ABSTRACT

The mechanism by which thermal/pressure processing influences the allergenicity of shrimp (Penaeus vannamei) was explored by anaphylaxis in mice, the protein structure, gastrointestinal digestion, and linear epitopes. Roasting induced the unfolding of the structure, which may reduce the allergenicity, but it made more linear epitopes to be exposed, causing mice to exhibit similar systemic anaphylaxis as mice fed with the raw shrimp protein (p > 0.05). However, the roasted + reverse-pressure-sterilized shrimp can significantly reduce specific antibodies, mast cell degranulation, vascular permeability, and histopathological morphology in mice compared with the raw and roasted shrimp (p < 0.05) because reverse-pressure sterilization causes protein to aggregate, hiding the heat/digested stable epitopes of arginine kinase (Glu59-Ser63, Asn112-Lys118, Leu131-Phe136, and Ser158-Glu162) and sarcoplasmic calcium-binding protein (Asn57-Phe67, Ser159-Cys165, and Glu126-Ala130) inside a 3D structure, while gastrointestinal digestion can destroy immunodominant, minor epitopes and the epitopes exposed by roasting. Meanwhile, the low binding frequency of IgE to troponin C was also responsible for maintaining the hypoallergenicity of shrimp.

ABSTRACT

Angiosarcoma is a highly malignant endothelial tumor, accounting for 1% to 2% of soft tissue sarcomas. The most common site of the disease is the skin, accounting for approximately 60% of cases, especially the skin of the head. It can also occur in soft tissue, bone, and retroperitoneum. However, angiosarcomas of the breast and chest wall are rare. Here, we report a 44-year-old woman who was admitted to hospital with encapsulated empyema. She was subsequently diagnosed with angiosarcomas of the breast and chest wall with tuberculous empyema. At the same time, we also reviewed the current research progress in angiosarcoma of the breast and chest wall to explore better treatment options for these patients.

ABSTRACT

Dietary ω-3 PUFAs are highly prone to oxidation, and this may potentially limit their application in the health-promoting field. Here, we sought to investigate whether and how oxidized PUFAs modulate the susceptibility of mice to Salmonella typhimurium (S. Tm) infection. Algae oil (AO) and oxidized algae oil (ox-AO) were administered to the C57BL/6 mice prior to S. Tm infection. Compared to the S. Tm group, ox-AO increased bacterial burden in systemic and intestinal tissues, downregulated host anti-infection responses, and developed worse colitis. In macrophages, ox-AO decreased both phagocytosis of S. Tm and clearance of intracellular bacteria and dampened the activation of mitogen-activated protein kinase (MAPK), NF-κB, and autophagy pathways. Furthermore, ox-AO diminished LPS-induced inflammatory cytokine production and S. Tm induced NLRC4 inflammasome activation. This study reveals that oxidized PUFAs may contribute to the development of enteric infections and regular monitoring of the oxidation status in commercial PUFA supplements to prevent their potential adverse impact on human health.

ABSTRACT

Achieving mechanical robustness and highly efficient self-healing simultaneously at room temperature is always a formidable challenge for polymeric materials. Herein, a series of novel supramolecular polyurethane elastomers (SPUEs) are developed by incorporating dynamic covalent boronic ester and boron-nitrogen (B-N) coordination. The SPUEs demonstrate the highest tensile toughness (∼182.2 MJ m-3) to date for room-temperature self-healable polymers, as well as an excellent ultimate tensile strength (∼10.5 MPa) and ultra-high fracture energy (∼72 100 J m-2), respectively, owing to a synergetic quadruple dynamic mechanism. It is revealed that the B-N coordination not only facilitates the formation and dissociation of boronic ester at room temperature but also dramatically enhances the mechanical properties by the intermolecular coordinated chain crosslinking and intramolecular coordinated chain folding. Meanwhile, the B-N coordination and urethane hydrogen interaction also serve as sacrificial bonds, which rupture during stretching to dissipate energy and recover after release, leading to superior notch insensitiveness and recoverability. The SPUEs restore their mechanical robustness after self-healing at room temperature and the self-healing efficiency can be dramatically accelerated by surface wetting.

