important role in controlling glucagon secretion from α-cells and perhaps glucose homeostasis.

ABSTRACT

Effective treatment for recurrent, disseminated prostate cancer is notably limited. We have developed adenoviral vectors with a prostate-specific two-step transcriptional amplification (TSTA) system that would express therapeutic genes at a robust level to target metastatic disease. The TSTA system employs the prostate-specific antigen (PSA) promoter/enhancer to drive a potent synthetic activator, which in turn activates the expression of the therapeutic gene. In this study, we explored different configurations of this bipartite system and discovered that physical separation of the two TSTA components into E1 and E3 regions of adenovirus was able to enhance androgen regulation and cell-discriminatory expression. The TSTA vectors that express imaging reporter genes were assessed by noninvasive imaging technologies in animal models. The improved selectivity of the E1E3 configured vector was reflected in silenced ectopic expression in the lung. Significantly, the enhanced specificity of the E1E3 vector enabled the detection of lung metastasis of prostate cancer. An E1E3 TSTA vector that expresses the herpes simplex virus thymidine kinase gene can effectively direct positron emission tomography (PET) imaging of the tumor. The prostate-targeted gene delivery vectors with robust and cell-specific expression capability will advance the development of safe and effective imaging guided therapy for recurrent metastatic stages of prostate cancer.

ABSTRACT

The novel cell-cycle regulator p12(CDK2AP1) (p12) gene encodes a cyclin-dependent kinase 2 (CDK2) partner that participates in cell-cycle regulation, apoptosis, and proliferation. CDK2 has been implicated in maintenance of gonadal homeostasis, as knockout mice display reproductive abnormalities. To investigate the role of p12 in homeostasis of gonadal tissues in vivo, we generated a transgenic mouse model driven by the human keratin 14 promoter, reported to target transgene expression to gonadal tissues and also stratified epithelia. Overexpression of the transgene was associated with a gonadal atrophy phenotype in mice of both sexes, yet fertility was not impaired. Histological evaluation of testes showed seminiferous tubule degeneration and decreased tubule diameter. Female transgenic mice had small ovaries, with a higher number of atretic follicles/mm(2) as compared to control nontransgenic mice. Also observed was increased germ cell apoptosis in both sexes (TUNEL). These results suggest that overexpression of p12 leads to testicular and ovarian abnormalities, a phenotype closely related to that of cdk2-/- mice. In combination, these observations suggest that the p12/CDK2 signaling pathways are carefully orchestrated to maintain proper gonadal tissue homeostasis. We suggest that the mechanisms of this regulation may be through p12-mediated altered expression of gonadal-specific genes and apoptotic pathways.

ABSTRACT

Antibiotic susceptibility of the Staphylococcus spp. isolated from chicken carcass from the city of Recife, Pernambuco, was determined. Out of 90 strains of Staphylococcus spp., 51 were classified as Staphylococcus aureus and 39 as Staphylococcus negative coagulase. Samples were submitted to the disc diffusion technique for the antibiotic susceptibility test. Among the 17 antibiotics tested, the most efficient was vancomycin. Twenty (22.2 percent), 11 (12.2 percent) and 10 (11.1 percent) of the Staphylococcus spp. samples were resistant to five, six and 14 antibiotics, respectively.

ABSTRACT

The DNA-bending protein TF1 is the Bacillus subtilis bacteriophage SPO1-encoded homolog of the bacterial HU proteins and the Escherichia coli integration host factor. We recently proposed that TF1, which binds with high affinity (Kd was approximately 3 nM) to preferred sites within the hydroxymethyluracil (hmU)-containing phage genome, identifies its binding sites based on sequence-dependent DNA flexibility. Here, we show that two hmU-A base pair steps coinciding with two previously proposed sites of DNA distortion are critical for complex formation. The affinity of TF1 is reduced 10-fold when both of these hmU-A base pair steps are replaced with A-hmU, G-C, or C-G steps; only modest changes in affinity result when substitutions are made at other base pairs of the TF1 binding site. Replacement of all hmU residues with thymine decreases the affinity of TF1 greatly; remarkably, the high affinity is restored when the two hmU-A base pair steps corresponding to previously suggested sites of distortion are reintroduced into otherwise T-containing DNA. T-DNA constructs with 3-base bulges spaced apart by 9 base pairs of duplex also generate nM affinity of TF1. We suggest that twin hmU-A base pair steps located at the proposed sites of distortion are key to target site selection by TF1 and that recognition is based largely, if not entirely, on sequence-dependent DNA flexibility.

ABSTRACT

The case of a 66-year-old woman with several episodes of hemorrhage by esophageal varices and ascites due to portal hypertension is presented. Despite 4 sessions of endoscopic sclerotherapy bleeding recurred. A transjugular intrahepatic portosystemic shunt was performed. In the 10 months following the intervention, there was no further hemorrhage, the ascites disappeared and the shunt is open.
