ABSTRACT

The skin is considered the largest and most accessible organ in the human body, and allows the use of noninvasive and efficient strategies for drug administration, such as the transdermal drug delivery system (TDDS). TDDSs are systems or patches, with the ability and purpose to deliver effective and therapeutic doses of drugs through the skin. Regarding the specific interaction between hydrogels (HG) and microneedles (MNs), we seek to find out how this combination would be applied in the context of drug delivery, and we detail some possible advantages of the methods used. Depending on the components belonging to the HG matrix, we can obtain some essential characteristics that make the combination of hydrogels-microneedles (HG-MNs) very advantageous, such as the response to external stimuli, among others. Based on multiple characteristics provided by HGMNs that are depicted in this work, it is possible to obtain unique properties that include controlled, sustained, and localized drug release, as well as the possibility of a synergistic association between the components of the formulation and the combination of more than one bioactive component. In conclusion, a system based on HG-MNs can offer many advantages in the biomedical field, bringing to light a new technological and safe system for improving the pharmacokinetics and pharmacodynamics of drugs and new treatment perspectives.

ABSTRACT

Sclerotinia sclerotiorum is one of the most devastating and cosmopolitan plant pathogens. Rapid detection of S. sclerotiorum can provide growers an advantage in knowing what control measures should be taken to minimize crop damage and financial losses caused by it. Loop-mediated isothermal amplification (LAMP) is a fast, sensitive, and specific nucleic acid amplification method that does not require a thermal cycler. This study aimed to develop a LAMP-based assay for the specific detection of S. sclerotiorum (Ss-LAMP). A real-time quantitative LAMP reaction (Ss-qLAMP) and a calcein ion indicator-based LAMP reaction (Ss-cLAMP) were designed, optimized, and tested on fungi, plant, and soil samples. The Ss-LAMP reactions were very specific and sensitive. Applying the artificially inoculated soil samples with DNA purified by five protocols in the Ss-qLAMP reaction, it was possible to detect and quantify the pathogen DNA, regardless of the extraction protocol. Naturally infected soybean tissues had the pathogen detected by Ss-cLAMP directly in the reaction tube with no DNA extraction requirement. The assays should be applicable for many types of samples, such as soil, spore traps, and plant tissues from several crops, with no requirement for DNA extraction. The Ss-LAMP reactions took less than 1 h to complete, and they can be made directly in the field with real-time quantitative results (Ss-qLAMP) or qualitative naked-eye visual results (Ss-cLAMP). Results were obtained with 10 pg of DNA or 10 ng of crude mycelium, suggesting a detection limit close to a single DNA copy. Ss-LAMP reactions will allow rapid and accurate diagnosis of S. sclerotiorum and assist in pathogen management and control.

BACKGROUND: Soybean seed treatment with fungicides is a well-established disease management strategy. However, the movement of these fungicides within seedlings is not always well characterized. Thus, the objectives of this study were to determine the pattern of translocation of three fungicides with different modes of action applied as a seed treatment, and the effect of soil type on translocation. RESULTS: Most of the absorbed radioactivity was concentrated in the cotyledons and the maximum sum of the rates of absorption by roots, stems, and leaves of the plants was 15%. In most cases, absorption by roots, stems, and leaves were lower than 5% for 14 C-pyraclostrobin and 14 C-metalaxyl, and 1.6% for 14 C-carbendazim. Fungicides absorbed by the roots and the whole seedlings were higher when plants were grown in soil with lower organic matter content. Fungicides in the cotyledons are unlikely to be redistributed and are lost when cotyledons fall off the plants. CONCLUSION: Cotyledons are the part of the plant where fungicides are most absorbed, regardless of the fungicide. Soil type affects the absorption of fungicides, and in this study it was most likely caused by soil organic matter. These data improve knowledge of the movement of seed treatment fungicides in soybean seedlings and may help the development of seed treatment chemistry to manage seed and soilborne pathogens.

RESUMO O presente estudo teve por objetivo compreender a percepção dos enfermeiros quanto ao uso do sistema e-SUS AB no seu contexto de trabalho da Estratégia Saúde da Família (ESF). No tocante aos aspectos metodológicos, é uma pesquisa do tipo exploratória e descritiva, com abordagem qualitativa, realizada na cidade de Crato, Ceará. Participaram do estudo 18 enfermeiros que se enquadraram nos critérios de inclusão de estarem vinculados à ESF do município e que fizeram uso do sistema há pelo menos um mês. Os resultados obtidos a partir dos relatos dos participantes foram categorizados em: Processo de introdução e preparação dos profissionais para manuseio do sistema e-SUS AB; Potencialidades e desafios encontrados pelos enfermeiros diante do uso do sistema e-SUS AB; e Sentimentos dos enfermeiros em relação ao sistema e-SUS AB. Conclui-se que a utilização desse sistema, segundo as percepções dos enfermeiros, pode se constituir como uma ferramenta importante dentro do contexto da ESF, contribuindo para otimização dos processos de trabalho. Entretanto, ainda existem aspectos e concepções que dificultam a completa incorporação dessa tecnologia.


