ABSTRACT

The purpose of the present study was to document pathological findings on retromode imaging in choroidal nevi and evaluate its diagnostic validity, using the confocal scanning laser ophthalmoscope Nidek Mirante (cSLO). A total of 41 choroidal nevi from 41 patients were included. All patients underwent multicolor fundus (mCF), infrared reflectance (IR), green fundus autofluorescence (FAF), dark-field (DF) and retromode (RM) imaging and optical coherence tomography (OCT) scans. We investigated retromode images to evaluate choroidal nevus features by comparing the results with those of mCF, IR, FAF, DF and OCT. In 100% of available images, retromode scanning laser ophthalmoscopy was able to detect choroidal nevi with a characteristic "hypo-retro-reflective" pattern, even the cases not visible on mCF, IR and FAF images. It also made it possible to delineate the margins of lesions with the highest rate of sharpness and accuracy among the imaging modalities examined. These findings seem to demonstrate how RM-SLO is an innovative diagnostic tool to detect and follow up choroidal nevi in a fast, reliable and non-invasive way.

BACKGROUND: Biofortification of vegetables is an important innovation technique in the horticultural sector. Vegetables can be a vector of different minor elements that have beneficial effects on human health. Selenium (Se) is an important element for human nutrition and plays a significant role in defence mechanisms. The aim of this work was to investigate the effect of Se in the nutrient solutions on the crop biofortification ability, yield, and quality parameters of four baby leafy vegetables destined to the minimally processed industry. Experiments were performed on lamb's lettuce, lettuce, wild rocket, and spinach. These crops were cultivated in the floating systems with nutrient solution enriched with 0, 2.6, 3.9, and 5.2 µmol L-1 Se provided as sodium selenate. RESULTS: At harvest, Se concentrations, yield, nitrate concentration, sugars, and some mineral elements were measured. Data collected and analyses showed that yield, nitrate, sucrose, and reducing sugars were not affected by Se treatments, even if varied among species. Se concentrations linearly increased in leaves of different species by increasing the Se concentration in the nutrient solution. Rocket was the species with the highest accumulation ability and reached a concentration of 11 µg g-1 fresh weight Se in plants grown with 5.2 µmol L-1 Se. CONCLUSION: A floating system with Se-enriched nutrient solution is an optimal controlled growing biofortification system for leafy vegetables. The accumulation ability decreased in different species in the order wild rocket, spinach, lettuce, and lamb's lettuce, highlighting a crop-dependent behaviour and their attitude to biofortification. © 2023 The Authors. ABSTRACT

OBJECTIVES: To evaluate the outcomes of delayed intravitreal injections (IVIs) caused by the outbreak of coronavirus disease 2019 (COVID-19), in patients with neovascular age-related macular degeneration (nAMD). METHODS: nAMD patients with scheduled IVIs between March 1st and April 30th, 2020 were stratified through a risk-based selection into a non-adherent group (NA-group) if they skipped at least one IVI and an adherent group (A-group) if they followed their treatment schedule. During the pandemic visit (v0), if a significant worsening of the disease was detected, a rescue therapy of three-monthly IVIs was performed. Multimodal imaging and best-corrected visual acuity (BCVA) findings were evaluated after 6 months (v6), compared between groups and with the visit prior the lockdown (v-1). RESULTS: Two hundred fifteen patients (132 females, mean age: 81.89 ± 5.98 years) delayed their scheduled IVI while 83 (53 females, mean age: 77.92 ± 6.06 years) adhered to their protocol. For both groups, BCVA at v0 was significantly worse than v-1 (mean 4.15 ± 7.24 ETDRS letters reduction for the NA-group and 3 ± 7.96 for the A-group) but remained stable at v6. The two groups did not significantly differ in BCVA trends after 6 months and neither for development of atrophy nor fibrosis. CONCLUSIONS: A risk-based selection strategy and a rescue therapy may limit the long-term outcomes of an interruption of the treatment protocol in patients with nAMD.

