ABSTRACT

Background: The prevalence of peptic ulcers is increasing due to lifestyle changes and harmful diets. Objective: The aim of this study was to investigate the effect of fish oil (FO) on gastric ulcers induced by ethanol in rats. Methods: The pharmacological efficacy of FO with doses of 5 and 10 mg/kg investigated using the gastric ulcer index, the acidity of gastric secretions, pro-inflammatory cytokine assessment, and oxidative stress examination. Results: Ethanol-induced gastric ulcer improves with FO 5 or 10 mg/kg pretreatment (P<0.05). FO did have acid-neutralizing activity. FO also increased the levels of glutathione and catalase and decreased the malondialdehyde levels (P<0.05). Moreover, FO reduced the levels of tumour necrosis factor alpha (TNF-α) interleukin-6 (IL-6), through downregulation of nuclear factor kappa B (NF-κB) (P<0.05). Pretreatment with FO attenuates ethanol-induced gastric ulceration. Conclusion: The observed effects may be due to the role of FO in regulating gastric secretions, changes in the expression of NF-κB, and changes in the levels of oxidative stress factors.

ABSTRACT

OBJECTIVE: Penconazole is used in agriculture and human and veterinary medicine applications. It has been included in the acute toxicity hazard category by the WHO. This study examines the protective effect of selenium and vitamin C on the fertility of male rats given penconazole. METHODS: Nine groups of rats were given penconazole at concentrations of 50 and 75 mg/ml and selenium and vitamin C at concentrations of 0.5 and 100 mg/ml, respectively. Serum levels of LH and FSH were measured with ELISA kits; ß-actin, GPX4, AQP7, PRM2, and BAX gene expression was evaluated with real-time PCR performed on the left testis of each rat. RESULTS: LH, FSH, and testosterone levels were lower in the groups given penconazole (50 and 75 mg/kg). Histopathology showed that the groups given penconazole had the lowest number of spermatogonia and primary spermatocytes; these numbers were greater in the groups receiving penconazole together with selenium or vitamin C; and the highest counts were observed in separate groups given Se and vitamin C. GPX4, AQP7, PRM2 and BAX gene expression in the groups receiving penconazole was different from controls and was modulated by treatment with selenium or vitamin C. CONCLUSIONS: This study showed that antioxidant compounds have a strengthening effect on the reproductive system and can mitigate the destructive effects of chemical fungicides.

ABSTRACT

BACKGROUND: Probiotics and phytase are commonly used as dietary supplements in poultry diets. Phytase is involved in the release of phosphorus in plant grain ingredient of poultry feed, while probiotics provide beneficial organisms to the gastrointestinal tract. OBJECTIVE: The present study was performed to evaluate the effect of both commercial probiotic and phytase on chicken performance and biochemical indices. METHODS: A total of 300 chicks were divided into 4 groups that fed the basal diet, diet containing probiotic (Protexin®), Phytase (Meriphyze 5000®), and probiotic plus phytase all over the growing period. The growth indices were measured weekly, analysed at the 21 and 42 days of age. At 42 days of age, blood samples were collected from all chickens. The concentration of liver enzymes, lipid profiles and antioxidant status were measured in blood samples. RESULTS: The results showed that the weight gain and feed intake were significantly higher in chickens received phytase alone or phytase in combination with probiotic. The feed conversion ratio (FCR) significantly lower in all supplemented chickens in comparison with control chickens (p < 0.05). Although the addition of probiotic or phytase to chicken diet showed an extent variation in biochemical and antioxidant indices, the addition of phytase plus probiotic showed a significantly increase of blood total protein (TP), albumin (Alb), glutathione peroxidase (GPx), super oxide dismutase (SOD), catalysis (CAT) and total antioxidant capacity (TAC) level, while decrease triglyceride (TG), cholesterol (CHL), aspartate transferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP) and malondialdehyde (MDA) in comparison to control chickens. The supplementation of chicken diet with probiotic, phytase or probiotic plus phytase did not effect on low-density lipoprotein (LDL) and high-density lipoprotein (HDL). CONCLUSIONS: The simultaneous supplementation of probiotics and phytases seems to have a positive effect on growth indices in broilers, but they can cause changes in the serum biochemical profile, which sometimes lead to interference and do not always act synergistically.

ABSTRACT

Fungal diseases such as candidiasis are some of the deadliest diseases among immunocompromised patients. These fungi naturally exist on human skin and throughout the digestive system. When the microbiota balance becomes upset, these fungi become pathogenic and potentially lethal. At the pathogenesis of fungal diseases, host immune system response is diverse. At the early stages of fungal pathogenesis such as Candida albicans, it was shown that these fungi use the immune cells of the host body and cause malfunction the early induction of proinflammatory cytokines of the host body leading to a reduction in their numbers. However, at some stages of fungal diseases, the immune response is severe. Despite many treatments already being available, it seems that one of the best treatments could be an immune-stimulatory agent. Some of the subsets of MSCs and exosome-derived cells, as a cell-to-cell communicator agent, have many roles in the human body, including anti-inflammatory and immune-modulatory effects. However, the TLR4-primed and IL-17+ subsets of MSCs have been shown to have immune-stimulatory effects. These subsets of the MSCs produce pro-inflammatory cytokines and reduce immunosuppressive cytokines and chemokines. Thus, they could trigger inflammation and stop fungal pathogenesis. As some biological activities and molecules inherit elements of their exosomes from their maternal cells, the exosome-derived TLR4-primed and IL-17+ subsets of MSCs could be a good candidate for fighting against fungal diseases. The applications of exosomes in human diseases are well-known and expanding. It is time to investigate the exosomes application in fungal diseases. In this review, the probable role of exosomes in treating fungal diseases is explored.