ABSTRACT The present study aims to understand the nurses' perceptions regarding the use of the e-SUS AB system in their work context of the Family Health Strategy (FHS). Regarding the methodological aspects, it is an exploratory and descriptive research, with a qualitative approach, carried out in the city of Crato, Ceará. There were 18 nurses participating in the study, who met the inclusion criteria of being linked to the city's FHS and who had used the system for at least one month. The results obtained from the reports of the participants were categorized as: Process of introduction and preparation of professionals to handle the e-SUS AB system; Potentialities and challenges encountered by nurses regarding the use of the e-SUS AB system; and Nurses' feelings regarding the e-SUS system. It is concluded that the use of this system, according to the nurses' perceptions, can constitute an important tool within the context of the FHS, contributing to the optimization of the work processes. However, there are still aspects and conceptions that hinder the complete incorporation of that technology.

ABSTRACT

Fasciolosis is a zoonotic disease with a worldwide distribution caused by Fasciola hepatica, which leads to severe economic losses in cattle such as reducing meat and milk production, livers condemnation, growth retardation and increase in mortality. From October 2008 to April 2011, condemned bovine livers in slaughterhouses of different municipalities from São Paulo state, Brazil were evaluated for the presence of Fasciola hepatica. Out of 20,635 analyzed livers, 1422 were infected with F. hepatica. These cattle came from 33 municipalities, out of which 16 showed infected animals and where 7 municipalities did not show statistical difference between each month throughout the year: Tuiuti - 276/1408 (19,6%), Atibaia - 44/257 (17,1%), Joanópolis - 116/738 (15,7%), Bragança Paulista - 318/2316 (13,3%), Piracaia - 182/1442 (12,6%), Santo Antonio de Posse - 118/1005 (11,7%), Amparo 131/2003 (6,5%). The other nine municipalities, Monte Alegre do Sul, Descalvado, Campinas, Morungaba, Pedreira, Socorro, Munhoz, Jaguariúna and Itatiba showed a positive percentage varying from 5.08% to 1.46%. Our results demonstrated the presence of F. hepatica in this region was higher than official data, bringing the need for control measures. There is also an apparent increase in fasciolosis two to three months after low to medium precipitation, however high precipitation causes a decrease in fasciolosis prevalence.

ABSTRACT

White mold, caused by Sclerotinia sclerotiorum, is a yield-limiting disease of soybean in Brazil. Uniform fungicide trials have been conducted annually since 2009. Data from 74 cooperative field trials conducted over a 10-year period were assembled. We selected five fungicides applied two times around flowering: dimoxystrobin plus boscalid (DIMO+BOSC), carbendazim plus procymidone (CARB+PROC), fluazinam (FLUZ), fluopyram (FLUO), and procymidone (PROC). For comparison, thiophanate-methyl (TMET) applied four times was also included as a low-cost treatment. Network models were fitted to the log of white mold incidence (percentages) and log of sclerotia mass data (grams/hectare) and to the nontransformed yield data (kilograms/hectare) for each treatment, including the untreated check. Back-transformation of the meta-analytic estimates indicated that the lowest and highest mean (95% confidence interval [CI]) percent reductions in incidence and sclerotia mass were 54.2 (49.3 to 58.7) and 51.6% (43.7 to 58.3) for TMET and 83.8 (79.1 to 87.5) and 87% (81.9 to 91.6) for CARB+PROC, respectively. The overall mean (95% CI) yield responses ranged from 323 kg/ha (247.4 to 400.3) for TMET to 626 kg/ha (521.7 to 731.7) for DIMO+BOSC, but the variance was significantly reduced by a binary variable (30% threshold) describing disease incidence in the untreated check. On average, an increment of 352 kg/ha was estimated for trials where the incidence was >30% compared with the low-disease scenario. Hence, the probability of breaking even on fungicide costs for the high-disease scenario was >65% for the more effective, but more expensive fungicide (FLUZ) than TMET. For the low-disease scenario, profitability was less likely and depended more on variations in fungicide cost and soybean price.