ABSTRACT

PURPOSE: To report the incidence and risk factors for fibrosis at 10 years in a large cohort of persons with neovascular age-related macular degeneration (nAMD). DESIGN: Retrospective, multicenter, cohort study. METHODS: We included 225 naive nAMD eyes that underwent intravitreal anti-vascular endothelial growth factor treatment over 10 years of follow-up at two Italian referral centers. Demographic and clinical data were reviewed at baseline and on an annual basis. Onset of fibrosis was defined by clinically assessing photographs, fundus descriptions, or fluorescein angiograms. Optical coherence tomography (OCT) scans of fibrosis were inspected by an external reading center and graded as subretinal pigment epithelium (RPE), mixed, or subretinal. RESULTS: The mean age at baseline was of 72.1 ± 6.9 years. The incidence rate of fibrosis was estimated to be 8.9 per 100 person-years, with a cumulative incidence of 62.7% at 10 years. Fibrotic lesions were sub-RPE in 46.1%, mixed in 29.8%, and subretinal in 22.7%. Independent factors associated with fibrosis included the following: larger central subfield thickness variation (P < .001), submacular hemorrhages (P = .008), higher number of injections (P = .01), and worse baseline visual acuity (VA) (P = .03). Type 2 macular neovascularization was significantly associated with mixed and subretinal fibrosis. VA significantly declined over 10 years (-16.4 Early Treatment Diabetic Retinopathy Study [ETDRS] letters), particularly in eyes with mixed and subretinal fibrosis (P < .001). CONCLUSIONS: We identified a 62.7% cumulative incidence of fibrosis in a large nAMD cohort at 10 years. Fibrosis was more common with frequent reactivations and lower baseline VA; its onset had a significant impact on final VA. This supports the hypothesis that nAMD patients should be promptly treated with proactive regimens.

ABSTRACT

Corneal blindness is the fifth leading cause of blindness worldwide, and therapeutic options are still often limited to corneal transplantation. The corneal epithelium has a strong barrier function, and regeneration is highly dependent on limbal stem cell proliferation and basement membrane remodeling. As a result of the lack of corneal donor tissues, regenerative medicine for corneal diseases affecting the epithelium is an area with quite advanced basic and clinical research. Surgery still plays a prominent role in the treatment of epithelial diseases; indeed, innovative surgical techniques have been developed to transplant corneal and non-corneal stem cells onto diseased corneas for epithelial regeneration applications. The main goal of applying regenerative medicine to clinical practice is to restore function by providing viable cells based on the use of a novel therapeutic approach to generate biological substitutes and improve tissue functions. Interest in corneal epithelium rehabilitation medicine is rapidly growing, given the exposure of the corneal outer layers to external insults. Here, we performed a review of basic, clinical and surgical research reports on regenerative medicine for corneal epithelial disorders, classifying therapeutic approaches according to their macro- or microscopic target, i.e., into cellular or subcellular therapies, respectively.

ABSTRACT

Ageing is associated with a progressive decline and remodelling of the immune system. Also, the efficacy of COVID-19 vaccines has been observed to depend on subjects' age. The post-vaccination data about patients aged > 90 years old is scarcely represented in the literature. The antibody titre profiles of elderly vaccinated subjects (age > 90 years old) were evaluated and compared with profiles obtained in a younger population (age 23-69 years old). To the best of our knowledge, this is the first report providing post-vaccination serological data in subjects aged 90 + years old. This study suggests that distinct SARS-CoV-2 viral-specific antibody response profiles vary based on anti-N serostatus, age, and sex in the very elderly adults. The data obtained could impact the organisation of the vaccination campaign (i.e., prioritisation strategies, administration of additional doses) and the factors that facilitate intentions to receive the vaccination among elderly adults (i.e., vaccine effectiveness).

