ABSTRACT

Risk elements in blood matrices can affect human health status through associations with biomarkers at multiple levels. The aim of this study was to analyze 15 macro- and microelements in the blood serum of women with overweight (BMI of ≥25 kg/m2) and obesity (BMI of ≥30 kg/m2) and to examine possible associations with biochemical, liver enzymatic parameters, and markers of oxidative stress. Based on the power calculation, the study involved women (in the postmenopausal stage) with overweight (n = 26) and obesity (n = 22), aged between 50-65 years. Multifrequency bioelectrical impedance analysis was used to measure body composition parameters. Concentrations of elements were determined by inductively coupled plasma optical emission spectrometry, and Hg was measured using cold-vapor atomic absorption spectroscopy. Individuals with obesity, as indicated by a higher BMI, percentage of body fat, and visceral fat area, had elevated serum levels of Ca, Mg, Fe, Al, Sr, Pb, and Hg. Concentrations of Al, Cu, K, Sb, Zn, and Pb significantly affected biochemical and liver function markers in women with overweight or obesity. Elements such as Cu and Al were associated with increased total cholesterol. The correlation analysis between total antioxidant status and Cu, Al, and Ni confirmed associations in both groups. Our findings underscore the importance of addressing excess body weight and obesity in relation to risk elements. The results of the research could be beneficial in identifying potential targets for the treatment or prevention of comorbidities in people with obesity.

ABSTRACT

Chia seeds offer therapeutic properties that aid in the prevention of a variety of ailments, including cardiovascular disease, diabetes, obesity, and other risk factors. Arsenite, a common environmental chemical, has been identified as a reproductive toxin owing to its negative effects on male reproductive health. It has been shown to inhibit spermatogenesis and generate androgenic effects in men. The primary goal of this research was to look into the effect of Salvia hispanica on testicular toxicity caused by sodium arsenite in male rats. A set of 36 male albino rats was allocated to a negative control cohort. The individuals in this group were given a basic meal and orally given distilled water for a duration of 28 days. The other five groups were given a regular meal and received intra-peritoneal injections of sodium arsenite (NaAsO2) at a concentration of 4 mg/kg body weight that was diluted in a 0.9% NaCl solution. The injections were administered consecutively, with two doses given within a two-day period. Subsequently, the rats were categorized into several groups using the following classification: Group 2 consisted of a positive control cohort, in which the rats were given a typical baseline diet. Groups 3, 4, 5, and 6 were given a basic diet that included varying proportions of ground chia seeds, namely 5%, 10%, 15%, and 20% per 100 g of the diet. After the trial was completed, the rats were euthanized, and further biological examination was conducted. The measurements of the reproductive organs were documented and reported. The research assessed the following characteristics: sperm count, motility, progressive motility, and normal morphology. The research included examining serum sex hormones, namely luteinizing hormone (LH), follicle-stimulating hormone (FSH), and testosterone. An evaluation of the activity of antioxidant enzymes was performed in the tissue of the testicles. There were statistically significant improvements in the sperm parameters, serum sex hormone levels, and the activity of antioxidant enzymes, such as GPX, SOD, and CAT, in the therapy groups. The levels of malondialdehyde (MDA) exhibited a noteworthy decrease (p ≤ 0.05) when compared to the positive control group. Salvia hispanica seeds have demonstrated a significant level of effectiveness in reducing sodium arsenite-induced testicular toxicity, which leads to the conclusion. The flavonoid content and antioxidant properties of Salvia hispanica seeds may be to blame for the observed behavior. These indicated characteristics may have therapeutic significance in treating testicular harm induced by arsenite exposure.

ABSTRACT

Improvements in drinking water quality (DWQ) can lead, according to some estimates, to a 10% reduction of the world's disease load. The drinking water distribution system (DWDS) plays a crucial role in influencing DWQ and can contribute to the emergence of poultry-related epidemics. This study aims to monitor the variations in DWQ throughout the seasons within the DWDS of Egyptian poultry farms experiencing epidemics. The study assessed DWQ at four different points along the DWDS, including the water source (WS), water tank (WT), broiler drinker (BD), and layer drinker (LD), across 86 farms. Statistical analysis was employed to establish correlations between DWQ and the sampling points within the DWDS, as well as between water temperature (Tw.C°), ambient temperature (Ta.C°), and microbial DWQ. The survey revealed significant differences between Tw.C° and Ta.C°, with notable effect sizes (d = 0.89-1). Additionally, the results revealed significant differences in physicochemical DWQ between WS and house drinkers (HD), with medium to large effect sizes (d = 0.56-0.85). Furthermore, significant differences were identified in microbial DWQ between winter and summer, with a small to large effect size (d = 0.40-0.87). Notably, we recorded significant differences in microbial DWQ between WS and WT, with a small to medium effect size (d = 0.40-0.61), and between WT and BD, with a small to medium effect size (d = 0.48-0.53). Additionally, we found significant differences in microbial DWQ between WS and LD, with a medium effect size (d = 0.59-0.68). In conclusion, Tw.C° is influenced by seasonal variations in Ta.C°. While the physicochemical DWQ was unaffected by seasonal temperature variations, it was significantly impacted by the DWDS from WS to HD. In contrast, the microbial DWQ was strongly influenced by both seasonal temperature changes and DWDS.