ABSTRACT

An apparent decline of fungicide performance for the control of soybean rust in Brazil has been reported but the rate at which it has occurred has not been formally quantified. Control efficacy and yield response to three fungicides applied as single active ingredients (a.i.)-azoxystrobin (AZOX), cyproconazole (CYPR), and tebuconazole (TEBU)-and four applied as mixtures-AZOX+CYPR, picoxystrobin + CYPR, pyraclostrobin + epoxiconazole, and trifloxystrobin + prothioconazole (TRIF+PROT)-were summarized using network meta-analytic models fitted to mean severity and yield data from 250 trials (10-year period). The effect of year was tested on both variables in a meta-regression model. Overall control efficacy ranged from 56 to 84%; the three single-a.i. fungicides performed the poorest (56 to 62%). Yield increase for single-a.i. fungicides was as low as 30% but ranged from 47 to 65% for the premixes. Significant declines in both variables were detected for all fungicides except TRIF+PROT. For TEBU, control efficacy (yield response) declined the most: 78% (18%) to 54% (8%) from 2004-05 to 2013-14. The recent surge of resistant populations of Phakopsora pachyrhizi to both demethylation inhibitor and quinone outside inhibitor fungicides is likely the driving force behind a significant decline after 4 years of fungicide use.

The objective of this study was to verify the efficiency of the control of white mold on soybean with the use of fungicides applied alone and in rotation, at different growth stages and in a mixture of two active ingredients at three locations: Arapoti, PR, Mauá-da-Serra, PR and Pinhão, PR, Brazil. The fungicides used were carbendazim (Ca), thiophanate methyl (Tm), procymidone (Pr) and fluazinam (Fl). The experiments consisted on 17 treatments and 4 replications in a randomized block design. The analyzed variables were severity, incidence, number of sclerotia and yield. Mauá-da-Serra and Pinhão presented the highest incidences (31% and 29.8% in the control, respectively). At these two locations most of treatments with fungicides decreased the incidence and production of sclerotia, when compared to control; however, no differences in terms of yield were observed. Arapoti presented the lowest incidence (15.8% in the control) where most of treatments with fungicides did not present differences for the variables incidence, production of sclerotia and yield, when compared to the control. No differences were also observed for severity in any of three locations. In conclusion, fungicides applied in soybean areas with historically white mold incidence up to 31% can reduce the disease incidence and sclerotia production levels.


O objetivo deste estudo foi o de verificar a eficiência de controle do mofo-branco na soja com o uso de diferentes fungicidas aplicados isoladamente, alternados, em diferentes estádios fenológicos e em mistura de dois princípios ativos, em três locais: Arapoti/PR, Mauá-da-Serra/PR e Pinhão/PR, Brasil. Os fungicidas utilizados foram carbendazim (Ca), tiofanato metílico (Tm), fluazinam (Fl) e procimidona (Pr). Os experimentos foram compostos por 17 tratamentos e 4 repetições distribuídas em blocos aleatorizados. As variáveis analisadas foram incidência, severidade, produção de escleródios e o rendimento da cultura. Mauá-da-Serra e Pinhão apresentaram as maiores incidências, 31% e 29,8% no tratamento controle, respectivamente, onde a maioria dos tratamentos com fungicidas proporcionou menor incidência e produção de escleródios, quando comparados ao controle, porém sem diferenças quanto ao rendimento. Em Arapoti foi observada a menor incidência, 15,8% no controle, onde a maioria dos tratamentos com fungicidas não apresentou diferenças para incidência, produção de escleródios e rendimento, quando comparados ao controle. Em nenhum dos três locais foram verificadas diferenças entre os tratamentos e o controle para a variável severidade. Como conclusão, fungicidas aplicados em áreas de soja com histórico de incidência de mofo-branco de até 31% podem reduzir a incidência da doença e o nível de produção de escleródios.

White mold caused by the fungus Sclerotinia sclerotiorum is an important disease in relation to soybean. The use of less susceptible genotypes can be a productive strategy in the management of this disease, and the development of an appropriate methodology for soybean inoculation is useful for the differentiation of disease-resistant genotypes. The present study aimed to assess the susceptibility of 77 soybean genotypes based on their reaction to oxalic acid, as well as to determine correlations between three traditional disease assay methods (detached leaf, non-wounded stem and straw tests) and the results of the oxalic acid assay. Oxalic acid susceptibility was assessed by using a wilting score scale. For the other methods, the severity of disease symptoms was assessed. To compare methodologies, the values obtained for the genotypes using each method were categorized into classes, and a severity index was used to represent individuals within each class. All the methods used were efficient for the differentiation of soybean genotypes in terms of susceptibility to S. sclerotiorum; however, the behavior of the genotypes depended on the inoculation method adopted. Even though no significant relationship was identified between the severities of the damage resulting from the methodologies, the rankings acquired from the methods strongly agreed. The oxalic acid method was the most rapid, the least laborious, and was the cheapest compared with the other methods that were used.