ABSTRACT

A greenhouse experiment was conducted to study the effects of the application of a plant-derived biostimulant (Bioup® TF) on fruit quality and composition of two clusters (cluster II and cluster VI) of the cherry tomato cultivars 'Eletta', 'Kaucana', and 'Top Stellina'. The biostimulant application promoted fruit yield by 12% (up to 1.3 kg m-2 in 'Kaucana') and increased the concentrations of important functional constituents like phytoene, γ-tocopherol and ß-tocopherol by up to 16, 25, and 23%, respectively. Fruits from late-ripe cluster VI showed higher fruit weights, D-fructose, and total sugar contents than those from early-ripe cluster II (by 15, 7 and 5%, respectively), but reduced concentrations of acyclic carotenoids (phytoene and lycopene) and tocochromanols (mainly γ-tocopherol, -44%). 'Top Stellina' showed the highest responsiveness to the biostimulant, as particularly (all-E)-ß-carotene, phytofluene, and γ-tocopherol concentrations increased, indicating a genotype-dependent effect of the treatment. However, fruits of all treated genotypes showed a contextual decrease in D-fructose and total sugars in response to the biostimulant (on average by 7 and 10%, respectively), indicating a metabolic load burdening the accumulation of lipophilic antioxidants in cherry tomatoes at the expense of their taste-related C pool.

ABSTRACT

Common variable immunodeficiency (CVID) is the most frequent primary antibody deficiency whereby follicular helper T (Tfh) cells fail to establish productive responses with B cells in germinal centers. Here, we analyzed the frequency, phenotype, transcriptome, and function of circulating Tfh (cTfh) cells in CVID patients displaying autoimmunity as an additional phenotype. A group of patients showed a high frequency of cTfh1 cells and a prominent expression of PD-1 and ICOS as well as a cTfh mRNA signature consistent with highly activated, but exhausted, senescent, and apoptotic cells. Plasmatic CXCL13 levels were elevated in this group and positively correlated with cTfh1 cell frequency and PD-1 levels. Monoallelic variants in RTEL1, a telomere length- and DNA repair-related gene, were identified in four patients belonging to this group. Their blood lymphocytes showed shortened telomeres, while their cTfh were more prone to apoptosis. These data point toward a novel pathogenetic mechanism in CVID, whereby alterations in DNA repair and telomere elongation might predispose to antibody deficiency. A Th1, highly activated but exhausted and apoptotic cTfh phenotype was associated with this form of CVID.

PURPOSE: To compare the 2-year outcome to antivascular endothelial growth factor therapy for myopic choroidal neovascularization (CNV) in the eyes with or without dome-shaped macula (DSM). METHODS: Data from treatment-naive myopic CNV with a 2-year follow-up were retrospectively collected and divided into two groups according to the presence of DSM. The best-corrected visual acuity was acquired at baseline, 3, 12, and 24 months. The association between visual outcomes and CNV type and area, presence of scleral-derived feeder vessel, macular atrophy, and lacquer cracks at baseline was also evaluated. RESULTS: Fifty-four eyes of 54 patients were included; 18 eyes (33.4%) had DSM. Choroidal neovascularization was foveal in 10 DSM eyes (55.6%) and in 30 non-DSM eyes (83.9%), P = 0.033. At baseline, the mean best-corrected visual acuity was significantly higher in the DSM group (68.33 ± 12.04 Early Treatment Diabetic Retinopathy Study letters, 20/40 Snellen) compared with the non-DSM group (57.75 ± 13.46 Early Treatment Diabetic Retinopathy Study letters, 20/72 Snellen; P = 0.007). This difference disappeared after 3 months and did not reoccur afterward. All other parameters were not significantly associated with visual outcomes. CONCLUSION: Overall, DSM does not represent a negative prognostic factor in response to antivascular endothelial growth factor therapy in myopic CNVs after 2 years. However, in DSM eyes, CNVs tend to be extrafoveal, thus ensuring a good visual prognosis from the earliest stage of the disease.

ABSTRACT

Vegetables represent pillars of good nutrition since they provide important phytochemicals such as fiber, vitamins, antioxidants, as well as minerals. Biofortification proposes a promising strategy to increase the content of specific compounds. As minerals have important functionalities in the human metabolism, the possibility of enriching fresh consumed products, such as many vegetables, adopting specific agronomic approaches, has been considered. This review discusses the most recent findings on agronomic biofortification of vegetables, aimed at increasing in the edible portions the content of important minerals, such as calcium (Ca), magnesium (Mg), iodine (I), zinc (Zn), selenium (Se), iron (Fe), copper (Cu), and silicon (Si). The focus was on selenium and iodine biofortification thus far, while for the other mineral elements, aspects related to vegetable typology, genotypes, chemical form, and application protocols are far from being well defined. Even if agronomic fortification is considered an easy to apply technique, the approach is complex considering several interactions occurring at crop level, as well as the bioavailability of different minerals for the consumer. Considering the latter, only few studies examined in a broad approach both the definition of biofortification protocols and the quantification of bioavailable fraction of the element.