ABSTRACT

The beneficial influence of bacteriocin-producing, probiotic, mostly non-autochthonous bacteria has already been reported in various animals. However, their use in horses provides limited information, and results with autochthonous bacteria have not been reported. Therefore, the main objective of this model study was to test the effect of autochthonous, bacteriocin-producing faecal strain Enterococcus faecium EF 412 application in horses. One gram of freeze-dried EF 412 strain (109 CFU/mL for 21 days) was applied to horses in a small feed ball. Clinically healthy horses (12), Slovak warm-blood breed of various ages (5-13 years), were involved in a 35-day-long experiment, also functioning as control for themselves. They were stabled in separate boxes (university property), fed twice a day (hay, whole oats or grazed) with water access ad libitum. Sampling was performed at the start of the experiment, i.e. at days 0/1, 21 (3 weeks of EF 412 application) and at day 35 (2 weeks of EF 412 cessation). EF 412 colonized GIT of horses was 3.54 ± 0.75 CFU/g (log 10) at day 21. The eggs of the nematode Strongylus spp. were not found in horses after EF 412 application, and Eimeria spp. oocysts were similarly not found. The other microbiota were not reduced as evaluated by the use of standard method. Using next-generation sequencing, at phylum level, phyla Bacteroidetes and Firmicutes dominated and at family level, they were Bacteroidales BS11 and S24-7 gut goups and Lentisphaerae. In horses, the increasing tendency in phagocytic activity was noted after EF 412 application. Biochemical parameters were in the physiological range. Total protein value was significantly decreased at day 21 compared with day 0/1 as well as with day 35 (P < 0.05). Cholesterol and triglycerides were influenced (decreased) at day 21 compared with day 0/1 and day 35. Neither nematode eggs Strongylus spp. nor Eimeria spp. oocysts were found in faeces after EF 412 application. Autochthonous, faecal strain E. faecium EF 412 showed promising application potential.

ABSTRACT

The aim of this study was to monitor the effect of Bifidobacterium bifidum (BB) and the combination of Lactobacillus sporogenes, Enterococcus faecium, and Bifidobacterium bifidum (LEB) on the health status and weight gain of calves, and the utilisation of nitrogenous substances. The experiment was performed in the period from April 2020 to September 2020. A total of 90 Holstein heifers, which were one to 56 days old, were used as experimental animals. Differences in live weight gain were significant if we compared the LEB vs. BB group and the LEB vs. C, the control group (86.23 ± 5.49 kg vs. 84.72 ± 6.22 kg, p < 0.05; 86.23 ± 5.49 kg vs. 82.86 ± 5.35 kg, p < 0.01). Considering the live weight gain, group BB was heavier than group C only (84.72 ± 6.22 kg vs. 82.86 ± 5.35 kg, p < 0.05). An effect on reducing the incidence and duration of diarrheal diseases was not demonstrated in this study (p = 0.1957). The administration of feed additives had no statistically significant effect on the amount of N excreted in the feces. The values of hematological and biochemical parameters were unaffected except for the first sampling of urea. Other blood parameters were not affected by the addition of probiotic feed additives. The bacterial populations in the feces 5 days and 56 days after birth were not affected by the inclusion of feed additives.

Nowadays, there is an increased interest in coffee derivatives (green beans, roasted beans, and coffee by-products (Cascara and Silverskin)) due to their particular chemical composition. This study aimed to compare the content of dry matter, total fat, fatty acids, and fiber (ADF, NDF) of coffee by-products (Cascara and Silverskin) and coffee beans (green and roasted under different conditions). Coffee beans and their by-products were obtained from 100% C. arabica coffee cherries from Panama by dry process. The lowest concentrations of fat corresponded to Cascara 4.24 g·kg-1 and Silverskin 23.70 g·kg-1, respectively. The major fatty acids detected in all samples were palmitic, stearic, oleic, and linoleic acids, the latter two being essential fatty acids. LDA showed that 89.01% of the variability between beans and by-products was explained by lignoceric, myristic, behenic, tricosanoic, arachidic, and heneicosanoic acids. Silverskin appeared to be a good source of lignoceric, myristic, and behenic acids and had a higher concentration of dietary fiber (314.95 g·kg-1) than Cascara (160.03 g·kg-1). Coffee by-products (Silverskin and Cascara) are low-fat products enriched in dietary fiber. Their incorporation, after adjustment, into the global diet may contribute to nutrition security, the sustainability of the coffee sector, and human health.