O mofo branco causado pelo fungo Sclerotinia sclerotiorum é uma doença importante na cultura da soja. A utilização de genótipos menos suscetíveis pode ser uma estratégia interessante no manejo desta doença, e o desenvolvimento de um método de inoculação adequado para a soja é de grande valia para a discriminação de genótipos resistentes. O presente estudo objetivou avaliar a suscetibilidade de 77 genótipos de soja baseado na reação destes ao ácido oxálico, bem como através de três métodos tradicionais (folha destacada, inoculação na haste sem ferimento e utilizando ponteiras de micropipeta), a fim de verificar os resultados obtidos com o teste do ácido oxállico e correlações entre métodos. A suscetibilidade dos genótipos ao ácido oxálico foi avaliada através de uma chave de escores de murchamento. Para os demais métodos, a severidade de ataque do patógeno foi avaliada. Para permitir a comparação entre metodologias, os resultados obtidos para os genótipos foram organizados em classes, e um índice de severidade foi utilizado para representar os individuos de cada classe. Todos os métodos foram eficientes na diferenciação de genótipos de soja quanto à suscetibilidade à S. sclerotiorum, entretanto, o comportamento dos genótipos variou conforme o método empregado. Não foi observada relação entre as severidades e escores de murchamento obtidos com as metodologias, porém, os ranqueamentos dos genótipos obtidos dos diferentes métodos apresentaram forte correlação. O método baseado na reação ao ácido oxálico foi o mais rápido, menos trabalhoso e de menor custo dentre os métodos utilizandos neste trabalho.

ABSTRACT

The aim of this study was to evaluate different alternative products, foliar sprayed either alone or in combination with fungicide fluazinam, to control white mold in soybean at three locations: Arapoti, Mauá-da-Serra, and Pinhão, in Paraná state, southern Brazil. The following chemical products were used: sodium dichloroisocyanurate, benzalkonium chloride, pyroligneous extract, sodium hypochlorite, cobalt + molybdenum, sucrose, sodium bicarbonate and acibenzolar-S-methyl. Incidence, severity, yield, and number of sclerotia produced were analyzed. The majority of the variables did not follow the normal distribution of the data according to the Kolmogorov-Smirnov test; therefore, the use of non-parametric analysis was necessary. The analysis of all the treatments individually revealed no significant effects so it was decided to separate the analysis into the following groups of treatments: 'control', treatments using alternative products ('alternatives'), treatment only with fungicide ('fungicide') and treatments with alternative products and fungicide ('alternatives + fungicide'). In Arapoti, no significant differences for any variables were observed. In Mauá-da-Serra, the 'fungicide' and 'alternatives + fungicide' had the lowest incidence and number of sclerotia, with higher yield when compared to 'control' and 'alternatives'. No differences were observed for severity. In Pinhão, the 'fungicide' and 'alternatives + fungicide' were superior for incidence, number of sclerotia produced, and yield when compared to the 'control' and 'alternatives'. The 'alternatives + fungicide' showed significantly lower severity when compared to the control, 'alternatives' and 'fungicide' in Pinhão experiment. In conclusion, the alternative products applied in combination with fluazinam resulted in lower severity of white mold in soybean in the experiment conducted in Pinhão, and the application of fluazinam alone or in combination with alternative products, was efficient to control white mold in soybean in locations with high disease incidence.