PURPOSE: To describe our managing strategy for COVID-19 emergency, to evaluate the adherence to intravitreal treatment (AtT) rate during the outbreak in a referral hospital in Milan, and to correlate it with patients' clinical features. METHODS: The AtT rate of patients with scheduled intravitreal injections during the COVID-19 outbreak from February 23, 2020 to March 31, 2020 was compared with the previous trimester and with March 2019. The impact of age, sex, visual function, and diagnosis on the AtT rate during unlocked/locked weeks (from March 8th) was evaluated. RESULTS: Of 650 consecutive patients with scheduled intravitreal injections, the AtT rate during the COVID-19 outbreak was 0.37. This was significantly lower compared with AtT registered in the previous trimester (0.92) and in the same weeks in 2019 (0.90) (both P < 0.001). Patients adherent to treatment were significantly younger (P < 0.001) and had a lower best-corrected visual acuity in the fellow eye (P = 0.046). During the lockdown weeks, the AtT rate was significantly lower than in the two unlocked weeks (0.19 vs. 0.73, P < 0.001). In addition, the AtT rate in patients classified as "emergent" during the lockdown weeks was 0.60. CONCLUSION: These preliminary results can help the retina specialist community to foresee this unique scenario and to develop successful management strategies.

Grafting is a valuable tool for managing problems of tomato soil-borne pathogens and pests, but often generates unpredictable effects on crop yield and product quality. To observe these rootstocks-induced changes, experimental designs including many rootstock-scion combinations are required. To this end, a greenhouse experiment was conducted on 63 graft combinations, involving seven cherry tomato scions grouped in large, medium and small-fruited, and eight rootstocks with different genetic backgrounds (crosses between Solanum lycopersicum and S. habrochaites or S. peruvianum or S. pimpinellifolium, plus an intraspecific hybrid), using ungrafted controls. The response of the graft partners was firstly analyzed individually using the environmental variance (σ2E), then by grouping them by classes. When analyzed individually, the scion genotype influenced fruit L*, b*, shape index, total soluble solids (TSS) and its ratio with tritatable acidity (TSS/TA), whereas plant growth and yield were unpredictable. After clustering the graft partners, some of these responses were attributable to the imposed classes. The S. habrochaites-derived hybrids maximized plant biomass, unlike the S. pimpinellifolium ones. Both classes reduced fruit biomass in small- and medium-fruited scions (by 11 and 14%, respectively). The S. habrochaites and S. peruvianum hybrids reduced a* and TSS, whereas promoted TA. L-ascorbic acid was reduced by grafting (from -23 to -45%), in the S. pimpinellifolium group too, indicating, even in low vigor rootstocks, a dilution effect worsening this nutraceutical trait of tomatoes.

The experiment addressed the effects of two storage temperatures, namely 10 (T10) and 20 °C (T20), on main quality and functional traits of three cherry tomato cultivars ('Eletta', 'Sugarland' and 'Ottymo'), after 0 (S0), 7 (S7) and 14 (S14) days of storage. At T10 both fruit weight and firmness were better retained during storage. At S14, T10 promoted fruit Chroma and overall fruit color deviation (ΔE*ab). Total polyphenols content (TPC) of fruits peaked at S7 (4660 mg GAE kg-1 DW) then declined at S14 (by 16%), with the highest values recorded at T10. Lycopene showed a similar trend, but with a higher average concentration recorded at T20 (488 mg kg-1 DW). ß-carotene content peaked at S14, irrespective of the storage temperature. At S14, the concentrations of phytoene and phytofluene were higher at T20 (48.3 and 40.9 mg kg-1 DW, respectively), but the opposite was found at S7. 'Sugarland' and 'Ottymo' showed the highest ΔE*ab along storage, with the former cultivar proving the highest TPC and lycopene content, whereas 'Eletta' did so for phytoene and phytofluene. Our results suggest that unravelling the possible functional interactions among these three carotenoids would allow for a better orientation of breeding programs, targeting the phytochemical evolution of tomatoes during refrigerated storage.