ABSTRACT

The utilisation of different by-products from the food industry as nutrient sources for farm animals is both possible and beneficial. Grape pomace is a by-product that contains polyphenols and fatty acids, both of which have the potential to improve the nutritional quality of cow colostrum. This study aimed to explore how the addition of grape pomace to the diet of dry cows affects the concentration of nutrients and fatty acids of colostrum. Sixteen Slovak spotted cows in late pregnancy were used in this study. From the seventh day before expected calving to the day of calving, cows in the grape pomace group received a diet supplemented with dried grape pomace, at 0.116 kg/cow/day. Colostrum samples were analysed for basal nutrients and fatty acid concentrations. Between the control and experimental groups, the nutrient and fatty acid concentrations of all the colostrum samples did not show significant differences. The nutrient levels in the colostrum from both groups of cows were typical, as related to the time from calving. The addition of the grape pomace into the diet of dry cows had no effect on nutrient concentrations and the fatty acid composition of the colostrum. The somatic cell score of the colostrum sampled at the 12th hour after calving (4.2 versus 2.6) was positively affected by grape pomace addition. The results of this study revealed that grape pomace (fed in an amount of 0.116 kg/cow/day) had no positive or negative effect on the base nutrients and fatty acids of cow colostrum, and, therefore, grape pomace can be used as a nutrient source for dry cows in small amounts.

The aim of the present study was to determine the physico-chemical properties (dry matter content, combustion heat, electrical properties, total protein, ash, fat and crude fibre contents, selected amino acids, and trace elements), antioxidant content, and sensory profile of Linz biscuits. They were enriched by the addition of powdered carrot, nettle leaves and elderberry fruit, which is 3% of the product. For comparison of results, a control variant without the addition of these components was also prepared. The enriched biscuits showed slightly higher total ash and crude fibre contents in comparison to the control samples. Results for the antioxidant activity and total polyphenol, flavonoid, and phenolic acid contents of the enriched biscuits were higher in all observed parameters than in the control sample with the best results obtained for Linz biscuits enriched with elderberry and nettle powder. In enriched biscuits, higher contents of iron, zinc, and manganese were measured, especially in biscuits with nettle. Linz biscuits with nettle had higher combustion heat values than control samples; the other two sample types had lower values. We found that the resistance, capacitance, and relative permittivity of the enriched biscuits decreased with frequency according to the power regression function. On the contrary, the conductivity increased with an increasing frequency. Electrical properties were mainly influenced by the water content but also by added components.

Bee pollen has been successfully used as a feed additive with beneficial impacts on productive, reproductive, and immune conditions of animals. However, its effect on bone structure and bone health remains controversial. Therefore, the purpose of our study was to examine the impact of bee pollen supplementation on macroscopic and microscopic structure of a femoral bone using rats as suitable animal models. Male rats (1 month-old) were assigned into three groups: control (C group) that was fed a standard diet without bee pollen and two bee pollen supplemented groups (P1 and P2 groups) that received an experimental diet including 0.5% and 0.75% of bee pollen, respectively, for 3 months. A number of unfavorable effects of 0.75% bee pollen administration on bone weight, cortical bone thickness, calcium content, alkaline phosphatase activity, sizes of primary osteons' vascular canals, Haversian canals and secondary osteons in the cortical bone have been recorded, whereas these bone parameters were significantly decreased in the P2 group versus the C group. On the contrary, the concentration of 0.5% did not affect any of bone features mentioned above. In conclusion, the impact of bee pollen supplementation on femoral bone structure of rats depends on the dose used.