O objetivo deste estudo foi avaliar diferentes produtos alternativos pulverizados via foliar, isoladamente ou em combinação com o fungicida fluazinam para o controle do mofo branco na cultura da soja, em três locais: Arapoti, PR, Mauá-da-Serra, PR e Pinhão, PR, Brasil. Os produtos utilizadas foram: dicloroisocianurato de sódio, cloreto de benzalcônio, extrato pirolenhoso, hipoclorito de sódio, cobalto + molibdenio, sacarose, bicarbonato de sódio e acibenzolar-S-metil. As variáveis analisadas foram incidência, severidade, rendimento e o número de escleródios produzidos. A maioria das variáveis não segue a distribuição normal dos dados conforme o teste Kolmogorov-Smirnov, assim sendo, o uso de testes não-paramétricos se fez necessário. A análise de todos os tratamentos individualmente não revelou efeitos significativos, onde optou-se por separar a análise em grupos de tratamentos, onde a testemunha foi denominada "controle"; os tratamentos com os produtos alternativos como "alternativos", o tratamento com o fungicida isolado como "fungicida" e os tratamentos com produtos alternativos e fungicida foi denominado "alternativos + fungicidas". Em Arapoti, não foram observadas diferenças estatisticamente significantes em nenhuma das variáveis analisadas. Em Mauá-da-Serra, os grupos 'fungicida' e 'alternativos + fungicida', apresentaram a menor incidência e número de escleródios, com maior rendimento quando comparados aos grupos 'controle' e 'alternativos'. Não foram observadas diferenças na severidade. Em Pinhão, os grupos 'fungicida' e 'alternativos + fungicida' foram estatisticamente superiores para incidência, número de escleródios produzidos e rendimento, quando comparados aos grupos 'controle' e 'alternativos'. O grupo 'alternativo + fungicida' apresentou severidade significativamente menor quando comparado aos grupos 'controle', 'alternativos' e 'fungicida' no experimento conduzido em Pinhão. Como conclusões, observou-se que os produtos alternativos aplicados em associação com o fungicida fluazinam proporcionaram menor severidade do mofo branco na cultura da soja no experimento conduzido em Pinhão, PR, e a aplicação do fungicida fluazinam isolado ou em associação com os produtos alternativos foi eficiente no controle do mofo branco na cultura da soja nos locais com alta incidência da doença.

ABSTRACT

The Sclerotinia sclerotiorum (Lib.) de Bary fungus is a necrotrophic and polyphagous pathogen with soybean crop as one of its most important hosts. However in order to occur epidemics caused by such a pathogen it is necessary that the climatic conditions, mainly air temperature and relative humidity be favorable to the occurrence and development of the disease. The current research aimed to assess the incidence and severity of S. sclerotiorum in soybean plants grown in Arapoti, PR, Brazil, as a function of different plant populations, row spacing, and microclimate. The experiment was conducted in a naturally infested area. The experimental design adopted was a randomized block in a factorial combination with 4 row spacing (0.35, 0.45, 0.60, 0.75 m) and 4 plant populations (150, 200, 250; 300 thousand plants per hectare), totaling 16 treatments and 4 replications. Throughout the current study we performed 4 assessments of incidence and severity. Local climatic conditions were favorable for the incidence and severity of the pathogen. Mean air temperature, rainfall, relative humidity and leaf wetness duration, monitored during all experimental period, favored the development of the pathogen in the experimental field. Yield and 100 seed weight did not show fluctuations as a function of variations on the incidence and severity of white mold in soybean crop fields. Nevertheless, crop yield was higher under a reduced spacing, whereas the 100 seed weight showed the highest values under the lowest plant population treatment.


O fungo Sclerotinia sclerotiorum (Lib.) de Bary é um patógeno necrotrófico, polífago que tem a planta da soja como um dos seus principais hospedeiros. Porém, para que ocorram epidemias em decorrência desse patógeno é necessário que as condições climáticas, principalmente de umidade e temperatura do ar, sejam favoráveis para a ocorrência e o desenvolvimento da doença. O presente trabalho teve como objetivo avaliar a incidência e severidade de S. sclerotiorum em plantas de soja cultivadas em Arapoti, PR, Brasil, em função de diferentes densidades populacionais, espaçamentos entrelinhas e microclima. O experimento foi conduzido em área naturalmente infestada. O delineamento experimental adotado foi o de blocos casualizados em esquema fatorial, combinando 4 espaçamentos entrelinhas (0,35; 0,45; 0,60; 0,75 metros) e 4 densidades populacionais (150, 200, 250, 300 mil plantas ha-1), totalizando 16 tratamentos e 4 repetições. Ao longo do experimento foram efetuadas 4 avaliações de incidência e severidade. A temperatura média do ar, precipitação, umidade relativa do ar e a duração do molhamento foliar, monitoradas durante todo o período experimental, foram favoráveis para o desenvolvimento do patógeno na área experimental. A produção e a massa de 1000 grãos não apresentaram oscilações em função das variações nos níveis de incidência e severidade de mofo-branco na cultura da soja cultivada na condição climática estudada. Contudo, a produtividade da cultura foi maior nos tratamentos com espaçamentos reduzidos. Já a massa de 1000 grãos apresentou maiores valores em tratamentos com reduzida densidade populacional.