BACKGROUND: The clinical features of adult-onset atopic dermatitis (AD) are heterogeneous and the diagnosis can be a challenge. A new biologic drug (dupilumab) has been approved for moderate to severe AD in adult patients. The efficacy and safety have been demonstrated in clinical trials, but these studies do not reflect conditions in daily practice and do not consider the different clinical manifestations of AD. OBJECTIVES: Analyzing the dupilumab activity in a real-world setting and comparing its efficacy on different AD phenotypes. METHODS: We retrospectively evaluated 221 AD patients treated with dupilumab, stratified into six clinical phenotypes: classic, generalized eczema inflammatory and lichenoid patterns, prurigo, nummular eczema, and erythroderma. At baseline and at weeks 4, 16, and 52, the disease severity was assessed through the Eczema Area and Severity Index (EASI) and the quality of life was assessed through the Dermatology Life Quality Index (DLQI) questionnaire, Peak Pruritus Numerical Rating Scale (itch NRS), and Peak Sleep NRS. RESULTS: We found a significant improvement after 16 weeks of treatment (p < 0.0001) in all six phenotypes for all the assessed scores mentioned above, persisting up to week 52. The best improvement was seen in the more severe phenotypes, particularly the erythrodermic one. CONCLUSIONS: The present study confirmed the efficacy and safety of dupilumab in the treatment of severe AD. Its strength was in the stratification of AD patients in six different phenotypes based on their clinical presentation, all of whom markedly improved in terms of both clinically evident and reported symptoms, as well as their quality of life.

In this study, five allometric models were used to estimate the single leaf area of three well-known medicinal and aromatic plants (MAPs) species, namely basil (Ocimum basilicum L.), mint (Mentha spp.), and sage (Salvia spp.). MAPs world production is expected to rise up to 5 trillion US$ by 2050 and, therefore, there is a high interest in developing research related to this horticultural sector. Calibration of the models was obtained separately for three selected species by analyzing (a) the cultivar variability-i.e., 5 cultivars of basil (1094 leaves), 4 of mint (901 leaves), and 5 of sage (1103 leaves)-in the main two traits related to leaf size (leaf length, L, and leaf width, W) and (b) the relationship between these traits and single leaf area (LA). Validation of the chosen models was obtained for each species using an independent dataset, i.e., 487, 441, and 418 leaves, respectively, for basil (cv. 'Lettuce Leaf'), mint (cv. 'Comune'), and sage (cv. 'Comune'). Model calibration based on fast-track methodologies, such as those using one measured parameter (one-regressor models: L, W, L2, and W2) or on more accurate two-regressors models (L × W), allowed to achieve different levels of accuracy. This approach highlighted the importance of considering intra-specific variability before applying any models to a certain cultivar to predict single LA. Eventually, during the validation phase, although modeling of single LA based on W2 showed a good fitting (R2basil = 0.948; R2mint = 0.963; R2sage = 0.925), the distribution of the residuals was always unsatisfactory. On the other hand, two-regressor models (based on the product L × W) provided the best fitting and accuracy for basil (R2 = 0.992; RMSE = 0.327 cm2), mint (R2 = 0.998; RMSE = 0.222 cm2), and sage (R2 = 0.998; RMSE = 0.426 cm2).

ABSTRACT

The evaluation of optical properties of biological samples is gaining increasing interests both in scientific and commercial fields concerning agriculture and food processing. The optical techniques can indeed be able to provide information on quality assessment in a fast and non-destructive way. This feature makes them suitable for automatic management of control processes. In this paper, we propose to use the Delayed Luminescence, a ultra-weak and photo-induced emission of optical photons, as a tool for a rapid evaluation of germination performance, the principal index reflecting seed quality. Two lots of 'Mirella' F1 watermelon dried seeds, of 96 seeds each, were considered. The seeds were analyzed in the conditions as provided by seed/breeding company. Characteristics of Delayed Luminescence emission from each single seed were correlated to the different germination levels as assessed by International Seed Testing Procedures. Parametric differences in the two lots were determined, based on the relaxation kinetics of some spectral components. A control test, with the aim to construct a calibration model, was conceived and successfully tested. Time decays of Delayed Luminescence spectral components at central wavelengths 450 nm, 550 nm and 650 nm, corresponding to spectrum region where natural biomarkers as NADH, flavins and lipopigments, protoporphyrin and ROS respectively emit, have been evaluated. The results show that such time decays are strictly connected to the biological state of the system under analysis and allow also proposing Delayed Luminescence measurement as a quick, cheap and non-destructive test for seed viability analysis.

