ABSTRACT

The role of upflow velocity and Ca2+ concentration in controlling the type and rate of CaCO3 crystallization and their impacts on the anaerobic granular sludge (AnGS) formation and performance in an expanded granular sludge bed (EGSB) reactor were studied. The results showed that an improved upflow velocity could promote metastable CaCO3 crystals and achieve the optimized portion of vaterite with a value of 84 % at 10 m/h with a small amount of aragonite, thus limiting the scaling in the reactor. The removal efficiency of Ca2+ was to some extent positively correlated to the influent Ca2+ concentration, but declined when Ca2+ exceeded a specific threshold. Vaterite was dominant with the increase of Ca2+ concentrations of the influent. Compared with granules in R1 (Ca2+ 10 mg/L) and R2 (Ca2+ 100 mg/L), granules cultivated in R3 (Ca2+ 800 mg/L) revealed maximum amount of biomass with biggest particle size distribution and fastest average settling rate, with relative stable COD removal efficiency and the fast optimized reactor capacity at OLR of 16 kgCOD/m3d. A low upflow velocity and a higher Ca2+ concentration promoted nucleus formation and granules growth at the initial cultivation stage of the EGSB reactor. The Ca2+ concentration had a significant impact on the bacterial community and favoured the growth of Tolumonas and Anaeromousa Anaeroarcus. Archaea, rather than bacteria, was strengthened to contribute more to methane production at a relatively high Ca2+ concentration.

ABSTRACT

Gestational diabetes mellitus (GDM) represents a common carbohydrate metabolism disorder during pregnancy. The objective of this study was to evaluate the expression levels of mitofusin 2 (MFN2) expression in placentae of GDM patients compared to that in the placental tissues from normal uncomplicated pregnancies. A total of 70 subjects were enrolled from September 2014 to June 2016, including 42 patients with GDM (the GDM group) and 28 normal uncomplicated pregnancies (the control group). Immunohistochemical staining and qRT-PCR were used for the detection of the expression levels and distribution of MFN2 in the placentae of GDM patients and normal controls. Kolmogorov-Smirnov test was used for statistical analysis. P < 0.05 and P < 0.01 were used for assessing statistical significance. The baseline characteristics were comparable in both groups. The 1-h and 2-h postprandial glucose levels (PPG) were 7.94±1.26 versus 6.88±0.51 mmol/L and 7.01±1.34 versus 6.14±0.63 mmol/L, respectively,for the GDM group and the control group (P < 0.05). The relative expression levels of MFN2 mRNA were 0.982±1.242 for GDM and 1.257 ± 0.815 for control, respectively, with significant between group difference (P < 0.01). Immunohistochemical staining analysis showed that MFN2 was mostly distributed in the cytoplasm of syncytiotrophoblasts under optical microscopy. Additionally,about 50% of samples of the GDM group were within the intensity of moderate staining of MFN2 and more than 50% of patients in the control group were within the intensity of strong staining of MFN2. The expression levels of MFN2 in GDM placentae was significantly lower compared to that of placentae from normal uncomplicated pregnancies.

ABSTRACT

OBJECTIVE: To study the significance of Fas and Fas-L expression in adenocarcinoma of uterine cervix. METHODS: Both carcinoma tissue and their surrounding tissues from 36 patients with adenocarcinoma of uterine cervix, previously untreated either by radiation or chemotherapy, were studied for the expression of Fas and Fas-L by immunohistochemical stain with DNA apoptosis fragment detected by TUNEL. RESULTS: The TUNEL labeling index was negatively correlated with differentiation of adenocarcinoma of cervix. Compared to highly differentiated and moderately differentiated tumor, the TUNEL labeling index was reduced obviously in poorly differentiated adenocarcinoma (P < 0.01). Fas expression was detected in 31 cases (86%) while there were only 3 weakly stained in the normal endocervical glands around the carcinoma. The 5 unstained carcinomas were 3 highly differentiated and 2 moderately differentiated. The positively stained Fas was associated with differentiation; the stronger the stain, the less differentiation there was. The Fas-L expression was detected in all adenocarcinomas while there was only 1 weakly stained in the normal ones. No significant difference was found in the expression of Fas-L in carcinomas with different degrees of differentiation. No correlation was observed between Fas and Fas-L expression. CONCLUSIONS: The Fas expression is positively correlated with the different degrees of differentiation and Fas-L expression may be associated with the escape from of immunal surveillance.