ABSTRACT

O Mofo Branco, Sclerotinia sclerotiorum, deve ser manejado por métodos de controle integrado, entre eles a resistência genética. O objetivo do trabalho foi avaliar e definir um método confiável na seleção de genótipos de soja com resistência ao mofo branco. Os testes foram conduzidos em dois ambientes distintos: em ambiente controlado, onde as plantas foram cultivadas em casa-de-vegetação e levadas para o laboratório para serem inoculadas e incubadas em câmara de crescimento; e no campo, onde a doença está sujeita às condições ambientais adversas. Observou-se que a temperatura, condições de cultivo, e o método de inoculação podem influenciar na seleção dos genótipos quanto à resistência à podridão da haste da soja. Definiu-se que o melhor método de avaliação à resistência de genótipos de soja foi a inoculação realizada com ferimento no terço médio das plantas em fase final de florescimento e início de enchimento dos grãos, com discos de BDA e avaliadas 14 dias após a inoculação. A cultivar que pode ser considerada como padrão de resistência foi a Emgopa 316, e os padrões de suscetibilidade foram a BRSMG Garantia e BRSMG 68 Vencedora.


The Soybean stem white rot should be managed by integrated control methods, including genetic resistance. This study evaluated and developed a reliable method for the selection of soybean genotypes resistant to stem white rot. The tests were conducted in two different environments, a controlled one, where plants were grown in a greenhouse and brought into the laboratory to be inoculated and incubated in a growth chamber where the temperature is favorable for fungal growth, and a field, where the pathogen is subjected to adverse environment conditions. Was observed that temperature growth conditions, and inoculation method may affect the selection of resistant soybean genotypes against stem rot. The best method to evaluate soybean resistance was by wound inoculating the middle third of the plant when the plants were at the end of flowering and early grain filling, with PDA disks and evaluating 14 days later. The resistance standard was Emgopa 316, while the susceptible standards were BRSMG Garantia and BRSMG68 Vencedora.

ABSTRACT

Sclerotinia sclerotiorum é um fungo polífago apresentando mais de 400 espécies hospedeiras. No Brasil relata-se que sua ocorrência tenha aumentado a partir da safra 2003/2004 em função do uso de sementes infectadas, falta de rotação de culturas, entre outras causas. Nos EUA, há relatos que o aumento da doença também tenha ocorrido devido à redução no uso de herbicidas do grupo das triazinas na cultura do milho, em rotação. Alguns trabalhos relatam que esse herbicida atua na formação de apotécios anormais e na inibição da produção de ascósporos. Frente a isso o presente trabalho objetivou o estudo do efeito de 12 herbicidas e 5 fungicidas sobre a germinação carpogênica de S. sclerotiorum. O trabalho foi conduzido em laboratório na Universidade Estadual de Ponta Grossa. Em caixas tipo gerbox foram colocados 80 gramas de substrato e 20 escleródios. A aplicação foi realizada utilizando um volume de calda de 400 L ha-1. Após a aplicação as caixas foram acondicionadas em ambiente controlado (20°C e fotoperíodo de 12 horas). Os tratamentos utilizados foram: glifosato, lactofen, atrazina, setoxidim, diuron, diuron + paraquat, imazaquim, trifluralina, diclofop metílico, clorimuron etílico, metribuzim e imazetapir (herbicidas) e fluazinam, procimidone, carbendazim, dimoxystrobin + boscalid e tiofanato metílico (fungicidas). Aos 36 dias após a aplicação foi realizada uma contagem do número de escleródios germinados, número de estipes e número de apotécios por escleródio. Para os dados de germinação de escleródios, foi constatado que somente dimoxystrobin + boscalid apresentou germinação inferior a 70%. Os demais tratamentos apresentaram germinação superior a 83%. Para número de estipes formadas por escleródio foi constatado que todos os tratamentos apresentaram de duas a quatro estipes por escleródio. Foi observado que os tratamentos com dimoxystrobin + boscalid, tiofanato metílico e carbendazim apresentaram as menores taxas de diferenciação de estipes em apotécios, ou seja, constatou-se a formação de estipes inviáveis. No tratamento com dimoxystrobin + boscalid foi constatado uma viabilidade de apenas 34,56% de suas estipes. Os resultados aqui apresentados mostram que a aplicação antecipada desses tratamentos pode ser realizada visando atingir a fonte de inóculo (escleródios) no solo, reduzindo o potencial de infecção dos mesmos e consequentemente sua incidência sobre culturas.