BACKGROUND: Cultivation of cauliflower is diffused in Mediterranean areas where water salinity results in the need to identify alternative irrigation sources or management strategies. Using saline water during two growth phases (from transplanting to visible appearance of inflorescence or from appearance of inflorescence to head harvest), the present study aimed to identify the growth period that is more suitable for irrigation with low quality water in relation to cauliflower production and quality. RESULTS: Salinity affected cauliflower growth mainly when imposed in the first growth phase. The growth reduction depended mainly on ion-specific effects, although slight nutrient imbalances as a result of Na+ and Cl- antagonisms were observed. The use of non-saline water in the first or second growth period reduced both the osmotic and toxic effects of salinity. When salinity was applied during inflorescence growth, yield was reduced because of a restriction of water accumulation in the head. CONCLUSION: The results of the present study demonstrate the possibility of producing marketable cauliflower heads under conditions of salinity by timing the application of the best quality water during the first growth phase to improve fruit quality and during the second phase to reduce the negative effects of salinity on yield. © 2016 Society of Chemical Industry.

ABSTRACT

Grafting can enhance the tolerance of vegetable crops to soilborne diseases. The aim of this study was to investigate whether different tomato scion-rootstock combinations may affect the plant susceptibility to Fusarium oxysporum f. sp. radicis-lycopersici (FORL), the causal agent of crown and root rot. A proteomic approach was used to investigate whenever the protein repertoire of the rootstock can be modified by FORL infection, in relation to cultivar susceptibility/tolerance to the disease. To this purpose, plants of tomato hybrids with different vigor, "Costoluto Genovese" (less vigorous) and "Kadima" (more vigorous), were grafted onto "Cuore di Bue" and "Natalia" hybrids, sensitive and tolerant versus FORL infections, respectively. Disease symptoms, plant biomasses, and protein expression patterns were evaluated 45 days after FORL inoculation. The extent of vascular discoloration caused by FORL in tomato plants grafted on "Natalia" rootstock (0.12-0.37 cm) was significantly lower than that of plants grafted on sensitive "Cuore di Bue" (1.75-6.50 cm). FORL symptoms significantly differed between "Costoluto Genovese" and "Kadima" scions only when grafted on sensitive rootstock. Shoot FW of non-inoculated "Kadima"/"Cuore di Bue" combination was 35% lower than "Kadima"/"Natalia", whereas no difference was manifested in inoculated plants. Shoot FW of inoculated "Costoluto Genovese"/"Cuore di Bue" combination was decreased of 39%, whereas that of "Costoluto Genovese"/"Natalia" of 11%, compared to control plants. Proteomic results showed a higher representation of proteins associated with pathogen infection in the tolerant rootstock, compared to the sensitive one, meaning a direct involvement of plant defence mechanisms in the tomato response to the pathogen challenge.

ABSTRACT

The average content of some classes of antioxidants is generally higher in cherry tomatoes than in normal-sized berries. The aim of this work was to assess the nutritional value of cherry tomato (cv. Naomi F1) by investigating the compositional pattern of berries harvested at different ripening stages and evaluating, in particular, all of the main antioxidants (carotenoids, ascorbic acid, phenolic compounds, and alpha-tocopherol) and the antioxidant activity of the water-soluble and water-insoluble fractions. Results confirmed the relatively high level of carotenoids in cherry tomato but showed that not all biologically active compounds necessarily increase in tomatoes picked at later stages of ripeness. Cherry tomatoes harvested at full ripeness exhibited the highest level of carotenoids and antioxidant activity in the water-insoluble fraction. On the other hand, no significant differences in ascorbic acid content were observed at different ripening stages, whereas the main phenolics content and the antioxidant activity of water-soluble fraction showed slight, but significant, decreases at later stages of ripeness.
