ABSTRACT

Acacia concinna leaves are beneficial as medicinal for treating many symptoms and vegetable plants in Thai cuisine. The objective of the study was to analyze proximate compositions and micronutrients value, as well as, to evaluate biological activities including, antioxidant and anticancer activities of A. concinna leaves in Thailand. Proximate composition and micronutrients value were determined relative to the Association of Official Analytical Chemists standard methodology. Antioxidant capacity was examined following oxygen radical absorbance capacity (ORAC) and ferric reducing antioxidant power (FRAP) assay. Anticancer activity was determined following resazurin microplate assay. Proximate composition of A. concinna leaves including crude protein and dietary fiber constituted 6.73 ± 0.03 g and 9.15 ± 0.03 g/100 g sample. Energy constituted 113.00 ± 3.61 kcal/100 g sample. Its leaves also had the highest micronutrients value on calcium, with constituted 321.38 ± 4.29 mg/100 g sample. A. concinna leaves hold antioxidant capacity with ORAC and FRAP values of 20407.55 ± 74.23 µmoles and 1016.38 ± 29.29 µmoles TE/100 g sample, respectively. Its ethanolic extract holds slightly anticancer effect against tested cancerous cell lines. A. concinna can be a healthy alternative source of nutrients with good antioxidant capacity and slightly anticancer properties.

ABSTRACT

Acanthopanax trifoliatus has been used as both traditional plant food and medicinal plant in Thailand. This study aimed to evaluate proximate, vitamin, and mineral compositions of A. trifoliatus leaf samples together with antioxidant and anticancer activities of ethanolic leaf extract of A. trifoliatus. For leaf samples, quantitative determination of proximate composition was evaluated comprising moisture, crude protein, total fat, total carbohydrate, dietary fiber, ash, as well as energy. Quantitative determination of vitamin and mineral composition including Vitamin A, Vitamin B1, Vitamin B2, Vitamin C, calcium, sodium, and iron was also assessed. For ethanolic leaf extract, antioxidant activity was investigated using oxygen radical absorbance capacity (ORAC) and ferric reducing antioxidant power (FRAP) methods. Anticancer activity was determined against human ductal, bronchogenic, liver, gastric, and colon cancer cell lines using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide method. Proximate composition of A. trifoliatus was found to be 74.62 ± 0.38, 5.01 ± 0.05, 0.95 ± 0.04, 16.47 ± 0.40, 8.54 ± 0.06, and 2.95 ± 0.04 g/100 g sample, respectively, and energy was found to be 94.48 ± 1.30 kcal/100 g sample. Vitamin and mineral composition was found to be 428.47 ± 3.00 µg/100 g sample, 0.41 ± 0.01, 0.17 ± 0.00, 11.95 ± 0.86, 675.35 ± 46.57, 13.46 ± 0.95, and 4.79 ± 0.15 mg/100 g sample, respectively. A. trifoliatus ethanolic leaf extract revealed antioxidant capacity with ORAC value of 9057.29 ± 43.08 µmol TE/100 g and FRAP value of 1230.88 ± 19.51 µmol TE/100 g. Its extract also showed cytotoxic potential against all tested cancer cell lines. A. trifoliatus leaf is a good source of essential nutrients, which had antioxidant and anticancer potential.

ABSTRACT

Diabetes and cancer are a major global public health problem. Plant-derived agents with undesirable side-effects were required. This study aimed to evaluate antidiabetic and anticancer activities of the ethanolic leaf extract of Mangifera indica cv. Okrong and its active phytochemical compound, mangiferin. Antidiabetic activities against yeast α-glucosidase and rat intestinal α-glucosidase were determined using 1 mM of p-nitro phenyl-α-D-glucopyranoside as substrate. Inhibitory activity against porcine pancreatic α-amylase was performed using 1 mM of 2-chloro-4 nitrophenol-α-D-maltotroside-3 as substrate. Nitrophenol product was spectrophotometrically measured at 405 nm. Anticancer activity was evaluated against five human cancer cell lines compared to two human normal cell lines using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Mango leaf extract and mangiferin exhibited dose-dependent inhibition against yeast α-glucosidase with the IC50 of 0.0503 and 0.5813 mg/ml, respectively, against rat α-glucosidase with the IC50 of 1.4528 and 0.4333 mg/ml, respectively, compared to acarbose with the IC50 of 11.9285 and 0.4493 mg/ml, respectively. For anticancer activity, mango leaf extract, at ≥200 µg/ml showed cytotoxic potential against all tested cancer cell lines. In conclusion, mango leaf possessed antidiabetic and anticancer potential in vitro.

ABSTRACT

Thai mango cultivars are classified into six groups plus one miscellaneous group according to germplasm database for mango. Characterization is important for conservation and the development of Thai mango cultivars. This study investigated macroscopic, microscopic leaf characteristics, and genetic relationship among 17 cultivars selected from six groups of mango in Thailand. Selected mango samples were obtained from three different locations in Thailand (n = 57). They were observed for their leaf and fruit macroscopic characteristics. Leaf measurement for the stomatal number, veinlet termination number, and palisade ratio was evaluated under a microscope attached with digital camera. DNA fingerprint was performed using CTAB extraction of DNA and inter-simple sequence repeat (ISSR) amplification. Forty-five primers were screened; then, seven primers that amplified the reproducible band patterns were selected to amplified and generate dendrogram by Unweighted Pair-Group Method with Arithmetic Average. These selected 17 Thai mango cultivars had individually macroscopic characteristics based on fruits and leaves. For microscopic characteristics, the stomatal number, veinlet termination number, and palisade ratio were slightly differentiable. For genetic identification, 78 bands of 190-2660 bps were amplified, of which 82.05% were polymorphic. The genetic relationship among these cultivars was demonstrated and categorized into two main clusters. It was shown that ISSR markers could be useful for Thai mango cultivar identification.