Sclerotinia sclerotiorum is a pathogenic fungus with over 400 host species. In Brazil, it is reported that its occurrence has increased since the 2003-2004 growing season due to use of infected seed, lack of crop rotation, and several other factors. In the United States, reports suggest that the disease increase has occurred due to reduced use of herbicides from the triazine group on corn in rotation with soybean. Reports show that these herbicides affect S. sclerotiorum, causing the formation of abnormal apothecia and inhibiting ascospore production. Because of the possible linkage with herbicide use, a study was designed to examine the effect of 12 herbicides and 5 fungicides on the carpogenic germination of S. sclerotiorum. This work was performed at Universidade Estadual de Ponta Grossa. Eighty grams of sterilized field soil and 20 sclerotia, placed on the soil surface, were added to "gerbox" boxes. Chemicals were applied once using a rate of 400 L ha-1. After application, boxes were incubated in a controlled environment (20°C with a photoperiod of 12 hours). The treatments used were: glyphosate, lactofen, atrazine, sethoxydim, diuron, diuron + paraquat, imazaquin, trifluralin, diclofop methyl, chrlorimuron ethyl, metribuzin and imazethapyr (herbicides) and fluazinam, procymidone, carbendazim, dimoxystrobin + boscalid and thiophanate methyl (fungicides). The number of germinated sclerotia, stipes and apothecia was recorded 36 days after chemical application. For sclerotia germination, only dimoxystrobin + boscalid reduced germination below 70%. All other treatments had greater than 83% sclerotia germination. The number of stipes produced per sclerotium was constant for all treatments (two to four stipes per sclerotium). Dimoxystrobin + boscalid, thiophanate methyl and carbendazim had the lowest level of apothecia formation, which means inviable stipes were formed. For the treatment of dimoxystrobin + boscalid, only 34.56% of stipes developed into mature apothecia. The results of this study demonstrate that application of herbicides did not affect the sclerotia of S. sclerotiorum. Three of the five fungicide tested in this trial, dimoxystrobin + boscalid, thiophanate methyl and carbendazim, can reduce inoculum on the soil, which, in turn, would likely reduce disease incidence in the following crop.

O objetivo do trabalho foi avaliar o comportamento de genótipos transgênicos e convencionais de soja frente ao patógeno S. sclerotiorum, agente etiológico da podridão branca da haste. O primeiro experimento avaliou a resistência parcial de genótipos convencionais de soja de ciclo semiprecoce e médio/semitardio usando inoculação de disco de BDA contendo micélio do fungo na haste mais jovem da planta. O segundo experimento avaliou a resistência parcial de genótipos transgênicos de ciclo semiprecoce, médio/semitardio, através da inoculação natural das plantas pelo patógeno. Os genótipos convencionais GOBR03-2776-4SFGO e Emgopa 316 e os genótipos transgênicos BRY08-1.812Y, BRY08-1.018Y e GO04-5014 B2GO foram selecionados como padrões de resistência para trabalhos futuros. Como padrões de suscetibilidade, foram selecionados os genótipos convencionais GOBR03-3151-34GO e GOBR01-1252-23 GO2 e os genótipos transgênicos BR05-73615Y e BRBIGO03-20023-30GO.


The objective of this study was to evaluate the performance of transgenic and conventional soybean genotypes against the S. sclerotiorum, causal agent white mold. The first experiment evaluated partial resistance of conventional soybean genotypes of semi-early and medium/late cycles, by the inoculation of PDA disk, containing mycelium of the fungus, on the young stem of the plant. The second study evaluated the partial resistance of transgenic genotypes of early and medium/late cycles, by natural inoculation of the plant by pathogen. Results from the first experiment demonstrated varietal differences among genotypes of semi-early and medium/medium late cycles for the severity of sclerotinia stem rot. Conventional genotypes GOBR03- 2776-316 and Emgopa 4SFGO and transgenic genotypes BRY08-1.812Y, BRY08-1.018Y and GO.04 B2GO5014 were selected as the resistance standard for future work. As susceptibility standards were selected genotypes conventional GOBR03-34GO-3151 and 1252-23 GOBR01-GO2 and transgenic genotypes BR05- 20023-73615Y and BRBIGO03-30GO.

This work had as objectives the study of the physical and mechanical characteristics of maize kernels in relation to the contamination by Fusarium sp and by zearalenone in twenty landraces of maize from the southern region of Brazil. From the analyzed samples, 60 percent has been considered to have a hard endosperm type and 40 percent an intermediary one. A correlation between the physical and mechanical variables was observed as an indication that the higher is the proportion of hard endosperm, more dense will be the kernel and more force for its rupture will be necessary. The level of contamination by Fusarium sp was between 5.5 and 24.75 percent among the analyzed grains, correlating positively with the flotation index, indicating that the landraces of maize with a softer endosperm can present a higher contamination by this genus. The presence of zearalenone was verified in 75 percent of the samples, in concentrations varying from 50 to 640 µg kg-1.