Coconut oil has a high content of lauric acid, which has selective antibacterial activity. This study aimed to explore the effect of coconut oil ingestion on the gastrointestinal microbiomes of pigs. A 14-day-long feeding experiment included 19 pigs in two groups (9 on a normal diet and 10 on a diet supplemented with coconut oil). At the start and end of the experiment, a rectal swab sample was taken from each pig in both groups, and total bacterial DNA was extracted. We used 16S rRNA high-throughput amplicon sequencing to evaluate the microbiome changes during the feeding experiment. A total of 446 operational taxonomic units (OTUs) were detected in the whole sample set. Shannon's indices of bacterial diversity did not change significantly during the experiment. Changes in the bacterial community during the study period and in response to the coconut oil treatment were highly significant (p < 0.001). During the study, an increase in the abundance of Lactobacillus was detected in the group treated with coconut oil. An increase in Alloprevotella, Bifidobacteriales, and Lactobacillales and a decrease in Corynebacterium, Mitsuokella, Psychrobacter, and Pseudomonadales were attributed to the coconut oil treatment. Although the addition of coconut oil to pig feed did not affect Shannon's index of diversity, it had a positive effect on the abundance of bacterial groups that are considered to be commensal and/or probiotic.

Quercetin is a dietary bioflavonoid used widely as a food supplement and is generally recognized as safe. The aim of this in vitro study was to examine the steroid hormone (progesterone and 17- ß estradiol) release, proliferation (PCNA and cyclin B1) and apoptosis (caspase 3 and p53) of porcine ovarian granulosa cells after the addition of quercetin at concentrations 0.01, 0.1, 1, 10 and 100 µmol L-1. Progesterone release was stimulated at the concentration 10 µmol L-1. Quercetin neither had any impact on 17-ß estradiol secretion nor on the presence of PCNA. However, a significant enhancement of the occurrence of cyclin B1 was noted except for the lowest concentration 0.01 µmol L-1. Quercetin did not have any influence on the number of granulosa cells containing caspase 3, but at the concentration 10 µmol L-1 it inhibited p53 occurrence. Results confirm the safety of quercetin in porcine ovarian granulosa cell model and further suggest its possible concentration-dependent influence on ovarian functions through pathway that may involve progesterone, cyclin B1 and p53.

The general objective of this in vitro study was to examine the effect of bee pollen on the release of insulin-like growth factor I (IGF-I) and steroid hormone progesterone, and expression of markers of proliferation (PCNA) and apoptosis (caspase-3) in porcine ovarian granulosa cells. Concentrations of IGF-I and progesterone were determined by RIA method and expression of PCNA and caspase-3 by immunocytochemistry. Bee pollen addition at the dose of 10 ng/mL significantly (P<0.05) inhibited IGF-I release by porcine ovarian granulosa cells. This growth factor was not influenced by 100 and 1000 ng/mL doses of bee pollen. Progesterone release by cells was not influenced by bee pollen addition at the doses of 10, 100 and 1000 ng/mL as used in our study. Similarly expression of PCNA and caspase-3 was not affected by bee pollen addition. The present study shows dose-dependent regulation of IGF-I by experimental bee pollen addition in vitro. Progesterone release, expression of PCNA and caspase-3 in porcine ovarian granulosa cells was not induced by pollen. Our results contribute to new insights regarding the possible effect of bee pollen on IGF-I release, which is important for regulation of porcine ovarian functions.

ABSTRACT

Contamination by microscopic fungi and mycotoxins in high moisture corn (HMC) silages conserved by chemical additives was investigated. The samples were examined for the concentration and identification of microscopic fungi able to grow on Malt and Czapek-Dox agar and for mycotoxins content (deoxynivalenol, T-2 toxin, zearalenone and total aflatoxins, fumonisins, ochratoxins) by direct competitive enzyme-linked immunosorbent assays. The average fungal counts were 3.37 +- 2.52 log cfu/g in control HMC silages, 2.91 +- 0.51 log cfu/g in HMC silages treated by organic salts and inorganic salt, 3.62 +- 1.46 log cfu/g in HMC ensiled with organic acids and 3.49 +- 1.12 log cfu/g of HMC silages treated by organic acids along with organic salt. In this study, 740 isolates belonging to 10 fungal species representing 9 genera were recovered. The genera of microscopic fungi most frequently found in HMC were Penicillium (56.49 percentages) and Paecilomyces (32.16 percentages). T-2 toxin was the secondary metabolite with the highest concentration ranging from 179.13 +- 3.04 to 249.40 +- 24.69 micrograms/kg, followed by deoxynivalenol and total fumonisins. The highest mean of deoxynivalenol level was 0.13 +- 0.02 mg/kg and concentration of total fumonisins ranged from 20.13 +- 2.53 to 90.33 +- 10.35 micrograms/kg. This study indicated that application of chemical additives containing organic acids, organic salts and inorganic salt was sufficient to inhibit mycotoxins formation. The use of calcium formiate, sodium benzoate and sodium nitrite resulted in high hygienic quality of HMC silages and significantly reduced the concentration of zearalenone, deoxynivalenol and total ochratoxins and fumonisins.