ABSTRACT

Chinese sturgeon (Acipenser sinensis) is a critically endangered species, and waters downstream from Gezhouba Dam are the only known spawning ground. To optimize the velocity conditions in the spawning ground by controlling the opening mode of Gezhouba Dam generator units, a mathematical model of Chinese sturgeon spawning ground was established in FLOW-3D. The model was evaluated with velocity measurements, and the results were determined to be in good agreement. By inverting the 2016-2019 field monitoring results, the model shows that the preferred velocity range for Chinese sturgeon spawning is 0.6-1.5 m/s. Velocity fields of different opening modes of the generator units were simulated with identical discharge. The suitable-velocity area was maximal when all units of Dajiang Plant of Gezhouba Dam were open. For discharges below 12,000 m3/s, most of the area was suitable; for discharges above 12,000 m3/s, the suitable area rapidly decreased with increasing discharge. A comparison of suitable areas under high-flow showed that at discharges of 12,000-15,000 m3/s, opening 11-13 units on the left side was optimal. For discharges above 15,000 m3/s, all units should be open. We used these results to recommend a new operation scheme to support the conservation of Chinese sturgeon.

ABSTRACT

Complete mitochondrial genomes (mitogenomes) are important molecular markers for understanding the phylogenetics of various species. Although recent studies on the mitogenomes of the Scorpaeniformes species have been greatly advanced, information regarding molecular studies and the taxonomic localization of Platycephalidae is still sparse. To further analyze the phylogeny of Platycephalidae, we sequenced the complete mitogenome of Cociella crocodilus of the Platycephalidae family within Scorpaeniformes for the first time. The mitogenome was 17,314 bp in length, contained two ribosomal RNA genes (rRNAs), 22 transfer RNA genes (tRNAs), 13 protein-coding genes (PCGs), and two typical noncoding control regions (the control region (CR) and origin of the light strand (OL)). All PCGs used standard initiation codons ATG, apart from cox1. The majority of the tRNA genes could be folded into cloverleaf secondary structures, whereas the secondary structure of tRNASer (AGN) lacked a dihydrouridine (DHU) stem. The CR contained several conserved sequence blocks (CSBs) and eight tandem repeats. In addition, the phylogenetic relationship based on the concatenated nucleotides sequences of 13 PCGs indicated that the Platycephalidae species are relatively basal in the phylogenetic relationships of Scorpaeniformes. Our results may not only advance the origin and the evolution of Scorpaeniformes, but also provide information for the genetic evolution and taxonomy of the teleostean species.

ABSTRACT

The mitochondrial genome (mitogenome) can provide useful information for analyzing phylogeny and molecular evolution. Scorpaeniformes is one of the most diverse teleostean orders and has great commercial importance. To develop mitogenome data for this important group, we determined the complete mitogenomes of two gurnards Chelidonichthys kumu and Lepidotrigla microptera of Triglidae within Scorpaeniformes for the first time. The mitogenomes are 16,495 bp long in C. kumu and 16,610 bp long in L. microptera. Both the mitogenomes contain 13 protein-coding genes (PCGs), 2 ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes and two non-coding regions. All PCGs are initiated by ATG codons, except for the cytochrome coxidase subunit 1 (cox1) gene. All of the tRNA genes could be folded into typical cloverleaf secondary structures, with the exception of tRNASer(AGN) lacks a dihydrouracil (DHU) stem. The control regions are both 838 bp and contain several features common to Scorpaeniformes. The phylogenetic relationships of 33 fish mitogenomes using Bayesian Inference (BI) and Maximum Likelihood (ML) based on nucleotide and amino acid sequences of 13 PCGs indicated that the mitogenome sequences could be useful in resolving higher-level relationship of Scorpaeniformes. The results may provide more insight into the mitogenome evolution of teleostean species.