Este trabalho teve por objetivos verificar as características físicas, mecânicas, contaminação por Fusarium sp e por zearalenona e suas relações, em vinte variedades crioulas de milho da região sul do Brasil. Verificou-se que das amostras analisadas, 60 por cento foram consideradas como possuidoras de endosperma do tipo duro e 40 por cento com endosperma do tipo intermediário. As variáveis físicas e mecânicas se correlacionaram, indicando que quanto maior a proporção de endosperma vítreo, mais denso e maior a força necessária até a ruptura do grão. A contaminação por Fusarium sp esteve entre 5,5 por cento e 24,75 por cento nos grãos analisados, correlacionando-se positivamente com o índice de flotação, indicando que as variedades crioulas de milho com uma maior proporção de endosperma macio podem apresentar uma maior contaminação por Fusarium sp, sugerindo-se a utilização de variedades crioulas com a textura do endosperma predominantemente vítrea. A presença de zearalenona foi verificada em 75 por cento das amostras, em concentrações que variaram de 50 a 640 µg kg-1.

Biological control consists of using one organism to attack another that may cause economic damage to crops. Integrated Pest Management (IPM) is a very common strategy. The white mold produced by Sclerotiniasclerotiorum (Lib.) causes considerable damage to bean crops. This fungus is a soil inhabitant, the symptoms of which are characterized by water-soaked lesions covered by a white cottony fungal growth on the soil surface and/or the host plant. Possible biological control agents taken from plants are being investigated as phytopathogen inhibitors. These are endophytic microorganisms that inhabit the intercellular spaces of vegetal tissues and are often responsible for antimicrobial production. The objective of the present study was to select endophytic fungi isolated from comfrey (Symphytumofficinale L.) leaves with in vitro antagonist potential against the phytopathogenic fungus S. sclerotiorum. Twelve isolates of endophytic fungi and a pathogenic strain of S. sclerotiorum were used in the challenge method. With the aid of this method, four endophytes with the best antagonistic activity against S. sclerotiorum were selected. Pathogen growth inhibition zones were considered indicative of antibiosis. The percentages of pathogenic mycelia growth were measured both with and without the antagonist, resulting in growth reductions of 46.7% to 50.0% for S. sclerotiorum. These analyses were performed by evaluating the endophytic/pathogenic mycelia growth in mm/day over an eight-day period of antagonistic tests.


O controle biológico consiste no uso de organismos que atacam outros que causam danos a culturas de plantas. Esta é uma estratégia muito utilizada no Controle Integrado de Pragas (CIP). O mofo branco, causado por Sclerotiniasclerotiorum (Lib.), causa danos em culturas de feijão. Este fungo é encontrado no solo e seus sintomas são caracterizados por lesões úmidas cobertas por micélios algodonosos, crescidos a partir do solo e/ou da planta hospedeira. Há relatos de pesquisas buscando agentes potenciais de controle biológico isolados de plantas para controlar fungos fitopatogênicos. Entre estes agentes encontram-se os microrganismos endofíticos, habitantes de espaços intercelulares de tecidos vegetais, muitas vezes responsáveis pela produção de substâncias antimicrobianas. Este trabalho teve por objetivo selecionar linhagens endofíticas isoladas de folhas de confrei (Symphytumofficinale L.), com potencial de antagonismo in vitro contra a linhagem fitopatogênica S. sclerotiorum. Doze linhagens de fungos endofíticos foram utilizadas na técnica de desafio em placa contra um isolado patogênico de S. sclerotiorum. Com o auxílio desta técnica, quatro linhagens com melhor atividade antagonística contra S. sclerotiorum foram selecionadas. Zonas de inibição no crescimento da linhagem patogênica foram consideradas como indicativo de antibiose. Foram efetuadas análises da porcentagem de elongação micelial com e sem antagonismo, mostrando resultados de 46,7% a 50,0% de redução no crescimento micelial do fitopatógeno. Estas análises consistiram de avaliações das medidas do crescimento em mm/dia das linhagens endofíticas/patogênica em testes de antagonismo.

Discute-se a importância do oftalmologista no diagnóstico e encaminhamento precoces de pacientes com esta síndrome e propõe-se uma forma mais adequada de acompanhamento desses pacientes.

Os autores tem por objetivo descrever o emprego da colonoscopia como método diagnóstico e terapeutico, suas principais indicaçöes e limitaçöes com as complicaçöes do método. A ampla utilizaçäo da colonoscopia terapeutica e ressaltada na polipectomia, na identificaçäo de areas de sangramento e na hemostasia. Säo discutidas as intercorrencias como a hemorragia, a perfuraçäo e a sindrome pos polipectomia.
